目的:探讨脂肪来源间充质干细胞(ADSC)对瘘管型克罗恩病(CD)模型大鼠瘘管创面的修复作用以及其对miR-146a/白细胞介素-1受体相关激酶(IRAK)信号轴的调节机制。方法:RT-PCR检测80例瘘管治疗的瘘管型CD患者术后血清中miR-146a和IRAK的mRNA表达。双荧光素酶基因报告分析miR-146a和IRAK的关系。将成功制备的CD瘘管病变模型大鼠分为对照组(Con)、ADSC干预组(ADSC)、ADSC+miR-146a-antagomir干预组(ADSC+antagomir)、ADSC+miR-146a-antagomir+siRNA-IRAK干预组(ADSC+antagomir+si),每组10只;造模成功后1 h,ADSC组大鼠以2×10^(6)个/ml ADSC悬液注入瘘口周围区域,并皮下注射2μg/kg miR-146a-antagomir-NC;ADSC+antagomir组大鼠以2×10^(6)个/ml ADSC悬液注入瘘口周围区域,并皮下注射2μg/kg miR-146a-antagomir;ADSC+antagomir+si组大鼠以2×10^(6)个/ml ADSC悬液注入瘘口周围区域,并皮下注射2μg/kg miR-146a-antagomir+siRNA-IRAK,Con组大鼠以等剂量的生理盐水注入瘘口周围区域,并皮下注射2μg/kg miR-146aantagomir-NC;每组每日定时处理2次,连续处理14 d。流式细胞术检测大鼠瘘管创面组织细胞凋亡;试剂盒检测大鼠血清中TNF-α和IL-6水平;免疫组化检测各组大鼠瘘管创面组织中的血管新生;Western blot检测瘘管创面组织中IRAK、凋亡蛋白B淋巴细胞瘤-2(Bcl-2)、Bcl-2相关x蛋白(Bax)、血管内皮生长因子(VEGF)表达。结果:miR-146a表达在CD瘘管病变患者术后的血清中明显降低,IRAK的mRNA表达明显升高,差异均具有统计学意义(均P<0.05);双荧光素酶结果显示miR-146a能靶向调控IRAK表达;与Con组相比,ADSC组、ADSC+antagomir组、ADSC+antagomir+si组大鼠瘘管创面组织的细胞凋亡率、血清中TNF-α和IL-6含量、瘘管组织中IRAK、Bax表达明显降低,瘘管组织中新生血管的数量以及Bcl-2、VEGF表达明显升高;与ADSC组相比,ADSC+antagomir组、ADSC+antagomir+si组大鼠瘘管创面组织细胞凋亡率、血清中TNF-α和IL-6含量、瘘管组织中IRAK、Bax表达明显升高,瘘管组织中新生血管的数量以及Bcl-2、VEGF的表达明显降低;与ADSC+antagomir组相比,ADSC+antagomir+si组大鼠瘘管创面组织中细胞凋亡率、血清中TNF-α和IL-6含量、瘘管组织中IRAK、Bax表达明显降低,瘘管组织中新生血管数量以及Bcl-2、VEGF表达明显升高,差异均有统计学意义(均P<0.05)。结论:ADSC促进CD模型大鼠瘘管病变术后的瘘管愈合,这可能与调控miR-146a/IRAK的表达,抑制细胞凋亡,促进创面血管新生有关。

在社会转型和人口老龄化的双重影响下,年龄刻板印象和社会偏见导致了包容性下降、代际脱节等系列问题,人们面临越来越多的“代际鸿沟”。作为一种跨学科和跨年龄的活动,代际学习被认为是解决代际困境、促进代际和谐的有效路径和支持机制。文章梳理了代际概念的学科渊源及演变过程,并以身份识别、文化形塑和世代差异为切入口,厘清了代际学习的核心要素与特征。并在此基础上,依据“血缘—地缘—业缘”的社会场域框架,描绘出代际学习的实践样态与发展趋势,以期为老龄化背景下代际学习的高质量发展提供学理依据和实践参考。

