Influence of ground effect on flow field structure and aerodynamic noise of high-speed trains

The simulation of the ground effect has always been a technical difficulty in wind tunnel tests of high-speed trains.In this paper,large eddy simulation and the curl acoustic integral equation were used to simulate the flow-acoustic field results of high-speed trains under four ground simulation systems(GSSs):“moving ground+rotating wheel”,“stationary ground+rotating wheel”,“moving ground+stationary wheel”,and“stationary ground+stationary wheel”.By comparing the fluid-acoustic field results of the four GSSs,the influence laws of different GSSs on the flow field structure,aero-acoustic source,and far-field radiation noise characteristics were investigated,providing guidance for the acoustic wind tunnel testing of high-speed trains.The calculation results of the aerodynamic noise of a 350 km/h high-speed train show that the moving ground and rotating wheel affect mainly the aero-acoustic performance under the train bottom.The influence of the rotating wheel on the equivalent sound source power of the whole vehicle was not more than 5%,but that of the moving ground slip was more than 15%.The average influence of the rotating wheel on the sound pressure level radiated by the whole vehicle was 0.3 dBA,while that of the moving ground was 1.8 dBA.

The integer least squares(ILS)estimation is commonly used for carrier phase ambiguity resolution(AR).More recently,the best integer equivariant(BIE)estimation has also attracted an attention for complex application scenarios,which exhibits higher reliability by a weighted fusion of integer candidates.However,traditional BIE estimation with Gaussian distribution(GBIE)faces challenges in fully utilizing the advantages of BIE for urban low-cost positioning,mainly due to the presence of outliers and unmodeled errors.To this end,an improved BIE estimation method with Laplacian distribution(LBIE)is proposed,and several key issues are discussed,including the weight function of LBIE,determination of the candidates included based on the OIA test,and derivation of the variance of LBIE solutions for reliability evaluation.The results show that the proposed LBIE method has the positioning accuracy similar to the BIE using multivariate t-distribution(TBIE),and significantly outperforms the ILS-PAR and GBIE methods.In an urban expressway test with a Huawei Mate40 smartphone,the LBIE method has positioning errors of less than 0.5 m in three directions and obtains over 50%improvements compared to the ILS-PAR and GBIE methods.In an urban canyon test with a low-cost receiver STA8100 produced by STMicroelectronics,the positioning accuracy of LBIE in three directions is 0.112 m,0.107 m,and 0.252 m,respectively,with improvements of 17.6%,27.2%,and 26.1%compared to GBIE,and 23.3%,28.2%,and 30.6%compared to ILS-PAR.Moreover,its computational time increases by 30–40%compared to ILS-PAR and is approximately half of that using TBIE.

Stress and illness connection is complex and involves multiple physiological systems.Panax ginsengs,reputed for their broad-spectrum“cure-all”effect,are widely prescribed to treat stress and related illnesses.However,the identity of ginseng’s“cure-all”medicinal compounds that relieve stress remains unresolved.Here,we identify ginsentides as the principal bioactives that coordinate multiple systems to restore homeostasis in response to stress.Ginsentides are disulfide-rich,cell-penetrating and proteolytic-stable microproteins.Using affinity-enrichment mass spectrometry target identification together with in vitro,ex vivo and in vivo validations,we show that highly purified or synthetic ginsentides promote vasorelaxation by producing nitric oxide through endothelial cells via intracellular PI3K/Akt signaling pathway,alleviate a1-adrenergic receptor overactivity by reversing phenylephrine-induced constriction of aorta,decrease monocyte adhesion to endothelial cells via CD166/ESAM/CD40 and inhibit P2Y12 receptors to reduce platelet aggregation.Orally administered ginsentides were effective in animal models to reduce ADP-induced platelet aggregation,to prevent collagen and adrenalineinduced pulmonary thrombosis as well as anti-stress behavior of tail suspension and forced swimming tests in mice.Together,these results strongly suggest that ginsentides are the principal panacea compounds of ginsengs because of their ability to target multiple extra-and intra-cellular proteins to reverse stress-induced damages.

