Anti-allergic Effects of Ginsenosides Extracted by High Temperature and High Pressure Method

Ginseng（Panax ginseng C. A. Mey.） is a traditional medicinal herb in Asia. Studies have shown that ginsenosides significantly affect immnnoregulation and rare ginsenosides have anti-allergic effects. In this research, a high temperature and high pressure method was utilized to increase the contents of rare ginsenosides in the ginseng extract（GE）. The anti-allergic effects of this extract were investigated in vivo. Water was used as the extraction solvent in extracting the rare ginsenosides via the high temperature and high pressure method. Extraction time and temperature were investigated in order to increase the contents of rare ginsenosides. Rare ginsenosides were qualitatively analyzed by HPLC-ESI-MS and quantitatively analyzed by HPLC-UV. Anti-allergic effects of the extracts were assessed using the ovalbumin（OVA）-induced allergic asthma model in vivo. An extraction temperature of 145 ℃ and extraction time of 2.0 h were chosen as the optimal conditions. Compared with traditional method, the contents of total rare ginsenosides extracted were considerably higher using the new method, that is, 14.74 times that extracted by the traditional method. In our in vivo experiments, treatment with high concentration GE may have anti-allergic erects in decreasing the total amount of IgE in serum and IL-4 in bronchoalveolar lavage fluid（BALF）, aud in improving the ratio of CD4^＋ to CD8^＋ T cells. The high temperature and high pressure method was an effective method to obtain GE containing more rare ginsenosides, which maybe become anti-allergic agents.

Influence of heat treatment on microstructure and mechanical properties of Al/Al-Cu graded materials

Five-layered Al/Al-Cu functionally graded material （FGM） was prepared by powder metallurgy technology, and the subsequent heat treatment was carried out for the graded material. The microstructures and distribution of Cu element under pressure sintering （F）, solution treatment （T4） and artificial aging treatment （T6） were investigated, and the Vickers hardness and flexural properties of different states were tested. The results showed that sintered compact with dense structure and compositional continuous change was obtained. The second-phase CuAl 2 was dispersively distributed along grain boundary of Al matrix. After solution treatment at 503 C for 3 h, CuAl 2 phase obviously decreased and dissolved into the Al matrix, and the flexural strength was thereupon enhanced to 228.5 MPa. With the subsequent aging treatment at 150 C for 15 h, the majority of flake shaped precipitates θ phases were uniformly distributed in the matrix. And the distribution of Cu element became gradual continuous compared to sintered compact. Meanwhile, the flexural strength increased further, which accompanied with the decline of plasticity.

Food allergy has become a worldwide problem. To find an effective way to reduce the allergic effect of allergen is an interesting topic. In this work, the interaction between ovalbumin（OVA） and tangeretin was studied by means of spectroscopy, molecular docking calculation and animal experiment. As the results show, static quenching of fluorescence intensity happened due to the interaction between OVA and tangeretin. Binding constants, binding sites, and thermometric parameters were obtained. The conformation changing of OVA occurred due to the interaction between it and tangeretin. The molecular docking calculation results show that the epitopes Arginine 84（ARG84）, Leucine 87（LEU87）, Asparagine 88（ASN88）, Serine 103（SER103）, Arginine 104（ARG104）, Leucine 124（LEUI24）, Arginine 126（ARG126）, Glycine 127（GLY127）, Glycine 128（GLY128） and Tryptohan 148（TRP148） of OVA were occupied by tangeretin after the docking, which means tangeretin may have some inhibition effects. As the results of animal experiments, the amount of IgE and lung tissue sections of female BALB/c mice in different groups show that tangeretin with different concentrations has better effects than dexamethasone. In conclusion, the tangeretin may have the effects to reduce the allergic effects of OVA via occupying the IgE-binding epitopes of OVA.