The fronts of a subject could be perceived from the publications of top journals. We investigated four top journals on chemical engineering: AIChE Journal, Chemical Engineering Science, Industrial & Engineering Chemistry Research and Chemical Engineering Journal. Based on Web of Science, in the last five years, these four journals have published 21062 papers, including 590 highly cited papers and 31 “hot” papers among which only 7 are from Industrial & Engineering Chemistry Research and all the others from Chemical Engineering Journal (data obtained on November 17, 2018). The traditional research “Pure and pseudo-pure fluid thermophysical property evaluation and the open-source thermophysical property library CoolProp” is the most cited paper published in 2014 [1]. Both highly cited and “hot” papers were published in 2017 on electrodes of supercapacitors [2]. But researches on environmental science and engineering have been paid most attention with 17 of 27 both highly cited and “hot” papers on this research area. The most cited and “hottest” paper is “Boron nitride-based materials for the removal of pollutants from aqueous solutions: A review” in 2018 [3]. Over the past decades, research and technologies advancement in chemical science and engineering in China has expanded in a lightning speed globally. To meet the growing needs of knowledge exchange and understanding of the underlying scientific principles, we launched in 2007 a new journal named Frontiers of Chemical Engineering in China with all papers translated from the Chinese journals. Original research articles or reviews have been published since 2009. The new name Frontiers of Chemical Science and Engineering (FCSE) has begun only since 2011 and FCSE became one of the Transactions of the Chinese Academy of Engineering (CAE) in 2014. The potential of Arctic routes(ARs)has attracted global attention,and exploiting the Arctic has become an important strategy for many ***,there are still some challenges for ships sailing in Arctic ice zones,including harsh marine environments and the insufficient service capacity of sea ice information service *** better understand the route changes in the Arctic and extract real-time ship navigation routes,we developed an online interactive route planning system(RouteView)for ships sailing in the Arctic based on big Earth *** includes two main features:(1)an online calculation interface is provided for optimal routes along the Arctic Northeast Passage(NEP)60 days into the future by utilizing reinforcement learning(RL)based on sea ice and meteorological data,and(2)an online ice-water classification is established based on synthetic aperture radar(SAR)data and deep learning to help users extract the sea ice distribution in real *** work can potentially enhance the safety of shipping navigation along the NEP and improve information extraction methods for ARs.

The journal Science China Chemistry published 63 papers in polymer fields in 2010, leading to a percentage increase of 473% compared to the year 2008 and 70% to the year 2009, respectively （Figure 1）. In this year, three polymer Special Topics were designed and published： Biomedical Polymer （Number 3） [1], Advances in Principles of Polymerization （Number 8） [2] and Highly Branched Polymers-Promising Architectural Macromolecules （Number 12） [3]. Several high qualified papers were published and great achievements have been obtained in the year 2010.

Living cells are constantly threatened by endogenous and environmental agents that can induce various DNA lesions including 8-oxoguanine(8-oxoG).Increasing evidence has suggested that 8-oxoG is not only a biomarker of oxidative stress,but also a novel epigenetic-like modification involved in transcriptional regulation in mammalian *** of DNA damage and repair is useful for both basic research and clinical applications,but current methods for 8-oxoG detection still suffer from some problems such as poor selectivity,time consuming and being ***,we developed a fast and simple biosensing approach for quantitative analysis of 8-oxoG in DNA,which was based on the selective chemical biotinylation of 8-oxoG in conjunction with biotin-streptavidin enzyme-linked immunosorbent *** have also successfully applied this method to achieve efficient detection of the repair activities of DNA glycosylases Fpg and hOGG1 toward 8-oxoG in vitro and in human *** newly developed biosensing assay should be generally applicable for rapid detection of 8-oxoG and its repair in other organisms.
