Cardiac fibroblasts(CFs)are the primary cells tasked with depositing and remodeling collagen and significantly associated with heart failure(HF).TEAD1 has been shown to be essential for heart development and homeostasis.However,fibroblast endogenous TEAD1 in cardiac remodeling remains incompletely understood.Transcriptomic analyses revealed consistently upregulated cardiac TEAD1 expression in mice 4 weeks after transverse aortic constriction(TAC)and Ang-l infusion.Further investigation revealed that CFs were the primary cell type expressing elevated TEAD1 levels in response to pressure overload.Conditional TEAD1 knockout was achieved by crossing TEAD1-floxed mice with CFs-and myofibroblasts-specific Cre mice.Echocardiographic and histological analyses demonstrated that CFs-and myofibroblasts-specific TEAD1 deficiency and treatment with TEAD1 inhibitor,VT103,ameliorated TAC-induced cardiac remodeling.Mechanistically,RNA-seq and ChiP-seq analysis identified Wnt4 as a novel TEAD1 target.TEAD1 has been shown to promote the fibroblast-to-myofibroblast transition through the Wnt signalling pathway,and genetic Wnt4 knockdown inhibited the pro-transformation phenotype in CFs with TEAD1 overexpression.Furthermore,coimmunoprecipitation combined with mass spectrometry,chromatin immunoprecipitation,and luciferase assays demonstrated interaction between TEAD1 and BET protein BRD4,leading to the binding and activation of the Wnt4 promoter.In conclusion,TEAD1 is an essential regulator of the pro-fibrotic CFs phenotype associated with pathological cardiac remodeling via the BRD4/Wnt4 signallingpathway.

The Investigator 24plex QS Kit(QIAGEN,Hilden,Germany)is a 6-dye fluorescent chemistry short tandem repeat(STR)polymerase chain reaction(PCR)amplification system that simultaneously amplifies 20 of the expanded Combined DNA Index System(CODIS)core STR loci,SE33,DYS391,and the standard sex-determining locus,amelogenin,as well as two special internal performance quality sensor controls(QS1 and QS2),which are included in the primer mix to check the PCR performance.This study was designed to be a pilot evaluation of this STR-PCR kit in a Chinese Han population regarding the PCR conditions,sensitivity,precision,accuracy,repeatability,reproducibility,and concordance;tolerance to PCR inhibitors;applicability to real“forensic-type”samples;species specificity;mixture,balance and stutter analyses,and utility in a population investigation.The exhaustive validation studies demonstrated that the Investigator 24plex QS system is accurate,sensitive and robust for STR genotyping.In addition,these genetic markers in the population data in our study indicated that they can also be useful for forensic identification and paternity testing in the Chinese Han population.

Molecular level understanding of CO_(2) capture in ionic liquid/polyimide composite membrane

Ionic liquid(IL)/polyimide(PI)composite membranes demonstrate promise for use in CO_(2)separation applications.However,few studies have focused on the microscopic mechanism of CO_(2)in these composite systems,which is important information for designing new membranes.In this work,a series of systems of CO_(2)in 1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide composited with 4,4-(hexafluoroisopropylidene)diphthalic anhydride(6FDA)-based PI,6FDA-2,3,5,6-tetramethyl-1,4-phenylene-diamine,at different IL concentrations were investigated by all-atom molecular dynamics simulation.The formation of IL regions in PI was found,and the IL regions gradually became continuous channels with increasing IL concentrations.The analysis of the radial distribution functions and hydrogen bond numbers demonstrated that PI had a stronger interaction with cations than anions.However,the hydrogen bonds among PI chains were destroyed by the addition of IL,which was favorable for transporting CO_(2).Furthermore,the self-diffusion coefficient and free energy barrier suggested that the diffusion coefficient of CO_(2)decreased with increasing IL concentrations up to 35 wt-%due to the decrease of the fractional free volume of the composite membrane.However,the CO_(2)selfdiffusion coefficients increased when the IL contents were higher than 35 wt-%,which was attributed to the formation of continuous IL domain that benefitted the transportation of CO_(2).

