Core–Shell Microfiber Encapsulation Enables Glycerol‑Free Cryopreservation of RBCs with High Hematocrit

Cryopreservation of red blood cells(RBCs)provides great potential benefits for providing transfusion timely in emergencies.High concentrations of glycerol(20%or 40%)are used for RBC cryopreservation in current clinical practice,which results in cytotoxicity and osmotic injuries that must be carefully controlled.However,existing studies on the low-glycerol cryopreservation of RBCs still suffer from the bottleneck of low hematocrit levels,which require relatively large storage space and an extra concentration process before transfusion,making it inconvenient(time-consuming,and also may cause injury and sample lose)for clinical applications.To this end,we develop a novel method for the glycerol-free cryopreservation of human RBCs with a high final hematocrit by using trehalose as the sole cryoprotectant to dehydrate RBCs and using core–shell alginate hydrogel microfibers to enhance heat transfer during cryopreservation.Different from previous studies,we achieve the cryopreservation of human RBCs at high hematocrit(>40%)with high recovery(up to 95%).Additionally,the washed RBCs post-cryopreserved are proved to maintain their morphology,mechanics,and functional properties.This may provide a nontoxic,high-efficiency,and glycerol-free approach for RBC cryopreservation,along with potential clinical transfusion benefits.

Early Devonian Post-collisional Granitic Magmatism in the North Qilian Orogenic Belt,Western China:Insights into Lithospheric Delamination and Orogenic Collapse

Post-collisional magmatism contains important clues for understanding the reworking and growth of continental crust,as well as lithospheric delamination and orogenic collapse.Early Devonian magmatism has been identified in the North Qilian Orogenic Belt(NQOB).This paper reports an integrated study of petrology,whole-rock geochemistry,Sm-Nd isotope and zircon U-Pb dating,as well as Lu-Hf isotopic data,for two Early Devonian intrusive plutons.The Yongchang and Chijin granites yield zircon U-Pb ages of 394-407 Ma and 414 Ma,respectively.Both of them are characterized by weakly peraluminous to metaluminous without typical aluminium-rich minerals,LREE-enriched patterns with negative Eu anomalies and a negative correlation between P_(2)O_(5) and SiO_(2) contents,consistent with geochemical features of I-type granitoids.Zircons from the studied granites display negative to weak positive ε_(Hf)(t)values(−5.7 to 2.1),which agree well with those of negative ε_(Nd)(t)values(−6.4 to−2.9)for the whole-rock samples,indicating that they were derived from the partial melting of Mesoproterozoic crust.Furthermore,low Sr/Y ratios(1.13-21.28)and high zircon saturation temperatures(745℃ to 839℃,with the majority being>800℃)demonstrated a relatively shallow depth level below the garnet stability field and an additional heat source.Taken together,the Early Devonian granitic magmatism could have been produced by the partial melting of ancient crustal materials heated by mantle-derived magmas at high-temperature and low-pressure conditions during postcollisional extensional collapse.The data obtained in this study,when viewed in conjunction with previous studies,provides more information about the tectonic processes that followed the closure of the North Qilian Ocean.The tectonic transition from continental collision to post-collisional delamination could be constrained to~430 Ma,which is provided by the sudden decrease of Sr/Y and La/Yb ratios and an increase in zircon ε_(Hf)(t)values for granitoids.A two-stage tectonic evolution model

Acupotomy alleviates knee osteoarthritis in rabbit by regulating chondrocyte mitophagy via Pink1-Parkin pathway

OBJECTIVE: To investigate the effect of acupotomy, on mitophagy and the Pink1-Parkin pathway in chondrocytes from rabbits with knee osteoarthritis(KOA).METHODS: A KOA model was established via the modified Videman method. Rabbits were randomly divided into a control group(CON), KOA group and KOA + acupotomy group(Acu). Rabbits in the acupotomy group were subjected to acupotomy for 4 weeks after model establishment. The behavior of the rabbits before and after intervention was recorded. Cartilage degeneration was evaluated by optical microscopy and fluorescence microscopy. The level of mitophagy was evaluated by transmission electron microscopy,immunofluorescence and enzyme-linked immunosorbent assay(ELISA). The expression of phosphatase and tensin homolog(PTEN)-induced kinase 1(Pink1)-Parkin mitophagy pathway components was evaluated by immunofluorescence, Western blotting and real-time polymerase chain reaction.RESULTS: In rabbits with KOA, joint pain, mobility disorders and cartilage degeneration were observed, the Mankin score was increased, collagen type Ⅱ(Col-Ⅱ)expression was significantly decreased, mitophagy was inhibited, mitochondrial function was impaired, and factors associated with the Pink1-Parkin pathway were inhibited. Acupotomy regulated the expression of Pink1-Parkin pathway-related proteins, the mitophagy-related protein microtubule-associated protein-1 light chain-3,the translocase of the outer membrane, and the inner mitochondrial membrane 23; increased the colocalization of mitochondria and autophagosomes; promoted the removal of damaged mitochondria;restored mitochondrial adenosine-triphosphate(ATP) production;and alleviated cartilage degeneration in rabbits with KOA.CONCLUSIONS: Acupotomy played a role in alleviating KOA in rabbits by activating mitophagy in chondrocytes via the regulation of proteins that are related to the Pink1-Parkin pathway.

