In the intricate process of natural product biosynthesis, a metabolon can enhance metabolic flux by associating sequential enzymes. A fungal metabolon, comprising of flavin-dependent monooxygenase Spe F and P450 monooxygenase Spe G, is identified in the biosynthesis of spiro polycyclic alkaloids(+)-notoamide B and its diastereomer(+)-versicolamide B. Using notoamide E as a substance, Spe F/Spe G metabolon can control the stereoselectivity of its 2,3-epoxidation, followed by hydrogen atom ion at C-17 to generate reactive epoxide tau-m A with dienyl iminium unit. Subsequently,(+)-notoamide B and(+)-versicolamide B are produced via tandem nonenzymatic inverse-electron-demand Diels-Alder reaction and semipinacol rearrangement. This provides the first example of metabolon in the biosynthesis of spiro-prenylated indole alkaloids.

By the condensation of 2,6-bis（4-amino-5-mercapto-[1,2,4]-triazoles-2）pyridine with aromatic acid in the presence of phosphorus oxychloride. Compounds of 2,6-bis（6-aryl-[1,2,4]-triazolo[3,4-b][1,3,4]-thiadiazole-3-yl）pyridines were synthesized. Their structures were confirmed by IR, ^1H NMR spectroscopies and elemental analysis. Their electrochemical behavior and cyclic voltammogram also were be studied. The results showed that they have high ionization potentials and good affinity.

A complex community of human-associated microorganisms plays diversely important roles in maintaining health,and the metabolites help to define whether a particular human-associated microbe is a friend or a *** work shows that Aspergillus brunneoviolaceus FB-2,derived from the stool of a healthy individual,produces a family of antiosteoporotic xanthone dinners including penibishexahydroxanthone A(1)and brunneoxanthones A-E(2-6,undescribed previously).The structures of 1-6 were elucidated by MS and NMR spectroscopic analysis,and the absolute configuration of 2 was established by X-ray *** 4-6 are relatively rare examples of highly oxidized heterodimeric xanthones with an 8-ketone ***,the prednisolone-induced osteoporotic zebrafish enabled our demonstration that the antiosteoporotic effects of 1-6 are dose-dependent.

Polyelectrolyte brushes(PEBs)are commonly used to modify surface that have attracted great research *** dielectric permittivity of the grafted surface is typically significantly different from that of solution,which results in surface polarization(SP)effect with a jump of electric *** is thus important to study how SP alters the PEB’s structure and *** this work,the SP effects on PEB structure was studied using a statistical thermodynamic *** free energy functional to describe SP effect was constructed by using the image-charge ***,the electrostatic potential was solved from a modified Poisson-Boltzmann equation taking the ion solvation effect into *** the absence of SP,the thickness of PEB exhibited a continuous collapse transition when decreasing the solvent *** the presence of SP,the collapse became a jump-like *** energy analysis showed that the long-range Coulombic interaction dominated the transition because of the enhanced counterion condensation in the presence of *** theory provides an effective tool to study SP effect on PEBs,and the results explain the underlying physics in PEB collapse transition.

BACKGROUND In recent years,the mechanical concept of intervertebral disc regeneration has become more and more popular due to the increasing awareness of the importance of preservation of spine ***,there is increasing evidence,however,that dynamic stabilization systems may compensate nonphysiological loads,limit pathological movement,normalize disc height and intradiscal pressure,and provide an adaptive environment for disc *** SUMMARY The patient was a 54-year-old man,who presented with a 10-year history of mechanical back pain,which had become progressively serious and radiated into the left lower limb with numbness 3 mo *** had decreased muscle strength(class IV)of the left dorsal extensor and plantar *** resonance imaging scans showed L3-S1 disc degeneration and L4-L5 disc *** the patient did not respond to various conservative treatments,he underwent a posterior L4-5 discectomy with fixation of the BioFlex dynamic stabilization system(Bio-Spine,Seoul,Korea).Preoperative symptoms were relieved and lumbar function was markedly improved after the operation.L4-L5 disc rehydration of instrumented segment was noted on magnetic resonance imaging at the 2-year *** Rehydration of the degenerated disc in our patient indicates that the BioFlex dynamic stabilization system may promote disc *** research is needed to provide more evidence to support lumbar disc rehydration in the bridged segment using this system.

Some drugs exert curative effects intracellu- larly, but their hydrophilic property prohibits the mem- brane-penetrating process and thus limits the curative efficacies, although this property guarantee their solubility in aqueous phase. An example is sodium tanshinone II-A sulfonate （DS-201）, a derivative of Chinese medical herb Danshen （Salvia miltiorrhiza） which is a BKc~ channel opener and a vasodilator. This study established and opti- mized a liposome delivery system which could pack and deliver DS-201 into HEK293 cells transfected with BKca channels, and DS-201 given this way significantly increased the open probability of BKca channel from baseline 0.013 ~ 0.004 to 0.036 -4- 0.011 at -t-40 mV membrane potential （P 〈 0.05） in single-channel attached study, and also increased the current density from baseline 23.2 ＋ 4.4 to 66.0 4- 15.2 pA/pF at ＋40 mV membrane potential （P 〈 0.05）, compared with the direct extracellular administration of this drug. Moreover, showing a ~ 60 % inhibition of the PE or PGF2a induced vascular constric- tion, the DS-201 liposomes did posses significantly enhanced vasorelaxant effect on rat mesenteric artery, compared with 20 % inhibition of the directly administration of this drug （P 〈 0.05）, suggesting that DS-201 delivered by liposomes significantly improved the drug＇s vasorelaxing effect. Taken together, the optimized DS-201 liposomes in our study successfully delivered DS-201 into cells and thus significantly activated BKca channels to reverse the contraction induced by PE and PGF2a, attesting the enhanced bioavailability.
