Prognostic value of lactate transporter SLC16A1 and SLC16A3 as oncoimmunological biomarkers associating tumor metabolism and immune evasion in glioma

Background:Hypoxic microenvironment is immunosuppressive and protu-morigenic,and elevated lactate is an intermediary in the modulation of immune responses.However,as critical lactate transporters,the role of SLC16A1 and SLC16A3 in immune infiltration and evasion of glioma is not fully elucidated.Methods:gene expression in low‐and high‐grade glioma(LGG and GBM)was evaluated with TCGA database.The TISIDB,TIMER and CIBERSORT databases were utilized for the analysis of the correlation between SLC16A1 or SLC16A3 and immunocyte infiltration as well as immune checkpoints.Results:Compared with normal tissues,a significant increase of both SLC16A1 and SLC16A3 was found in LGG and GBM,and closely related to the poor prognosis only in LGG.Cancer SEA indicated that SLC16A1 was involved in hypoxia while SLC16A3 contributed to metastasis and inflamma-tion in glioma.The SLC16A3 expression was significantly correlated with neutrophil activation by GO analysis.TISCH showed the distribution of SLC16A1 on glioma cells and SLC16A3 on immune cells,which was correlated to tumor‐associated macrophages and neutrophils that are immunosuppressive.SLC16A1 and SLC16A3 were identified to tightly interacted with diverse immune checkpoints(especially PD1,PD‐L1,PD‐L2,Tim‐3)and immunosuppressive factors(TGF‐βand IL‐10)in glioma.Furthermore,SLC16A3 had a positive correlation to activation markers of tumor‐associated neutrophils and chemokines such as CCL2,CCL22,CXCR2,CXCR4 in LGG and CCL7,CCL20 CXCL8 in GBM,which could enhance infiltration of immunosuppressive cells to the tumor microenvironment.Conclusion:In general,our results suggest that SLC16A1 and SLC16A3 act as a bridge between tumor metabolism and immunity by promoting immuno-suppressive cell infiltration,which contributes to immune evasion and a worse prognosis in glioma.Targeting SLC16A1 and SLC16A3 may provide novel therapeutic strategy for immunotherapy in glioma.

江苏某地区稻虾蟹综合种养模式下养殖水和产品中农药残留与风险评价

为掌握江苏省某地区稻田综合种养基地内(“稻-蟹”及“稻-虾”)养殖水环境及水产品中的农药残留特征与生态风险及健康风险,利用三重四极杆气质联用仪(GC-MS/MS),选择6个养殖区,根据生长周期,跟踪监测养殖水环境及同期水产品(虾、蟹)中208种农药残留情况,并根据商值法对养殖水环境进行生态风险评价,利用食用安全风险指数对水产品进行食用安全分析。结果表明:综合种养模式下,水环境中检出农药的主要类型为除草剂、杀菌剂、植物生长调节剂等,平均质量分数为0.33μg·L^(-1),同时有少量杀虫剂、杀螨剂、增效剂有检出;在整个生长周期内,稻虾、稻蟹农药残留的主要类型为除草剂、杀菌剂、杀虫剂和植物生长调节剂,平均质量分数为6.1μg·kg^(-1)(以湿质量计)。养殖水环境中农药主要来源于养殖前期环境中残留或外源性污染,随着养殖时间延长,农药含量整体趋向于减少,通过对养殖水环境生态风险进行分析,发现各抽样点位联合风险商值(RQ)均在0~1之间,属于中风险,对环境仍具有一定的压力,除草剂等类型农药应引起重视;在监测周期内,水产品中大部分农药的残留量逐渐减少直至未检出,仅有部分点位杀菌剂有少量存在,通过对同期虾、蟹的健康风险进行分析发现,平均食品安全指数远小于1,农药残留的食品安全风险可以接受。

目的评价原型株、Beta株、Gamma株和Delta株SARS-CoV-2灭活疫苗在大鼠体内的免疫原性。方法分别将原型株、Beta株、Gamma株和Delta株SARS-CoV-2灭活疫苗经大腿肌内免疫5只雌性Wistar大鼠2次,间隔14 d,免疫剂量均为3μg病毒蛋白/(0.5 mL·只),初次免疫后14、28和42 d,经静脉采集血,分离血清。间接ELISA法检测血清IgG抗体水平;微量中和试验检测血清中和抗体效价;计算血清中和抗体效价的抗原比,以评价2株病毒间的抗原性差异。结果初次免疫后14 d,全部免疫血清中均可检测到针对4株病毒的IgG抗体,28 d的IgG抗体水平较14 d提高10倍以上,42 d时效价略有下降。首次免疫后14 d,免疫原型株或Delta株疫苗的大鼠血清中,针对4株病毒的中和抗体全部阳转;免疫Beta株和Gamma株疫苗的大鼠血清中,针对Beta株和Gamma株病毒的中和抗体全部阳转,针对原型株或Delta株病毒的中和抗体部分阳转。初次免疫后28 d,4种病毒免疫血清中和抗体均阳转,中和抗体效价较14 d大幅升高;初次免疫42 d时中和抗体效价较28 d总体略有下降。Beta株与Gamma株间、原型株与Gamma株间抗原性差异较小,原型株与Beta株间抗原性差异较大。结论原型株、Beta株、Gamma株和Delta株SARSCoV-2灭活疫苗均能刺激大鼠产生较高效价的中和抗体,但免疫原性有差异。

