circRNA3669 promotes goat endometrial epithelial cells proliferation via miR-26a/RCN2 to activate PI3K/AKT-mTOR and MAPK pathways

The development of receptive endometrium(RE) from pre-receptive endometrium(PE) for successful embryo implantation is a complex dynamic process in which the morphology and physiological states of the endometrial epithelium undergo a series of significant changes, including cell proliferation and apoptosis. However, the molecular mechanisms are not yet fully understood. In this study, a higher circRNA3669 level was observed in PE than in RE of goats. Functional assays revealed that this overexpression promoted the proliferation of goat endometrial epithelial cells(GEECs) by activating the expression of genes related to the PI3K/AKT-mTOR and MAPK pathways,thereby inhibiting apoptosis in vitro. Furthermore, circRNA3669 functioned as a competing endogenous RNA(ceRNA) to upregulate Reticulocalbin-2(RCN2) expression at the post-transcriptional level by interacting with and downregulating miR-26a in GEECs. In addition, RCN2, which is highly expressed in the PE of goats, was found to be regulated by β-estradiol(E2) and progesterone(P4). Our results demonstrated that RCN2 also affected the key proteins PI3K, AKT, mTOR, JNK, and P38 in the PI3K/AKT-mTOR and MAPK pathways, thereby facilitating GEECs proliferation and suppressing their apoptosis in vitro. Collectively, we constructed a new circRNA3669-miR-26aRCN2 regulatory network in GEECs, which further provides strong evidence that circRNA could potentially play a crucial regulatory role in the development of RE in goats.

Much effort has been made to uncover the cellular heterogeneities of human hearts by single-nucleus RNA sequencing.However,the cardiac transcriptional regulation networks have not been systematically described because of the limitations in detecting transcription factors.In this study,we optimized a pipeline for isolating nuclei and conducting single-nucleus RNA sequencing targeted to detect a higher number of cell signal genes and an optimal number of transcription factors.With this unbiased protocol,we characterized the cellular composition of healthy human hearts and investigated the transcriptional regulation networks involved in determining the cellular identities and functions of the main cardiac cell subtypes.Particularly in fibroblasts,a novel regulator,PKNOX2,was identified as being associated with physiological fibroblast activation in healthy hearts.To validate the roles of these transcription factors in maintaining homeostasis,we used single-nucleus RNA-sequencing analysis of transplanted failing hearts focusing on fibroblast remodelling.The trajectory analysis suggested that PKNOX2 was abnormally decreased from fibroblast activation to pathological myofibroblast formation.Both gain-and loss-of-function in vitro experiments demonstrated the inhibitory role of PKNOX2 in pathological fibrosis remodelling.Moreover,fibroblast-specific overexpression and knockout of PKNOX2 in a heart failure mouse model induced by transverse aortic constriction surgery significantly improved and aggravated myocardial fibrosis,respectively.In summary,this study established a high-quality pipeline for single-nucleus RNA-sequencing analysis of heart muscle.With this optimized protocol,we described the transcriptional regulation networks of the main cardiac cell subtypes and identified PKNOX2 as a novel regulator in suppressing fibrosis and a potential therapeutic target for future translational studies.

Plant-based dietary patterns and risk of esophageal cancer:A prospective cohort study spanning 17 years

Objective: Plant-based diets have multiple health benefits for cancers;however, little is known about the association between plant-based dietary patterns and esophageal cancer(EC).This study presents an investigation of the prospective associations among three predefined indices of plant-based dietary patterns and the risk of EC.Methods: We performed endoscopic screening for 15,709 participants aged 40-69 years from two high-risk areas of China from January 2005 to December 2009 and followed the cohort until December 31, 2022. The overall plant-based diet index(PDI), healthful plant-based diet index(h PDI), and unhealthful plant-based diet index(u PDI), were calculated using survey responses to assess dietary patterns. We applied Cox proportional hazard regression to estimate the multivariable hazard ratios(HRs) and 95% confidence intervals(95% CIs) of EC across 3plant-based diet indices and further stratified the analysis by subgroups.Results: The final study sample included 15,184 participants in the cohort. During a follow-up of 219,365person-years, 176 patients with EC were identified. When the highest quartile was compared with the lowest quartile, the pooled multivariable-adjusted HR of EC was 0.50(95% CI, 0.32-0.77) for h PDI. In addition, the HR per 10-point increase in the h PDI score was 0.42(95% CI, 0.27-0.66) for ECs. Conversely, u PDI was positively associated with the risk of EC, and the HR was 1.80(95% CI, 1.16-2.82). The HR per 10-point increase in the u PDI score was 1.90(95% CI, 1.26-2.88) for ECs. The associations between these scores and the risk of EC were consistent in most subgroups. These results remained robust in sensitivity analyses.Conclusions: A healthy plant-based dietary pattern was associated with a reduced risk of EC. Emphasizing the healthiness and quality of plant-based diets may be important for preventing the development of EC.

