Therapeutic strategies of targeting nonapoptotic regulated cell death (RCD) with smallmolecule compounds in cancer

Regulated cell death(RCD)is a controlled form of cell death orchestrated by one or more cascading signaling pathways,making it amenable to pharmacological *** subroutines can be categorized as apoptotic or non-apoptotic and play essential roles in maintaining homeostasis,facilitating development,and modulating *** evidence has recently revealed that RCD evasion is frequently the primary cause of tumor *** non-apoptotic RCD subroutines have garnered attention as promising cancer therapies due to their ability to induce tumor regression and prevent relapse,comparable to ***,they offer potential solutions for overcoming the acquired resistance of tumors toward apoptotic *** an increasing understanding of the underlying mechanisms governing these non-apoptotic RCD subroutines,a growing number of small-molecule compounds targeting single or multiple pathways have been discovered,providing novel strategies for current cancer *** this review,we comprehensively summarized the current regulatory mechanisms of the emerging non-apoptotic RCD subroutines,mainly including autophagy-dependent cell death,ferroptosis,cuproptosis,disulfidptosis,necroptosis,pyroptosis,alkaliptosis,oxeiptosis,parthanatos,mitochondrial permeability transition(MPT)-driven necrosis,entotic cell death,NETotic cell death,lysosome-dependent cell death,and immunogenic cell death(ICD).Furthermore,we focused on discussing the pharmacological regulatory mechanisms of related small-molecule *** brief,these insightful findings may provide valuable guidance for investigating individual or collaborative targeting approaches towards different RCD subroutines,ultimately driving the discovery of novel small-molecule compounds that target RCD and significantly enhance future cancer therapeutics.

Two-dimensional(2D)semiconductors with intrinsic ferromagnetism are highly desirable for potential applications in nextgeneration spintronic and optoelectronic ***,controllable synthesis of intrinsic 2D magnetic semiconductor on a substrate is still a challenging ***,large-area 2D non-layered rock salt(α-phase)MnSe nanosheets were grown on mica substrates,with the thickness changing from 54.2 to 0.9 nm(one unit cell),by chemical vapour *** X-ray diffraction,Raman spectroscopy,transmission electron microscopy,and X-ray photoelectron spectroscopy measurements confirmed that the resulting 2Dα-MnSe nanosheets were obtained as high-quality single *** magnetic hysteresis loops and synchrotron X-ray measurements directly indicated the anomalous magnetic properties inα-MnSe *** analysis of the reasons for magnetic property revealed that the low-temperature phase transition,small number of stacking differences in crystals,and surface weak oxidation in(111)-orientedα-MnSe were the main ***,α-MnSe nanosheets exhibited broadband photoresponse from 457 to 671 nm with an outstanding detectivity and responsivity *** study presents the detailed growth process of ultrathin 2D magnetic semiconductorα-MnSe,and its outstanding magnetic properties and broadband photodetection,which provide an excellent platform for magneto-optical and magneto-optoelectronic research.

Detwinning and Anneal-Hardening Behaviors of Pre-Twinned AZ31 Alloys under Cryogenic Loading

Detwinning behavior of a pre-twinned magnesium alloy AZ31 at cryogenic temperature was investigated,also with a focus on the annealing hardening behavior of samples with different fractions of ***-compression along the transverse direction with strains of 1.7%,3.0%,and 6.0% was applied to generated[1012]*** behavior,microstructure,and texture evolution during subsequent tension were *** results show that low temperature did not change the fact that detwinning still pre-dominated in the pre-twinned samples under reverse ***,a relatively harder migration of twin boundaries was found at cryogenic *** annealing hardening of 27-40 MPa was observed in the pre-twinned samples,and such a hardening effect shows a close relation with the fraction of pre-twins or the level of *** annealing hardening effect disappeared if the matrix was consumed by twins along with the increased pre-compression *** corresponding reasons for the annealing hardening behavior were discussed.

