Factors controlling organic-rich shale development in the Liushagang Formation,Weixinan Sag,Beibu Gulf Basin:Implications of structural activity and the depositional environment

The mechanisms of lacustrine organic-rich shale formation have attracted attention due to its association with global shale oil and shale gas exploration.Samples of general-quality and excellent-quality source rocks,and oil shale from the Beibu Gulf Basin were analyzed to investigate their organic geochemistry,palynofacies,and trace elements.Hydrocarbon potential was higher in the oil shale(29.79 mg/g)than in the general-quality source rock(3.82 mg/g),and its kerogen type wasⅠ-Ⅱ2.Hydrogen-rich liptinite(cutinite and sporinite)components derived from terrigenous higher plants provided most of the hydrocarbon potential of excellent-quality source rock and oil shale.Under the influence of depressioncontrolling fault activity,a deeper subsidence center promotes the deposition of excellent-quality source rock and oil shale in brackish-hypoxic bottom water.A shallower subsidence center,due to subsag-controlling fault activity,promotes the formation of excellent-quality source rock under freshbrackish and weak oxidation-weak reducing conditions.The local uplift and shallow-slope led to the formation of general-quality source rock,under freshwater weak-oxidation conditions.A model was established for organic matter(OM)accumulation in organic-rich shales,accounting for fault activity,terrigenous hydrogen-rich OM,and the preservation conditions,to predict the development of excellent-quality source rock from areas with low levels of exploration.

Prognostic value of a nomogram model for postoperative liver metastasis of colon cancer

BACKGROUND Colon cancer is one of the most common malignant tumors of the digestive system.Liver metastasis after colon cancer surgery is the primary cause of death in patients with colon cancer.AIM To construct a novel nomogram model including various factors to predict liver metastasis after colon cancer surgery.METHODS We retrospectively analyzed 242 patients with colon cancer who were admitted and underwent radical resection for colon cancer in Zhejiang Provincial People’s Hospital from december 2019 to december 2022.Patients were divided into liver metastasis and non-liver metastasis groups.Sex,age,and other general and clinicopathological data(preoperative blood routine and biochemical test indexes)were compared.The risk factors for liver metastasis were analyzed using singlefactor and multifactorial logistic regression.A predictive model was then constructed and evaluated for efficacy.RESULTS Systemic inflammatory index(SII),C-reactive protein/albumin ratio(CAR),red blood cell distribution width(RDW),alanine aminotransferase,preoperative carcinoembryonic antigen level,and lymphatic metastasis were different between groups(Pder the curve was 0.93 for the column-line diagram prediction model constructed based on these risk factors to distinguish whether liver metastasis occurred postoperatively.The actual curve of the column-line diagram predicting the risk of postoperative liver metastasis was close to the ideal curve,with good agreement.The prediction model curves in the decision curve analysis showed higher net benefits for a larger threshold range than those in extreme cases,indicating that the model is safer.CONCLUSION Liver metastases after colorectal cancer surgery could be well predicted by a nomogram based on the SII,CAR,and RDW.

Genetic Analysis of Two Novel GPI Variants Disrupting H Bonds and Localization Characteristics of 55 Gene Variants Associated with Glucose-6-phosphate Isomerase deficiency

Objective:Glucose-6-phosphate isomerase(GPI)deficiency is a rare hereditary nonspherocytic hemolytic anemia caused by GPI gene variants.This disorder exhibits wide heterogeneity in its clinical manifestations and molecular characteristics,often posing challenges for precise diagnoses using conventional methods.To this end,this study aimed to identify the novel variants responsible for GPI deficiency in a Chinese family.Methods:The clinical manifestations of the patient were summarized and analyzed for GPI deficiency phenotype diagnosis.Novel compound heterozygous variants of the GPI gene,c.174C>A(p.Asn58Lys)and c.1538G>T(p.Trp513Leu),were identified using whole-exome and Sanger sequencing.The AlphaFold program and Chimera software were used to analyze the effects of compound heterozygous variants on GPI structure.Results:By characterizing 53 GPI missense/nonsense variants from previous literature and two novel missense variants identified in this study,we found that most variants were located in exons 3,4,12,and 18,with a few localized in exons 8,9,and 14.This study identified novel compound heterozygous variants associated with GPI deficiency.These pathogenic variants disrupt hydrogen bonds formed by highly conserved GPI amino acids.Conclusion:Early family-based sequencing analyses,especially for patients with congenital anemia,can help increase diagnostic accuracy for GPI deficiency,improve child healthcare,and enable genetic counseling.

