Comparison of fondaparinux sodium and low molecular weight heparin in the treatment of hypercoagulability secondary to traumatic infection

Purpose: To compare the effects and side-effects of fondaparinux sodium and low molecular weight heparin in patients with hypercoagulability accompanied with traumatic infection. Methods: Thirty-six patients with post-traumatic infections in our hospital intensive care center were diagnosed with hypercoagulability from February 2012 to February 2013. These patients were randomly divided into 2 groups. In group F (18 patients), the patients were treated with fondaparinux sodium, 2.5 mg, 1/d for 11 d. In group L (18 patients), the patients were treated with low molecular weight heparin, 4100 U, 1/12 h for 11 d. The incidence of deep vein thrombosis, bleeding events and multiple organ dysfunction syndrome (MODS) and mortality of two groups after anticoagulation therapy were analyzed. Fibrinogen, D-dimer level and activity of antithrombin III were measured by the coagulation analyzer. Results: The incidence of deep vein thrombosis, MODS incidence and mortality were not significantly different between the two groups. The rate of bleeding evens in group F was lower than group L ( p 0.05). D-dimer was significantly decreased after anticoagulant therapy for 5 d ( p 0.05). Conclusion: Fondaparinux sodium and low molecular weight heparin can effectively improve coagulopathy in patients with traumatic infection. Compared with low molecular weight heparin, fondaparinux sodium may reduce the risk of bleeding events in patients with hypercoagulability accompanied by traumatic infection.

Invasive fungal infections are life-threatening,and neutrophils are vital cells of the innate immune system that defend against them.The role of LTA4H-LTB_(4)-BLT1 axis in regulation of neutrophil responses to fungal infection remains poorly understood.Here,we demonstrated that the LTA4H-LTB_(4)-BLT1 axis protects the host against Candida albicans and Aspergillus fumigatus,but not Cryptococcus neoformans infection,by regulating the antifungal activity of neutrophils.Our results show that deleting Lta4h or Blt1 substantially impairs the fungal-specific phagocytic capacity of neutrophils.Moreover,defective activation of the spleen tyrosine kinase(Syk)and extracellular signal-related kinase(ERK1/2)pathways in neutrophils accompanies this impairment.Mechanistically,BLT1 regulates CR3-mediated,β-1,3-glucan-induced neutrophil phagocytosis,while a physical interaction with CR3 with slight influence on its dynamics is observed.Our findings thus demonstrate that the LTA4H-LTB_(4)-BLT1 axis is essential for the phagocytic function of neutrophils in host antifungal immune response against Candida albicans and Aspergillus fumigatus.

More Cycles of Intravenous Chemotherapy are Associated with Reduced Growth in Children with Retinoblastoma

Background Chemotherapy can have a negative impact on the growth of children with different cancers.However,few studies have examined whether intravenous chemotherapy(IVC)affects the growth of children with retinoblastoma(RB).The present study evaluated the height differences(actual height compared to the age standardized value)and survival of pediatric RB patients treated with IVC.Methods This was an observational cohort study.A total of 87 pediatric RB patients were included.The study population was stratified into two groups based on the number of chemotherapy cycles administered(≤4 versus>4).The height at baseline(before IVC),height after IVC and overall survival were compared between the two groups.Results Before IVC,no height differences were observed between the two groups(P=0.585).After IVC,all of the patients had a reduced height compared to the age standardized height(P=0.035).Patients who underwent more cycles of chemotherapy had a greater height difference compared to those who received fewer cycles(P=0.008).For those who had reduced height,the difference was positively associated with the number of chemotherapy cycles(r=0.279,P=0.043).Among the patients who exhibited a greater height difference,those who underwent more than four cycles of chemotherapy had a decreased overall survival(P=0.042).Conclusions Pediatric RB patients who underwent more cycles of chemotherapy were more likely to have a reduced height.Further studies are needed to determine the optimal treatment strategy to prevent the reduced growth while maintaining the benefits of chemotherapy.

