BACKGROUND： Electrophysiological properties of the song nucleus have been revealed using conventional techniques, such as intracellular and extracellular recording. Research concerning the neuronal activation properties and regulations of the song system at the cellular and ion channel level may help reveal the neural mechanism of song learning. OBJECTIVE： To perform whole-cell recording of robust nucleus of the arcopallium （RA） neurons in brain slices from adult zebra finches （Taeniopygia guttata） and observe the action potential, sodium/potassium current and the spontaneous postsynaptic current of RA neurons. DESIGN, TIME AND SETTING： Self-controlled, neuroelectrophysiological experiment. The study was performed at the Neurophysiology Laboratory of South China Normal University from April to September 2008. MATERIALS： Flaming/Brown puller P-97 was purchased from Sutter Ins, USA; Axopatch 700B amplifier and Digidata 1332A converter were purchased from Axon Instrument, USA; pClamp software was provided by Axon Instrument, USA. METHODS： RA neurons were acutely isolated from 24 healthy male zebra finches. The action potential, voltage-gate sodium/potassium current and spontaneous postsynaptic current were recorded by whole-cell recording technology. Data were analyzed by pClamp software. MAIN OUTCOME MEASURES： The amplitude and frequency of the action potential, and the amplitude of the voltage-dependent and spontaneous postsynaptic currents, were measured. RESULTS：（1） Testing of action potential： Cells exhibited a stable current-voltage relationship following a series of hyperpolarization stepped currents, and an action potential was triggered by the spike threshold. All the recorded cells displayed repetitive firing following depolarizing current injection, with a frequency beyond 100 Hz. （2） Testing of voltage-gate currents： The inward and outward whole-cell currents were observed after a series of depolarizing voltage steps. Estimation of Ecological Water Requirements Based on Habitat Response to Water Level in Huanghe River Delta,China

In recent years,wetland ecological water requirements (EWRs) have been estimated by using hydrological and functional approaches,but those approaches have not yet been integrated for a whole *** paper presents a new method for calculating wetland EWRs,which is based on the response of habitats to water level,and determines water level threshold through the functional integrity of *** show that in the Huanghe (Yellow) River Delta water levels between 5.0 m and 5.5 m are required to maintain the functional integrity of the wetland at a value higher than *** of the dominant plants in the delta,Phragmites australis,tolerates water level fluctuation of about ± 0.25 m without the change in wetland functional *** minimum,optimum and maximum EWRs for the Huanghe River Delta are 9.42×106 m3,15.56×106 m3 and 24.12×106 m3 with water levels of 5.0 m,5.2 m and 5.5 m,corresponding to functional integrity indices of 0.70,0.84 and 0.72,respectively.A wetland restoration program has been performed,which aims to meet these EWRs in attempt to recover from losses of up to 98% in the delta's former wetland area.

本研究以烤烟品种云烟87为试验对象,设置不同碳基有机肥施用量,研究其对烤烟生长及产质量的影响。结果表明:施用碳基有机肥可以提高烟叶产量,改善农艺性状及品质,烤后烟叶均符合优质烟叶指标要求;与常规施肥对照相比,烟叶总产量提升52.3%~96.6%,优质烟叶产量提升44.5%~54.0%,中等质量烟叶产量提升68.7%~144.0%。综合来看,以施用碳基有机肥1500 kg/hm^(2)对烤烟生长及产质量的提升效果最好。

Construction of a microsatellite-based genetic linkage map for half-smooth tongue sole Cynoglossus semilaevis

The half-smooth tongue soleCynoglossus semilaevisis an important cultured marine fish and a promising model fish for the study of sex determination. Sex-specific genetic linkage maps of half-smooth tongue sole were developed with 567 markers (565 microsatellite markers and two SCAR markers). The parents and F1 progeny (92 individuals) were used as segregating populations. The female map was composed of 480 markers in 21 linkage groups, covering a total of 1388.1 cM, with an average interval 3.06 cM between markers. The male map consisted of 417 markers in 21 linkage groups, spanning 1480.9 cM, with an average interval of 3.75 cM. The female and male maps had 474 and 416 unique positions, respectively. The genome length of half-smooth tongue sole was estimated to be 1522.9 cM for females and 1649.1cM for males. Based on estimations of map length, the female and male maps covered 91.1% and 89.8% of the genome, respectively. Furthermore, two female-specific SCAR markers, f-382 and f-783, were mapped on LG15f (linkage group 15 in female maps). The present study presents a mid-density genetic linkage map for half-smooth tongue sole. These improved genetic linkage maps may facilitate systematic genome searches to identify quantitative trait loci (QTL), such as disease resistance, growth and sex-related traits, and are very useful for marker-assisted selection breeding programs for economically important traits in half-smooth tongue sole.