Research and Exploration of Ideological and Political Education in the Course of Pathophysiology

Course based ideological and political education (CIPE) is an important way to improve the quality of ideological and political work and talent cultivation. This study explores for the first time the implementation of ideological and political education in the teaching of pathophysiology courses, and also analyzes the evaluation of student psychological status and the importance of mental health education in the implementation of IPE courses. A survey was conducted on 211 students at Yangtze University to understand their motivation and behavior towards learning ideological, political, and pathophysiological courses. In addition, a questionnaire survey was used to explore the relationship between pathophysiology and ideological and political courses, as well as the importance of their satisfaction with the implementation of ideological and political courses in pathophysiology and mental health education. The research results indicate that factors such as educational background and gender differences affect the learning of CIPE. Graduate students are more interested in ideological and political courses, while female students find it difficult to study pathophysiology;In addition, the results of one-way ANOVA indicate that the implementation effect of IPE in pathophysiology courses depends on the level of interest in IPE and pathophysiology courses, the level of consideration for the importance of professional courses, the professional gains after studying pathophysiology, and the level of understanding of the relationship between IPE and CIPE. 81.04% of students believe that in the CIPE process, telling stories by teachers themselves is the most popular way of communication and education. This reflects the importance of mental health education from the perspective of CIPE. In addition, this study also indicates that PBL and flipped classroom teaching models are popular teaching models in CIPE. Effects of sgRNA length and number on gene editing efficiency and predicted mutations generated in rice

CRISPR-Cas9 is a common tool for gene editing, and appropriate sg RNAs are the key factor for successful editing. In this study, the effect of sg RNA length and number on editing efficiency was analyzed in rice using CYP81 A6 as the target gene. A series of CRISPR-Cas9 plant expression vectors containing single sg RNAs with different lengths(17, 18, 19, 20, 21, 22, 23 nt) or two sg RNAs were constructed and introduced into rice cultivar Zhonghua11 by Agrobacterium-mediated transformation. Analysis of the editing status of 1283 transgenic rice plants showed that 371 were successfully edited with base preference.Single A or T insertions were the most frequent among the six edited types. The editing efficiency of transgenic rice with two sg RNAs was higher than that with a single sg RNA. Editing efficiency and sg RNA length showed a normal distribution with 20 nt sg RNA(25%) being the most efficient. The editing efficiency decreased slightly with decreases of 1–2 bases(19 nt 20%, 18 nt 21%), but decreased significantly with a decrease of 3 bases(17 nt 4.5%). Editing efficiency was significantly reduced by adding 1 to 3 bases(21 nt 16.8%, 22 nt 13%, 23 nt 13%) to the sg RNA. These results provide data for successful gene editing or rice by CRISPR-Cas9.

Virus-induced gene silencing of SlPYL4 decreases the drought tolerance of tomato

The abscisic acid(ABA)receptor PYR/PYL/RCAR,located upstream of the ABA signaling pathway,recognizes ABA signals and initiates the primary process of signal transduction.In our previous study,we found that the PYL4 gene in tomato plants was significantly upregulated after exogenous treatment with the hormone ABA.The present study used virus-induced gene silencing(VIGS)to further elucidate the role of the PYL4 gene in response to drought in tomato(Solanum lycopersicum)plants.Under the drought treatment,the SlPYL4-silenced plants wilted faster than the control plants did.Physiological indicators showed that the total reactive oxygen species(ROS)content and superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT)activities of the SlPYL4-silenced plants were lower than those of the control plants after 3 and 6 h of drought stress treatment.After 3 h of drought treatment,hydrogen peroxide(H_(2)O_(2)) and superoxide anion(O_(2)^(−).)accumulatedmuch more in the leaves of the SlPYL4-silenced plants than in the leaves of the control plants.These results indicated that PYL4 plays a positive role in drought tolerance in tomato plants.Our results also suggest that SlPYL4 is located upstream of the ABA pathway to regulate the expression of PP2C,SnRK,and AREB/ABF genes.

