Targeting stroma and tumor,silencing galectin 1 treats orthotopic mouse hepatocellular carcinoma

This study examines inhibiting galectin 1(Gal1)as a treatment option for hepatocellular carcinoma(HCC).Gal1 has immunosuppressive and cancer-promoting roles.Our data showed that Gal1 was highly expressed in human and mouse HCC.The levels of Gal1 positively correlated with the stages of human HCC and negatively with survival.The roles of Gal1 in HCC were studied using overexpression(OE)or silencing using Igals1 siRNA delivered by AAV9.Prior to HCC initiation induced by RAS and AKT mutations,lgals1-OE and silencing had opposite impacts on tumor load.The treatment effect of lgals1 siRNAwas further demonstrated by intersecting HCC at different time points when the tumor load had already reached 9%or even 42%of the body weight.Comparing spatial transcriptomic profiles of Gal1 silenced and OE HCC,inhibiting matrix formation and recognition of foreign antigen in CD45t cell-enriched areas located at tumor-margin likely contributed to the anti-HCC effects of Gal1 silencing.Within the tumors,silencing Gal1 inhibited translational initiation,elongation,and termination.Furthermore,Gal1 silencing increased immune cells as well as expanded cytotoxic T cells within the tumor,and the anti-HCC effect of lgals1 siRNAwas CD8-dependent.Overall,Gal1 silencing has a promising potential for HCC treatment.

Spark plasma sintering of tungsten-based WTaVCr refractory high entropy alloys for nuclear fusion applications

W-based WTaVCr refractory high entropy alloys (RHEA) may be novel and promising candidate materials for plasma facing components in the first wall and diverter in fusion reactors. This alloy has been developed by a powder metallurgy process combining mechanical alloying and spark plasma sintering (SPS). The SPSed samples contained two phases, in which the matrix is RHEA with a body-centered cubic structure, while the oxide phase was most likely Ta2VO6through a combined analysis of X-ray diffraction (XRD),energy-dispersive spectroscopy (EDS), and selected area electron diffraction (SAED). The higher oxygen affinity of Ta and V may explain the preferential formation of their oxide phases based on thermodynamic calculations. Electron backscatter diffraction (EBSD) revealed an average grain size of 6.2μm. WTaVCr RHEA showed a peak compressive strength of 2997 MPa at room temperature and much higher micro-and nano-hardness than W and other W-based RHEAs in the literature. Their high Rockwell hardness can be retained to at least 1000°C.

An Effective Approach for the Preparation of High Performance Thermal Conductive Polymer Composites Containing Liquid Metal

The preparation of high-performance thermal conductive composites containing liquid metals(LM)has attracted significant attention.However,the stable dispersion of LM within polymer solution and effective property contribution of liquid metals remains significant challenges that need to be overcome.Inspired by the properties of the dendritic structure of the tree root system in grasping the soil,“shear-induced precipitation-interfacial reset-reprotonation”processing strategy is proposed to prepare nanocomposites based on aramid micron fibers(AMFs)with hierarchical dendritic structure.Thanks to the combination of van der Waals force provided by hierarchical dendritic structure,electrostatic interaction between AMFs and LM,coordinative bonding of―NH to LM,together with interfacial re-setting and multi-step protonation,several features can be achieved through such strategy:conducive to the local filler network construction,improvement of interfacial interaction,improvement of the stability of filler dispersion in the solvent,and enhancement of mechanical and thermal properties of the films.The resulting AMFsp H=4/LM films demonstrate a thermal conductivity of 10.98 W·m^(-1)·K^(-1)at 70%filler content,improvement of 126.8%compared to ANFs/LM film;while maintaining a strength of~85.88 MPa,improvement of 77%compared to AMFs/LM film.They also possess insulation properties,enable heat dissipation for high power electronics.This work provides an effective strategy for the preparation of high performance polymer composites containing liquid metal.

Incidence of surgical site infection in minimally invasive colorectal surgery

BACKGROUND Surgical site infection(SSI)is a common complication of colorectal surgery.Minimally invasive surgery notably reduces the incidence of SSI.This study aimed to compare the incidences of SSI after robot-assisted colorectal surgery(RACS)vs that after laparoscopic assisted colorectal surgery(LACS)and to analyze associated risk factors for SSI in minimally invasive colorectal surgery.AIM To compare the incidences of SSI after RACS and LACS,and to analyze the risk factors associated with SSI after minimally invasive colorectal surgery.METHODS Clinical data derived from patients who underwent minimally invasive colorectal surgery between October 2020 and October 2022 at the First Affiliated Hospital of Soochow University were collated.Differences in clinical characteristics and surgeryrelated information associated with RACS and LACS were compared,and possible risk factors for SSI were identified.RESULTS A total of 246 patients(112 LACS and 134 RACS)were included in the study.Fortythree(17.5%)developed SSI.The proportions of patients who developed SSI were similar in the two groups(17.9%vs 17.2%,P=0.887).Diabetes mellitus,intraoperative blood loss≥100 mL,and incision length were independent risk factors for SSI.Possible additional risk factors included neoadjuvant therapy,lesion site,and operation time.CONCLUSION There was no difference in SSI incidence in the RACS and LACS groups.Diabetes mellitus,intraoperative blood loss≥100 mL,and incision length were independent risk factors for postoperative SSI.