Colorectal cancer(CRC)is the second leading cause of cancer-related deaths worldwide1.Surgical radical resection with adjuvant chemotherapy remains the primary treatment choice for CRC,but the 5-year postoperative survival rate is only approximately 60%,and approximately one-third of patients with CRC experience recurrence within 2 years of surgery2.Fortunately,the transformation of high-throughput sequencing has accelerated the development of precision medicine.For example,KRAS mutations indicate resistance to anti-epidermal growth factor receptor(EGFR)-targeted therapies in CRC3.Furthermore,molecular-guided individualized therapy has brought new promise in major clinical areas and challenges,such as novel biomarkers predicting sensitivity and resistance to immunotherapy for microsatellite stable(MSS)CRC.

Learning to sample initial solution for solving 0-1 discrete optimization problem by local search

Local search methods are convenient alternatives for solving discrete optimization problems(DOPs).These easy-to-implement methods are able to find approximate optimal solutions within a tolerable time limit.It is known that the quality of the initial solution greatly affects the quality of the approximated solution found by a local search method.In this paper,we propose to take the initial solution as a random variable and learn its preferable probability distribution.The aim is to sample a good initial solution from the learned distribution so that the local search can find a high-quality solution.We develop two different deep network models to deal with DOPs established on set(the knapsack problem)and graph(the maximum clique problem),respectively.The deep neural network learns the representation of an optimization problem instance and transforms the representation to its probability vector.Experimental results show that given the initial solution sampled from the learned probability distribution,a local search method can acquire much better approximate solutions than the randomly-sampled initial solution on the synthesized knapsack instances and the Erd?s-Rényi random graph instances.Furthermore,with sampled initial solutions,a classical genetic algorithm can achieve better solutions than a random initialized population in solving the maximum clique problems on DIMACS instances.Particularly,we emphasize that the developed models can generalize in dimensions and across graphs with various densities,which is an important advantage on generalizing deep-learning-based optimization algorithms.

deepsea mining has been proposed since the 1960s to alleviate the lack of resources on land.Vertical hydraulic transport of collected ores from the seabed to the sea surface is considered the most promising method for industrial applications.In the present study,an indoor model test of the vertical hydraulic transport of particles was conducted.A noncontact optical method has been proposed to measure the local characteristics of the particles inside a vertical pipe,including the local concentration and particle velocity.The hydraulic gradient of ore transport was evaluated with various particle size distributions,particle densities,feeding concentrations and mixture flow velocities.During transport,the local concentration is larger than the feeding concentration,whereas the particle velocity is less than the mixture velocity.The qualitative effects of the local concentration and local fluid velocity on the particle velocity and slip velocity were investigated.The local fluid velocity contributes significantly to particle velocity and slip velocity,whereas the effect of the local concentration is marginal.A higher feeding concentration and mixture flow velocity result in an increased hydraulic gradient.The effect of the particle size gradation is slight,whereas the particle density plays a crucial role in the transport.