4种杀螺剂对福寿螺防效及对螃蟹安全性初探

选用4种杀螺剂在水稻田与室内2种环境下开展药效试验,并评价了2种药剂对螃蟹的安全性。结果表明:在水稻田未保水的情况下,70%杀螺胺WP 600 g/hm^(2)、750 g/hm^(2),26%四聚·杀螺胺SC 1800 g/hm^(2)对福寿螺表现出良好的防效,药后7 d的防效分别为96.45%、97.17%、97.87%,同时对稻田福寿螺表现出较好的速效性。室内条件下,26%四聚·杀螺胺SC 1800 g/hm^(2)、6%四聚乙醛GR 9000 g/hm^(2)对福寿螺的防效良好,药后24 h的防效分别为100.00%、95.83%。室内条件下,四聚乙醛、杀螺胺对螃蟹敏感,安全性低。

The Contribution of United States Aircraft Reconnaissance Data to the China Meteorological Administration Tropical Cyclone Intensity Data:An Evaluation of Homogeneity

This paper investigates the homogeneity of United States aircraft reconnaissance data and the impact of these data on the homogeneity of the tropical cyclone(TC)best track data for the seasons 1949-1987 generated by the China Meteorological Administration(CMA).The evaluation of the reconnaissance data shows that the minimum central sea level pressure(MCP)data are relatively homogeneous,whereas the maximum sustained wind(MSW)data show both overestimations and spurious abrupt changes.Statistical comparisons suggest that both the reconnaissance MCP and MSW were well incorporated into the CMA TC best track dataset.Although no spurious abrupt changes were evident in the reconnaissance-related best track MCP data,two spurious changepoints were identified in the remainder of the best-track MCP data.Furthermore,the influence of the reconnaissance MSWs seems to extend to the best track MSWs unrelated to reconnaissance,which might reflect the optimistic confidence in making higher estimates due to the overestimated extreme wind“observations”.In addition,the overestimation of either the reconnaissance MSWs or the best track MSWs was greater during the early decades compared to later decades,which reflects the important influence of reconnaissance data on the CMA TC best track dataset.The wind-pressure relationship(WPR)used in the CMA TC best track dataset is also evaluated and is found to overestimate the MSW,which may lead to inhomogeneity within the dataset between the aircraft reconnaissance era and the satellite era.

Discovery of strong bulk Dzyaloshinskii-Moriya interaction in composition-uniform centrosymmetric magnetic single layers

Dzyaloshinskii-Moriya interaction(DMI)is the key ingredient of chiral spintronic phenomena and the emerging technologies based on such phenomena.A nonzero DMI usually occurs at magnetic interfaces or within non-centrosymmetric single crystals.Here,we report the observation of a strong unexpected DMI within a centrosymmetric polycrystalline ferromagnet that has neither a crystal inversion symmetry breaking nor a composition gradient.This DMI is a bulk effect,increases with the thickness of the magnetic layer,and is insensitive to the symmetry of the interfaces or the neighboring materials.We observe a total DMI strength that is a factor of>2 greater than the highest interfacial DMI in the literature.This DMI most likely arises from the strong spin-orbit coupling,strong orbital hybrization,and a“hidden”long-range asymmetry in the material.Our discovery of the strong unconventional bulk DMI in centrosymmetric,composition-uniform magnetic single layers provides fundamental building blocks for the emerging field of spintronics and will stimulate the exploitation of unconventional spin-orbit phenomena in a wide range of materials.

Attribute Reduction of Hybrid Decision Information Systems Based on Fuzzy Conditional Information Entropy

