A Stable Open-Shell Conjugated Diradical Polymer with Ultra-High Photothermal Conversion Efficiency for NIR-Ⅱ Photo-Immunotherapy of Metastatic Tumor

Massive efforts have been concentrated on the advance of eminent near-infrared(NIR) photothermal materials(PTMs) in the NIR-Ⅱ window(1000–1700 nm), especially organic PTMs because of their intrinsic biological safety compared with inorganic PTMs. However, so far, only a few NIR-Ⅱresponsive organic PTMs was explored, and their photothermal conversion efficiencies(PCEs) still remain relatively low. Herein, donor–acceptor conjugated diradical polymers with open-shell characteristics are explored for synergistically photothermal immunotherapy of metastatic tumors in the NIR-Ⅱ window. By employing side-chain regulation, the conjugated diradical polymer TTB-2 with obvious NIR-Ⅱ absorption was developed, and its nanoparticles realize a record-breaking PCE of 87.7% upon NIR-Ⅱ light illustration. In vitro and in vivo experiments demonstrate that TTB-2 nanoparticles show good tumor photoablation with navigation of photoacoustic imaging in the NIR-Ⅱ window, without any side-effect. Moreover, by combining with PD-1 antibody,the pulmonary metastasis of breast cancer is high-effectively prevented by the efficient photo-immunity effect. Thus, this study explores superior PTMs for cancer metastasis theranostics in the NIR-Ⅱ window, offering a new horizon in developing radical-characteristic NIR-Ⅱ photothermal materials.

The E3 ubiquitin ligase NEDD4-1 protects against acetaminophen-induced liver injury by targeting VDAC1 for degradation

Acetaminophen(APAP)overdose is a major cause of liver injury.Neural precursor cell expressed developmentally downregulated 4—1(NEDD4-1)is an E3 ubiquitin ligase that has been implicated in the pathogenesis of numerous liver diseases;however,its role in APAP-induced liver injury(AILI)is unclear.Thus,this study aimed to investigate the role of NEDD4-1 in the pathogenesis of AILI.We found that NEDD4-1 was dramatically downregulated in response to APAP treatment in mouse livers and isolated mouse hepatocytes.Hepatocyte-specific NEDD4-1 knockout exacerbated APAP-induced mitochondrial damage and the resultant hepatocyte necrosis and liver injury,while hepatocyte-specific NEDD4-1 overexpression mitigated these pathological events both in vivo and in vitro.Additionally,hepatocyte NEDD4-1 deficiency led to marked accumulation of voltage-dependent anion channel 1(VDAC1)and increased VDAC1 oligomerization.Furthermore,VDAC1 knockdown alleviated AILI and weakened the exacerbation of AILI caused by hepatocyte NEDD4-1 deficiency.Mechanistically,NEDD4-1 was found to interact with the PPTY motif of VDAC1 through its WW domain and regulate K48-linked ubiquitination and degradation of VDAC1.Our present study indicates that NEDD4-1 is a suppressor of AILI and functions by regulating the degradation of VDAC1.

Reversible solid-liquid conversion enabled by self-capture effect for stable non-flow zinc-bromine batteries

Non-flow aqueous zinc-bromine batteries without auxiliary components(e.g.,pumps,pipes,storage tanks)and ion-selective membranes represent a cost-effective and promising technology for large-scale energy storage.Unfortunately,they generally suffer from serious diffusion and shuttle of polybromide(Br^(-),Br^(3-))due to the weak physical adsorption between soluble polybromide and host carbon materials,which results in low energy efficiency and poor cycling stability.Here,we develop a novel self-capture organic bromine material(1,10-bis[3-(trimethylammonio)propyl]-4,4'-bipyridinium bromine,NVBr4)to successfully realize reversible solid complexation of bromide components for stable non-flow zinc-bromine battery applications.The quaternary ammonium groups(NV^(4+)ions)can effectively capture the soluble polybromide species based on strong chemical interaction and realize reversible solid complexation confined within the porous electrodes,which transforms the conventional“liquid-liquid”conversion of soluble bromide components into“liquid-solid”model and effectively suppresses the shuttle effect.Thereby,the developed non-flow zinc-bromide battery provides an outstanding voltage platform at 1.7 V with a notable specific capacity of 325 mAh g^(-1)NVBr4(1 A g^(-1)),excellent rate capability(200 mAh g^(-1)NVBr4 at 20 A g^(-1)),outstanding energy density of 469.6 Wh kg^(-1)and super-stable cycle life(20,000 cycles with 100%Coulombic efficiency),which outperforms most of reported zinc-halogen batteries.Further mechanism analysis and DFT calculations demonstrate that the chemical interaction of quaternary ammonium groups and bromide species is the main reason for suppressing the shuttle effect.The developed strategy can be extended to other halogen batteries to obtain stable charge storage.

