Network pharmacology approaches for research of Traditional Chinese Medicines

Pharmacodynamics material basis and effective mechanisms are the two main issues to decipher the mechnisms of action of Traditional Chinese medicines(TCMs)for the treatment of diseases.TCMs,in“multi-component,multi-target,multi-pathway”paradigm,show satisfactory clinical results in complex diseases.New ideas and methods are urgently needed to explain the complex interactions between TCMs and diseases.Network pharmacology(NP)provides a novel paradigm to uncover and visualize the underlying interaction networks of TCMs against multifactorial diseases.The development and application of NP has promoted the safety,efficacy,and mechanism investigations of TCMs,which then reinforces the credibility and popularity of TCMs.The current organcentricity of medicine and the“one disease-one target-one drug”dogma obstruct the understanding of complex diseases and the development of effective drugs.Therefore,more attentions should be paid to shift from“phenotype and symptom”to“endotype and cause”in understanding and redefining current diseases.In the past two decades,with the advent of advanced and intelligent technologies(such as metabolomics,proteomics,transcriptomics,single-cell omics,and artificial intelligence),NP has been improved and deeply implemented,and presented its great value and potential as the next drug-discovery paradigm.NP is developed to cure causal mechanisms instead of treating symptoms.This review briefly summarizes the recent research progress on NP application in TCMs for efficacy research,mechanism elucidation,target prediction,safety evaluation,drug repurposing,and drug design.

Preliminary calibration results for Jason-3 and Sentinel-3 altimeters in the Wanshan Islands

We present preliminary calibration results for Jason-3 and Sentinel-3A altimeters that we set up in the Wanshan Islands in Guandong Province,China.Two campaigns were carried out in 2018,from March 8 to April 16 and from November 3 to December 11,2018.Three GPS reference stations and tide gauges were established on the islands of Zhiwan,Dangan,and Wailingding during the campaigns.The in-situ sea surface height(SSH)of the altimeter footprint was determined using the tide gauge.The tidal and geoid connection between the tide gauge locations and the altimeter footprints were computed with the NAO.99Jb tidal prediction system and the EGM 2008 geoid,respectively.The data of the tide gauges were defi ned using the GPS buoy and GPS reference stations during the campaigns.The results show that the waveform of the altimeters was slightly contaminated by the small islands.The bias associated with Jason-3 and Sentinel-3A amounted to approximately+20.7±49.7 mm and+30.1±39.4 mm,respectively,which agreed well with the results from other in-situ calibration fi elds.This indicates that the Wanshan area was very suitable as an in-situ calibration/validation fi eld.The wet zenith delay(WZD)determined from the Microwave Radiometer(MWR)and the GPS measurements diff ered from each other for the Jason-3 and Sentinel-3A by−6.6±7.4 mm and 0±6.8 mm,respectively.

3D model retrieval virtual reality applications. In can benefit many downstream this paper, we propose a new sketch-based 3D model retrieval framework by coupling local features and manifold ranking. At technical fronts, we exploit spatial pyramids based local structures to facilitate the efficient construction of feature descriptors. Meanwhile, we propose an improved manifold ranking method, wherein all the categories between arbitrary model pairs will be taken into account. Since the smooth and detail-preserving line drawings of 3D model are important for sketch-based 3D model retrieval, the Difference of Gaussians （DOG） method is employed to extract the line drawings over the projected depth images of 3D model, and Bezier Curve is then adopted to further optimize the extracted line drawing. On that basis, we develop a 3D model retrieval engine to verify our method. We have conducted extensive experiments over various public benchmarks, and have made comprehensive comparisons with some state-of-the-art 3D retrieval methods. All the evaluation results based on the widely-used indicators prove the superiority of our method in accuracy, reliability, robustness, and versatility.

Background:Understanding the spatial pattern and driving factors of forest carbon density in mountainous terrain is of great importance for monitoring forest carbon in support of sustainable forest management for mitigating climate change.Methods:We collected the forest inventory data in 2015 in Shanxi Province,eastern Loess Plateau of China, to explore the spatial pattern and driving factors of biomass carbon density(BCD) for natural and planted coniferous forests using Anselin Local Moran’s I,Local Getis-Ord G~* and semivariogram analyses,and multi-group structural equation modeling,respectively.Results:The result of spatial pattern of BCDs for natural forests showed that the BCD was generally higher in the north but lower in the south of Shanxi.The spatial pattern for planted forests was substantially different from that for natural forests.The results of multi-group SEM suggested that elevation(or temperature as the alternative factor of elevation) and stand age were important driving factors of BCD for these two forest types.Compared with other factors,the effects of latitude and elevation on BCD showed much greater difference between these two forest types.The difference in indirect effect of latitude(mainly through affecting elevation and stand age) between natural and planted forests was to some extent a reflection of the difference between the spatial patterns of BCDs for natural and planted forests in Shanxi.Conclusions:The natural coniferous forests had a higher biomass carbon density,a stronger spatial dependency of biomass carbon density relative to planted coniferous forests in Shanxi.Elevation was the most important driving factor,and the effect on biomass carbon density was stronger for natural than planted coniferous forests.Besides,latitude presented only indirect effect on it for the two forest types.

The effect of extrusion conditions on the properties and textures of AZ31B alloy

The effect of extrusion conditions on the tensile properties and texture of AZ31B alloy has been investigated by means of optical microscopy(OM),scanning electron microscopy(SEM),electron backscattered diffraction(EBSD)and tensile tests.It is found that the ultimate tensile strength(UTS),the yield strength(YS)and elongation(EN)of the extruded AZ31B alloy are more significantly influenced by extrusion velocities in contrast with temperature.Although the extrusion conditions are different,the {1102} texture is the chief texture in the AZ31B after extrusion.Moreover,the extrusion textures become scattered with increasing the temperatures at the same extrusion velocity.As the extrusion velocity is raised at the same temperature,the orientation density of textures increases and the separated textures become relatively concentrated.This leads to the changes of tensile properties at different extrusion conditions.©2017 Production and hosting by Elsevier B.V.on behalf of Chongqing University.