The presence of numerous uncertainties in hybrid decision information systems(HDISs)renders attribute reduction a formidable task.Currently available attribute reduction algorithms,including those based on Pawlak attribute importance,Skowron discernibility matrix,and information entropy,struggle to effectively manages multiple uncertainties simultaneously in HDISs like the precise measurement of disparities between nominal attribute values,and attributes with fuzzy boundaries and abnormal values.In order to address the aforementioned issues,this paper delves into the study of attribute reduction withinHDISs.First of all,a novel metric based on the decision attribute is introduced to solve the problem of accurately measuring the differences between nominal attribute values.The newly introduced distance metric has been christened the supervised distance that can effectively quantify the differences between the nominal attribute values.Then,based on the newly developed metric,a novel fuzzy relationship is defined from the perspective of“feedback on parity of attribute values to attribute sets”.This new fuzzy relationship serves as a valuable tool in addressing the challenges posed by abnormal attribute values.Furthermore,leveraging the newly introduced fuzzy relationship,the fuzzy conditional information entropy is defined as a solution to the challenges posed by fuzzy attributes.It effectively quantifies the uncertainty associated with fuzzy attribute values,thereby providing a robust framework for handling fuzzy information in hybrid information systems.Finally,an algorithm for attribute reduction utilizing the fuzzy conditional information entropy is presented.The experimental results on 12 datasets show that the average reduction rate of our algorithm reaches 84.04%,and the classification accuracy is improved by 3.91%compared to the original dataset,and by an average of 11.25%compared to the other 9 state-of-the-art reduction algorithms.The comprehensive analysis of these research results clearly indicates that

Background:Inflammatory bowel disease(IBD),comprising Crohn's disease(CD)and ulcerative colitis(UC),is a heterogeneous state of chronic intestinal inflammation.Intestinal innate immunity,including innate immune cells,defends against pathogens and excessive entry of gut microbiota,while preserving immune tolerance to resident intestinal microbiota,and may be characterized by its capacity to produce a rapid and nonspecific reaction.The association between microbiota dysbiosis and the pathogenesis of IBD is complex and dynamic.When the intestinal ecosystem is in dysbiosis,the reduced abundance and diversity of intestinal gut microbiota make the host more vulnerable to the attack of exogenous and endogenous pathogenic gut microbiota.The aim of our study was to comprehensively assess the relationship between microbial populations within UC,the signaling pathways of pathogenic gut microbe therein and the inflammatory response,as well as to understand the effects of using PE&AFWE(poppy extract[Papaver nudicaule L.]and Artemisia frigida Willd.extract)on UC modulation.Methods:A UC mouse model was established by inducing SPF-grade C57BL/6 mice using dextrose sodium sulfate(DSS).Based on metagenomic sequencing to characterize the gut microbiome,the relationship between gut microbiota dysbiosis and gut microbiota was further studied using random forest and Bayesian network analysis methods,as well as histopathological analysis.Results:(1)We found that the 5 gut microbiota with the highest relative abundance of inflammatory bowel disease UC model gut microbiota were consistent with the top 5 ranked natural bacteria.There were three types of abundance changes in the model groups:increases(Chlamydiae/Proteobacteria and Deferribacteres),decreases(Firmicutes),and no significant changes(Bacteroidetes).The UC model group was significantly different from the control group,with 1308 differentially expressed species with abundance changes greater than or equal to 2-fold.(2)The proportion of the fecal flora in the UC group decreased by

C1QTNF5 is a novel attachment factor that facilitates the entry of influenza A virus

Influenza A virus(IAV)binds sialic acid receptors on the cell surface to enter the host cells,which is the key step in initiating infection,transmission and pathogenesis.Understanding the factors that contribute to the highly efficient entry of IAV into human cells will help elucidate the mechanism of viral entry and pathogenicity,and provide new targets for intervention.In the present study,we reported a novel membrane protein,C1QTNF5,which binds to the hemagglutinin protein of IAV and promotes IAV infection in vitro and in vivo.We found that the HA1 region of IAV hemagglutinin is critical for the interaction with C1QTNF5 protein,and C1QTNF5 interacts with hemagglutinin mainly through its N-terminus(1–103 aa).In addition,we further demonstrated that overexpression of C1QTNF5 promotes IAV entry,while blocking the interaction between C1QTNF5 and IAV hemagglutinin greatly inhibits viral entry.However,C1QTNF5 does not function as a receptor to mediate IAV infection in sialic acid-deficient CHO-Lec2 cells,but promotes IAV to attach to these cells,suggesting that C1QTNF5 is an important attachment factor for IAV.This work reveals C1QTNF5 as a novel IAV attachment factor and provides a new perspective for antiviral strategies.

Low-frequency oscillation of train-network system considering traction power supply mode

The low-frequency oscillation(LFO)has occurred in the train-network system due to the introduction of the power electronics of the trains.The modeling and analyzing method in current researches based on electrified railway unilateral power supply system are not suitable for the LFO analysis in a bilateral power supply system,where the trains are supplied by two traction substations.In this work,based on the single-input and single-output impedance model of China CRH5 trains,the node admittance matrices of the train-network system both in unilateral and bilateral power supply modes are established,including three-phase power grid,traction transformers and traction network.Then the modal analysis is used to study the oscillation modes and propagation characteristics of the unilateral and bilateral power supply systems.Moreover,the influence of the equivalent inductance of the power grid,the length of the transmission line,and the length of the traction network are analyzed on the critical oscillation mode of the bilateral power supply system.Finally,the theoretical analysis results are verified by the time-domain simulation model in MATLAB/Simulink.