Externallyvalidated nomogramfor predicting short-term pregnancy outcome of singleton pregnancies with fetal growth restriction(FGR)

Objective This studyaimed at developing an available predictive model of singleton pregnancies with fetal growth restriction(FGR)for accurate and individualised prognosis assessment.Methods The prediction nomogram was developed by using multivariable Cox regression with data for 301 singleton FGR pregnancies at Peking University People's Hospital.External validation was performed in 321eligible singleton FGR pregnancies at the Affiliated Hospital of Qingdao University.Results Absent umbilical arterial flow,fetal anomaly,history of abnormal pregnancy,non-cephalic presentation and historyof caesarean sectionwere independent prognostic factors foradverse perinatal outcomes in singleton FGR pregnancies in the training set.In the training cohort of the internal validation set,the nomogram estimated pregnancy prognosis of FGR singleton pregnancies based on these five variables,with a concordance index(C-index)of 0.859(95%Cl:0.81 to 0.90)for predicting termination of pregnancy(TOP),which included intrauterine fetal death and therapeutic lethal induction,with a C-index of 0.92(95%Cl:0.86 to 0.98)for predicting stillbirth,and a C-index of 0.87(95%Cl:0.83 to 0.92)for predicting therapeutic lethal induction with indications.Encouragingly,consistent results wereobserved in the external validation set,witha C-index of 0.776(95%Cl:0.71 to 0.84)for predicting TOP,which included intrauterine fetal death and therapeutic lethal induction,with a C-index of 0.773(95%Cl:0.70 to 0.84)for predicting stillbirth,and a C-index of 0.776(95%Cl:0.70 to 0.85)for predicting therapeutic lethal induction with indications.Furthermore,the calibrations of the nomograms predicting the 28th and 34th TOPfreegestation week strongly corresponded to theactual survivaloutcome.Conclusion This prediction model may help clinicians indecision-making for singleton pregnancies with FGR,especially for patients with a single abnormal umbilical arterial flow orfetal anomaly,without induced labour indications for these abnormalities.

Comparative Study of Genetic Structure and Genetic Diversity betweenWild and Cultivated Populations of Taxus cuspidata,Northeast China

Taxus cuspidata is a rare plant with important medicinal and ornamental value.Aiming at the obvious differences between wild and cultivated populations of T.cuspidata from Northeast China,a total of 61 samples,that is,33 wild yews and 28 cultivated yews were used to analyze the differences and correlations of the kinship,genetic diversity,and genetic structure between them by specific length amplified fragment sequencing(SLAF-seq).Finally,470725 polymorphic SLAF tags and 58622 valid SNP markers were obtained.Phylogenetic analysis showed that 61 samples were classified into 2 clusters:wild populations and cultivated populations,and some wild yews were categorized into the cultivated populations;the genetic diversity analysis showed that the Nei diversity index of wild populations(0.4068)was smaller than that of cultivated populations(0.4414),and the polymorphic information content(PIC)of wild populations(0.2861)was smaller than that of cultivated populations(0.3309).The genetic differentiation analysis showed that the total populations of gene diversity(H_(t))of cultivated and wild populations were respectively 0.8159 and 0.5685,the coefficient of gene differentiation(G_(st))of cultivated and wild populations was respectively 0.3021 and 0.1068,and the gene flow(N_(m))(2.4967)of wild populations was larger than cultivated populations(0.8199).The molecular variance(AMOVA)revealed that inter-population variation accounted for 29.57%of the total genetic variation,while intra-population variation accounted for 70.42% of the total genetic variation(p<0.001),this suggested that the genetic variation in the T.cuspidata is mainly attributed to within-population factors.In conclusion,the genetic distance between geographical ecological groups of wild populations was generally smaller than that of cultivated populations,and the degree of genetic diversity and genetic differentiation was smaller than that of cultivated populations.As evident,the utilization of SLAF-seq technology enables efficient and accurate development of

Partial aging can counter-intuitively couple with sulfidation to improve the reactive durability of zerovalent iron

Sulfated zero-valent iron(SZVI)has shown promising applications in wastewater treatment.However,the rapid decline in the reactivity of SZVI with time limits its real practice.To mediate this problem,partial aging was proposed to improve the reactive durability of SZVI.Taking Cr(VI)as the target contaminant,we found that the aged ZVI(AZVI)gradually lost reactivity as aging time increased from 0.5 to 2 d.Counter-intuitively,the partially aged SZVI(ASZVI)showed greater reactivity than SZVI when exposed to oxygenated water for a period ranging from 0.5 to 14 d.In addition,the ASZVI with 0.5 d of aging time(ASZVI-0.5)not only maintained reactivity in successive runs but also increased the Cr(VI)removal capacity from 9.1 mg/g by SZVI to 19.1 mg/g by ASZVI-0.5.Correlation analysis further revealed that the electron transfer from the Fe0 core to the shell was mediated by the conductive FeS and FeS2 in the subshell of ASZVI.Meanwhile,the lepidocrocite and magnetite on the surface of ASZVI facilitated Cr(VI)adsorption and subsequent electron transfer for Cr(VI)reduction.Moreover,the iron(hydr)oxide shell could retain the conductive FeS and FeS2 in the subshell,allowing ASZVI to reduce Cr(VI)efficiently and sustainably.In general,partial aging can enhance the reactive durability of ZVI when coupled with sulfidation and this synergistic effect will be beneficial to the application of SZVI-based technology for wastewater treatment.

