Thrombospondin 1 Promotes Endoplasmic Reticulum Stress and Apoptosis in HK-2 Cells by Upregulating ATF6-CHOP

Objective The goal of this study is to investigate the role and mechanism of endoplasmic reticulum stress and apoptosis regulated by thrombospondin 1(TSP1)in human renal tubular epithelial cells(HK-2 cells).Methods HK-2 cells were exposed to high concentrations of glucose(HG).The endoplasmic reticulum stress inhibitor 4-phenylbutyric acid(4-PBA)was administered by transfecting TSP1 or an empty vector to explore the mechanism of the endoplasmic reticulum response regulated by TSP1 and stress in renal cell apoptosis.The effects of TSP1 and 4-PBA on the proliferation and apoptosis of HK-2 cells under HG conditions were assessed using Cell counting kit-8 and flow cytometry.Western blotting was used to detect the apoptosis-and endoplasmic reticulum stress-related protein expression regulated by TSP1 and 4-PBA.Results HG treatment induced high cell apoptosis,abundantly expressed TSP1 level and restrained viability in HK-2 cells.Overexpression of TSP1 significantly inhibited the proliferation of and facilitated apoptosis of HK-2 cells under HG conditions.Administration of endoplasmic reticulum stress inhibitor 4-PBA after overexpression of TSP1 antagonized the inhibitory proliferation and promoted apoptosis rate in HG-triggered HK-2 cells induced by TSP1 overexpression.4-PBA treatment significantly hindered the expression of endoplasmic reticulum stress markers,such as PERK,ATF4,ATF6,p-eIF2α,IRE1,CHOP and XBP1,suggesting that the administration of 4-PBA was successful.Conclusion Overexpression of TSP1 activated endoplasmic reticulum stress by regulating the ATF6-CHOP axis.TSP1 restrained cell proliferation,and promoted apoptosis and endoplasmic reticulum stress by activating the ATF6-CHOP axis.

Numerous nanocarriers have been currently developed for intracellular delivery.The potential cytotoxicity of these very small inorganic nanocarriers has raised great consideration.Thus,it becomes of utmost importance to conduct the intracellular trace of nanocarriers.Among many analytical techniques,surface enhanced Raman scattering(SERS)method is one of the current state-of-the-art techniques for cell visualization and trace.In this work,a novel stellate porous silica based gene delivery system has been designed for SERS trace purpose.A stellate porous silica nanoparticle modified with many small Au nanoparticles is designed to replace common metallic SERS tags.The results show that the designed system not only could deliver si RNA into cells for therapy,but also could realize SERS trace with high sensitivity and non-invasive features.The constructed delivery system has considerable potential to trace the dynamic gene delivery in living cells.

Both the prevalence of atrial fibrillation(AF)and frailty are increasing with age,and they often occur simultaneously,leading them to become the central concerns in this aging society.The incidence of frailty in patients with AF is highly variable,ranging from 4.4%to 75.4%,depending on different evaluating instruments used.Moreover,the incidence of frailty among patients with AF is on the rise,which indicated that patients with AF are more prone to frailty compared to patients without AF.The relationship between AF and frailty is complicated.Frailty elevates the risk of stroke and mortality in AF patients and is also associated with longer hospitalizations.On the other hand,it may reduce the appropriate anticoagulation in AF patients.However,the evidence of the effects of frailty on anti-arrhythmic and interventional therapy in patients with AF is scarce.Frailty affects both the management and the prognosis of AF in the geriatric population.Vice versa,AF could worsen the frail state and may represent a marker of frailty.However,there are still questions need to be resolved,for example,the impact of frailty on the interventional therapy of patients with AF.Therefore,the geriatric assessment of frailty should be considered when planning individualized management of AF in older patients.