Quantification of Central and Eastern China's atmospheric CH_(4) enhancement changes and its contributions based on machine learning approach

Methane is the second largest anthropogenic greenhouse gas,and changes in atmospheric methane concentrations can reflect the dynamic balance between its emissions and sinks.Therefore,the monitoring of CH_(4) concentration changes and the assessment of underlying driving factors can provide scientific basis for the government’s policy making and evaluation.China is the world’s largest emitter of anthropogenic methane.However,due to the lack of ground-based observation sites,little work has been done on the spatial-temporal variations for the past decades and influencing factors in China,especially for areas with high anthropogenic emissions as Central and Eastern China.Here to quantify atmospheric CH_(4) enhancements trends and its driving factors in Central and Eastern China,we combined the most up-to-date TROPOMI satellite-based column CH_(4)(xCH_(4))concentration from 2018 to 2022,anthropogenic and natural emissions,and a random forest-based machine learning approach,to simulate atmospheric xCH_(4) enhancements from 2001 to 2018.The results showed that(1)the random forest model was able to accurately establish the relationship between emission sources and xCH_(4) enhancement with a correlation coefficient(R^(2))of 0.89 and a root mean-square error(RMSE)of 11.98 ppb;(2)The xCH_(4) enhancement only increased from 48.21±2.02 ppb to 49.79±1.87 ppb from the year of 2001 to 2018,with a relative change of 3.27%±0.13%;(3)The simulation results showed that the energy activities and waste treatment were the main contributors to the increase in xCH_(4) enhancement,contributing 68.00% and 31.21%,respectively,and the decrease of animal ruminants contributed-6.70% of its enhancement trend.

High cerium content(Nd-Pr-Ce)_(2)Fe_(14)B melt-spun ribbons with high coercivity

The effects of wheel speeds on the magnetic properties and microstructures of[(Nd,Pr)_(1-x)Ce_(x)]-Fe-B meltspun ribbons were investigated.Compared to melt-spun ribbons with low cerium(Ce)content(x=0.2),amorphous formation can be suppressed at high wheel speed in the ribbons with a relatively high Ce content(x=0.8),and with the increase of the wheel speed,the magnetic properties of the ribbons with high Ce content raise continuously.At high wheel speed,the coercivity mechanism of melt-spun ribbons is mainly pinning field,and the high wheel speed introduces a large number of defects into the matrix as pinning sites.Furthermore,ribbons with high Ce content form a fine and uniform grain structure,so the grain boundary area enlarges,which further hinders the movement of domain walls.The combined effects of the above factors enable ribbons with high Ce content to have excellent magnetic properties(Hcj=0.977 T,Mr=64 m^(2) A/kg)at the wheel speed of 35 m/s.

Highly sensitive nanozyme strip:An effective tool for forensic material evidence identification

During criminal case investigations,blood evidence tracing is critical for criminal investigation.However,the blood stains are often cleaned or covered up after the crime,resulting in trace residue and difficult tracking.Therefore,a highly sensitive and specific method for the rapid detection of human blood stains remains urgent.To solve this problem,we established a nanozyme-based strip for rapid detection of blood evidence with high sensitivity and specificity.To construct reliable nanozyme strips,we synthesized CoFe_(2)O_(4) nanozymes with high peroxidase-like activity by scaling up to gram level,which can be supplied for six million tests,and conjugated antibody as a detection probe in nanozyme strip.The developed CoFe_(2)O_(4) nanozyme strip can detect human hemoglobin(HGB)at a concentration as low as 1 ng/mL,which is 100 times lower than the commercially available colloidal gold strips(100 ng/mL).Moreover,this CoFe_(2)O_(4) nanozyme strip showed high generality on 12 substrates and high specificity to human HGB among 13 animal blood samples.Finally,we applied the developed CoFe_(2)O_(4) nanozyme strip to successfully detect blood stains in three real cases,where the current commercial colloidal gold strip failed to do.The results suggest that the CoFe_(2)O_(4) nanozyme strip can be used as an effective on-scene detection method for human blood stains,and can further be used as a long-term preserved material evidence for traceability inquiry.

