Chinese dwarf cherry (Cerasus humilis) is a fruit unique to China， which is considered to have osteoprotective effects. However， no systematic experimental characterization was available. In this study， the osteoprotective activity and mechanism of Chinese dwarf cherry polyphenol extract (OPE) was studied. In vitro， OPE stimulated the alkaline phosphatase activity in the early differentiation stage， increased the osteocalcin level in the middle differentiation stage， and induced the formation of more bone-mineralized nodules in the late osteogenic stage. In vivo， OPE improved cancellous bone structure and maximum load of the femur in ovariectomized (OVX) rats. The balance between bone formation and resorption was regulated. Oxidative stress levels in the peripheral blood， liver and femur were reduced. OPE alleviated the disturbance in energy metabolism， muscle development， and muscle regulation-related signaling pathways caused by OVX and activated the calcium/adenosine monophosphate-activated protein kinase signaling pathway. Therefore， OPE is a potential dietary supplement for the prevention and treatment of osteoporosis.

High mobility group box-1 protein(HMGB1)is an evolutionarily ancient protein,which,as an important non-histone chromosome-binding protein in organism cells,is involved in a variety of important biological processes,including DNA repair,gene transcription,cellular inflammatory response,and autophagy.In this study,we established an eccentric exercise model to observe the effect of HMGB1 on skeletal muscle autophagy and to investigate the underlying molecular mechanisms.Forty-eight male 8-week-old SD rats were randomly divided into control group(C)and exercise group(E).Group E was subjected to a bout of eccentric exercise on a treadmill and sampled soleus at 0 h,12 h,24 h,48 h,and 72 h post-exercise.The speed of the exercise protocol in this study was 16 m/min,the slope was−16°,and the time was 90 min.The ultrastructural changes of skeletal muscle were observed by transmission electron microscopy.The protein expressions of HMGB1,Beclin1,and LC3 were detected by Western Blot.The co-localizations of Beclin1/Bcl-2,Beclin1/HMGB1,and Beclin1/Vps34 were measured by immunofluorescence.The results show that eccentric exercise leads to abnormal changes in the ultrastructure of skeletal muscle,and the protein levels of Beclin1,LC3-II/LC3-I,and the content of HMGB1 in nuclear and cytoplasm were significantly increased at 24 h post-exercise(P<0.05).The co-localization of Beclin1/Bcl-2 and Beclin1/HMGB1 were increased significantly at 0 h post-exercise and then decreased,while the co-localization of Beclin1/Vps34 showed the highest level at 24 h post-exercise.In conclusion,HMGB1 facilitates the separation of Beclin1 from Bcl-2 and promotes Beclin1 binding to Vps34,which may play an important role in eccentric exercise-induced skeletal muscle autophagy.

Ancient dolomite reservoirs play an increasingly important role in deep oil and gas exploration.The mechanism of formation and preservation of dolomite reservoirs is complex,which is always the key issue.With the discovery of deep oil and gas in the Ediacaran dolomites of the world,the upper Ediacaran Qigebrak Formation in the Tabei area has begun to attract attention,but its reservoir space difference and formation mechanism have yet to be clarified.Based on ultra-deep drilling cores and field outcrops in the Tabei area,the lithofacies,reservoir space,and formation mechanism are systematically analyzed by macro to micro,and qualitative to quantitative petrology:(1)The types of dolomite can be divided into five major categories,including microbial dolomite,granular dolomite,residual granular dolomite,crystalline dolomite and karst breccias.(2)The main types of reservoir space are microbial-framework pores,microbial-mold pores,and non-fabric selective dissolution pores.Spongiomicrobialite,karst breccias,and fine-grained dolomite are the dominant reservoir rock types.(3)High-frequency sedimentary cycles and meteoric dissolution are the key factors of reservoir formation.Two sets of large-scale reservoirs are present:the first set is mainly controlled by the supergene karst of the Keping movement,and the second set is mainly controlled by high-frequency sedimentary cycles in the penecontemporaneous period.The reservoirs formed at the shallow burial stage and were preserved until the deep burial stage.(4)The quality of a deep reservoir depends on the geological events that affect the processes of pore reduction and increase.Cementation,compaction and pressure solution are the main destructive diagenetic processes;however,the reservoir space can still be effectively preserved under the influence of constructive diagenetic processes,such as meteoric dissolution and early dolomitization.This research has important theoretical and practical significance for revealing the formation mechanism of upper Ediacaran deep dolomi

Apple(Malus domestica)fruit generally undergoes a climacteric.During its ripening process,there is a peak in ethylene release and its firmness simultaneously decreases.Although more in-depth research into the mechanism of climacteric-type fruit ripening is being carried out,some aspects remain unclear.In this study,we compared the transcriptomes of 0-Pre and 15-Post(pre-and post-climacteric fruit),and 15-Post and 15-MCP[fruit treated with 1-MCP(1-methylcyclopropene)].Various transcription factors,such as MADS-box,ERF,NAC,Dof and SHF were identified among the DEGs(differential gene expressions).Furthermore,these transcription factors were selected for further validation analysis by qRT-PCR.Moreover,yeast one hybrid(Y1H),β-glucuronidase(GUS)transactivation assay and dual-luciferase reporter assay showed that MdAGL30,MdAGL104,MdERF008,MdNAC71,MdDof1.2,MdHSFB2a and MdHSFB3 bound to MdACS1 promoter and directly regulated its transcription,thereby regulating ethylene biosynthesis in apple fruit.Our results provide useful information and new insights for research on apple fruit ripening.

