Chinese dwarf cherry (Cerasus humilis) is a fruit unique to China， which is considered to have osteoprotective effects. However， no systematic experimental characterization was available. In this study， the osteoprotective activity and mechanism of Chinese dwarf cherry polyphenol extract (OPE) was studied. In vitro， OPE stimulated the alkaline phosphatase activity in the early differentiation stage， increased the osteocalcin level in the middle differentiation stage， and induced the formation of more bone-mineralized nodules in the late osteogenic stage. In vivo， OPE improved cancellous bone structure and maximum load of the femur in ovariectomized (OVX) rats. The balance between bone formation and resorption was regulated. Oxidative stress levels in the peripheral blood， liver and femur were reduced. OPE alleviated the disturbance in energy metabolism， muscle development， and muscle regulation-related signaling pathways caused by OVX and activated the calcium/adenosine monophosphate-activated protein kinase signaling pathway. Therefore， OPE is a potential dietary supplement for the prevention and treatment of osteoporosis.

The subsynchronous oscillations(SSOs)related to renewable generation seriously affect the stability and safety of the power systems.To realize the dynamic monitoring of SSOs by utilizing the high computational efficiency and noise-resilient features of the matrix pencil method(MPM),this paper propos es an improved MPM-based parameter identification with syn chrophasors.The MPM is enhanced by the angular frequency fitting equations based on the characteristic polynomial coeffi cients of the matrix pencil to ensure the accuracy of the identi fied parameters,since the existing eigenvalue solution of the MPM ignores the angular frequency conjugation constraints of the two fundamental modes and two oscillation modes.Then,the identification and recovery of bad data are proposed by uti lizing the difference in temporal continuity of the synchropha sors before and after noise reduction.The proposed parameter identification is verified with synthetic,simulated,and actual measured phase measurement unit(PMU)data.Compared with the existing MPM,the improved MPM achieves better accuracy for parameter identification of each component in SSOs,better real-time performance,and significantly reduces the effect of bad data.

Gastric cancer(GC)is the third most common cause of cancer death globally and a large portion of patients are diagnosed at advanced stages with cancer invasion and metastasis1,2.However,the mechanisms underlying the invasion and metastasis of GC remain to be delineated.ZYX plays critical roles in cell mobility via cytoskeleton regulation in various cell types.3 In this study,we further reported that ZYX promoted migration,invasion,and metastasis of GC cells.Mechanistically,ZYX promoted WNK1 activation and SNAl1 up-regulation,inducing epithelial-mesenchymal transition(EMT)to enhance the mobility of GC cells.Inhibition of WNK1 impaired the mobility of GC cells.Therefore,ZYX/WNK1 could be potential therapeutic targets for GC treatment.

Regulatory T(T_(reg))cells play an essential role in maintaining immune balance across various physiological and pathological conditions.However,the mechanisms underlying T_(reg)homeostasis remain incompletely understood.Here,we report that RIPK1 is crucial for T_(reg) cell survival and homeostasis.We generated mice with T_(reg) cell-specific ablation of Ripk1 and found that these mice developed fatal systemic autoimmunity due to a dramatic reduction in the Treg cell compartment caused by excessive cell death.Unlike conventional T cells,Treg cells with Ripk1 deficiency were only partially rescued from cell death by blocking FADD-dependent apoptosis.However,simultaneous removal of both Fadd and Ripk3 completely restored the homeostasis of Ripk1-deficient Treg cells by blocking two cell death pathways.Thus,our study highlights the critical role of RIPK1 in regulating Treg cell homeostasis by controlling both apoptosis and necroptosis,thereby providing novel insights into the mechanisms of Treg cell homeostasis.

Privacy leakage is one of the most critical issues in machine learning and has attracted growing interest for tasks such as demonstrating potential threats in model attacks and creating model defenses.In recent years, numerous studies have revealed various privacy leakage risks(e.g., data reconstruction attack, membership inference attack, backdoor attack, and adversarial attack) and several targeted defense approaches(e.g., data denoising, differential privacy, and data encryption). However, existing solutions generally focus on model parameter levels to disclose(or repair) privacy threats during the model training and/or model interference process, which are rarely applied at the model architecture level. Thus, in this paper, we aim to exploit the potential privacy leakage at the model architecture level through a pioneer study on neural architecture search(NAS) paradigms which serves as a powerful tool to automate a neural network design.By investigating the NAS procedure, we discover two attack threats in the model architecture level called the architectural dataset reconstruction attack and the architectural membership inference attack. Our theoretical analysis and experimental evaluation reveal that an attacker may leverage the output architecture of an ongoing NAS paradigm to reconstruct its original training set, or accurately infer the memberships of its training set simply from the model architecture. In this work, we also propose several defense approaches related to these model architecture attacks. We hope our work can highlight the need for greater attention to privacy protection in model architecture levels(e.g., NAS paradigms).

