Rheumatoid arthritis(RA)is a chronic autoimmune disorder marked by persistent synovial inflammation and joint degradation,posing challenges in the development of effective treatments.Nuciferine,an alkaloid found in lotus leaf,has shown promising anti-inflammatory and anti-tumor effects,yet its efficacy in RA treatment remains unexplored.This study investigated the antiproliferative effects of nuciferine on the MH7A cell line,a human RA-derived fibroblast-like synoviocyte,revealing its ability to inhibit cell proliferation,promote apoptosis,induce apoptosis,and cause G1/S phase arrest.Additionally,nuciferine significantly reduced the migration and invasion capabilities of MH7A cells.The therapeutic potential of nuciferine was further evaluated in a collagen-induced arthritis(CIA)rat model,where it markedly alleviated joint swelling,synovial hyperplasia,cartilage injury,and inflammatory infiltration.Nuciferine also improved collagen-induced bone erosion,decreased pro-inflammatory cytokines and serum immunoglobulins(IgG,IgG1,IgG2a),and restored the balance between T helper(Th)17 and regulatory T cells in the spleen of CIA rats.These results indicate that nuciferine may offer therapeutic advantages for RA by decreasing the proliferation and invasiveness of FLS cells and correcting the Th17/Treg cell imbalance in CIA rats.

An efficient reliability-based design optimization method for the support structures of monopile offshore wind turbines is proposed herein.First,parametric finite element analysis(FEA)models of the support structure are established by considering stochastic variables.Subsequently,a surrogate model is constructed using a radial basis function(RBF)neural network to replace the time-consuming FEA.The uncertainties of loads,material properties,key sizes of structural components,and soil properties are considered.The uncertainty of soil properties is characterized by the variabilities of the unit weight,friction angle,and elastic modulus of soil.Structure reliability is determined via Monte Carlo simulation,and five limit states are considered,i.e.,structural stresses,tower top displacements,mudline rotation,buckling,and natural frequency.Based on the RBF surrogate model and particle swarm optimization algorithm,an optimal design is established to minimize the volume.Results show that the proposed method can yield an optimal design that satisfies the target reliability and that the constructed RBF surrogate model significantly improves the optimization efficiency.Furthermore,the uncertainty of soil parameters significantly affects the optimization results,and increasing the monopile diameter is a cost-effective approach to cope with the uncertainty of soil parameters.

[目的/意义]提升老年人数字素养是弥合社会数字鸿沟、促进数字包容的必要之举。新加坡政府提升老年人数字素养的举措与成功经验可为我国政府顶层设计、项目开发与实施优化提供参考。[方法/过程]文章在文献调研基础上,深入调查新加坡政府及老年人服务相关网站,获取政府发布的老年人数字素养提升政策文本及其主导的项目资料,系统梳理和归纳关键举措。[结果/结论]新加坡政府提升老年人数字素养的核心举措有:制定政策、组建专门机构和服务团队、搭建数字技能学习交流平台、提供资金与设备支持。其特点是政府主导并协同社会多方力量参与、重视代际与同辈的作用、线上线下结合多种学习模式、获得多方资金与技术支持等。据此提出我国政府未来实践可参考的方向:进一步发挥政府职能整合社会各方优势资源,设计具有全国范围影响力的实践项目和学习模式,拓展老年人数字技能学习平台和交流空间,增加资金支持和老年友好型智能设备供给。

数字记忆平台是数字时代再现和传播特定社群珍贵文化的重要渠道,而资源是数字记忆构建的基础要素,构建以资源为中心数字记忆平台建设评价指标体系,可为平台建设质量评价提供参考,有助于发现资源建设问题并对问题进行优化。首先,基于文献调研初步确定指标构成;其次,运用德尔菲法,由领域专家对初步构建的指标体系进行评价和修正;再次,结合德尔菲法与层次分析法对优化后的指标体系进行权重测算,形成完整的数字记忆平台建设评价指标体系;最后,选取典型的数字记忆平台“中国传统音乐录音档案”进行实证评估。最终构建由5个一级指标和23个二级指标构成的、以资源为中心的数字记忆平台建设评价指标体系;利用该指标体系评估“世界的记忆·中国传统音乐录音档案”平台建设水平,验证其有效性和客观性;依据评价指标权重和实践现状,对我国数字记忆平台未来建设与资源优化提出建议。

Viruses are representative of a global threat to agricultural production. Genetic resistance is the preferred strategy for the control of viral infection and against loss of crop yield. Viral protein synthesis requires host cellular factors for translating their viral RNAs, and for regulating their replication and cell to cell systemic movement. Therefore, the viruses are dependent on cellular translation factors. Mutations in the gene encoding eIF4E and eIF4G or their isoforms, eIFiso4 E, eIFiso4 G and eIF2Bβ have been mapped as a source of plant potyvirus while other genus of plant virus recessive resistance genes in many species are originated from these loci. Some of other plant translation factors, such as eIF3,eIF4 A-like helicases, eEF1A and eEF1B, which are required in interacting with viral RNAs and regulating various aspects of the infection cycle,have also been identified. Here, we summarized the mechanisms utilized by RNA viruses of eukaryotic plants and the essential roles of e IFs in virus infection. Moreover, we discussed the potential of e IFs as a target gene in the development of genetic resistance to viruses for crop improvement. This review highlighted newly revealed examples of abnormal translational strategies and provided insights into natural host resistance mechanisms that have been linked to 3 cap-independent translational enhancer activity.

The development of new copper-based catalysts in photoredox transformations has become a rapidly growing area of research and has been actively explored in the past several years.Herein,we have developed a new copper-based catalyst,[(binap)(tpy)Cu]Cl,and applied it in the visible-light promoted Sonogashira coupling reactions.This mild protocol results in various substituted alkynes from simple sulfonium salts which can be easily prepared from commercially available and inexpensive thiols,alcohols,and alkyl halides.This study will broaden the still limited strategies of copper photocatalysts in organic transformations.

Introduction: The transmission pathways of coronavirus disease 2019 (COVID-19) remain not completely clear. In this case study the test for the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in pharyngeal swab and anal swab were compared.Case presentation: A 3-month-old girl was admitted to our hospital with COVID-19. Her parents had both been diagnosed with COVID-19. The results of pharyngeal swab and anal swab of the little girl were recorded and compared during the course of the disease. The oropharyngeal specimen showed negative result for SARS-CoV-2 on the 14th day after onset of the illness. However, the anal swab was still positive for SARS-CoV-2 on the 28th day after the onset of the illness.Conclusion: The possibility of fecal-oral transmission of COVID-19 should be assessed. Personal hygiene during home quarantine merits considerable attention.
