DNA barcodes,short and unique DNA sequences,play a crucial role in sample identification when processing many samples simultaneously,which helps reduce experimental ***,the low quality of long-read sequencing makes it difficult to identify barcodes accurately,which poses significant challenges for the design of barcodes for large numbers of samples in a single sequencing ***,we present a comprehensive study of the generation of barcodes and develop a tool,PRO,that can be used for selecting optimal barcode sets and *** formulate the barcode design problem as a combinatorial problem and prove that finding the optimal largest barcode set in a given DNA sequence space in which all sequences have the same length is theoretically *** practical applications,we developed the novel method PRO by introducing the probability divergence between two DNA sequences to expand the capacity of barcode kits while ensuring demultiplexing ***,the maximum size of the barcode kits designed by PRO is 2,292,which keeps the length of barcodes the same as that of the official ones used by Oxford Nanopore Technologies(ONT).We validated the performance of PRO on a simulated nanopore dataset with high error *** demultiplexing accuracy of PRO reached 98.29%for a barcode kit of size 2,922,4.31%higher than that of Guppy,the official demultiplexing *** the size of the barcode kit generated by PRO is the same as the official size provided by ONT,both tools show superior and comparable demultiplexing accuracy.

Many different factors,such as species traits,socio-economic factors,geographical and environmental factors,can lead to specimen collection *** study aims to determine whether grassland specimen collection in China is preferred by species traits(i.e.,plant height,flowering and fruiting period),environmental range(i.e.,the temperature and precipitation range)and geographical range(i.e.,distribution range and altitudinal range).Ordinary least squares models and phylogenetic generalized linear mixed models were used to analyze the relationships between specimen number and the explanatory *** Forest models were then used to find the most parsimonious multivariate *** results showed that interannual variation in specimen number between 1900 and 2020 was *** number of these species in southeast China was notably lower than that in northwest *** range and geographical range of species had significant positive correlations with specimen *** addition,there were relatively weak but significant associations between specimen number and species trait(i.e.,plant height and flowering and fruiting period).Random Forest models indicated that distribution range was the most important variable,followed by flowering and fruiting period,and altitudinal *** findings suggest that future floristic surveys should pay more attention to species with small geographical range,narrow environmental range,short plant height,and short flowering and fruiting *** correction of specimen collection preference will also make the results of species distribution model,species evolution and other works based on specimen data more accurate.

In long-cavity edge-emitting diode lasers, longitudinal spatial hole burning (LSHB), two-photon absorption (TPA) and free carrier absorption (FCA) are among the key factors that affect the linear increase in output power at high injection currents. In this paper, a simplified numerical analysis model is proposed for 1. 06 mu m long-cavity diode lasers by combining TPA and FCA losses with one-dimensional (1D) rate equations. The effects of LSHB, TPA and FCA on the output characteristics are systematically analyzed, and it is proposed that adjusting the front facet reflectivity and the position of the quantum well (QW) in the waveguide layer can improve the front facet output power.

Sulfurized polyacrylonitrile(SPAN)with the exceptional stability,safety,low cost,and high capacity have been positioned as a highly promising cathode material for next-generation lithium-ion ***,in the market,polyacrylonitrile(PAN)sourced from different suppliers and available at varying prices exhibits significant variations in physical and chemical properties,resulting in diverse behaviors in Li-SPAN *** studying the mechanism,we found that the PAN copolymerization structure leads to the stacking of chain segments which obstructs the embedding of sulfur and lithium ***,we propose a universal strategy for the refined frameworks by an exogenous additive to modify various PAN raw materials,and the battery capacity and cycling performance are obviously *** a result,the copolymerized SPAN with a poor original capacity is nearly doubled to over 500 mAh g^(-1),almost comparable to high-quality yet expensively imported products;for the sample with a high initial capacity but fading in ether-based electrolytes,it can be modified to maintain stability over 400 *** strategy offers an alternative approach for SPAN modification that is characterized by its simplicity and low cost,thereby facilitating the large-scale development of Li-SPAN batteries.

新月形沙丘的移动特征可以反映区域内风沙运移和地貌演化过程,对于科学开展风沙灾害防治具有重要意义。本文利用卫星遥感影像与气象再分析数据,研究了塔克拉玛干沙漠南缘6个新月形沙丘群2012—2022年的形态变化与移动特征。结果表明:(1)研究区域内新月形沙丘平均移动速率为6.86 m·a^(-1),移动方向与对应区域内盛行风方向基本一致;(2)不同区域内新月形沙丘形态参数之间的关联性呈现显著差异;(3)沙丘高度、下垫面植被覆盖、年均降雨量以及地表温度与沙丘移动速率呈负相关关系,分区拟合结果有差异;(4)且末西南区域的年平均输沙通量约为77120.5 kg·m^(-1)。研究结果反映了新月形沙丘移动规律的复杂性,不同地理环境因子的影响程度或存在显著差异,需结合实地情况进行具体分析。同时,在沙丘演化过程中,形态参数的动态变化反映了沙丘迁移的内在机理,进一步为新月形沙丘的发育机理提供了实证数据,也为沙丘危害防治和土地规划提供了参考依据。

Perspective of Calcium Imaging Technology Applied to Acupuncture Research

Acupuncture, a therapeutic treatment defined as the insertion of needles into the body at specific points(ie, acupoints), has growing in popularity world-wide to treat various diseases effectively, especially acute and chronic pain. In parallel, interest in the physiological mechanisms underlying acupuncture analgesia, particularly the neural mechanisms have been increasing. Over the past decades, our understanding of how the central nervous system and peripheral nervous system process signals induced by acupuncture has developed rapidly by using electrophysiological methods. However, with the development of neuroscience, electrophysiology is being challenged by calcium imaging in view field, neuron population and visualization in vivo. Owing to the outstanding spatial resolution, the novel imaging approaches provide opportunities to enrich our knowledge about the neurophysiological mechanisms of acupuncture analgesia at subcellular, cellular, and circuit levels in combination with new labeling, genetic and circuit tracing techniques. Therefore, this review will introduce the principle and the method of calcium imaging applied to acupuncture research. We will also review the current findings in pain research using calcium imaging from in vitro to in vivo experiments and discuss the potential methodological considerations in studying acupuncture analgesia.
