Regulations of m^(6)A and other RNA modifications and their roles in cancer

RNA modification is an essential component of the epitranscriptome,regulating RNA metabolism and cellular *** types of RNA modifications have been identified to date;they include N^(6)-methyladenosine(m^(6)A),N^(1)-methyladenosine(m1A),5-methylcytosine(m5C),N^(7)-methylguanosine(m^(7)G),N^(6),2′-O-dimethyladenosine(m^(6)Am),N4-acetylcytidine(ac^(4)C),*** modifications,mediated by regulators including writers,erasers,and readers,are associated with carcinogenesis,tumor microenvironment,metabolic reprogramming,immunosuppression,immunotherapy,chemotherapy,etc.A novel perspective indicates that regulatory subunits and post-translational modifications(PTMs)are involved in the regulation of writer,eraser,and reader functions in mediating RNA modifications,tumorigenesis,and anticancer *** this review,we summarize the advances made in the knowledge of different RNA modifications(especially m^(6)A)and focus on RNA modification regulators with functions modulated by a series of factors in cancer,including regulatory subunits(proteins,noncoding RNA or peptides encoded by long noncoding RNA)and PTMs(acetylation,SUMOylation,lactylation,phosphorylation,etc.).We also delineate the relationship between RNA modification regulator functions and carcinogenesis or cancer ***,inhibitors that target RNA modification regulators for anticancer therapy and their synergistic effect combined with immunotherapy or chemotherapy are discussed.

Qualitative and quantitative analysis of trace heavy metals in aqueous environment are rapidly assuming significance along with the rapid development of *** this paper,gold microelectrode array(MEA)plated with mercury film was used for simultaneous voltammetric detection of zinc,cadmium,lead and copper ions in *** electrochemical behavior and the actual surface area of the MEA were investigated by cyclic voltammetry in K_(3)[Fe(CN)_(6)].Electrochemical impedance spectrum(EIS)was utilized to examine the deposition of mercury on the electrode *** on anodic stripping voltammetry,mercury filmAu MEA was applied to the detection of heavy metals in artificial analyte,where good calibrate linearity was obtained for cadmium,lead and copper ions,but with zinc exhibiting poor linearity.

For decades,researchers have known that when neurons lose their activating inputs,they can atrophy and even *** example,removing sensory nerve inputs from the arm to the spinal cord after amputation or cutting the spinal nerves can shrink the post-synaptic target zones(e.g.,Woods et al.,2000).From touch receptors for the hand and arm,primary nerve afferents enter the spinal cord,and axons travel in the dorsal columns to target the cuneate nucleus(Cu)of the brainstem on the same side of the body(Figure 1).When sensory loss is unilateral,the overwhelming result is for the associated primary target zone to shrink in total size(cross-sectional area),as compared to the size of the opposite side.

Water inrush is a common disaster in submarine *** key to preventing this disaster is to restrict the water-conducting fissure propagation induced by mining from forming a flow channel to communicate with the overlying *** mechanical behavior of hydraulic fractures under stress disturbance lies at the heart of the ***,the multiple parallel bond-grain-based model(multi Pb-GBM)is introduced to explore the hydraulic fracture evolution law of crystalline granite under the influence of stress *** results show that:hydraulic fracturing under stress disturbance is clearly affected by the stress wave frequency;the higher the frequency is,the more difficult it is to crack,but the crack propagation speed is faster after crack initiation;the propagation direction of a crack is deflected towards the propagation direction of the stress wave and the crack dip angle is controlled by the maximum principal stress;the internal crystal boundary of the same mineral is the most stable one among the three contacts,the contact boundary between different minerals being the most fragile *** research not only has a practical application value to seabed mining engineering,but also has important theoretical significance in enriching deep rock mechanics theory.