Aldehyde oxidase(AOX)is a molybdoenzyme that is primarily expressed in the liver and is involved in the metabolism of drugs and other xenobiotics.AOX-mediated metabolism can result in unexpected outcomes,such as the production of toxic metabolites and high metabolic clearance,which can lead to the clinical failure of novel therapeutic agents.Computational models can assist medicinal chemists in rapidly evaluating the AOX metabolic risk of compounds during the early phases of drug discovery and provide valuable clues for manipulating AOX-mediated metabolism liability.In this study,we developed a novel graph neural network called AOMP for predicting AOX-mediated metabolism.AOMP integrated the tasks of metabolic substrate/non-substrate classification and metabolic site prediction,while utilizing transfer learning from 13C nuclear magnetic resonance data to enhance its performance on both tasks.AOMP significantly outperformed the benchmark methods in both cross-validation and external testing.Using AOMP,we systematically assessed the AOX-mediated metabolism of common fragments in kinase inhibitors and successfully identified four new scaffolds with AOX metabolism liability,which were validated through in vitro experiments.Furthermore,for the convenience of the community,we established the first online service for AOX metabolism prediction based on AOMP,which is freely available at https://***.

Eco-physiological characteristics of Tetracentron sinense Oliv.saplings in response to different light intensities

The regeneration of Tetracentron sinense Oliv.is poor in the understory and in open areas due to the charac-teristics of natural regeneration of the species on forest edges and in gaps.It is unclear whether different light intensities in various habitats affect eco-physiological characteristics of saplings and their natural regeneration.In this study,the light intensity in T.sinense habitats was simulated by artificial shading(L1:100%NS(natural sunlight)in the open;L2:50%NS in a forest gap or edge;L3:10%NS in understory)to investigate differences in morphology,leaf structure,physiology,and photosynthesis of 2-year-old sap-lings,and to analyze the mechanism of light intensity on sapling establishment.Significant differences were observed in morphology(including leaf area,and specific leaf area)under different light intensities.Compared to L1 and L3,chloroplast structure in L2 was intact.With increasing time,superoxide dismutase(SOD)and catalase(CAT)activities in L2 became gradually higher than under the other light intensities,while malondialdehyde(MDA)content was opposite.Shading decreased osmoregulation substance contents of leaves but increased chlorophyll.The results suggest that light intensities significantly affect the eco-physiological characteristics of T.sinense saplings and they would respond most favorably at intermediate levels of light by optimizing eco-physiological characteristics.Therefore,50%natural sunlight should be created to promote saplings establishment and population recovery of T.sinense during in situ conservation,including sowing mature seeds in forest edges or gaps and providing appropriate shade protection for seedlings and saplings in the open.

Transmucosal drug administration represents a potential strategy for enhancing treatment efficacy and reducing side effects by avoiding the first-pass effect into the systemic circulation and delivering therapeutics directly to the target disease site.However,many challenges still remain in its clinical application,including low drug availability and limited retention time in the mucosa.The burgeoning advancement of nanotechnologies offers great potential to overcome the above limitations,leveraging their distinct advantages of high drug-loading capacity and strong permeability.In this review,the latest developments of nanoparticles(NPs)in transmucosal drug delivery as well as their clinical applications are discussed.

SETDB1-mediated CD147-K71 di-methylation promotes cell apoptosis in non-small cell lung cancer

Protein post-translational modifications(PTMs)are at the heart status of cellular signaling events and broadly involved in tumor progression.CD147 is a tumor biomarker with various PTMs,promoting tumor metastasis and metabolism reprogramming.Nevertheless,the relationship between the PTMs of CD147 and apoptosis has not been reported.In our study,we produced a specific anti-CD147-K71 di-methylation(CD147-K71me2)antibody by immunizing with a di-methylated peptide and observed that the level of CD147-K71me2 in non-small cell lung cancer(NSCLC)tissues were lower than that in NSCLC adjacent tissues.SETDB1 was identified as the methyltransferase catalyzing CD147 to generate CD147-K71me2.RNA-seq showed that FOSB was the most significant differentially expressed gene(DEG)between wild-type CD147(CD147-WT)and K71-mutant CD147(CD147-K71R)groups.Subsequently,we found that CD147-K71me2 promoted the expression of FOSB by enhancing the phosphorylation of p38,leading to tumor cell apoptosis.In vivo experiments showed that CD147-K71me2 significantly inhibited tumor progression by promoting cell apoptosis.Taken together,our findings indicate the inhibitory role of CD147-K71me2 in tumor progression from the perspective of post-translational modification,which is distinct from the pro-cancer function of CD147 itself,broadening our perspective on tumor-associated antigen CD147.