Thermal stable intermetallic particles are important for the heat resistance of magnesium(Mg)*** this work,many lath-like particles formed in α-Mg grains of a Mg-8Gd-3Sm-0.7Al casting alloy when heat-treated at 873 ***-resolution high-angle annular dark field scanning transmission electron microscopy(HAADF-STEM) characterizations indicate that most of them are Mg-containing Al_(2)(Gd,Sm),with the atomic ratio of Mg:Al:(Gd,Sm) being ~1:1:1;a small part of them with relatively wider thickness are long-period stacking ordered(LPSO) phases simultaneously containing both 14H and 18R *** followed common orientation relationships with Mg matrix as those reported in previous *** addition,many Mg laths were observed in the primary blocky Al_(2)(Gd,Sm) phase at grain boundaries,where the atomic ratio of Al:(Gd,Sm) in the Al_(2)(Gd,Sm) matrix was 2:***,density functional theory(DFT) calculations illustrated the detail structure of the re-constructed Mg/Al_(2)RE interface and simultaneously deduced the underlying reason for the re-dissolution of the newly formed Mg-containing Al_(2)(Gd,Sm) plates in α-Mg matrix.

Hydrogen storage thermodynamics and kinetics of as-cast Ce-Mg-Ni-based alloy

The reaction kinetics of alloys based on magnesium are known to be greatly improved by the partial substitution of Mg with rare earths and transition metals,particularly *** enhanced superficial hydrogen dissociation rate,the weakened Mg-H bond and the lower activation energy following element replacement are thought to be related to the better *** experimental alloys Ce5Mg_(95-x)Ni_(x)(x=5,10,15)were smelted by the vacuum induction *** phase transformation and structural evolution of experimental alloys before and after reaction with hydrogen were char-acterized by X-ray diffraction,scanning electron microscopy and transmission electron *** cast specimens contain CeMg_(12),Mg and Mg_(2)Ni phases,and the increase in Ni content results in an obvious growth of Mg_(2)Ni *** isothermal and non-isothermal hydrogenation and dehydrogenation kinetics of the experimental specimens were investi-gated using the Sievert apparatus,differential scanning calorimetry and thermal gravimetric *** activation energy may be calculated using the Arrhenius and Kissinger *** experimental alloys have been shown to have good activation properties,with a reversible hydriding and dehydriding capacities of around 5.0 wt.%in the first *** initial dehydrogenation temperature of MgH_(2) decreases from 557.5 to 537.7 K with changing Ni content from 5 to 15 at.%.The dehydrogenation activation energy also reduces from 77.09 to 62.96 kJ/mol,which explains the improved hydrogen storage performance caused by Ni *** can be shown that the impact of Ni on the decomposition enthalpy of MgH_(2) is quite modest,with the absolute enthalpy(ΔHr)only decreasing from 78.48 to 76.15 kJ/mol.

An electron-transfer[Fe_(2)Co_(2)]square complex exhibiting unprecedented wide room-temperature hysteresis

Switchable molecules with magnetic bistability that can function at standard conditions are highly desirable from the perspective of practical *** the last decade,significant progress has been made in modulating thermally induced electrontransfer-coupled spin transition(ETCST)events between diamagnetic{Fe_(LS)^(Ⅱ)(μ-CN)Co_(LS)^(Ⅲ)}and paramagnetic{Fe_(LS)^(Ⅲ)(μ-CN)-Co_(HS)^(Ⅱ)}electron configurations(named LS and HS,respectively,for short)within the Fe/Co mixed-valence cyanometallate ***,we prepared a novel[Fe_(2)Co_(2)]square complex[(pzTp)Fe(CN)_(3)Co(bnbpen)]_(2)[NO_(3)]_(2)·4CH_(3)OH·2.5H_(2)O(1·Solv)and its desolvated phase[(pzTp)Fe(CN)_(3)Co(bnbpen)]_(2)[NO_(3)]_(2)(1),obtained via single-crystal-to-single-crystal(SCSC)*** 1·Solv remaining in the diamagnetic state below room temperature exhibits an irreversible ETCST behavior accompanied by desolvation upon ***,complex 1 displays an unprecedented rate-dependent ETCST behaviour involving with two diamagnetic LS states(a dynamically favoured LS1 state and a thermodynamically favoured LS2state),between which a net one-way transition from LS1 to LS2 is accessible likely via a structural phase transition(SPT)above255 ***,two well separated transition channels(HS?LS1 at around 255 K and HS←→LS2 in the temperature range of 317-350 K)are achieved according to either fast(>20 K/min)or slow rate(<1 K/min),*** a moderate rate of 3 K/min,complex 1 shows a wide hysteresis of 66 K centered at 328 K,the largest yet reported for discrete Fe/Co cyanometallate *** 1 represents the first ETCST example coupled with dynamic SPT,providing a new strategy for manipulating bistability within this fascinating cyanometallate family.