EGCG and ECG induce apoptosis and decrease autophagy via the AMPK/mTOR and PI3K/AKT/mTOR pathway in human melanoma cells

Catechins have been proven to exert antitumor effects in different kinds of cancers.However,the underlying mechanisms have not been completely clarified yet.This study aimed to assess the effects and mechanisms of(-)-epigallocatechin-3-gallate(EGCG)and(-)-epicatechin-3-gallate(ECG)on human melanoma skin A375 cells.Results showed that EGCG and ECG inhibited the proliferation of A375 cells and ECG showed better inhibitory effect.Flow cytometry analysis had shown that EGCG and ECG induced apoptosis and led to cell cycle arrest.EGCG and ECG decreased Bcl-2 expression and upregulated Caspase-3 protein level,indicating the development of apoptosis.Furthermore,EGCG and ECG could decreased mitochondrial membrane potential of A375 cells.In addition,the expression of Beclin-1,LC3 and Sirt3 were downregulated at protein levels,which known to be associated with autophagy.After autophagy was increased by rapamycin,the apoptotic trend was not change,indicating that apoptosis and autophagy are independent.Mechanistically,EGCG and ECG treatments decreased phosphorylated-AMPK(p-AMPK)and increased the ratios of P-PI3K,p-AKT and p-mTOR in melanoma cells.Conclusively,EGCG and ECG induced apoptosis via mitochondrial signaling pathway,downregulated autophagy through modulating the AMPK/mTOR and PI3K/AKT/mTOR signaling pathway.It indicated that EGCG and ECG may be utilized in human melanoma treatment.

Introduction:High-resolution data is essential for understanding the complexity of the relationship between the spread of coronavirus disease 2019(COVID-19),resident behavior,and interventions,which could be used to inform policy responses for future prevention and control.Methods:We obtained high-resolution human mobility data and epidemiological data at the community level.We propose a metapopulation Susceptible-Exposed-Presymptomatic-Infectious-Removal(SEPIR)compartment model to utilize the available data and explore the internal driving forces of COVID-19 transmission dynamics in the city of Wuhan.Additionally,we will assess the effectiveness of the interventions implemented in the smallest administrative units(subdistricts)during the lockdown.Results:In the Wuhan epidemic of March 2020,intra-subdistrict transmission caused 7.6 times more infections than inter-subdistrict transmission.After the city was closed,this ratio increased to 199 times.The main transmission path was dominated by population activity during peak evening hours.Discussion:Restricting the movement of people within cities is an essential measure for controlling the spread of COVID-19.However,it is difficult to contain intra-street transmission solely through citywide mobility restriction policies.This can only be accomplished by quarantining communities or buildings with confirmed cases,and conducting mass nucleic acid testing and enforcing strict isolation protocols for close contacts.

Additive manufacturing(AM)with particular processing characteristics shows excellent potential to innovate the manufacturing industry and the production mode.AM offers a considerable degree of design freedom and allows manufacturi ng of geometries that are not feasible by conventional manufacturing and is treated as a superb solution reducing lead time to market for new products.Moreover,the pointwise forming characteristics empower this technique to produce refined and above supernormal microstructures owing to rapid cooling and non-equilibrium solidification,creating more possibilities for material microstructure modification.

Bel 10 is required for the development and suppressive function of Foxp3^(+)regulatory T cells

Foxp3^(+)regulatory T(Treg)cells play a critical role in peripheral tolerance.Bcl1O,acting as a scaffolding protein in the Carma1-Bcl10-Malt1(CBM)complex,has a critical role in TCR-induced signaling,leading to NF-kB activation and is required for T-cell activation.The role of Bcl1O in conventional T(Tconv)cells has been well characterized;however,the role of Bcl1 in the development of Treg cells and the maintenance of the suppressive function and identity of these cells has not been well characterized.In this study,we found that Bcl10 was required for not only the development but also the function of Treg cells.After deleting Bcl1O in T cells,we found that the development of Treg cells was significantly impaired.When Bcl1O was specifically deleted in mature Treg cells,the suppressive function of the Treg cells was impaired,leading to lethal autoimmunity in Bcl10^(fl/fl)Foxp3^(cre) mice.Consistently,in contrast to WT Treg cells,Bcl10-deficient Treg cells could not protect Rag1-deficient mice from T-cell transfer-induced colitis.Furthermore,Bcl1O-deficient Treg cells downregulated the expression of a series of Treg-cell effector and suppressive genes and decreased effector Treg-cell populations.Moreover,Bcl1O-deficient Treg cells were converted into IFNγ-producing proinflammatory cells with increased expression of the transcription factors T-bet and HIF-1α.Together,our study results provide genetic evidence,indicating that Bcl1O is required for the development and function of Treg cells.
