miR-200b-3p accelerates progression of pituitary adenomas by negatively regulating expression of RECK

MicroRNA(miR)-200b-3p has been associated with many tumors,but its involvement in pituitary adenoma is unclear.This study investigated the molecular mechanism underlying miR-200b-3p regulation in pituitary adenomas to provide a theoretical basis for treatment.Bioinformatics was used to analyze pituitary adenoma-related genes and screen new targets related to RECK and miRNA.As well,the relationship between miR-200b-3p and RECK protein was verified using a double-luciferase reporter gene assay.The expression of miR-200b-3p in clinical samples was analyzed by in situ hybridization.Transfection of the miR-200b-3p inhibitor and small interfering-RECK(si-RECK)was verified by qPCR.GH3 cell viability and proliferation were detected using CCK8 and EdU assays.Apoptosis was detected by flow cytometry and western blotting.Wound healing and Transwell assays were used to detect cell migration and invasion.The effects of miR-200b-3p and RECK on GH3 cells were verified using salvage experiments.miR-200b-3p was highly expressed in pituitary tumor tissue.Inhibitors of miR-200b-3p inhibited cell proliferation promoted cell apoptosis,inhibited invasion and migration,and inhibited the expression of matrix metalloproteinases.Interestingly,miR-200b-3p negatively regulated RECK.The expression of RECK in pituitary adenoma tissues was lower than that in neighboring tissues.Si-RECK rescued the function of miR-200b-3p inhibitors in the above cellular behaviors,and miR-200b-3p accelerated the development of pituitary adenoma by negatively regulating RECK expression.In summary,this study investigated the molecular mechanism by which miR-200b-3p regulates the progression of pituitary adenoma through the negative regulation of RECK.The findings provide a new target for the treatment of pituitary adenoma.

Bioinspired Tactile Sensation Based on Synergistic Microcrack-Bristle Structure Design toward High Mechanical Sensitivity and Direction-Resolving Capability

Natural tactile sensation is complex,which involves not only contact force intensity detection but also the perception of the force direction,the surface texture,and other mechanical parameters.Nevertheless,the vast majority of the developed tactile sensors can only detect the normal force,but usually cannot resolve shear force or even distinguish the directions of the force.Here,we present a new paradigm of bioinspired tactile sensors for resolving both the intensity and the directions of mechanical stimulations via synergistic microcrack-bristle structure design and cross-shaped configuration engineering.The microcrack sensing structure gives high mechanical sensitivity to the tactile sensors,and the synergistic bristle structure further amplifies the sensitivity of the sensors.The cross-shaped configuration engineering of the synergistic microcrack-bristle structure further endows the tactile sensors with good capability to detect and distinguish the directions of the applied mechanical forces.The as-fabricated tactile sensors exhibit a high sensitivity(25.76 N^(−1)),low detection limit(5.4 mN),desirable stability(over 2,500 cycles),and good capability to resolve both mechanical intensity and directional features.As promising application scenarios,surface texture recognition and biomimetic path explorations are successfully demonstrated with these tactile sensors.This newly proposed tactile sensation strategy and technology have great potential applications in ingenious tactile sensation and construction of various robotic and bionic prostheses with high operational dexterity.

Coordination complex of a copper cyanurate(Cu(Ⅱ)-CA) was transformed into coordination polymers upon the stimulus of extra Cu(Ⅱ) through “directed Ostwald ripening”. By increasing the molar ratio of Cu(Ⅱ) to CA, we obtained two coordination polymers with selective coordination sites: Cu(Ⅱ)-κ N(HCA)κ NCu(Ⅱ) and Cu(Ⅱ)-κ N(HCA)κ O-Cu(Ⅱ), which display disparate magnetic interactions.

