When the loop-mediated isothermal amplification(LAMP)assay is used for detecting target genes,DNA extraction is unnecessary in many *** pretreatment(***)is enough to obtain rather sensitive *** test samples without any pretreatment can be used as *** feature suggests that LAMP is superior to PCR in developing point-of-care test *** this study,using Stx1 gene from *** as model,we verified that viable cells,dead cells and extracellular DNA could function as template in the LAMP *** the incubation at 63℃,viable bacteria in the LAMP reaction mixture lysed completely within 2 min,providing DNA template for nucleic acid *** Stx1 gene in diluted culture medium,spiked tap water,spiked seawater and real seawater all could be detected,with or without the step of DNA *** found that the complex substances in real sample(*** seawater)exhibited considerable inhibitory effect on the sensitivity of the LAMP *** outcomes are meaningful for building a point-of-care strategy by employing the LAMP assay for environmental monitoring,bio-resource surveys,food safety,*** particular those based on environmental DNA.

Laparoscopic skills training has always been crucial for novice surgeons. Readily accessible equipment, aswell as structured training curriculum should be provided to guarantee adequate practice hours and skillproficiency. Dry-lab training is typically adopted before animal model surgery, usually comprising ofpurpose-built bulky simulators that is neither accessible nor portable. In this technical note, we designed ahome-made simulator, using two 4 L water jugs as operating space that are communicated inside, plus anobservation hole taped in between to mimic the triangular working space of laparoscopic surgery. Imagingwas achieved via smartphone camera, which was wirelessly connected to a laptop and a projector for realtime display on multiple screens, using built-in multi-screen collaboration software. A self-regulated andproficiency-based training curriculum was adopted. This dry-lab simulator is low-cost, highly portable andeasily replicable for basic laparoscopic skills training for the beginners to intermediate surgeons, whichmay serve as a good way for the standardized residency and specialist training program.

Aging is associated with a progressive decline in physiological capacities and an increased risk of aging-associated *** increasing body of experimental evidence shows that aging is a complex biological process coordinately regulated by multiple factors at diferent molecular ***,it is difcult to delineate the overall systematic aging changes based on single-layer ***,multimodal omics approaches,in which data are acquired and analyzed using complementary omics technologies,such as genomics,transcriptomics,and epigenomics,are needed for gaining insights into the precise molecular regulatory mechanisms that trigger *** recent years,multimodal omics sequencing technologies that can reveal complex regulatory networks and specifc phenotypic changes have been developed and widely applied to decode aging and age-related *** review summarizes the classifcation and progress of multimodal omics approaches,as well as the rapidly growing number of articles reporting on their application in the feld of aging research,and outlines new developments in the clinical treatment of age-related diseases based on omics technologies.

Apples are popular fruits worldwide and rich in phenolic compounds that can alleviate obesity and related metabolic diseases. However， the mechanisms underlying the anti-obesity actions of apple polyphenols (AP) like phlorizin (PZ) and procyanidin B2 (PB2) on transplanted obese patient fecal microbiota (TOPFM)-induced obesity and related syndromes have not yet been fully examined in vivo. Herein， a commercial AP product， PZ compound or PB2 compound was used to ameliorate TOPFM-induced obesity in mice. The results indicated that the AP， PZ or PB2 supplementation markedly alleviate TOPFM-induced obesity in mice through effectively suppressing body weight gain and fat accumulation， alleviating insulin resistance and liver inflammation， regulating gut microecology and lipid synthesis/metabolism， and improving gut barrier function and antioxidant capacity. The gut barrier function and integrity were improved through regulating the expression of intestinal pro-inflammatory cytokines， TNF-α， IL-1β and IL-6， and gut barrier function-related genes， ZO-1 and Occludin， and raising the GLP-2 level via increasing the contents of short-chain fatty acids (SCFAs). Interestingly， the AP， PZ or PB2 supplementation could significantly improve the production of SCFAs and restore the microbial community structure and diversity in mice with TOPFM-induced obesity， in particular， increased the abundance of Lachnospiraceae and Bifidobacteriaceaca possibly by inhibiting Blautia and Bifidobacterium phages. The influences of AP， PZ or PB2 on gut microorganisms and phases of the mice upon TOPFM were species-specific. This study was the first report on the ability of an AP， PZ or PB2 supplementation to promote the production of SCFAs by modulating gut microbiota possibly via regulating gut phages.