提出了玄武岩筋回收轮胎钢纤维混凝土(BRC)短柱新型材料构件,为研究BRC短柱偏压性能,制作6根混凝土短柱,并结合非接触式全场应变位移测量技术-数字图像(3D-DIC)技术进行试验测量,研究了回收轮胎钢纤维(RTSF)体积掺量和偏心距对短柱偏压性能和破坏模式的影响.研究结果表明:BRC短柱受压破坏为混凝土压碎、玄武岩纤维增强复合材料(BFRP)筋压断破坏,RTSF提高了混凝土短柱的受压性能,对限制试验柱裂缝和变形的发展具有显著作用,并能有效提高其开裂荷载、极限荷载、混凝土极限压应变和抗压强度.在相同偏心距下,与玄武岩筋普通混凝土短柱相比,BRC混凝土短柱的开裂荷载分别提高了20.46%,23.19%和38.36%,极限承载力分别提高了15.39%,21.55%和30.69%.由于现有的计算模型不足以准确计算BRC短柱受压承载力,提出了考虑RTSF混凝土抗拉强度的承载力计算模型,BRC短柱试验值与预测值符合较好.

YES proto-oncogene 1(YES1)is an SRC family kinase(SFK)that plays a key role in cancer cell proliferation,adhesion,invasion,survival,and angiogenesis during tumorigenesis and tumor development.Reports suggest that YES1 amplification is associated with resistance to chemotherapeutic drugs and tyrosine kinase inhibitors(TKIs)in human malignancies.However,the mechanisms of drug resistance have not been fully elucidated.In this article,we review the literature on YES1 and discuss the implications of YES1 signaling for targeted therapy and chemotherapy resistance in malignancies.Moreover,recent advances in targeted therapy for YES1-amplified malignancies are summarized.Finally,we conclude that targeting YES1 may reverse drug resistance and serve as a valuable tumor treatment strategy.

Radiomics is widely used in adult tumors but has been rarely applied to the field of pediatrics.Promoting the application of radiomics in pediatric diseases,especially in the early diagnosis and stratified treatment of tumors,is of great value to the realization of the WHO 2030“Global Initiative for Childhood Cancer.”This paper discusses the general characteristics of radiomics,the particularity of its application to pediatric diseases,and the current status and prospects of pediatric radiomics.Radiomics is a data-driven science,and the combination of medicine and engineering plays a decisive role in improving data quality,data diversity,and sample size.Compared with adult radiomics,pediatric radiomics is significantly different in data type,disease spectrum,disease staging,and progression.Some progress has been made in the identification,classification,stratifica-tion,survival prediction,and prognosis of tumor diseases.In the future,big data applications from multiple centers and cross-talent training should be strengthened to improve the benefits for clinical workers and children.

Multi-locus identification of Psilocybe cubensis by high-resolution melting(HRM)

Hallucinogenic mushroom is a kind of toxic strain containing psychoactive tryptamine substances such as psilocybin,psilocin and ibotenic acid,etc.The mushrooms containing hallucinogenic components are various,widely distributed and lack of standard to define,which made a great challenge to identification.Traditional identification methods,such as morphology and toxicology analysis,showed shortcomings in old or processed samples,while the DNA-based identification of hallucinogenic mushrooms would allow to identify these samples due to the stability of DNA.In this paper,four primer sets are designed to target Psilocybe cubensis DNA for increasing resolution of present identification method,and the target markers include largest subunit of RNA polymerase II(marked as PC-R1),psilocybin-related phosphotransferase gene(marked as PC-PT),glyceraldehyde 3-phosphate dehydrogenase(marked as PC-3)and translation EF1α(marked as PC-EF).Real-time PCR with high-resolution melting(HRM)assay were used for the differentiation of the fragments amplified by these primer sets,which were tested for specificity,reproducibility,sensitivity,mixture analysis and multiplex PCR.It was shown that the melting temperatures of PC-R1,PC-PT,PC-3 and PC-EF of P.cubensis were(87.93±0.12)℃,(82.21±0.14)℃,(79.72±0.12)℃ and(80.11±0.19)℃ in our kinds of independent experiments.Significant HRM characteristic can be shown with a low concentration of 62.5pg/µL DNA sample,and P.cubensis could be detected in mixtures with Homo sapiens or Cannabis sativa.In summary,the method of HRM analysis can quickly and specifically distinguish P.cubensis from other species,which could be utilized for forensic science,medical diagnosis and drug trafficking cases.