Ethanol extract of Abelmoschus manihot suppresses endoplasmic reticulum stress in contrast-induced nephropathy

Objective:To explore the efficacy and potential mechanisms of the ethanol extract of Abelmoschus manihot(L.)Medic in contrast-induced nephropathy(CIN).Methods:CIN rat models and human renal proximal tubular cells(HK-2)with iopromide-induced injury were employed to mimic CIN *** effect of Abelmoschus manihot extract on the rat models and HK-2 cells was *** rat models,kidney function,histology,oxidative stress and apoptosis were *** HK-2 cells,cell viability,apoptosis,mitochondrial membrane potential,and endoplasmic reticulum stress were ***:Abelmoschus manihot extract significantly improved structural and functional impairments in the kidneys of CIN ***,the extract effectively mitigated the decline in cellular viability and reduced iopromide-induced oxidative stress and lipid *** investigations revealed that Abelmoschus manihot extract prominently attenuated acute endoplasmic reticulum stress-mediated apoptosis by downregulating GRP78 and CHOP protein ***:Abelmoschus manihot extract can be used as a promising therapeutic and preventive agent in the treatment of CIN.

Unique Azolyl Acylhydrazonyl Hybridization of Aloe Emodins to Access Potential Antibacterial Agents

A type of unique azole-hybridized acylhydrazonyl aloe emodins(AAEs)were developed as new antibacterial agents for combating bacterial *** target AAEs showed strong antibacterial activities,especially,tetrazolylthioether AAE 27a exhibited broad antibacterial spectrum with 16-256 folds and 8-64 folds more active antibacterial efficacy than the reference drugs aloe emodin and norfloxacin,*** AAE 27a also gave low hemolysis and cytotoxicity,as well as favorable *** mechanism explorations revealed that tetrazolylthioether AAE 27a could cause bacterial membrane depolarization and damage the cell membrane,resulting in nucleic acid ***,compound 27a could intercalate into DNA to impede its replication and form supramolecular 27a-DNA gyrase complex to disturb the function of DNA *** findings would provide valuable insights for the further exploration of azolyl acylhydrazonyl aloe emodins as new potential antibacterial candidates.

Evolutionary Neural Architecture Search and Its Applications in Healthcare

Most of the neural network architectures are based on human experience,which requires a long and tedious trial-and-error *** architecture search(NAS)attempts to detect effective architectures without human *** algorithms(EAs)for NAS can find better solutions than human-designed architectures by exploring a large search space for possible *** multiobjective EAs for NAS,optimal neural architectures that meet various performance criteria can be explored and discovered ***,hardware-accelerated NAS methods can improve the efficiency of the *** existing reviews have mainly focused on different strategies to complete NAS,a few studies have explored the use of EAs for *** this paper,we summarize and explore the use of EAs for NAS,as well as large-scale multiobjective optimization strategies and hardware-accelerated NAS *** performs well in healthcare applications,such as medical image analysis,classification of disease diagnosis,and health *** for NAS can automate the search process and optimize multiple objectives simultaneously in a given healthcare *** neural network has been successfully used in healthcare,but it lacks *** data is highly sensitive,and privacy leaks are frequently reported in the healthcare *** solve these problems,in healthcare,we propose an interpretable neuroevolution framework based on federated learning to address search efficiency and privacy ***,we also point out future research directions for evolutionary ***,for researchers who want to use EAs to optimize NNs in healthcare,we analyze the advantages and disadvantages of doing so to provide detailed guidance,and propose an interpretable privacy-preserving framework for healthcare applications.