Effect of solvents on the morphology and structure of barium titanate synthesized by a one-step hydrothermal method

Tetragonal barium titanate was synthesized from barium hydroxide octahydrate and titanium tetrachloride through a simple one-step hydrothermal method.The effect of different solvents on the crystal structure and morphology of barium titanate nanoparticles during the hy-drothermal process was investigated.Except for ethylene glycol/water solvent,impurity-free barium titanate was synthesized in pure water,methanol/water,ethanol/water,and isopropyl alcohol/water mixed solvents.Compared with other alcohols,ethanol promotes the formation of a tetragonal structure.In addition,characterization studies confirm that particles synthesized in methanol/water,ethanol/water,and isopropyl al-cohol/water mixed solvents are smaller in size than those synthesized in pure water.In the case of alcohol-containing solvents,the particle size decreases in the order of isopropanol,ethanol,and methanol.Among all the media used in this study,ethanol/water is considered the optimum reaction media for barium titanate with high tetragonality(defined as the ratio of two lattice parameters c and a,c/a=1.0088)and small aver-age particle size(82 nm),which indicates its great application potential in multilayer ceramic capacitors.

Interface engineering of FeCo LDH@NiCoP nanowire heterostructures for highly efficient and stable overall water splitting

Developing efficient and inexpensive OER electrocatalysts is a challenge for overall water splitting.Herein,the heterostructured Fe Co LDH@NiCoP/NF nanowire arrays with high performance were rationally designed and prepared using an interface engineering strategy.Benefitting from the special heterostructure between Fe Co LDH and Ni Co P,the as-synthesized Fe Co LDH@Ni Co P/NF electrocatalyst exhibits outstanding OER performance with an exceptionally low overpotential of 206 mV to achieve 20 mA/cm^(2)current density in an alkaline electrolyte.Importantly,a cell constructed using the FeCo LDH@NiCoP/NF electrocatalyst as cathode and anode just needs a voltage of 1.48 V at 10 mA/cm^(2),and shows excellent stability over 80 h.Experimental and theoretical results verified that the introduction of Ni Co P efficiently regulates the electronic structure of Fe Co LDH,which tremendously boosts the conductivity and intrinsic catalytic activity of FeCo LDH@NiCoP/NF electrocatalyst.The present work provides guidance for the preparation of other efficient and cheap electrocatalytic materials.

Anode materials are an essential part of lithium-ion batteries(LIBs),which determine the performance and safety of LIBs.Currently,graphite,as the anode material of commercial LIBs,is limited by its low theoretical capacity of 372 mA·h·g^(−1),thus hindering further development toward high-capacity and large-scale applications.Alkaline earth metal iron-based oxides are considered a promising candidate to replace graphite because of their low preparation cost,good thermal stability,superior stability,and high electrochemical performance.Nonetheless,many issues and challenges remain to be addressed.Herein,we systematically summarize the research progress of alkaline earth metal iron-based oxides as LIB anodes.Meanwhile,the material and structural properties,synthesis methods,electrochemical reaction mechanisms,and improvement strategies are introduced.Finally,existing challenges and future research directions are discussed to accelerate their practical application in commercial LIBs.

目的:在我国青春期至成年早期人群中探索自制力与抑郁症状和超重肥胖共病风险的关联,为将来针对不同风险人群开展个性化干预提供依据。方法:基于一项长达10年的队列研究——中国家庭追踪调查(China family panel studies,CFPS),纳入2010年10~19岁、中国标准下处于正常体质量、无抑郁症状、有自制力得分,且2010—2020年至少有两次抑郁症状得分和体重指数(body mass index,BMI)、每个家庭中唯一或年龄最小的608名青少年作为研究对象。用整个随访期间平均标准化抑郁症状得分和BMI Z评分均处于高水平,或者基于潜分类轨迹模型(latent class trajectory modeling,LCTM)得到的抑郁症状和BMI随时间的轨迹分类同时属于“风险型”,或者最后一次随访调查时有抑郁症状且超重肥胖三种方式定义抑郁症状和超重肥胖共病人群。采用无序多分类Logistic回归模型分析标准化自制力得分与抑郁症状和超重肥胖共病风险的关联。结果:校正年龄、性别、城乡、每周体力活动时长、父母文化程度、父母超重肥胖和抑郁情况后,无论哪种共病定义方式,当以健康人群为参照组时,自制力得分每升高一个标准差,个体患抑郁症状和超重肥胖共病的风险降低33%(95%CI:14%~48%,基于整个随访期间平均水平)~78%(95%CI:6%~95%,基于抑郁症状和BMI随时间变化的联合轨迹)。此外,自制力得分每升高一个标准差,个体仅患抑郁症状和仅超重肥胖的风险分别降低25%(95%CI:4%~42%,仅基于整个随访期间平均水平)和21%(95%CI:1%~37%,仅基于抑郁症状和BMI随时间变化的联合轨迹)。本研究主要结果与根据世界卫生组织(World Health Organization,WHO)标准定义个体是否超重肥胖时的敏感性分析结果一致。结论:青春期和成年早期个体自制力越高,患抑郁症状和超重肥胖共病的风险越低,提示未来可基于自制力开展个性化的抑郁症状和超重肥胖共病干预。

