Fluorescence imaging of single molecules is becoming a powerful tool to examine biological processes at the molecular *** total internal reflection fluorescence microscopy (TIRFM),it has been possible to study the dynamic behavior of single molecules on living cell ***,we briefly review the application of TIRFM-based single-molecule imaging in studies of membrane receptors involved in signal ***,we discuss several examples of our own research on growth factor receptors,including TGF-β receptors,HER2,and EGFR,and speculate possible applications of this technique to investigate other cellular events occurring on or near the plasma-membrane.

BACKGROUND X-linked agammaglobulinemia is a primary immunodeficiency disease caused by gene mutations of Bruton’s tyrosine kinase(BTK).We found a new mutation point and summarized the correlation analysis and performed a literature *** SUMMARY The proband was a 5-year-old *** was admitted to our hospital due to a recurrent cough and a fever that had persisted for a *** had a history of multiple respiratory infections and *** was no immunodeficiency or recurrent infection history among his family *** was characterized as follows:Immunoglobulin(Ig)A,90.0 mg/dL(90-450 mg/dL);IgG,20.0 mg/dL(800-1800 mg/dL);and IgM,18.0 mg/dL(60-280 mg/dL).Notably,the assessment of IgG subtypes revealed the following very low levels:Subtype 1,0.26 g/L(3.62-12.28 g/L);subtype 2,0.10 g/L(0.57-2.9 g/L);subtype 3,0.009 g/L(0.129-0.789 g/L);and subtype 4,0.003 g/L(0.013-1.446 g/L).Cellular immunological test results were as follows:CD3,74.6%(50%-84.0%);CD4,47.3%(27.0%-51.0%);and CD8,24.9%(15.0%-44.0%).A de novo hemizygous deletion in BTK was detected:c.902_c.904delAAG/*** levels of the mutant BTK were similar to those of the wild-type gene,though overexpression resulted in markedly reduced levels of mutant BTK(9.49%±1.58%),relative to the wildtype BTK(75.8%±2.98%,P<0.01).CONCLUSION This case of X-linked agammaglobulinemia was attributed to a de novo hemizygous deletion mutation in BTK(c.902_c.904delAAG/p.E301del).The mutation resulted in markedly reduced BTK protein stability in vitro.

Effects of Imports and Exports on China's PM2.5 Pollution

This paper examines the effects o f imports and exports on Chinas PM2.5 pollution using data from 31 provinces during the period *** the aggregate level,our analysis shows that exports have a pollution-generating effect while imports have a pollution-shifting effect on provincial PM2.5 ***-sectional analysis reveals that imports and exports exhibit opposite effects on manufacturing and high-tech industries,but demonstrate the same pollution-generating effect for the heavy industry *** the regional level,the effects are similar to those at the aggregate *** also examine the impacts o f other factors on PM2.5 *** empirical evidence shows that PM2.5 pollution is positively correlated with domestic sales,population density,economic growth,urbanization rate,and transportation,but negatively correlated with energy efficiency and industry *** paper suggests that reducing exports and increasing imports will help to reduce PM2.5 pollution.

In recent years,with the extensive application of immunotherapy in clinical practice,it has achieved encouraging therapeutic *** enhancing clinical efficacy,however,it can also cause autoimmune damage,triggering immunerelated adverse events(irAEs).Reports of immunotherapy-induced gastritis have been increasing annually,but due to its atypical clinical symptoms,early diagnosis poses a certain ***,it can lead to severe complications such as gastric bleeding,elevating the risk of adverse outcomes for solid tumor patients if immunotherapy is ***,gaining a thorough understanding of the pathogenesis,clinical manifestations,diagnostic criteria,and treatment of immune-related gastritis is of utmost importance for early identification,diagnosis,and ***,the treatment of immune-related gastritis should be personalized according to the specific condition of each *** patients with grade 2-3 irAEs,restarting immune checkpoint inhibitors(ICIs)therapy may be considered when symptoms subside to grade *** restarting ICIs therapy,it is often recommended to use different types of *** grade 4 irAEs,permanent discontinuation of the medication is necessary.

This paper studies the effect of high-speed rail(HSR)on urban economic growth using a panel data comprising 285 Chinese cities in *** the endogenous growth model with a difference-in-difference analysis,we extend the horse-mass theory to explain how China may use HSR to avoid the so-called middle-income *** paper also examines the efficient boundaries of HSR and simultaneously studies HSR timespace compression as well as the city neighboring ejfects on economic *** is found that HSRs efficient boundaries are within the range of 200-1,200 km for provincial capitals and 50-300 km for prefecture-level *** stimulates economic growth by approximately 0.6 percent,and the neighboring effect accounts for one-quarter of economic *** policy implications are drawn:(i)China needs tofurther reduce the travel times between the inland provincial cities and Beijing,Shanghai or Guangzhou;(ii)China should build a denser HSR network to maximize its economic impact on the vast majority of cities;(Hi)China needs to develop some powerful economic growth centers in the inland areas to lead the development of their neighboring cities.

BACKGROUND In most cases of yellow nail syndrome(YNS),the classic triad of yellow nails,lymphedema and respiratory manifestations rarely manifest ***,diagnosis is delayed or frequently *** SUMMARY We report a 62-year-old YNS patient presenting with bilateral pleural,pericardial and peritoneal effusions who,2 mo later,developed minimal-change nephrotic *** treatment with vitamin E,clarithromycin and prednisone for 3 mo,effusions in the chest,pericardium and abdominal cavity decreased while urine protein levels returned to within normal *** Clinicians should consider the possibility of YNS for patients presenting with multiple serous effusions and nephrotic syndromes.