Immune-mediated liver injury (ILI) is a condition where an aberrant immune response due to various triggers causes the destruction of hepatocytes. Fibroblast growth factor 4 (FGF4) was recently identified as a hepatoprotective cytokine;however, its role in ILI remains unclear. In patients with autoimmune hepatitis (type of ILI) and mouse models of concanavalin A (ConA)- or S-100-induced ILI, we observed a biphasic pattern in hepatic FGF4 expression, characterized by an initial increase followed by a return to basal levels. Hepatic FGF4 deficiency activated the mitochondria-associated intrinsic apoptotic pathway, aggravating hepatocellular apoptosis. This led to intrahepatic immune hyper-reactivity, inflammation accentuation, and subsequent liver injury in both ILI models. Conversely, administration of recombinant FGF4 reduced hepatocellular apoptosis and rectified immune imbalance, thereby mitigating liver damage. The beneficial effects of FGF4 were mediated by hepatocellular FGF receptor 4, which activated the Ca^(2+)/calmodulin-dependent protein kinasekinase 2 (CaMKKβ) and its downstream phosphatase and tensin homologue-induced putative kinase 1 (PINK1)-dependent B-cell lymphoma 2-like protein 1-isoform L (Bcl-XL) signalling axis in the mitochondria. Hence, FGF4 serves as an early response factor and plays a protective role against ILI, suggesting a therapeutic potential of FGF4 and its analogue for treating clinical immune disorder-related liver injuries.

The development of a selective catalyst for the conversion of biomass and plastics into H2by steam reforming can combat the energy crisis and global warming.In this work,support Ni-Fe-Ca/H-Al bifunctional catalysts were prepared by loading Ni and Fe into pretreatment CaO/Al_(2)O_(3)(Ca/H-Al)carriers and showed high catalytic activity for the steam reforming of biomass and plastic.Moreover,the idea of bidirectional degradation was exploited to strengthen the pyrolysis of plastic with a high H/C and biomass with a high O/C.Interestingly,the products presented high H2selective(1302.10 m L/g)and low CO_(2)yield(120.23 m L/g)in 7Ni-5Fe-Ca/H-Al(2:4)catalyst compared with current reports.Here,the abundant oxygen vacancies(Ov)in the H-Al carrier exhibited an electron-deficient nature,providing active sites for anchoring Ni O.Meanwhile,Ni O interacted with Ca_(2)Fe_(2)O_(5)to produce more defective Ovsites,which stabilized the NiO particles in the 7Ni-5Fe-Ca/H-Al(2:4)catalyst,and the interaction between the catalyst and the carrier was enhanced,leading to the reduction of weakly basic sites,this property promoted the strong adsorption of CO_(2)and H2O by the catalyst,contributing to the enhancement of efficient steam conversion and the promotion of conversion of by-products to H2.Notably,7Ni-5Fe-Ca/H-Al(2:4)catalysts maintained structural integrity after regeneration and exhibited excellent regenerability in H2selection and CO_(2)adsorption.The work provides a new idea for the study of efficient H2production from steam reforming of biomass and plastics.

Conventional self-assembled monolayer(SAM)hole transporters in organic solar cells(OSCs)generally suffer from poor uniformity and limited thickness tolerance,hindering their large-scale production.To overcome these limitations,we introduce a novel polymeric hole transporter synthesized by polymerizing carbazole phosphonic acid(PACz).The resultant Poly-2PACz material exhibits exceptional conductivity,high tolerance for variations in layer thickness,and improved film uniformity compared with conventional SAMs.Consequently,the OSCs utilizing Poly-2PACz achieve a remarkable power conversion efficiency of 19.1%and a high fill factor of 81.2%,surpassing the devices based on 2PACz.Moreover,Poly-2PACz-based OSCs demonstrate excellent operational stability,retaining over 80%of their initial efficiency after 1,400 h of continuous light exposure in ambient conditions.This work presents a novel strategy for designing hole transporters,paving the way for more efficient and stable OSCs.