High mobility group box-1 protein(HMGB1)is an evolutionarily ancient protein,which,as an important non-histone chromosome-binding protein in organism cells,is involved in a variety of important biological processes,including DNA repair,gene transcription,cellular inflammatory response,and autophagy.In this study,we established an eccentric exercise model to observe the effect of HMGB1 on skeletal muscle autophagy and to investigate the underlying molecular mechanisms.Forty-eight male 8-week-old SD rats were randomly divided into control group(C)and exercise group(E).Group E was subjected to a bout of eccentric exercise on a treadmill and sampled soleus at 0 h,12 h,24 h,48 h,and 72 h post-exercise.The speed of the exercise protocol in this study was 16 m/min,the slope was−16°,and the time was 90 min.The ultrastructural changes of skeletal muscle were observed by transmission electron microscopy.The protein expressions of HMGB1,Beclin1,and LC3 were detected by Western Blot.The co-localizations of Beclin1/Bcl-2,Beclin1/HMGB1,and Beclin1/Vps34 were measured by immunofluorescence.The results show that eccentric exercise leads to abnormal changes in the ultrastructure of skeletal muscle,and the protein levels of Beclin1,LC3-II/LC3-I,and the content of HMGB1 in nuclear and cytoplasm were significantly increased at 24 h post-exercise(P<0.05).The co-localization of Beclin1/Bcl-2 and Beclin1/HMGB1 were increased significantly at 0 h post-exercise and then decreased,while the co-localization of Beclin1/Vps34 showed the highest level at 24 h post-exercise.In conclusion,HMGB1 facilitates the separation of Beclin1 from Bcl-2 and promotes Beclin1 binding to Vps34,which may play an important role in eccentric exercise-induced skeletal muscle autophagy.

Enzymatic malonylation of natural glycosides provides a promising alternative method for drug-like malonylated glycosides supply.However,the catalytic potential and structural basis of plant malonyltransferase are far from being fully elucidated.This work identified a new malonyltransferase CtMaT1 from Cistanche tubulosa.It displayed unprecedented mono-and/or di-malonylation activity toward diverse glucosides with different aglycons.A“one-pot”system by CtMaT1 and a malonyl-CoA synthetase was established to biosynthesize nine new malonylated glucosides.Structural investigations revealed that CtMaT1 possesses an adequately spacious acyl-acceptor pocket capable of accommodating diverse glucosides.Additionally,it recognizes malonyl-CoA through strong electrotactic and hydrogen interactions.QM/MM calculation revealed the H167-mediated SN2 reaction mechanism of CtMaT1,while dynamic simulations detected the formation of stable hydrogen bonds between the glucose-6-OH group and H167,resulting in its high malonylation regiospecificity.Calculated energy profiles of two isomeric glycosides highlighted lower reaction energy barriers towards glucoside substrates,emphasizing CtMaT1's preference for glucosides.Furthermore,a mutant CtMaT1H36A with notably increased di-malonylation activity was obtained.The underlying molecular mechanism was illuminated through MM/GBSA binding free energy calculation.This study significantly advances the understanding of plant acyltransferases from both functional and protein structural perspectives,while also providing a versatile tool for enzymatic malonylation applications in pharmacology.

The Multi-access Edge Cloud(MEC) networks extend cloud computing services and capabilities to the edge of the networks. By bringing computation and storage capabilities closer to end-users and connected devices, MEC networks can support a wide range of applications. MEC networks can also leverage various types of resources, including computation resources, network resources, radio resources,and location-based resources, to provide multidimensional resources for intelligent applications in 5/6G.However, tasks generated by users often consist of multiple subtasks that require different types of resources. It is a challenging problem to offload multiresource task requests to the edge cloud aiming at maximizing benefits due to the heterogeneity of resources provided by devices. To address this issue,we mathematically model the task requests with multiple subtasks. Then, the problem of task offloading of multi-resource task requests is proved to be NP-hard. Furthermore, we propose a novel Dual-Agent Deep Reinforcement Learning algorithm with Node First and Link features(NF_L_DA_DRL) based on the policy network, to optimize the benefits generated by offloading multi-resource task requests in MEC networks. Finally, simulation results show that the proposed algorithm can effectively improve the benefit of task offloading with higher resource utilization compared with baseline algorithms.

Urinary kidney injury molecule 1(uKIM-1)serves as a reliable marker for the early diagnosis of acute kidney injury(AKI).The rapid and facile detection of changes in uKIM-1 is essential for early AKI diagnosis,ultimately improving the prognosis of patients.In this study,we developed a fully printed photonic crystal-integrated microarray with photonic crystal-enhanced fluorescence properties,which can detect uKIM-1 levels at the point-of-care.We confirmed its efficacy in the early diagnosis of AKI using clinical urine specimens.Direct quantitative detection of uKIM-1 was achieved within 10 min.The lowest limit of detection is 8.75 pg·mL^(-1) with an accuracy of 94.2%.The diagnostic efficacy was validated using 86 clinical urine samples,highlighting the high sensitivity and stability of the photonic crystal microarray.Consequently,a facile and reliable immunoassay was designed and prepared for the rapid quantitative detection of uKIM-1,which is crucial for the early identification and convenient detection of AKI in hospital or community settings.Rapid,convenient,cost-effective,and long-term monitoring of changes in uKIM-1 levels can assist clinicians in making timely adjustments to treatment regimens,preventing the transition from AKI to chronic kidney disease(CKD),improving the quality of life of patients with AKI,and reducing healthcare costs.It highlights the advantages of utilizing urine samples as a noninvasive and easily accessible medium for early detection and monitoring of kidney-related conditions.
