Acid phosphatase(ACP)is a ubiquitous phosphatase in living organisms.The abnormal variation of ACP is related to various diseases.Herein,we propose a colorimetric method based on CeO_(2)-modified gold core shell nanoparticles(Au@CeO_(2)NPs)to analyze ACP activity with high sensitivity and specificity.In this design,2-phospho-L-ascorbic acid trisodium salt(AAP)is dephosphorylated by ACP and produces reductive ascorbic acid(AA),which makes the CeO_(2)shell decomposition.A remarkable blue shift of localized surface plasmon resonance peak(LSPR,from yellow to green)along with the scattering intensity ratio changes from individual Au@CeO_(2)NPs are observed.ACP activity can be quantified by calculating the ratio changes of individual Au@CeO_(2)NPs.This assay reveals limit of detection(LOD)of 0.044 mU/mL and the linear range of 0.05–5.0 mU/mL,which are much lower than most of spectroscopic measurements in bulk solution.Furthermore,the recovery measurements in real samples are satisfactory and the capacity for practical application is demonstrated.As a consequence,Au@CeO_(2)NPs used in this assay will find new applications for the ultrasensitive detection of enzyme activity.

Epithelial-mesenchymal transition(EMT)and proliferation play important roles in epithelial cancer formation and progression,but what molecules and how they trigger EMT is largely unknown.Here we performed spatial transcriptomic and functional analyses on samples of multistage esophageal squamous-cell carcinoma(ESCC)from mice and humans to decipher these critical issues.By investigating spatiotemporal gene expression patterns and cell–cell interactions,we demonstrated that the aberrant epithelial cell interaction via EFNB1-EPHB4 triggers EMT and cell cycle mediated by downstream SRC/ERK/AKT signaling.The aberrant epithelial cell interaction occurs within the basal layer at early precancerous lesions,which expands to the whole epithelial layer and strengthens along the cancer development and progression.Functional analysis revealed that the aberrant EFNB1-EPHB4 interaction is caused by overexpressedΔNP63 due to TP53 mutation,the culprit in human ESCC tumorigenesis.Our results shed new light on the role of TP53-TP63/ΔNP63-EFNB1-EPHB4 axis in EMT and cell proliferation in epithelial cancer formation.

Objective:This study aimed to investigate the incidence of enteral nutrition intolerance(ENI)in patients with sepsis and explore potential risk factors.Methods:A case-control study was conducted in patients with sepsis who were receiving enteral nutrition(EN)at a tertiary hospital in China.The included patients were divided into the ENI group and the non-ENI group.Univariate and multivariate analyses were performed to identify the risk factors for ENI.Results:A total of 859 patients were included in the study.Among them,288(33.53%)patients experienced symptoms of ENI,including diarrhea,vomiting,bloating,and gastric retention.Logistic regression analysis revealed that the Acute Physiology and Chronic Health Evaluation H(APACHE H)score,thoracocentesis,and usage of cardiotonic drugs(namely,inotropes)were independent predictors of the ENI.Conclusion:The incidence of ENI is relatively high in patients with sepsis,especially in those who have higher APACHE H scores,have undergone thoracocentesis,and have received inotropes.

