Cerebral furin deficiency causes hydrocephalus in mice

Furin is a pro-protein convertase that moves between the trans-Golgi network and cell surface in the secretory pathway.We have previously reported that cerebral overexpres-sion of furin promotes cognitive functions in mice.Here,by generating the brain-specific furin conditional knockout(ckO)mice,we investigated the role of furin in brain development.We found that furin deficiency caused early death and growth retardation.Magnetic resonance im-aging showed severe hydrocephalus.In the brain of furin cko mice,impaired ciliogenesis and the derangement of microtubule structures appeared along with the down-regulated expres-sion of RAB28,a ciliary vesicle protein.In line with the widespread neuronal loss,ependymal cell layers were damaged.Further proteomics analysis revealed that cell adhesion molecules including astrocyte-enriched ITGB8 and BCAR1 were altered in furin cKO mice;and astrocyte overgrowth was accompanied by the reduced expression of sox9,indicating a disrupted differ-entiation into ependymal cells.Together,whereas alteration of RAB28 expression correlated with the role of vesicle trafficking in ciliogenesis,dysfunctional astrocytes might be involved in ependymal damage contributing to hydrocephalus in furin ckO mice.The structural and mo-lecular alterations provided a clue for further studying the potential mechanisms of furin.

The increasing awareness of environmental concerns has prompted a surge in the exploration of leadfree,high-power ceramic capacitors.Ongoing efforts to develop leadfree dielectric ceramics with exceptional energystorage performance(ESP)have predominantly relied on multicomponent composite strategies,often accomplished under ultrahigh electric fields.However,this approach poses challenges in insulation and system downsizing due to the necessary working voltage under such conditions.Despite extensive study,bulk ceramics of(Bi_(0.5)Na_(0.5))TiO_(3)(BNT),a prominent lead-free dielectric ceramic family,have seldom achieved a recoverable energy-storage(ES)density(Wrec)exceeding 7 J cm^(−3).This study introduces a novel approach to attain ceramic capacitors with high ESP under moderate electric fields by regulating permittivity based on a linear dielectric model,enhancing insulation quality,and engineering domain structures through chemical formula optimization.The incorporation of SrTiO_(3)(ST)into the BNT matrix is revealed to reduce the dielectric constant,while the addition of Bi(Mg_(2/3)Nb_(1/3))O_(3)(BMN)aids in maintaining polarization.Additionally,the study elucidates the methodology to achieve high ESP at moderate electric fields ranging from 300 to 500 kV cm^(−1).In our optimized composition,0.5(Bi_(0.5)Na_(0.4)K_(0.1))TiO_(3)–0.5(2/3ST-1/3BMN)(B-0.5SB)ceramics,we achieved a Wrec of 7.19 J cm^(−3) with an efficiency of 93.8%at 460 kV cm^(−1).Impressively,the B-0.5SB ceramics exhibit remarkable thermal stability between 30 and 140℃ under 365 kV cm^(−1),maintaining a Wrec exceeding 5 J cm^(−3).This study not only establishes the B-0.5SB ceramics as promising candidates for ES materials but also demonstrates the feasibility of optimizing ESP by modifying the dielectric constant under specific electric field conditions.Simultaneously,it provides valuable insights for the future design of ceramic capacitors with high ESP under constraints of limited electric field.

The existing automated wastewater treatment control systems encounter challenges such as the utilization of specialized testing instruments, equipment repair complications, high operational costs, substantial operational errors, and low detection accuracy. An effective soft measure model offers a viable approach for real-time monitoring and the development of automated control in the wastewater treatment process. Consequently, a novel hybrid deep learning CNN-BNLSTM-Attention (CBNLSMA) model, which incorporates convolutional neural networks (CNN), bidirectional nested long and short-term memory neural networks (BNLSTM), attention mechanisms (AM), and Tree-structure Parzen Estimators (TPE), has been developed for monitoring effluent water quality during the wastewater treatment process. The CBNLSMA model is divided into four stages: the CNN module for feature extraction and data filtering to expedite operations;the BNLSTM module for temporal data’s temporal information extraction;the AM module for model weight reassignment;and the TPE optimization algorithm for the CBNLSMA model’s hyperparameter search optimization. In comparison with other models (TPE-CNN-BNLSTM, TPE-BNLSTM-AM, TPE-CNN-AM, PSO-CBNLSTMA), the CBNLSMA model reduced the RMSE for effluent COD prediction by 25.4%, decreased the MAPE by 32.9%, and enhanced the R2 by 14.9%. For the effluent SS prediction, the CBNLSMA model reduced the RMSE by 26.4%, the MAPE by 21.0%, and improved the R2 by 35.7% compared to other models. The simulation results demonstrate that the proposed CBNLSMA model holds significant potential for real-time effluent quality monitoring, indicating its high potential for automated control in wastewater treatment processes.