Modeling of spatial and temporal variations of ozone-NO_(x)-VOC sensitivity based on photochemical indicators in China

Comprehensive air quality model with extensions(CAMx)-decoupled direct method(DDM)was used to simulate ozone-NO_(x)-VOCs sensitivity of for May-November in 2016-2018 in China.Based on the relationship between the simulated ozone(O_(3))sensitivity values and the ratio of formaldehyde(HCHO)to NO_(2)(FNR)and the ratio of production rate of hydrogen peroxide(H_(2)O_(2))to production rate of nitric acid(HNO_(3))(P_(H_(2)O_(2))/P_(HNO_(3))),the localized range of FNR and P_(H_(2)O_(2))/P_(HNO_(3))thresholds in different regions in China were obtained.The overall simulated FNR values are about 1.640-2.520,and P_(H_(2)O_(2))/P_(HNO_(3))values are about 0.540-0.830 for the transition regime.Model simulated O_(3)sensitivities or region specific FNR or P_(H_(2)O_(2))/P_(HNO_(3))thresholds should be applied to ensure the accurate local O3 sensitivity regimes.Using the tropospheric column FNR values from ozone monitoring instrument(OMI)satellite data as an indicator with the simulated threshold values,the spatial distributions of O_(3)formation regimes in China are determined.The O_(3)sensitivity regimes from eastern to central China are gradually from VOC-limited,transition to NO_(x)-limited spatially,and moving toward to transition or NO_(x)-limited regime from 2005 to 2019 temporally.

Polar codes become the coding scheme for control channels of enhanced mobile broadband(eMBB)scenarios in the fifth generation(5 G)communication system due to their excellent decoding performance.For the cell search procedure in 5 G system,some common information bits(CIBs)are transmitted in consecutive synchronization signal blocks(SSBs).In this paper,a dual-cyclic redundancy check(dual-CRC)aided encoding scheme is proposed,and the corresponding dual-successive cancellation flip(dual-SCFlip)algorithm is given to further improve the performance of polar codes in the low signal-to-noise ratio(SNR)environment.In dual-CRC aided encoding structure,the information bits of polar codes in different transmission blocks add cyclic redundancy check(CRC)sequences respectively according to CIBs and different information bits(DIBs).The structure enlarges the size of CIBs to improve the block error ratio(BLER)performance of the system.The dual-SCFlip decoder can perform bit flip immediately once CIBs is decoded completely,and then decode DIBs or terminate decoding in advance according to the CRC result,which reduces the delay of decoding and mitigates the error propagation effect.Simulation results show that the dual-CRC aided encoding scheme and dual-SCFlip decoder have significant performance improvement compared to other existing schemes with low SNR.

The self-lubricating ceramic coatings that can control friction and wear have attracted researchers’widespread attention.However,the poor interfacial bonding between lubricants and ceramics and the deterioration of mechanical properties due to a tribological design limit their practical applications.Here,a robust self-lubricating coating was fabricated by an in-situ synthesis of MoS_(2)/C within inherent defects of thermally sprayed yttria-stabilized zirconia(YSZ)coatings.The edge-pinning by noncoherent endows hybrid coatings with excellent interfacial strength,increasing their hardness(HV)and cohesive strength.Furthermore,owing to the formation of a well-covered robust lubricating film at a frictional interface,a coefficient of friction(COF)can be reduced by 79.6%to 0.15,and a specific wear rate(W)drops from 1.36×10^(−3) to 6.27×10^(−7) mm^(3)·N^(−1)·m^(−1).Combining outstanding mechanical properties and tribological performance,the hybrid coating exhibits great application potential in controlling friction and wear.Importantly,this strategy of introducing the target materials into the inherent defects of the raw materials to improve the relevant properties opens new avenues for the design and preparation of composite materials.

Traditional Chinese medicine (TCM) is a medical system that has collected and summarized abundant clinical experience over its long history of more than 2000 years. However, the frequent occurrence of TCM-induced adverse reactions has hindered the modernization and internationalization of TCM, while attracting increasing attention from around the world. Unlike chemical drugs and biological agents, the difficulties involved in research on the toxicity and safety of TCM mainly include the complexity of its components and the unpredictability of drug–body interactions. Much of TCM, which has overall therapeutic effects, has the typical mechanisms of multiple components, multiple pathways, and multiple targets. While considering the gradualness and unpredictability of TCM toxicity, the ambiguity of toxicants and safe dosage, and individual differences during long-term TCM administration, we have systematically established key techniques for the toxicity assessment of TCM. These techniques mainly include TCM toxicity discovery in an early phase, based on a combination of drug toxicology genomics and metabolomics;methods to identify dose–toxicity relationships in TCM;and integrated techniques for the exploration of TCM interactions, such as fast-screening tests based on drug-metabolizing enzymes and receptor pathways. In particular, we have developed a new technical system for TCM safety evaluation using molecular toxicology, which has been validated well in research on TCM compatibility contraindication, quality control, and allergen discovery. The application of this key technical platform is introduced here in detail. This application includes model organisms, toxicant biomarkers, a magnetic suspension technique, and the application of network toxicology and computational toxicology in research on the toxicity of Fructus toosendan, Semen cassiae, Polygonum multiflorum, and Fructus psoraleae.