Analysis of conductance and interfacial polarisation behaviours of silicone rubber based on frequency domain spectroscopy

Silicone rubber(SR)is widely used for cable insulation and operates in multi-stress coupling conditions.Its dielectric properties and ageing performance are the essential factors affecting cable operation.In this study,the frequency domain spectroscopy of unaged SR and SR aged under various conditions is measured.The dielectric responses before and after ageing are analysed in depth by the conjoint analysis via multiple complex planes(permittivity,conductivity,and modulus).Typical conductivity and interfacial polarisation properties,as well as property changes resulting from thermal and tensile ageing,are examined.Experimental results show that tensile ageing promotes conductance and interfacial polarisation,while thermal ageing shows the opposite effects.Based on the multi-core model and charge carrier hopping transport model,thermal and tensile ageing mechanisms on the conductance and interfacial polarisation behaviours of SR are demonstrated.Both behaviours exhibit positively correlated changing characteristics,suggesting that the two behaviours may originate from a common process.The carrier transport process,promoted by the tensile effect and slightly suppressed by the thermal effect,is the primary factor and considered an important indicator to evaluate the dielectric properties of SR insulation.

Fast,simple,efficient Agrobacterium rhizogenes-mediated transformation system to non-heading Chinese cabbage with transgenic roots

Non-heading Chinese cabbage, a variety of Brassica campestris, is an important vegetable crop in the yangtze River Basin of China. However,the immaturity of its stable transformation system and its low transformation efficiency limit gene function research on non-heading Chinese cabbage. Agrobacterium rhizogenes-mediated(ARM) transgenic technology is a rapid and effective transformation method that has not yet been established for non-heading Chinese cabbage plants. Here, we optimized conventional ARM approaches(one-step and two-step transformation methods) suitable for living non-heading Chinese cabbage plants in nonsterile environments. Transgenic roots in composite non-heading Chinese cabbage plants were identified using phenotypic detection, fluorescence observation, and PCR analysis. The transformation efficiency of a two-step method on four five-day-old non-heading Chinese cabbage seedlings(Suzhouqing, Huangmeigui, Wuyueman, and Sijiu Caixin) was 43.33%-51.09%, whereas using the stout hypocotyl resulted in a transformation efficiency of 54.88% for the 30-day-old Sijiu Caixin.The one-step method outperformed the two-step method;the transformation efficiency of different varieties was above 60%, and both methods can be used to obtain transgenic roots for functional studies within one month. Finally, optimized ARM transformation methods can easily,quickly, and effectively produce composite non-heading Chinese cabbage plants with transgenic roots, providing a reliable foundation for gene function research and non-heading Chinese cabbage genetic improvement breeding.

Excitonic devices based on two-dimensional transition metal dichalcogenides van der Waals heterostructures

Excitonic devices are an emerging class of technology that utilizes excitons as carriers for encoding, transmitting, and storing information. Van der Waals heterostructures based on transition metal dichalcogenides often exhibit a type II band alignment, which facilitates the generation of interlayer excitons. As a bonded pair of electrons and holes in the separation layer, interlayer excitons offer the chance to investigate exciton transport due to their intrinsic out-of-plane dipole moment and extended exciton lifetime. Furthermore, interlayer excitons can potentially analyze other encoding strategies for information processing beyond the conventional utilization of spin and charge. The review provided valuable insights and recommendations for researchers studying interlayer excitonic devices within van der Waals heterostructures based on transition metal dichalcogenides. Firstly, we provide an overview of the essential attributes of transition metal dichalcogenide materials, focusing on their fundamental properties, excitonic effects, and the distinctive features exhibited by interlayer excitons in van der Waals heterostructures. Subsequently, this discourse emphasizes the recent advancements in interlayer excitonic devices founded on van der Waals heterostructures, with specific attention is given to the utilization of valley electronics for information processing, employing the valley index. In conclusion, this paper examines the potential and current challenges associated with excitonic devices.

