Objective To evaluate levels of lead(Pb) and cadmium(Cd) in the breast milk in the second postpartum month,to investigate the relationship between Pb/Cd levels in breast milk and some sociodemographic parameters,and to explore whether these levels affect the infants' physical status or the mothers' psychological status(postpartum depression).Methods A cross-sectional study was conducted between November 2009 and December *** 170 healthy mothers were enrolled from Nanjing Maternity and Child Health Care *** inclusion criteria were:voluntary to participate in this study,healthy,with no chronic disease,breastfeeding in the second postpartum month,living in a suburban but not non-industrial area of Nanjing,and not occupationally exposed to toxic *** the mothers completed a questionnaire and were evaluated based on the Edinburgh Postpartum Depression Scale(EPDS) to identify the risk of postpartum *** and Cd levels in breast milk were determined by inductively coupled plasma mass *** infants of these mothers were examined for their z scores of weight for age,length for age,head circumference for age,and body mass index for *** The median breast milk levels of Pb and Cd were 40.6 μg/L and 0.67 μg/L,*** 164(96.5%) of the 170 samples,Pb levels were higher than the limit reported by the World Health Organization(> 5 μg/L).Breast milk Cd level was > 1 μg/L in 54(31.8%) *** mothers with a history of anemia had a higher breast milk Pb level than those without a history of anemia(41.1 μg/L vs.37.9 μg/L,P = 0.050).The median breast milk Cd level in those who were active and passive smokers during pregnancy was significantly higher than that in non-smokers(0.88 μg/L vs.0.00 μg/L,P = 0.025).The breast milk Cd level in the mothers not taking iron and vitamin supplements for 2 months postpartum was higher than in those taking the supplements(iron supplement:0.74 μg/L vs.0.00 μg/L,P = 0.025;vitamin supplemen

Background Tumor hypoxia, one of the features of solid tumors, is associated with chemo-resistance. Recently, nuclear factor-KB （NF-κB） was found to be activated during hypoxia. However, the impact of NF-κB activation on chemo-resistance during hypoxia remains unknown. Methods Human lung adenocarcinoma A549 cells were transfected with NF-κB p65siRNA and treated with cobalt chloride （COCI2） to mimic hypoxia in the presence or absence of cisplatin. NF-KB expression was measured by Western blotting, immune-fluorescence and real-time PCR. Hypoxia-inducible factor-1α （HIF-1α） and Bcl-2 expression were determined by Western blotting. Cell apoptosis and survival with half-maximum inhibitory concentration （IC50） of cisplatin were determined by Annexin V-FITC/PI and 3-（4, 5-dimethylthiazol-2-yl）-2, 5-diphenyltetrazolium bromide （MTT）, respectively. Results Exposure ofA549 cells to COCl2 increased nuclear HIF-1α protein expression, and enhanced NF-KB p65 protein nuclear accumulation （the mark of NF-κB activation） in a time and dose dependant manner. COCI2 did not promote apoptosis in A549 cells; on the contrary, it reduced cisplatin-induced apoptosis and increased the IC50 of cisplatin. However, when we inhibited CoCI2-induced activation of NF-κB through NF-κB p65siRNA, cisplatin-induced apoptosis was increased and IC50 of cisplatin was reduced to levels similar to those in control cells. Meanwhile, CoCI2-induced Bcl-2 over- expression was down-regulated in the presence of cisplatin when NF-κB activity was inhibited. Conclusion Up-regulating Bcl-2 might be involved in NF-κB activation induced resistance to cisplatin in A549 cells under CoCl2-induced chemical hypoxia.

Objective: EphB4 is a member of the largest known family of receptor protein tyrosine kinases, which plays an important role in a variety of processes during embryonic development including pattern formation, cell aggregation and migration, segmentation, neural development, angiogenesis, and vascular network assembly. EphB4 selectively binds Ephrin-B2 and not other Ephrin B ligands, interaction between EphB4 and Ephrin-B2 can provoke bidirectional signaling,then can regulates cell proliferation. We focused on EphB4 for its potential role in tumor biology because of its location on chromosome 7q22, a locus frequently amplified in cancers including prostate cancer, and because of its expression in various cancers, However, little is known about the role of EphB4 in prostate cancer development and *** the study was mainly investigate the expression of EphB4 in human prostate cancer and their relationship with tumor lymphangiogenesis and metastasis. Methods: Expression of LYVE-1,EphB4 receptor was examined with immunohistochemistry in tumor specimens from 40 prostate cancer cases and 10 benign prostatic hyperplasia (BPH) *** vessel density (MLD) was counted according to the result of LYVE-1 staining. Results: Using immunohistochemical techniques ,we showed that the EphB4 receptor was significantly higher in prostate cancer Compared with benign prostatic hyperplasia ( BPH ) ,and EphB4 receptor was produced significantly in increased amounts in human prostate cancer of greater Gleason score compared with lower Gleason score tumor group. there also appeared to be a trend towards increased expression of EphB4 receptor with higher grade disease. Compared with EphB4 negative group, the MLD was significantly greater in EphB4 positive group. Conclusion:The expression of EphB4 receptor might contribute to tumor lymphangiogenesis and tumor invasion as well as metastasis in prostate cancer. Single-cell omics represent a groundbreaking advancement in the biomedical field,offering profound insights into the understanding of complex diseases, including cancers. Here, we comprehensively summarize recent advances in single-cell omics technologies, with a specific focus on the methodology section. This overview aims to guide researchers in selecting appropriate methods for single-cell sequencing and related data analysis.

