Dysfunction of synaptic endocytic trafficking in Parkinson's disease

Parkinson's disease is characterized by the selective degeneration of dopamine neurons in the nigrostriatal pathway and dopamine deficiency in the *** precise reasons behind the specific degeneration of these dopamine neurons remain largely *** investigations have identified over 20 causative PARK genes and 90 genomic risk loci associated with both familial and sporadic Parkinson's ***,several of these genes are linked to the synaptic vesicle recycling process,particularly the clathrinmediated endocytosis *** suggests that impaired synaptic vesicle recycling might represent an early feature of Parkinson's disease,followed by axonal degeneration and the eventual loss of dopamine cell bodies in the midbrain via a"dy.ng back"***,several new animal and cellular models with Parkinson's disease-linked mutations affecting the endocytic pathway have been created and extensively *** models faithfully recapitulate certain Parkinson's disease-like features at the animal,circuit,and cellular levels,and exhibit defects in synaptic membrane trafficking,further supporting the findings from human genetics and clinical *** this review,we will first summarize the cellular and molecular findings from the models of two Parkinson's disease-linked clathrin uncoating proteins:auxilin(DNAJC6/PARK19)and synaptojanin 1(SyNJ1/PARK20).The mouse models carry.ng these two PARK gene mutations phenocopy each other with specific dopamine terminal pathology and display a potent synergistic ***,we will delve into the involvement of several clathrin-mediated endocytosis-related proteins(GAK,endophilin A1,SAC2/INPP5 F,synaptotagmin-11),identified as Parkinson's disease risk factors through genome-wide association studies,in Parkinson's disease *** will also explore the direct or indirect roles of some common Parkinson's disease-linked proteins(alpha-synuclein(PARK1/4),Parkin(PARK2),and LRRK2(PARK

The oyster mushroom(Pleurotus spp.)is one of the most widely cultivated mushroom species *** present study investigated the effect of synbiotics on the growth and quality of Pleurotus ostreatus and Pleurotus *** synbiotics formulations were applied by spray.ng mushroom samples daily and measuring their growth parameters,yield,biological efficiency,proximate composition,mineral content,total phenolic content(TPC),and diphenyl-1-picryl-hydrazyl(DPPH)radical scavenging *** demonstrated that the most significant yield of oyster mushrooms was harvested from synbiotics sprayed with inulin and Lactobacillus casei(56.92 g).Likewise,the highest biological efficiency obtained with a similar synbiotic was 12.65%.Combining inulin and *** was the most effective method of improving the mushrooms'growth performance and nutrient content in both ***,synbiotics that combined inulin and *** resulted in the highest TPC(20.550 mg gallic acid equivalent(GAE)/g dry extract(DE))in white oyster mushrooms(***).In comparison,in grey mushroom(***)the highest TPC was yielded by ***(1.098 mg GAE/g DE)followed by inulin and ***(1.079 mg GAE/g DE).The DPPH results indicated that the oyster mushroom could be an efficient *** results revealed that apply.ng synbiotics improved the mushrooms'quality by increasing their antioxidant capacity with higher amounts of phenolic compounds and offering better health benefits with the increased levels of mineral ***,these studies demonstrated the potential of using synbiotics as a biofertilizer,which is helpful for mushroom cultivation;therefore,it might solve the challenge of inconsistent quality mushroom growers face.

Effect of Interface Form on Creep Failure and Life of Dissimilar Metal Welds Involving Nickel-Based Weld Metal and Ferritic Base Metal