Highly efficient through-space charge transfer TADF molecule employed in TADF-and TADF-sensitized organic light-emitting diodes

The inception and harnessing of excitons are paramount for the electroluminescence performance of organic light-emitting devices(OLEDs).Through-space charge transfer(TSCT)via intramolecular interaction has proved to be one of the most potent techniques employed to achieve 100% internal quantum efficiency.However,molecular strategies utilized to comprehensively enhance the electroluminescent performance of TSCT emitters regarding improving the photoluminescence quantum yield(PLQY)and elevating the light out-coupling efficiency remain arduous.To surmount this challenge,we deliberately designed and synthesized a proof-of-concept TSCT emitter called CzO-TRZ by incorporating an extra carbazole donor into spiroheterocyclic architecture.The introduction of rigid spiral fragments can immensely boost the horizontal orientation dipole ratio and establish an extra through-bond charge transfer(TBCT)radiative decay channel.As a result,a very high PLQY of 98.7%,fast kRISCof 2.2×10^(5)s^(-1)and high k_(r)^(s) of 2.2×10^(7)s^(-1),and an ultrahigh horizontal dipolar ratio of 90%were concurrently achieved for Cz O-TRZ blended films.Furthermore,corresponding thermally activated delayed fluorescence(TADF)-and TADF-sensitized fluorescence(TSF)-OLEDs based on CzO-TRZ demonstrated external quantum efficiencies(EQEs)of 33.4% and 30.3%,respectively,highlighting its versatile applications as both an emitter and sensitized host.

Background:Abnormal lipids are strong predictors of cardiovascular disease in type 1 diabetes mellitus(T1DM)and type 2 diabetes mellitus(T2DM).However,the potential associations of insulin resistance(IR)and beta-cell function(BCF)with abnormal lipids in newly diagnosed T1DM or T2DM patients are not fully understood.Methods:A cross-sectional survey of 15,928 participants was conducted.Homeostasis model assessment and postprandial C-peptide levels were used to estimate IR and BCF.A restricted cubic spline(RCS)nested in binary logistic regression was used to examine the associations of IR and BCF with abnormal lipids.Results:High triglyceride(TG),low high-density lipoprotein cholesterol,and high low-density lipoprotein cholesterol(LDL-C)accounted for 49.7%,47.8%,and 59.2%of the participants,respectively.In multivariable analysis,high IR was associated with an increased risk of high TGs(P for trend<0.001)in T1DM and is associated with an elevated risk of high TG and low HDL-C(all P for trend<0.01)in T2DM.Low BCF was not associated with risks of dyslipidemia in patients with T1DM or T2DM after adjustment for potential confounders.Conclusion:High IR had different associations with the risk of dyslipidemia in newly diagnosed T1DM and T2DM patients,suggesting that early treatment that improves IR may benefit abnormal lipid metabolism.

Prokaryotes play a fundamental role in global ocean biogeochemical cycles.However,how the abundance and metabolic activity of ecologically distinct subgroups(i.e.,high nucleic acid(HNA)and low nucleic acid(LNA)cells),and their regulating factors,change in response to changing marine environmental conditions remains poorly understood.Here,we delved into the time-evolving dynamic responses of the HNA and LNA prokaryotic subgroups to declining resource availability and selective grazing by protozoa by conducting a 73-day incubation experiment in a large-volume(117,000 L)macrocosm that facilitates community-level exploration.We found that the metabolic activity of the HNA subgroup was higher than that of the LNA subgroup when the macrocosm was resource replete but that the HNA subgroup declined more rapidly than the LNA subgroup as the resources became increasingly scarce,leading to a steadily increasing contribution of LNA cells to prokaryotic activity.Meanwhile,as resources in the macrocosm became limited,protozoan grazing preference shifted from the HNA to the LNA subgroup and the contributions of the LNA subgroup to the carbon flow within the macrocosm increased.The findings highlight the resilience of LNA cells in resource-limited environments,illuminate the critical role of selective grazing by protozoa in balancing distinct prokaryotic subgroups under changing resource conditions,and demonstrate the complex and adaptive interactions between protozoa and prokaryotes across diverse environmental contexts.

Deep learning-enabled virtual histological staining of biological samples

Histological staining is the gold standard for tissue examination in clinical pathology and life-science research,which visualizes the tissue and cellular structures using chromatic dyes or fluorescence labels to aid the microscopic assessment of tissue.However,the current histological staining workflow requires tedious sample preparation steps,specialized laboratory infrastructure,and trained histotechnologists,making it expensive,time-consuming,and not accessible in resource-limited settings.Deep learning techniques created new opportunities to revolutionize staining methods by digitally generating histological stains using trained neural networks,providing rapid,cost-effective,and accurate alternatives to standard chemical staining methods.These techniques,broadly referred to as virtual staining,were extensively explored by multiple research groups and demonstrated to be successful in generating various types of histological stains from label-free microscopic images of unstained samples;similar approaches were also used for transforming images of an already stained tissue sample into another type of stain,performing virtual stain-to-stain transformations.In this Review,we provide a comprehensive overview of the recent research advances in deep learning-enabled virtual histological staining techniques.The basic concepts and the typical workflow of virtual staining are introduced,followed by a discussion of representative works and their technical innovations.We also share our perspectives on the future of this emerging field,aiming to inspire readers from diverse scientifc fields to further expand the scope of deep learning-enabled virtual histological staining techniques and their applications.