Aqueous zinc-ion batteries(AZIBs)are regarded as promising electrochemical energy storage devices owing to its low cost,intrinsic safety,abundant zinc reserves,and ideal specific capacity.Compared with other cathode materials,manganese dioxide with high voltage,environmental protection,and high theoretical specific capacity receives considerable attention.However,the problems of structural instability,manganese dissolution,and poor electrical conductivity make the exploration of high-performance manganese dioxide still a great challenge and impede its practical applications.Besides,zinc storage mechanisms involved are complex and somewhat controversial.To address these issues,tremendous efforts,such as surface engineering,heteroatoms doping,defect engineering,electrolyte modification,and some advanced characterization technologies,have been devoted to improving its electrochemical performance and illustrating zinc storage mechanism.In this review,we particularly focus on the classification of manganese dioxide based on crystal structures,zinc ions storage mechanisms,the existing challenges,and corresponding optimization strategies as well as structure-performance relationship.In the final section,the application perspectives of manganese oxide cathode materials in AZIBs are prospected.

Toxin-antitoxin(TA)systems are ubiquitous in bacteria and archaea.Most are composed of two neighboring genetic elements,a stable toxin capable of inhibiting crucial cellular processes,including replication,transcrip-tion,translation,cell division and membrane integrity,and an unstable antitoxin to counteract the toxicity of the toxin.Many new discoveries regarding the biochemical properties of the toxin and antitoxin components have been made since the first TA system was reported nearly four decades ago.The physiological functions of TA systems have been hotly debated in recent decades,and it is now increasingly clear that TA systems are important immune systems in prokaryotes.In addition to being involved in biofilm formation and persister cell formation,these modules are antiphage defense systems and provide host defenses against various phage infec-tions via abortive infection.In this review,we explore the potential applications of TA systems based on the recent progress made in elucidating TA functions.We first describe the most recent classification of TA systems and then introduce the biochemical functions of toxins and antitoxins,respectively.Finally,we primarily focus on and devote considerable space to the application of TA complexes in synthetic biology.

Coral reefs have high primary productivity and are one of the most important ecosystems in the ocean. However, the health and stability of coral reefs are constantly threatened by climate change and human activities. The coral holobiont is a complex symbiosis between the coral animal, zooxanthellae, and the associated community of microorganisms including bacteria, archaea, viruses, etc. Coral-associated microorganisms are found to be important for the maintenance of coral health,and they are proposed to contribute to the acclimatization and adaptation of reef-building corals under rapid climate change. The coral-microbe interaction mediated by chemical signalling molecules is an important driving force for shaping the microbial communities. Herein, we summarize our current knowledge of the chemical signalling molecules involved in the interaction of the coral holobiont. Specifically, the cooperation and competition among microbes mediated by N-acyl homoserine lactones(AHLs), the interrelationship between microbes and hosts mediated by dimethylsulfoniopropionate(DMSP) and nitric oxide(NO), and the response of corals and microorganisms to reactive oxygen species(ROS) under environmental stresses are highlighted in this review. We further discuss the potential of manipulating the coral microbiome using signalling molecules to restore and protect coral reefs.

OASs play critical roles in immune response against virus infection by polymerizing ATP into 2-5 As,which initiate the classical OAS/RNase L pathway and induce degradation of viral RNA.OAS members are functionally diverged in four known innate immune pathways(OAS/RNase L,OASL/IRF7,OASL/RIG-I,and OASL/cGAS),but how they functionally diverged is unclear.Here,we focus on evolutionary patterns and explore the link between evolutionary processes and functional divergence of Tetrapod OAS1.We show that Palaeognathae and Primate OAS1 genes are conserved in genomic and protein structures but differ in function.The former(i.e.,ostrich)efficiently synthesized long 2-5 A and activated RNase L,while the latter(i.e.,human)synthesized short 2-5 A and did not activate RNase L.We predicted and verified that two in-frame indels and one positively selected site in the active site pocket contributed to the functional divergence of Palaeognathae and Primate OAS1.Moreover,we discovered and validated that an in-frame indel in the C-terminus of Palaeognathae OAS1 affected the binding affinity of ds RNA and enzymatic activity,and contributed to the functional divergence of Palaeognathae OAS1 proteins.Our findings unravel the molecular mechanism for functional divergence and give insights into the emergence of novel functions in Tetrapod OAS1.

高压下的稀土金属超氢化物因具有高温超导电性而受到广泛关注。由于实验只能部分地确定超氢化物中稀土金属原子的晶格结构,因此,第一性原理计算成为全面理解其结构与物性的重要方法。基于第一性原理计算,对氢含量不同但Ce晶格结构相同的面心立方CeH_(9)和CeH_(10)的弹性、晶格动力学、质子动力学性质进行了对比研究,发现低氢含量有利于面心立方超氢化铈的弹性和声子稳定向低压拓展。在100~140 GPa压强区间,室温下CeH_(9)和CeH_(10)不具有显著的质子扩散,但1 500 K时全面转变为超离子态,扩散系数分别为1.6×10^(-4)~1.2×10^(-4) cm^(2)/s和1.9×10^(-4)~1.5×10^(-4) cm^(2)/s;扩散系数与温度、氢含量正相关,但与压强负相关。所获得的压强、温度及氢含量对超氢化铈结构与动力学性质的影响规律可为其他超氢化物研究提供参考。
