Hepatocyte nuclear factor 4α in the pathogenesis of non-alcoholic fatty liver disease

Non-alcoholic fatty liver disease (NAFLD) is emerging as the most common chronic liver disease worldwide. It refers to a range of liver conditions affecting people who drink little or no alcohol. NAFLD comprises non-alcoholic fatty liver and non-alcoholic steatohepatitis (NASH), the more aggressive form of NAFLD. NASH is featured by steatosis, lobular inflammation, hepatocyte injury, and various degrees of fibrosis. Although much progress has been made over the past decades, the pathogenic mechanism of NAFLD remains to be fully elucidated. Hepatocyte nuclear factor 4α (HNF4α) is a nuclear hormone receptor that is highly expressed in hepatocytes. Hepatic HNF4α expression is markedly reduced in NAFLD patients and mouse models of NASH. HNF4α has been shown to regulate bile acid, lipid, glucose, and drug metabolism. In this review, we summarize the recent advances in the understanding of the pathogenesis of NAFLD with a focus on the regulation of HNF4α and the role of hepatic HNF4α in NAFLD. Several lines of evidence have shown that hepatic HNF4α plays a key role in the initiation and progression of NAFLD. Recent data suggest that hepatic HNF4α may be a promising target for treatment of NAFLD.

There is high expectation for significant improvements in cancer patient care after completion of the human genome project in 2003. Through pains-taking analyses of genomic profiles in cancer patients, a number of targetable gene alterations have been discovered, with some leading to novel therapies, such as activating mutations of EGFR, BRAF and ALK gene fusions. As a result, clinical management of cancer through targeted therapy has finally become a reality for a subset of cancers, such as lung adenocarcinomas and melanomas. In this review, we summarize how gene mutation discovery leads to new treatment strategies using non-small cell lung cancer （NSCLC） as an example. We also discuss possible future implications of cancer genome analyses.

Atomically dispersed metals on N-doped carbon supports(M-N_(xCs)) have great potential applications in various fields.However,a precise understanding of the definitive relationship between the configuration of metal single atoms and the dielectric loss properties of M-N_(xCs) at the atomic-level is still lacking.Herein,we report a general approach to synthesize a series of three-dimensional(3D)honeycomb-like M-N_xC(M=Mn,Fe,Co,Cu,or Ni) containing metal single atoms.Experimental results indicate that 3D M-N_(xCs) exhibit a greatly enhanced dielectric loss compared with that of the NC matrix.Theoretical calculations demonstrate that the density of states of the d orbitals near the Fermi level is significantly increased and additional electrical dipoles are induced due to the destruction of the symmetry of the local microstructure,which enhances conductive loss and dipolar polarization loss of 3D M-N_(xCs),respectively.Consequently,these 3D M-N_(xCs) exhibit excellent electromagnetic wave absorption properties,outperforming the most commonly reported absorbers.This study systematically explains the mechanism of dielectric loss at the atomic level for the first time and is of significance to the rational design of high-efficiency electromagnetic wave absorbing materials containing metal single atoms.

Copper is a microelement with important physiological functions in the body.However,the excess copper ion(Cu^(2+))may cause severe health problems,such as hair cell apoptosis and the resultant hearing loss.Therefore,the assay of Cu^(2+)is important.We integrate ionic imprinting technology(IIT)and structurally colored hydrogel beads to prepare chitosan-based ionically imprinted hydrogel beads(IIHBs)as a low-cost and high-specificity platform for Cu^(2+)detection.The IIHBs have a macroporous microstructure,uniform size,vivid structural color,and magnetic responsiveness.When incubated in solution,IIHBs recognize Cu^(2+)and exhibit a reflective peak change,thereby achieving label-free detection.In addition,benefiting from the IIT,the IIHBs display good specificity and selectivity and have an imprinting factor of 19.14 at 100μmol·L^(-1).These features indicated that the developed IIHBs are promising candidates for Cu^(2+)detection,particularly for the prevention of hearing loss.

Using first-principles calculations,hydrogen evolution reaction(HER) activity on two-dimensional(2 D) gallium chalcogenides monolayers GaX(X=O,S,Se,and Te) as well as the derived Janus monolayers Ga_(2) XY(X≠Y,X/Y=O,S,Se,and Te) were systematically examined.It was found that Ga_(2) OSe Janus monolayer with a 0.3% strain has the lowest ΔG_(H)*of 0.19 eV(modified to-0.01 eV including the solvation effect) because(ⅰ) O is the most electronegative among X/Y atoms,(ⅱ) the Ga_(2) OSe monolayer has a larger lattice parameter with respect to GaO monolayer,and(ⅲ) the built-in electric field is enhanced after H adsorption.The enhanced H adsorption with the lattice stretching is a result of the weaker Ga-O bond strength before H adsorption and the reduced electron fillings of anti-bonding molecular orbital formed by H 1 s and O2 p_(z) orbitals after H adsorption.The O-p_(z) band center can be served as a descriptor to describe the HER activity trend for these p-block materials.Moreover,Ga_(2) OSe monolayer has appropriate band alignment,distinguished optical absorption coefficient(10~5 cm^(-1)),low exciton binding energy(0.71 eV),and the spontaneous HER process,indicating that it is a highly potential candidate for near-infrared photocatalyst for hydrogen production.Our research provides a novel paradigm that forming Janus structure can effectively tune the HER activity,which would guide the searching for excellent HER photocatalysts for clean hydrogen production.

Zeolitie imidazolate framework-8(ZIF-8),composed of Zn ions and imidazolate ligands,is a class of metal-organic frameworks,which possesses a similar structure as conventional aluminosilicate zeolites.This material exhibits inherent porous property,high loading capacity,and pH-sensitive degradation,as well as excep-tional thermal and chemical stability.Extensive research effort has been devoted 10 relevant research aspects ranging from synthesis methods,property characterization to potential applications of ZIF-8.This review focuses on the recent development of ZIF-8 synthesis methods and its promising appications in drug delivery.The potential risks of using ZIF-8 for drug delivery are also summarized.

Pancreatic cancer,mostly pancreatic ductal adenocarcinoma(PDAC),is often metastatic upon the initial diagnosis,and has a high mortality rate.However,the primary and the matched metastatic cancers often share the same driver gene mutations.1,2 Gene expression comparison between primary and metastatic PDAC tissues revealed high expression of Gli2 and YAP1 in the metastatic PDAC.Gli2 is an important player in non-canonical hedgehog signaling,and YAP1 is an essential cell polarity gene.In this study,we investigated the effects of Gli2 and YAP1 on pancreatic cancer cell function in 3D culture and in mouse models.We further determined the relationship between Gli2 and YAP1.These studies further our understanding of pancreatic cancer metastasis.

Recently, in response to the major challenges in energy development and environmental issues, tremendous efforts are being devoted to developing electrochemical energy storage devices based on green sustainable resources. As a class of green materials, nanocellulose(NC) has received extensive attention. In this review, we summarize the research progress of NC derived materials in electrochemical energy storage. Specifically, we first introduce various synthesis methods based on NC and the pretreatment process to increase the conductivity. Then we focus on the specific application of NC in electrochemical energy storage devices. Finally, we summarize the previously reported work and put forward views on the further development of NC in the field of electrochemical energy storage.