The controlling factors of organic-rich shale accumulation is essential for the exploration and development of shale oil and gas resources. The sedimentary environment plays a vital role in the formation of organic-rich sediments in lacustrine facies. This article unravels the mineralogy, geochemistry, and paleoenvironmental evolution during the deposition of the Paleogene Shahejie Formation(Es3L). It discusses the effects of paleoclimate, paleosalinity, paleoredox conditions, paleowater depth, and paleoproductivity on organic matter(OM) enrichment. Finally, the OM enrichment model was established. The results show that the mineralogical compositions are mainly composed of calcite(avg. 40.13%), quartz(avg. 21.64%) and clay minerals(avg. 24.07%), accompanied by dolomite(avg. 7.07%), feldspar(avg. 6.36%)and pyrite(avg. 2.95%). The Es3Lshale has a high abundance of OM, with total organic carbon(TOC)ranging from 1.07% to 5.12%. The organic matter type is mainly composed of type I-II1kerogen, which is generally considered a good-quality source rock. The source of OM is a mixture of lower bacteria, algae,and plants. During the early sedimentary period, the paleoclimate was dry and cold, with high salinity,intense reducibility, and relatively low productivity. During the late sedimentary period, the climate became warmer and more humid. As a result, the salinity decreased to a level that was suitable for biological reproduction, and productivity increased gradually due to the input of terrigenous plants.Paleosalinity and paleoclimate determined the environment of the sedimentary period, in addition,paleoproductivity and paleoredox condition indicated the formation and preservation conditions of OM.The warm and humid climate, brackish water, suitable reduction conditions and high productivity are the favorable conditions for the generation and preservation of organic matter. Non-aromatic fluorescent and multi-responsive materials,exhibiting inherent fluorescence emission and controlled phase change,have garnered significant attention in recent years.However,the underlying interaction between their fluorescent properties and phase transition remains unclear.In this study,we synthesized a series of catalyst-free aza-Michael addition-based polyethyleneimine(RFPEI)materials by reacting polyethyleneimine(PEI)with N-isopropyl acrylamide(NIPAM).The resulting RFPEI was comprehensively characterized,and demonstrated dual-phase transition behavior(LCST and UCST)in water,which could be finely tuned by adjusting its composition or external factors such as pH.Notably,upon UV irradiation(365 nm),RFPEI exhibited strong fluorescence emission.We further investigated the effects of NIPAM grafting percentage to PEI,polymer concentration,and pH on the LCST/UCST and fluorescent properties of RFPEI aqueous solutions.Moreover,we showcased the great potential of RFPEI as a versatile tool for physiological cell imaging,trace detection,and controlled release of doxorubicin.Our study presents a novel class of stimuli-responsive fluorescent materials with promising applications in the field of biomedicine.

Objective This study employs the Geographically and Temporally Weighted Regression(GTWR)model to assess the impact of meteorological elements and imported cases on dengue fever outbreaks,emphasizing the spatial-temporal variability of these factors in border regions.Methods We conducted a descriptive analysis of dengue fever’s temporal-spatial distribution in Yunnan border areas.Utilizing annual data from 2013 to 2019,with each county in the Yunnan border serving as a spatial unit,we constructed a GTWR model to investigate the determinants of dengue fever and their spatio-temporal heterogeneity in this region.Results The GTWR model,proving more effective than Ordinary Least Squares(OLS)analysis,identified significant spatial and temporal heterogeneity in factors influencing dengue fever’s spread along the Yunnan border.Notably,the GTWR model revealed a substantial variation in the relationship between indigenous dengue fever incidence,meteorological variables,and imported cases across different counties.Conclusion In the Yunnan border areas,local dengue incidence is affected by temperature,humidity,precipitation,wind speed,and imported cases,with these factors’influence exhibiting notable spatial and temporal variation.

Brain injuries due to trauma or stroke are major causes of adult death and disability.Unfortunately,few interventions are effective for post-injury repair of brain tissue.After a long debate on whether endogenous neurogenesis actually happens in the adult human brain,there is now substantial evidence to support its occurrence.Although neurogenesis is usually significantly stimulated by injury,the reparative potential of endogenous differentiation from neural stem/progenitor cells is usually insufficient.Alternatively,exogenous stem cell transplantation has shown promising results in animal models,but limitations such as poor long-term survival and inefficient neuronal differentiation make it still challenging for clinical use.Recently,a high focus was placed on glia-to-neuron conversion under single-factor regulation.Despite some inspiring results,the validity of this strategy is still controversial.In this review,we summarize historical findings and recent advances on neurogenesis strategies for neurorepair after brain injury.We also discuss their advantages and drawbacks,as to provide a comprehensive account of their potentials for further studies.

Tumor cell clusters are regarded as critical factors in cancer pathophysiology,and increasing evidence of their higher treatment resistance and metastasis compared to single tumor cells has been obtained.However,existing cell separation methods that are designed for single tumor cells cannot be used to simultaneously purify tumor cell clusters.To address this problem,we demonstrated a microfluidic approach for the high-throughput,continuous-flow ternary separation of single tumor cells,tumor cell clusters,and WBCs from clinical pleural or abdominal effusions by coupling slanted spiral channels and periodic contraction-expansion arrays.We first systematically explored the influence of particle size and flow rate on particle focusing.The separation performance indicated that 94.0%of WBCs were removed and more than 97%of MDA-MB-231 tumor cells were recovered at a high flow rate of 3500µL/min.Moreover,more than 90%of tumor cell clusters were effectively preserved after separation.Finally,we successfully applied our device for the ternary separation of single tumor cells,tumor cell clusters,and WBCs from different malignant effusions collected from patients with metastatic cancer.Thus,our spiral-contraction-expansion device has potential as a sample pretreatment tool for the cytological diagnosis of malignant effusions.