For dissimilar metal welds(DMWs)involving nickel-based weld metal(WM)and ferritic heat resistant steel base metal(BM)in power plants,there must be an interface between WM and BM,and this interface suffers mechanical and microstructure mismatches and is often the rupture location of premature *** this study,a new form of WM/BM interface form,namely double y-type interface was designed for the *** behaviors and life of DMWs containing double y-type interface and conventional I-type interface were compared by finite element analysis and creep tests,and creep failure mechanisms were investigated by stress-strain analysis and microstructure *** apply.ng double y-type interface instead of conventional I-type interface,failure location of DMW could be shifted from the WM/ferritic heat-affected zone(HAZ)interface into the ferritic HAZ or even the ferritic BM,and the failure mode change improved the creep life of *** interface premature failure of I-type interface DMW was related to the coupling effect of microstructure degradation,stress and strain concentrations,and oxide notch on the WM/HAZ *** creep failure of double y-type interface DMW was the result of Type IV fracture due to the creep voids and micro-cracks on fine-grain boundaries in HAZ,which was a result of the matrix softening of HAZ and lack of precipitate pinning at fine-grain *** double y-type interface form separated the stress and strain concentrations in DMW from the WM/HAZ interface,preventing the trigger effect of oxide notch on interface failure and inhibiting the interfacial microstructure *** is a novel scheme to prolong creep life and enhance reliability of DMW,by means of optimizing the interface form,decoupling the damage factors from WM/HAZ interface,and then changing the failure mechanism and shifting the failure location.

Formation and transformation of metastable LPSO building blocks clusters in Mg-Gd-y-Zn-Zr alloys by spinodal decomposition and heterogeneous nucleation

To study the formation and transformation mechanism of long-period stacked ordered(LPSO)structures,a systematic atomic scale analysis was conducted for the structural evolution of long-period stacked ordered(LPSO)structures in the Mg-Gd-y-Zn-Zr alloy annealed at 300℃~500℃.Various types of metastable LPSO building block clusters were found to exist in alloy structures at different temperatures,which precipitate during the solidification and homogenization *** stability of Zn/y clusters is explained by the first principles of density functional *** LPSO structure is distinguished by the arrangement of its different Zn/y enriched LPSO structural units,which comprises local fcc stacking sequences upon a tightly packed *** presence of solute atoms causes local lattice distortion,thereby enabling the rearrangement of Mg atoms in the different configurations in the local lattice,and local HCP-FCC transitions occur between Mg and Zn atoms occupy.ng the nearest neighbor *** finding indicates that LPSO structures can generate necessary Schockley partial dislocations on specific slip surfaces,providing direct evidence of the transition from 18R to *** of the LPSO,devoid of any defects and non-coherent interfaces,was observed separately from other precipitated *** a result,the precipitation sequence of LPSO in the solidification stage was as follows:Zn/ycluster+Mg layers→various metastable LPSO building block clusters→18R/24R LPSO;whereas the precipitation sequence of LPSO during homogenization treatment was observed to be as follows:18R LPSO→various metastable LPSO building block clusters→14H *** these,14H LPSO was found to be the most thermodynamically stable structure.

A Gaussian Noise-Based Algorithm for Enhancing Backdoor Attacks

Deep Neural Networks(DNNs)are integral to various aspects of modern life,enhancing work ***-less,their susceptibility to diverse attack methods,including backdoor attacks,raises security *** aim to investigate backdoor attack methods for image categorization tasks,to promote the development of DNN towards higher *** on backdoor attacks currently faces significant challenges due to the distinct and abnormal data patterns of malicious samples,and the meticulous data screening by developers,hindering practical attack *** overcome these challenges,this study proposes a Gaussian Noise-Targeted Universal Adversarial Perturbation(GN-TUAP)*** approach restricts the direction of perturbations and normalizes abnormal pixel values,ensuring that perturbations progress as much as possible in a direction perpendicular to the decision hyperplane in linear *** limits anomalies within the perturbations improves their visual stealthiness,and makes them more challenging for defense methods to *** verify the effectiveness,stealthiness,and robustness of GN-TUAP,we proposed a comprehensive threat *** on this model,extensive experiments were conducted using the CIFAR-10,CIFAR-100,GTSRB,and MNIST datasets,comparing our method with existing state-of-the-art attack *** also tested our perturbation triggers using various defense methods and further experimented on the robustness of the triggers against noise filtering *** experimental outcomes demonstrate that backdoor attacks leveraging perturbations generated via our algorithm exhibit cross-model attack effectiveness and superior ***,they possess robust anti-detection capabilities and maintain commendable performance when subjected to noise-filtering methods.