Efficacy and safety of JAK inhibitor INC424 in patients with primary and post-polycythemia vera or post-essential thrombocythemia myelofibrosis in the Chinese population

A phase II study （A2202） was performed to evaluate the efficacy and safety of JAK inhibitor ruxolitinib in 63 Chinese MF patients. Ruxolitinib was given twice a day （bid） at a starting dose of 15 mg （n = 25） or 20 mg （n = 38） based on a baseline platelet count. About 94.7% of the patients achieved a reduction in spleen size, 27.0% of which exhibited significant reduction （≥35%） at week 24. Significant improvement in debilitating constitutional symptoms, as assessed by MFSAF v2.0, was observed in patients treated with ruxolitinib. Ruxolitinib treatment was generally weli tolerated by Chinese patients. Although the treatment was associated with an increase in certain adverse events （AEs） that were established as identified risks （anemia and thrombocytopenia）, these AEs were considered manageable in this clinical setting. Ruxolitinib provided substantial reductions in splenomegaly and improvements in symptoms, and was well-tolerated by Chinese patients with MF.

A Meta-analysis of Molecular Spectroscopy Databases,and Prospects of Molecule Detection with Some Future Facilities

Molecules reside broadly in the interstellar space and can be detected via spectroscopic observations.To date,more than 271 molecular species have been identified in interstellar medium or circumstellar envelopes.Molecular spectroscopic parameters measured in laboratory make the identification of new species and derivation of physical parameters possible.These spectroscopic parameters are systematically collected into databases,two of the most commonly used being the CDMS and JPL databases.While new spectroscopic parameters are continuously measured/calculated and added to those databases,at any point in time it is the existing spectroscopic data that ultimately limits what molecules can possibly be identified in astronomical data.In this work,we conduct a meta-analysis of the CDMS and JPL databases.We show the statistics of transition frequencies and their uncertainties in these two databases,and discuss the line confusion problem under certain physical environments.We then assess the prospects of detecting molecules in common ISM environments using a few facilities that are expected to be conducting spectroscopic observations in the future.Results show that CSST/HSTDM and SKA1-mid have the potential to detect some complex organic molecules,or even amino acids,with reasonable assumptions about ISM environments.

Preparation of a biomimetic superomniphobic hierarchical structure and analysis of droplet wettability

In this study,a bioinspired hierarchical structure was formed with superomniphobicity on a 7075 aluminium alloy using laser ablation.The morphology and wetting characteristics of the biomimetic sample surface were characterised using scanning electron microscopy,laser confocal microscopy,and contact angle measurements.The effect of the liquid properties and surface structures on the rolling behaviour was investigated.The results suggest that the fabricated biomimetic sample surface was a hierarchical structure.The prepared sample had superomniphobicity and low adhesion properties,and the contact angles of six different liquid droplets on the sample surface reached or approached 150°.Specifically,the largest contact angle was 152°,and the average rolling angle was 7.7°.This multi-hydrophobic surface provides a valuable reference for the research study and use of a variety of solid–liquid contacts.

Web page classification based on heterogeneous features and a combination of multiple classifiers

Precise web page classification can be achieved by evaluating features of web pages, and the structural features of web pages are effective complements to their textual features. Various classifiers have different characteristics, and multiple classifiers can be combined to allow classifiers to complement one another. In this study, a web page classification method based on heterogeneous features and a combination of multiple classifiers is proposed. Different from computing the frequency of HTML tags, we exploit the tree-like structure of HTML tags to characterize the structural features of a web page. Heterogeneous textual features and the proposed tree-like structural features are converted into vectors and fused. Confidence is proposed here as a criterion to compare the classification results of different classifiers by calculating the classification accuracy of a set of samples. Multiple classifiers are combined based on confidence with different decision strategies, such as voting, confidence comparison, and direct output, to give the final classification results. Experimental results demonstrate that on the Amazon dataset, 7-web-genres dataset, and DMOZ dataset, the accuracies are increased to 94.2%, 95.4%, and 95.7%, respectively. The fusion of the textual features with the proposed structural features is a comprehensive approach, and the accuracy is higher than that when using only textual features. At the same time, the accuracy of the web page classification is improved by combining multiple classifiers, and is higher than those of the related web page classification algorithms.