The Internet of Everything(IoE),which aims to realize information exchange and communications for anything with the Internet,has revolutionized our modern world.Serving as the driving force for devices in the IoE network,power supply systems play a fundamental role in the development of the IoE.However,due to the complexity,multifunctionality and wide-scale deployment of diverse applications,power supply systems face great challenges,including distribution,connection,charging technologies,and management.In this review,some challenges and advances in the development of both power supply systems and their units are presented.In the overall system-level field,establishing sustainable and maintenancefree power supply systems through wireless connections,efficient power management and integrated energy harvesting and storage systems is highlighted.Additionally,the main performance metrics of power supply units are discussed,including energy density,service life,and self-power ability.In addition,some directions of power quality assessment for both the system and unit levels of power supply systems are presented,aiming to provide insight into the future development of high-performance power supply systems for the IoE.

针对当前多模态情感识别算法在模态特征提取、模态间信息融合等方面存在识别准确率偏低、泛化能力较差的问题,提出了一种基于语音、文本和表情的多模态情感识别算法。首先,设计了一种浅层特征提取网络(Sfen)和并行卷积模块(Pconv)提取语音和文本中的情感特征,通过改进的Inception-ResnetV2模型提取视频序列中的表情情感特征;其次,为强化模态间的关联性,设计了一种用于优化语音和文本特征融合的交叉注意力模块;最后,利用基于注意力的双向长短期记忆(BiLSTM based on attention mechanism,BiLSTM-Attention)模块关注重点信息,保持模态信息之间的时序相关性。实验通过对比3种模态不同的组合方式,发现预先对语音和文本进行特征融合可以显著提高识别精度。在公开情感数据集CH-SIMS和CMU-MOSI上的实验结果表明,所提出的模型取得了比基线模型更高的识别准确率,三分类和二分类准确率分别达到97.82%和98.18%,证明了该模型的有效性。

目的探讨精浆氧化应激及生殖激素水平与畸形精子症及卵胞质内单精子注射技术(ICSI)结局的相关性。方法选取2021年8月—2022年12月我院生殖医学中心的男性不育症患者206例为研究对象,根据世界卫生组织第五版《世界卫生组织人类精液检查与处理实验室手册》,其中正常形态精子百分率<4%的120例,纳入畸形精子症组;正常形态精子百分率≥4%的86例,纳入正常组。两组均检测精浆氧化应激[活性氧(Reactive oxygen species,ROS),超氧化物歧化酶(SOD)]及生殖激素[卵泡刺激素(FSH)、黄体生成素(LH)、睾酮(T)]水平,分析畸形精子症影响因素。比较畸形精子症组不同精浆氧化应激及生殖激素水平患者ICSI受精率,分析畸形精子症程度与精浆氧化应激、生殖激素水平的相关性。结果畸形精子症组高龄、高体重指数以及吸烟、饮酒、精索静脉曲张、糖尿病发生率比例大于对照组,精子活动率、前向运动精子率小于对照组(P<0.05);畸形精子症组精浆ROS、FSH、LH高于对照组,SOD、T低于对照组(P<0.05);吸烟、饮酒、精索静脉曲张、ROS是畸形精子症的独立危险因素,SOD、T是畸形精子症的独立保护因素(P<0.05);畸形精子症组随着ROS水平升高,患者受精率呈降低趋势,随着精浆SOD、T水平升高,患者受精率呈升高趋势(P<0.05);Spearman相关系数分析,畸形精子症程度与精浆ROS水平呈正相关(r=0.832,P<0.001),与SOD水平呈负相关(r=-0.825,P<0.001),与T水平基本无相关性(r=0.206,P=0.024)。结论氧化应激反应、生殖激素异常表达与畸形精子症的发生密切相关,同时会影响其ICSI结局。