Microbial growth is an issue of concern that may cause hygienic and aesthetic problems during the transportation and usage of reclaimed water. Assimilable organic carbon （AOC） is an important parameter which determines the heterotrophic bacterial growth potential of water. Pseudomonas fluorescens P17 and Spirillum sp. NOX are widely used to measure AOC in drinking water. The AOC values of various reclaimed water samples determined by P 17 and NOX were compared with those determined by the new strains isolated from reclaimed water in this study. It showed that the conventional test strains were not suitable for AOC measurement of reclaimed water in certain cases. In addition to P17 and NOX, Stenotrophomonas sp. Z J2, Pseudomonas saponi- phila G3 and Enterobacter sp. G6, were selected as test strains for AOC measurement of reclaimed water. Key aspects of the bioassay including inoculum cell density, incubation temperature, incubation time and the pH of samples were evaluated for the newly selected test strains. Higher inoculum density （104 ***-1） and higher incubation temperature （25℃） could reduce the time required for the tests. The AOC results of various collected samples showed the advantages of the method proposed based on those five strains in evaluating the biologic stability of reclaimed water.

The purpose of this study was to give a worldwide overview of the concentrations of typical estrogenic endocrine disrupting chemicals （EDCs） in the effluent of sewage plants and then compare the concentra- tion distribution of the estrogenic EDCs in ten countries based on the survey data of the estrogenic EDCs research. The concentrations of three main categories （totally eight kinds） of estrogenic EDCs including steroidal estrogens （estrone （El）, estradiol （E2）, estriol （E3） and 17a- ethynylestradiol （EE2））, phenolic compounds （nonylphe- nol （NP） and bisphenol A （BPA）） and phthalate esters （dibutyl phthalate （DBP） and dibutyl phthalate （2- ethylhexyl） phthalate （DEHP）） in the effluents of sewage plants reported in major international journals over the past decade were collected. The statistics showed that the concentration distributions of eight kinds of EDCs were in the range of ng·L^-1 to μg·L^-1. The concentrations of steroidal estrogens mainly ranged within 50.00 ng. L-1, and the median concentrations of El, E2, E3 and EE2 were 11.00, 3.68, 4.90 and 1.00 ng·L^-1, respectively. Phenolic compounds and phthalate esters were found at pg. L-1 level （some individual values were at the high level of 40.00 μg·L^-1）. The median concentrations of BPA, NP, DBP and DEHP were 0.06, 0.55, 0.07 and 0.88 μg·L^-1, respectively. The concentrations of phenolic compounds and phthalate esters in the effluents were higher than that of steroids estrogens. The analysis of the concentration in various ten countries showed that steroids estrogens, phenolic compounds and phthalate esters in sewage plant effluents were detected with high concentration in Canada, Spain and China, respectively.

ObjectiveThis review aimed to summarize the role of the Hippo signaling pathway in renal cell carcinoma (RCC), a urologic malignancy with subtle initial symptoms and high mortality rates due to metastatic RCC. The Hippo signaling pathway, which regulates tissue and organ sizes, plays a crucial role in RCC progression and metastasis. Understanding the involvement of the Hippo signaling pathway in RCC provides valuable insights for the development of targeted therapies and improved patient *** this review, we explored the impact of the Hippo signaling pathway on RCC. Through an analysis of existing literature, we examined its role in RCC progression and metastasis. Additionally, we discussed potential therapeutic strategies targeting the Hippo pathway for inhibiting RCC cell growth and invasion. We also highlighted the importance of investigating interactions between the Hippo pathway and other signaling pathways such as Wnt, transforming growth factor-beta, and PI3K/AKT, which may uncover additional therapeutic *** Hippo signaling pathway has shown promise as a target for inhibiting RCC cell growth and invasion. Studies have demonstrated its dysregulation in RCC, with altered expression of key components such as yes-associated protein/transcriptional coactivator with PDZ-binding motif (YAP/TAZ). Targeting the Hippo pathway has been associated with suppressed tumor growth and metastasis in preclinical models of RCC. Furthermore, investigating crosstalk between the Hippo pathway and other signaling pathways has revealed potential synergistic effects that could be exploited for therapeutic *** the role of the Hippo signaling pathway in RCC is of paramount importance.