Objective To explore the clinical significance of the combined application of palpebral margin cleaning and antibiotic eye drops in inhibiting bacterial growth in the palpebral margin and conjunctival sacs before cataract extraction.Methods In this study,61 patients(97 eyes)with age-related cataract who underwent phacoemulsification and intraocular lens implantation were selected,and randomly grouped.In the experimental group,the combined application of palpebral margin cleaning with cotton pads and levofloxacin eye drops was given for three days before the surgery.In the control group,levofloxacin eye drops alone were applied for three consecutive days.Bacteria samples from the conjunctival sac and eyelid margins were cultivated and identified before and three days after taking antimicrobial measures,respectively.Results In the experimental group,the positive rates of the two bacteria samples were 100%(50/50)and 40%(20/50)before and 10%(5/50)and 0%(0/50)after the treatment.In the control group,the positive rates of the two bacteria samples were 97.9%(46/47)and 29.8%(14/47)before and 40.4%(19/47)and 10.6%(5/47)after the treatment.The positive rates between the two groups were not significantly different before taking antimicrobial measures(P=0.485 and 0.395),while they were significantly different after taking antimicrobial measures(P=0.001 and 0.024).Conclusion Combined application of eyelid and palpebral margin cleaning with cotton pads and antibiotic eye drops before cataract extraction imparted excellent antibacterial effects.

While nickel(II)complexes have been widely used as catalysts for carbon-carbon coupling reactions,the exploration of their photophysical and photochemical properties is still in the infancy.Here,a series of square-planar Ni(II)complexes[(diNHC)NiX2]bearing chelating benzimidazole-based bis(N-heterocyclic carbene)ligands and varying anionic coligands(1,X=Cl;2,X=Br;3,X=I)are synthesized and structurally characterized.In solid state,both 1 and 2 exhibit orange-red photoluminescence under ambient conditions.The photophysical and electrochemical measurements along with density functional theory(DFT)calculations reveal that the low-energy emissions can be attributed to singlet excited states with ligand-to-ligand charge-transfer(LLCT)character.This work suggests that strong-field N-heterocyclic carbene ligands play a crucial role to achieve the luminescence of Ni(II)complexes.

Background:The incidence of clear cell renal cell carcinoma(ccRCC)is globally high;however,despite the introduction of innovative drug therapies,there remains a lack of effective biomarkers for evaluating treatment response.Recently,Caspase recruiting domain-containing protein 11(CARD11)has garnered attention due to its significant association with tumor development and the immune system.Methods:The expression of CARD11 mRNA and protein in ccRCC were analyzed by public database and immunohistochemistry.The focus of this study is on the epigenomic modifications of CARD11,its expression of ccRCC immunophenotype,and its correlation with response to immunotherapy and targeted therapy.Furthermore,to investigate the mechanism of this molecule’s influence on different biological behaviors of cells,cell tests in vitro have been conducted to observe the impact of its expression level.Results:CARD11 expression was upregulated which may be mainly modified by body methylation and was correlated with poor prognosis in ccRCC.In the tumor microenvironment of ccRCC,CARD11 expression was positively correlated with increased T lymphocyte infiltration and increased expression of inhibitory immune checkpoints.Moreover,ccRCC patients with high CARD11 expression had a better response to immunotherapy and targeted therapy.The knockdown of CARD11 ultimately suppressed the proliferation,migration,and invasion capabilities of ccRCC cells while simultaneously enhancing tumor cell apoptosis.Conclusion:We identified CARD11 as a novel therapeutic biomarker for immunotherapy and targeted therapy in ccRCC.

To the Editor:Combined nuclei stimulation may have a synergistic effect on controlling the symptoms of nervous system diseases and improving quality of life.[1]However,increasing of leads may increase costs and complications.[2]Therefore,our study aims to investigate the safety and feasibility of the single curved lead,which can simultaneously reach both subthalamic nucleus(STN)and pedunculopontine nucleus(PPN).