Nonalcoholic fatty liver disease(NAFLD) is characterized by hepatic steatosis and insulin resistance and there are currently no approved drugs for its treatment.Hyperactivation of mTOR complex1(mTORCl) and subsequent impairment of the transcription factor EB(TFEB)-mediated autophagy-lysosomal pathway(ALP) are implicated in the development of NAFLD.Accordingly,agents that augment hepatic TFEB transcriptional activity may have therapeutic potential against NAFLD.The objective of this study was to investigate the effects of nuciferine,a major active component from lotus leaf,on NAFLD and its underlying mechanism of action.Here we show that nuciferine activated ALP and alleviated steatosis,insulin resistance in the livers of NAFLD mice and palmitic acid-challenged hepatocytes in a TFEB-dependent manner.Mechanistic investigation revealed that nuciferine interacts with the Ragulator subunit hepatitis B X-interacting protein and impairs the interaction of the Ragulator complex with Rag GTPases,thereby suppressing lysosomal localization and activity of mTORC1,which activates TFEB-mediated ALP and further ameliorates hepatic steatosis and insulin resistance.Our present results indicate that nuciferine may be a potential agent for treating NAFLD and that regulation of the mTORCl-TFEB-ALP axis could represent a novel pharmacological strategy to combat NAFLD.

因应平台经济领域初创企业并购的浪潮,强化反垄断并购审查正逐渐成为中国乃至全球的监管共识。以消除潜在竞争或创新为目标的杀式并购以及由此产生抑制投融资的创新杀戮地带的理论基础存在应用局限。作为平台市场中创新研发和商业化过程的有效机制,并购能够实现规模效益、激励风险投资并且产生创新乘数效应。目前中国和域外司法辖区针对初创企业并购制定的包括修正申报标准、创设无门槛申报义务、强化事后调查和倒置举证责任等变革措施,将抑制风险资本市场的并购货币化、增加并购审查的非必要成本以及削弱商业确定性。初创企业并购审查政策的革新应当保持一定的限度,摒弃专门申报标准,澄清创新竞争损害,以及维持举证责任分配。同时,应当结合平台并购特性对既有监管框架作出必要调适,强化排他行为的审查,建立双阶段竞争审查,细化行为性救济措施,以及明确事后调查规则。

Error Analysis of the Nonconforming P1 Finite Element Method to the Sequential Regularization Formulation for Unsteady Navier-Stokes Equations

In this paper we investigate the nonconforming P1finite element approximation to the sequential regularization method for unsteady Navier-Stokes equations. We provide error estimates for a full discretization scheme. Typically, conforming P1finite element methods lead to error bounds that depend inversely on the penalty parameter ?. We obtain an ?-uniform error bound by utilizing the nonconforming P1finite element method in this paper. Numerical examples are given to verify theoretical results.

本文旨在探讨人工智能背景下当代修辞学的发展及其面临的挑战。文章首先简要介绍大语言模型发展的过程,回顾西方修辞学和中国修辞学的传统理论框架,指出在大语言模型背景下当代修辞学研究的重新定位,内容包括:修辞学的本体研究、学科融合、情感转向、数字化、认知研究、生态转向。然后从语言理解和生成的角度对五家大语言模型进行了测试。文章发现大语言模型已经具备了一般的语言修辞能力,但是在理解复杂语义关系方面还存在一定的局限性。大语言模型的修辞理解和修辞生成能力给我们的启发是人工智能为修辞学带来了新的研究路径和变革的可能性。最后,文章展望了人工智能与修辞学未来的发展趋势,指出了人工智能大语言模型对修辞学研究可能带来的影响。

中国城乡代际收入流动性的测度及影响机制研究

基于CHIP1995—2013年调查数据以及CHFS2015与2017年数据,使用双样本两阶段最小二乘估计方法(TS2SLS)考察中国城乡居民家庭代际收入流动性的变化趋势及其影响机制。研究发现:城镇与农村居民家庭的代际收入流动性在较长的时期内均呈现上下波动的变化趋势;农村居民家庭的代际收入流动性持续高于城镇家庭;随着子代出生年份的推移,城镇家庭不同出生队列的代际收入流动性呈现先上升后下降的趋势,而农村家庭不同出生队列的代际收入流动性则呈现先下降后上升的趋势。影响机制分析结果表明,父代的非教育因素在城乡家庭代际收入传递过程中均发挥着主导作用,对农村家庭的影响甚至更大。得益于社会整体制度环境不断改善,父代的非教育因素在城镇家庭代际收入传递过程中的影响逐渐减弱,而教育因素的影响则逐渐上升,特别是城镇中、低收入家庭,父代的教育因素在代际收入传递过程中的影响大幅提高,但父代的非教育因素对农村家庭代际收入流动性的作用则一直保持较高水平。

运用我国299个城市的面板数据构建空间杜宾模型,实证研究绿色技术创新对城市碳排放影响的区域差异性和收敛性。结果表明:我国绿色技术创新可以降低碳排放水平,且存在空间关联效应。横向来看,不同区域的绿色技术创新对碳排放的影响具有明显的区域差异性,东部、长三角、珠三角等经济发达地区的影响效应更强,中西部、京津冀等地区相对较弱。纵向来看,2015年以来绿色技术创新对碳排放影响的区域差异持续收敛,根源在于区域经济发展差距的缩小。未来持续推进绿色技术创新、缩小区域发展差距是实现“双碳”目标的必由之路。