A Rendezvous Mission to the Second Earth Trojan Asteroid 2020 XL_5 with Low-Thrust Multi-Gravity Assist Techniques

As the second of Earth's Trojan asteroids, 2020 XL_(5) is worthy of rendezvous and even sample return missions in many aspects. In this paper, a rendezvous mission to Earth's second Trojan asteroid 2020 XL_(5) is proposed.However, due to its high inclination and large eccentricity, direct impulsive transfer requires large amounts of fuel consumption. To address this challenge, we explore the benefits of electric propulsion and multi-gravity assist techniques for interplanetary missions. These two techniques are integrated in this mission design. The design of a low-thrust gravity-assist(LTGA) trajectory in multi-body dynamics is thoroughly investigated,which is a complex process. A comprehensive framework including three steps is presented here for optimization of LTGA trajectories in multi-body dynamics. The rendezvous mission to 2020 XL_(5) is designed with this three-step approach. The most effective transfer sequence among the outcomes involves Earth–Venus–Earth–Venus-2020 XL_(5). Numerical results indicate that the combination of electric propulsion and multi-gravity assists can greatly reduce the fuel consumption, with fuel consumption of 9.03%, making it a highly favorable choice for this rendezvous mission.

Influence of transcatheter arterial embolization on symptom distress and fatigue in liver cancer patients

BACKGROUND Hepatocellular carcinoma(HCC)is a prevalent malignancy,and transcatheter arterial embolization(TAE)has emerged as a pivotal therapeutic modality.How-ever,TAE may induce symptom distress and fatigue,adversely affecting the quality of life of patients.AIM To investigate symptom distress,fatigue,and associated factors in HCC patients undergoing TAE.METHODS We used a cross-sectional design and purposive sampling to enroll HCC patients who underwent TAE at our institution from January to december 2022.Question-naires were utilized to collect data on symptom distress and fatigue scores from the first to the third day after TAE.RESULTS Our study revealed a significant reduction in fatigue and symptom distress among patients after TAE.Pain,fatigue,insomnia,fever and abdominal dis-tension were the most common symptoms troubling patients during the first 3 d post-TAE.Marital status,presence of family support,physical functional status,age,and symptom distress were identified as predictors of fatigue in patients.CONCLUSION Healthcare professionals should educate HCC patients on symptom distress and INTRODUCTION Hepatocellular carcinoma(HCC)ranks as the fifth most prevalent cancer and the third leading cause of cancer-related mortality globally.Surgical intervention remains the cornerstone of HCC treatment;however,due to the challenges associated with early diagnosis and the lack of specific diagnostic markers,a considerable proportion of patients are diagnosed at advanced stages,rendering them ineligible for surgical interventions.Transcatheter arterial embolization(TAE)is an interventional therapeutic approach involving the insertion of a fine catheter via the femoral artery to reach the vasculature near the tumor site.TAE aims to obstruct the arterial supply to the tumor by deploying embolic agents,thereby inducing necrosis in cancer cells.This procedure is suitable for patients with good liver function and overall health,particularly those with large HCCs that have not invaded the portal vein[1,2].Nonetheless,

Prostate cancer(PCa)is one of the most common malignancies in males worldwide,and its development and progression involve the regulation of multiple metabolic pathways.Alterations in lipid metabolism affect the proliferation and metastatic capabilities of PCa cells.Cancer cells increase lipid synthesis and regulate fatty acid oxidation to meet their growth and energy demands.Similarly,changes occur in amino acid metabolism in PCa.Cancer cells exhibit an increased demand for specific amino acids,and they regulate amino acid transport and metabolic pathways to fulfill their proliferation and survival requirements.These changes are closely associated with disease progression and treatment response in PCa cells.Therefore,a comprehensive investigation of the metabolic characteristics of PCa is expected to offer novel insights and approaches for the early diagnosis and treatment of this disease.

Cumulative evidence suggests that O-linkedβ-N-acetylglucosaminylation(OGlcNAcylation)plays an important regulatory role in pathophysiological processes.Although the regulatory mechanisms of O-GlcNAcylation in tumors have been gradually elucidated,the potential mechanisms of O-GlcNAcylation in bone metabolism,particularly,in the osteogenic differentiation of bone marrow mesenchymal stromal cells(BMSCs)remains unexplored.In this study,the literature related to O-GlcNAcylation and BMSC osteogenic differentiation was reviewed,assuming that it could trigger more scholars to focus on research related to OGlcNAcylation and bone metabolism and provide insights into the development of novel therapeutic targets for bone metabolism disorders such as osteoporosis.