Ground subsidence mechanism of a filling mine with a steeply inclined ore body

Long-term field monitoring finds that serious surface subsidence can still occur even if the high strength cemented fill method is *** the results of numerical simulations with global position system(GPS)monitoring,we took a typical filling mining mine with a steeply inclined ore body as an example,and explored its ground subsidence *** results show that the ground subsidence caused by the mining of steep ore body is characterized by two settlement centers and a significantly uneven spatial distribution,which is visibly different from ground subsidence characteristic of the coal *** subsidence on the hanging wall is much larger than that on the footwall,and the settlement center tends to move to the hanging wall with the increase of mining *** backfill improves the strength and surrounding rock bearing capacity,which leads to a lag of about 3 years of the ***,under the actions of continuous and repeated mining disturbances,the supporting effect of the backfill can only reduce the amplitude of the deformation,but it cannot prevent the occurrence of settlement.

Gli-1 siRNA induced apoptosis in Huh7 cells

AIM： To investigate the effects of Gli-1 small interference RNA （siRNA） on Huh7 cells, and the change of Bcl-2 expression in Huh7 cells. METHODS： Human hepatocellular carcinoma cells Huh7 were used. Cell viability was analyzed by 3-（4, 5-Dimethylthiazol-2-yl）-2, 5-diphenyl tetrazolium bromide （MTT） assay. The expressions of Gli-1 and Bcl-2 family members were detected by RT-PCR and Western blot. Apoptosis was detected by Flow cytometry using propidium iodide, measured by Hoechst 33258 staining using Advanced Fluorescence Microscopy and caspase-3 enzymatic assay. Cell growth was analyzed after treatment with Gli-1 siRNA and 5-fluorouracil （5-Fu）. RESULTS： Inhibition of Gli-1 mRNA in Huh7 cells through Gli-1 siRNA reduced cell viability. Gli-1 siRNA treatment also induced apoptosis by three criteria, increase in the sub-G1 cell cycle fraction, nuclear condensation, a morphologic change typical of apoptosis, and activation of caspase-3. Gli-1 siRNA was also able to down-regulate Bcl-2. However, Gli-1 siRNA resulted in no significant changes in Bcl-xl, Bax, Bad, and Bid. Furthermore, Gli-1 siRNA increased the cytotoxic effect of 5-Fu on Huh7 cell. CONCLUSION： Down-regulation of Bcl-2 plays an important role in apoptosis induced by Gli-1 siRNA in HCC cells. Combination Gli-1 siRNA with chemotherapeutic drug could represent a more promising strategy against HCC. The effects of the strategies need further investigation in vivo and may have potential clinical application.

Synergistic Optimization of Toughness and EMI Shielding Properties in Rigid Polyurethane Composites by Designing Ring Structures on the Surface of Carbon Fibers

How to achieve both toughness and enhanced electromagnetic interference shielding effectiveness(EMI SE)of carbon fibers(CFs)reinforced rigid polyurethane(RPU)composites is a significative challenge at *** this work,a ring-shaped zinc coating was deposited on the short CFs by electrodeposition *** is expected to improve the interfacial properties between the fibers and the resin matrix as well as enhance the EMI shielding properties of the composites by changing the surface morphology and roughness of the *** showed that the surface free energy of the ring-shaped zinc modified carbon fibers(RS-CFs)increased from 49.0 mJ/m^(2) to 53.2 mJ/m^(2),indicating that the surface roughness and wettability of the CFs were effectively *** comparison with the pristine short carbon fibers/rigid polyurethane(CFs/RPU)composites,tensile strength and tensile toughness of RS-CFs modified composites were increased by 27.1% and 121.4%,*** addition,the bending and impact strengths of RS-CFs reinforced RPU composites were also ***,the electrical conductivity of RS-CFs/RPU composites reached 1.2×10^(-5) S/m,which was much higher than that of CFs/RPU composites at 1.4×10^(-10) S/***,the EMI SE of the modified composites reached 22 dB,which was 152.9% higher than that of CFs/RPU *** enhanced electrical conductivity and EMI shielding properties of the composites could be attributed to the synergistic effect of the porous structure in the RPU matrix and the CFs modified by the metal coating.

A topological theory of liquid crystal films in the presence of defects is developed based on the Ф-mapping topological current theory. By generalizing the free-energy density in ＂one-constant＂ approximation, a covariant free- energy density is obtained, from which the U（1） gauge field and the unified topological current for monopoles and strings in liquid crystals are derived. The inner topological structure of these topological defects is characterized by the winding numbers of Ф-mapping.