Chemotherapy-induced complications,particularly lethal cardiovascular diseases,pose significant challenges for cancer survivors.The intertwined adverse effects,brought by cancer and its complication,further complicate anticancer therapy and lead to diminished clinical outcomes.Simple supplementation of cardioprotective agents falls short in addressing these challenges.Developing bifunctional co-therapy agents provided another potential solution to consolidate the chemotherapy and reduce cardiac events simultaneously.Drug repurposing was naturally endowed with co-therapeutic potential of two indications,implying a unique chance in the development of bi-functional agents.Herein,we further proposed a novel“trilogy of drug repurposing”strategy that comprises function-based,targetfocused,and scaffold-driven repurposing approaches,aiming to systematically elucidate the advantages of repurposed drugs in rationally developing bi-functional agent.Through function-based repurposing,a cardioprotective agent,carvedilol(CAR),was identified as a potential neddylation inhibitor to suppress lung cancer growth.Employing target-focused SAR studies and scaffold-driven drug design,we synthesized 44 CAR derivatives to achieve a balance between anticancer and cardioprotection.Remarkably,optimal derivative 43 displayed promising bi-functional effects,especially in various self-established heart failure mice models with and without tumor-bearing.Collectively,the present study validated the practicability of the“trilogy of drug repurposing”strategy in the development of bi-functional cotherapy agents.

Priority Encoder Based on DNA Strand Displacement

The slow development of traditional computing has prompted a search for new materials to replace silicon-based computers. Bio-computers, which use molecules as the basis of computation, are highly parallel and information capable, attracting a lot of attention. In this study, we designed a NAND logic gate based on the DNA strand displacement mechanism. We assembled a molecular calculation model, a 4-wire-2-wire priority encoder logic circuit, by cascading the proposed NAND gates. Different concentrations of input DNA chains were added into the system, resulting in corresponding output, through DNA hybridization and strand displacement. Therefore, it achieved the function of a priority encoder. Simulation results verify the effectiveness and accuracy of the molecular NAND logic gate and the priority coding system presented in this study. The unique point of this proposed circuit is that we cascaded only one kind of logic gate, which provides a beneficial exploration for the subsequent development of complex DNA cascade circuits and the realization of the logical coding function of information.

Near infrared emissions from both high efficient quantum cutting (173%) and nearly-pure-color upconversion in NaY(WO_(4))_(2):Er^(3+)/Yb^(3+) with thermal management capability for silicon-based solar cells

Raising photoelectric conversion efficiency and enhancing heat management are two critical concerns for siliconbased solar cells.In this work,efficient Yb^(3+) infrared emissions from both quantum cutting and upconversion were demonstrated by adjusting Er^(3+) and Yb^(3+) concentrations,and thermo-manage-applicable temperature sensing based on the luminescence intensity ratio of two super-low thermal quenching levels was discovered in an Er^(3+)/Yb^(3+) codoped tungstate system.The quantum cutting mechanism was clearly decrypted as a two-step energy transfer process from Er^(3+) to Yb^(3+).The two-step energy transfer efficiencies,the radiative and nonradiative transition rates of all interested 4 f levels of Er^(3+) in NaY(WO_(4))_(2) were confirmed in the framework of Föster-Dexter theory,Judd-Ofelt theory,and energy gap law,and based on these obtained efficiencies and rates the quantum cutting efficiency was furthermore determined to be as high as 173%in NaY(WO_(4))_(2):5 mol%Er^(3+)/50 mol%Yb^(3+) sample.Strong and nearly pure infrared upconversion emission of Yb3+under 1550 nm excitation was achieved in Er^(3+)/Yb^(3+)co-doped NaY(WO_(4))_(2) by adjusting Yb^(3+) doping concentrations.The Yb^(3+) induced infrared upconversion emission enhancement was attributed to the efficient energy transfer ^(4)I_(11/2)(Er^(3+))+^(2)F_(7/2)(Yb^(3+))→^(4)I_(15/2)(Er^(3+))+^(2)F_(5/2)(Yb^(3+))and large nonradiative relaxation rate of ^(4)I_(9/2).Analysis on the temperature sensing indicated that the NaY(WO_(4))_(2):Er^(3+)/Yb^(3+)serves well the solar cells as thermos-managing material.Moreover,it was confirmed that the fluorescence thermal quenching of ^(2)H_(11/2)/^(4)S_(3/2) was caused by the nonradiative relaxation of ^(4)S_(3/2).All the obtained results suggest that NaY(WO_(4))_(2):Er^(3+)/Yb^(3+) is an excellent material for silicon-based solar cells to improve photoelectric conversion efficiency and thermal management.