Plasticity changes of uninjured nerves can result in a novel neural circuit after spinal cord injury, which can restore sensory and motor functions to different degrees. Although processes of neural plasticity have been studied, the mechanism and treatment to effectively improve neural plasticity changes remain controversial. The present study reviewed studies regarding plasticity of the central nervous system and methods for promoting plasticity to improve repair of injured central nerves. The results showed that synaptic reorganization, axonal sprouting, and neurogenesis are critical factors for neural circuit reconstruction. Directed functional exercise, neurotrophic factor and transplantation of nerve-derived and non-nerve-derived tissues and cells can effectively ameliorate functional disturbances caused by spinal cord injury and improve quality of life for patients.

Developing nonflammable electrolyte with a wide electrochemical window has become an urgent demand for high-energy-density and high-safe lithium-ion batteries(LIBs).Herein,a fluorinated nonflammable phosphate electrolyte is developed to construct a safe 4.5 V-class LIB(Si-SiC-C/0.35Li2MnO3-0.65LiNi0.5Mn0.5O2).The proposed fluorinated phosphate electrolyte,0.8 M LiPF6/tris(2,2,2-trifluoroethyl)phosphate(TFEP)+5 vol%fluoroethylene carbonate(FEC)+5 vol%vinylene carbonate(VC),is not only completely nonflammable but also exhibits excellent oxidative/reductive stability on 0.35Li2MnO30.65LiNi0.5Mn0.5O2 cathode and Si-SiC-C *** in situ differential electrochemical mass spectrometry and X-ray photoelectron spectroscopy proved that TFEP-based electrolyte does not decompose into gases but forms a high-quality electrode-electrolyte interface on cathode surface at high working *** 4.5 V-class LIBs using 0.8 M LiPF6 TFEP-based nonflammable electrolyte shed some light on potential application for high-safe and low-cost larger-scale energy storage.

Objective: This study aims to explore the correlation between human papillomavirus (HPV) and Mycoplasma genitalium (CT) combined with TCT detection in cervical cancer screening. Method: A cross-sectional study design was adopted, and a total of 609 women who came to seek medical treatment were recruited as the study subjects. Combination testing was evaluated on cervical cancer screening by testing the women for HPV, CT with TCT detection and analyzing the relationship of cervical lesions with HPV and CT infection. Results: The study results showed that 21.57% of the subjects were infected with both HPV and CT, and 48.42% of the cases had abnormal TCT results at the same time. Further data analysis showed that HPV infection was significantly associated with abnormal TCT outcomes (p < 0.05), suggesting a possible synergistic effect of the two infections in cervical lesions. The combined sensitivity and specificity of HPV, CT and TCT detection were 21.57% and 48.42%, respectively, which were significantly higher than that of single detection. Conclusion: In summary, the results of this study support the importance of combined HPV, CT, and TCT testing in cervical cancer screening, and propose the hypothesis that combined testing may improve screening effectiveness. However, further large sample studies are needed to confirm this conclusion and explore the prospects of combined testing in clinical practice.

In recent years, with the continuous development of pathogen detection methods, the diagnosis and treatment of acute respiratory infections has received more and more attention from clinicians. The clinical diagnosis and treatment of acute respiratory infections in children and the research of laboratory detection methods have also been continuously developed. The manuscript presents a review of progress in the clinical diagnosis, treatment and laboratory testing of acute respiratory infections in children by collecting references.