Alkaline soils pose an increasing problem for agriculture worldwide,but using stress-tolerant plants as green manure can improve marginal land.Here,we show that the legume Sesbania cannabina is very tolerant to alkaline conditions and,when used as a green manure,substantially improves alkaline soil.To understand genome evolution and the mechanisms of stress tolerance in this allotetraploid legume,we generated the first telomere-to-telomere genome assembly of S.cannabina spanning~2,087 Mb.The assembly included all centromeric regions,which contain centromeric satellite repeats,and complete chromosome ends with telomeric characteristics.Further genome analysis distinguished A and B subgenomes,which diverged approximately 7.9 million years ago.Comparative genomic analysis revealed that the chromosome homoeologs underwent large-scale inversion events(>10 Mb)and a significant,transposon-driven size expansion of the chromosome 5A homoeolog.We further identified four specific alkali-induced phosphate transporter genes in S.cannabina;these may function in alkali tolerance by relieving the deficiency in available phosphorus in alkaline soil.Our work highlights the significance of S.cannabina as a green tool to improve marginal lands and sheds light on subgenome evolution and adaptation to alkaline soils.

Semantic separator learning and its applications in unsupervised Chinese text parsing

Grammar learning has been a bottleneck problem for a long time. In this paper, we propose a method of seman- tic separator learning, a special case of grammar learning. The method is based on the hypothesis that some classes of words, called semantic separators, split a sentence into sev- eral constituents. The semantic separators are represented by words together with their part-of-speech tags and other infor- mation so that rich semantic information can be involved. In the method, we first identify the semantic separators with the help of noun phrase boundaries, called subseparators. Next, the argument classes of the separators are learned from cor- pus by generalizing argument instances in a hypernym space. Finally, in order to evaluate the learned semantic separators, we use them in unsupervised Chinese text parsing. The exper- iments on a manually labeled test set show that the proposed method outperforms previous methods of unsupervised text parsing.

A numerical analysis of the shear behavior of granular soil with fines

Shear behavior of granular soil with fines is investigated using the discrete element method （DEM） and particle arrangements and inter-particle contacts during shear are examined. The DEM simulation reveals that fine particles play a vital role in the overall response of granular soil to shearing. The occurrence of liquefaction and temporary reduction of strength is ascribed mainly to the loss of support from the fine particle contacts （S-S） and fine particle-to-large particle contacts （S-L） as a consequence of the removal of fine particles from the load-carrying skeleton. The dilative strain-hardening response following the strain-softening response is associated with the migration of fine particles back into the load-carrying skeleton, which is thought to enhance the stiffness of the soil skeleton. During shear, the unit normal vector of the large particle-to-large particle （L-L） contact has the strongest fabric anisotropy, and the S-S contact unit normal vector possesses the weakest anisotropy, suggesting that the large particles play a dominant role in carrying the shear load. It is also found that, during shear, fine particles are prone to rolling at contacts while the large particles are prone to sliding, mainly at the S-L and L-L contacts.

Improving constraint on Ωfrom SDSS using marked correlation functions

Large-scale structure （LSS） surveys will increasingly provide stringent constraints on our cosmological models.Recently,the density-marked correlation function （MCF） has been introduced,offering an easily computable density-correlation statistic.Simulations have demonstrated that MCFs offer additional,independent constraints on cosmological models beyond the standard two-point correlation （2PCF）.In this study,we apply MCFs for the first time to SDSS CMASS data,aiming to investigate the statistical information regarding clustering and anisotropy properties in the Universe and assess the performance of various weighting schemes in MCFs,and finally obtain constraints on Ωm.Upon analyzing the CMASS data,we observe that,by combining different weights （α=[-0.2,0,0.2,0.6]）,the MCFs provide a tight and independent constraint on the cosmological parameter Ωm,yielding Ωm=0.293±0.006 at the 1σlevel,which represents a significant reduction in the statistical error by a factor of 3.4compared to that from 2PCF.Our constraint is consistent with recent findings from the small-scale clustering of BOSS galaxies(Zhai et al.Astronphys.J.948,99 （2023）) within the 1σlevel.However,we also find that our estimate is lower than the Planck measurements by about 2.6σ,indicating the potential presence of new physics beyond the standard cosmological model if all the systematics are fully corrected.The method outlined in this study can be extended to other surveys and datasets,allowing for the constraint of other cosmological parameters.Additionally,it serves as a valuable tool for forthcoming emulator analysis on the Chinese Space Station Telescope （CSST）.
