Nowadays, there are still many challenges to skin regeneration. As a new type of skin substitute, hydrogel has emerging gradually with its excellent properties. However, it is still a challenge to combine with biological active agents to facilitate skin regeneration. Under the circumstance, we synthesized argininebased poly(ester amide)(Arg-PEA) and hyaluronic acid(HA-MA), and combined them into new hybrid hydrogels via photo-crosslinking. We found that the internal structure and physicochemical properties of hybrid hydrogels were greatly improved with the increase of content of Arg-PEA. Therefore, we designed hybrid hydrogels with 5 wt% and 10 wt% of Arg-PEA content, respectively. Besides, we selected the corresponding anti-inflammatory(CRP, TNF-α) indicators to detect the anti-inflammatory properties of the hybrid hydrogels at the protein level, and the corresponding antioxidant indicators(SOD, GSH/GSSG, MDA)were selected to investigate the antioxidant properties of hybrid hydrogels at the cellular level in *** addition, we also selected relevant genes to test the effect of hybrid hydrogels on fibrosis and vascularization in the process of skin wound healing in vitro and verified them in vivo with a mouse dorsum wound model. The results confirmed that Arg-PEA/HA-MA(AH) hybrid hydrogel was a prospective scaffold material for skin regeneration.

Heterogeneous environment adaptation is critical to understand the species evolution and response to climate ***,how narrow-range species adapt to micro-geographic heterogeneity has been overlooked,and there is a lack of insights from metabolism and commensal ***,we studied the environmental adaptation for 3 geographic populations(>40 km apart)of Diploderma vela,a lizard endemic to dry-hot valleys of the Hengduan Mountain *** climatic boundary caused a cooler,droughtier,and barren environment for northernmost population(RM)than the middle(QZK)and southernmost populations(FS).Correspondingly,significant divergences in liver and muscle metabolism and commensal microbiota were detected between RM and QZK or FS individuals,but not between QZK and FS *** composition,coenzyme level(*** and NAD^(+)),and cholesterol metabolism(*** and estriol synthesis)constituted the major metabolic difference between RM and QZK/FS *** and QZK individuals kept abundant Proteobacteria and antifungal strains,while RM individuals maintained more Firmicutes and *** associations existed between varied host metabolite and gut *** were these interpopulation variations associated to the environment adaptation were *** results provided some novel insights into the environmental adaptation and implicated the consequence of climate change on narrow-range species.

Cartilage injuries are o?ten devastating and most cannot be cured because of the intrinsically low regenerative capacity of cartilage tissues. Although stem-cell therapy has shown enormous potential for cartilage repair, the therapeutic outcome has been restricted by low survival rates and poor chondrocyte differentiation in vivo. Here, we report an injectable hybrid inorganic(IHI) nanoscaffold that facilitates fast assembly, enhances survival and regulates chondrogenic differentiation of stem cells. IHI nanoscaffolds that strongly bind to extracellular matrix(ECM) proteins assemble stem cells through synergistic 3 D cell–cell and cell–matrix interactions, creating a favorable physical microenvironment for stem-cell survival and differentiation in vitro and in vivo. Additionally, chondrogenic factors can be loaded into nanoscaffolds with a high capacity, which allows deep, homogenous drug delivery into assembled 3 D stem-cell-derived tissues for effective control over the soluble microenvironment of stem cells. The developed IHI nanoscaffolds that assemble with stem cells are injectable. They also scavenge reactive oxygen species and timely biodegrade for proper integration into injured cartilage tissues. Implantation of stem-cell-assembled IHI nanoscaffolds into injured cartilage results in accelerated tissue regeneration and functional recovery. By establishing our IHI nanoscaffold-templated 3 D stem-cell assembly method, we provide a promising approach to better overcoming the inhibitory microenvironment associated with cartilage injuries and to advance current stem-cell-based tissue engineering.