Objective: The Hedgehog signaling pathway is important in vertebrate development and tumorigenesis, but the molecular mechanism is still remain largely unknown. Aim of the study is mainly investigate the effect of cyclopamine on growth and proliferation of human prostate cancer cells PC3. Methods: The PC3 cells were incubated with cyclopamine, which is the specific inhibitor of Hedghog signaling pathway, then the inverted microscope was uesed to observe the morphologic changes of the cells, MTT assay and flow cytometer (FCM) were used to examine the influence of cyclopamine on PC3 cells growth and DNA synthesis. Results: Compared with the control group, after the cells were incubated with cyclopamine, obvious morphologic changes of PC3 cells were observed;the growth of PC3 cells was inhibited in a concentration and time dependent manner;the DNA synthesis of PC3 cells was obviously inhibited;the number of PC3 cells in S phase decreased and the cells were blocked in G2 phase. Conclusion: Cyclopamine can effectually inhibit the growth and proliferation of PC3 cells, which indicated that the growth and survival of human prostate cancer cells PC3 rely upon sustained signaling from the Hedgehog pathway and Hedgehog signaling pathway may be inappropriately activated in prostate cancer. Inhibit Hedgehog signaling pathway can be a novel rational strategy for therapeutic treatment of prostate tumors, including metastatic tumors.

This study presents a novel method to fabricate metal-decorated,sulfur-doped layered double hydroxides （M/SLDH） through spontaneous redox and sulfurization *** developed Ag/SLDH and Pt/SLDH catalysts with abundant heterogeneous interfaces and hierarchical nanostructures demonstrated outstanding oxygen evolution reaction （OER） and hydrogen evolution reaction （HER） performance,achieving low overpotentials of 212 and 35 mV at 10 mA cm-2in 1 M KOH,*** both anode and cathode in water splitting,they required only 1.47 V to reach 10 mA cm-2and exhibited high structural robustness,maintaining stability at 1000 mA cm-2for 300 ***-situ Raman analysis revealed that the synergistic effects of metal nanoparticles and S doping significantly promote the transformation into the S-Co1-xFexOOH layer,which serves as the active phase for water ***,ultraviolet photoelectron spectroscopy （UPS） and density functional theory （DFT） analyses indicated that incorporating metal nanoparticles and S doping increase electron density near the Fermi level and reduce reaction energy barriers,thus enhancing intrinsic OER and HER *** study provides a scalable strategy for synthesizing high-performance electrocatalysts for water splitting,with promising potential for broader applications.

Several studies have shown that activation of unfolded protein response and endoplasmic reticulum(ER)stress plays a crucial role in severe cerebral ischemia/reperfusion *** occurs within hours after cerebral ischemia,but the relationship between ER stress and autophagy remains *** this study,we established experimental models using oxygen-glucose deprivation/reoxygenation in PC12 cells and primary neurons to simulate cerebral ischemia/reperfusion *** found that prolongation of oxygen-glucose deprivation activated the ER stress pathway protein kinase-like endoplasmic reticulum kinase(PERK)/eukaryotic translation initiation factor 2 subunit alpha(e IF2α)-activating transcription factor 4(ATF4)-C/EBP homologous protein(CHOP),increased neuronal apoptosis,and induced ***,inhibition of ER stress using inhibitors or by si RNA knockdown of the PERK gene significantly attenuated excessive autophagy and neuronal apoptosis,indicating an interaction between autophagy and ER stress and suggesting PERK as an essential target for regulating *** autophagy with chloroquine exacerbated ER stress-induced apoptosis,indicating that normal levels of autophagy play a protective role in neuronal injury following cerebral ischemia/reperfusion *** from this study indicate that cerebral ischemia/reperfusion injury can trigger neuronal ER stress and promote autophagy,and suggest that PERK is a possible target for inhibiting excessive autophagy in cerebral ischemia/reperfusion injury.

Peripheral nerve injury is a common neurological condition that often leads to severe functional limitations and *** on the pathogenesis of peripheral nerve injury has focused on pathological changes at individual injury sites,neglecting multilevel pathological analysis of the overall nervous system and target *** has led to restrictions on current therapeutic *** this paper,we first summarize the potential mechanisms of peripheral nerve injury from a holistic perspective,covering the central nervous system,peripheral nervous system,and target *** peripheral nerve injury,the cortical plasticity of the brain is altered due to damage to and regeneration of peripheral nerves;changes such as neuronal apoptosis and axonal demyelination occur in the spinal *** nerve will undergo axonal regeneration,activation of Schwann cells,inflammatory response,and vascular system regeneration at the injury *** damage to target organs can occur,including skeletal muscle atrophy and sensory receptor *** then provide a brief review of the research advances in therapeutic approaches to peripheral nerve *** main current treatments are conducted passively and include physical factor rehabilitation,pharmacological treatments,cell-based therapies,and physical ***,most treatments only partially address the problem and cannot complete the systematic recovery of the entire central nervous system-peripheral nervous system-target organ ***,we should further explore multilevel treatment options that produce effective,long-lasting results,perhaps requiring a combination of passive(traditional)and active(novel)treatment methods to stimulate rehabilitation at the central-peripheral-target organ levels to achieve better functional recovery.
