The prevalence of hyperlipidemia has increased significantly due to genetic, dietary, nutritional and pharmacological factors, and has become one of the most common pathological conditions in humans. Hyperlipidemia can lead to a range of diseases such as atherosclerosis, stroke, coronary heart disease, myocardial infarction, diabetes, and kidney failure, etc. High circulating low-density lipoprotein cholesterol (LDL-C) is one of the causes of hyperlipidemia. LDL-C in the blood binds to LDL receptor (LDLR) and regulates cholesterol homeostasis through endocytosis. In contrast, proprotein convertase subtilisin/kexin type 9 (PCSK9) mediates LDLR degradation via the intracellular and extracellular pathways, leading to hyperlipidemia. Targeting PCSK9-synthesizing transcription factors and downstream molecules are important for development of new lipid-lowering drugs. Clinical trials regarding PCSK9 inhibitors have demonstrated a reduction in atherosclerotic cardiovascular disease events. The purpose of this review was to explore the target and mechanism of intracellular and extracellular pathways in degradation of LDLR and related drugs by PCSK9 in order to open up a new pathway for the development of new lipid-lowering drugs.

Since determining the weight of pigs during large-scale breeding and production is challenging,using non-contact estimation methods is *** study proposed a novel pig weight prediction method based on a mod-ified mask region-convolutional neural network(mask R-CNN).The modified approach used ResNeSt as the backbone feature extraction network to enhance the image feature extraction *** feature pyramid net-work(FPN)was added to the backbone feature extraction network for multi-scale feature *** channel at-tention mechanism(CAM)and spatial attention mechanism(SAM)were introduced in the region proposal network(RPN)for the adaptive integration of local features and their global dependencies to capture global in-formation,ultimately improving image segmentation *** modified network obtained a precision rate(P),recall rate(R),and mean average precision(MAP)of 90.33%,89.85%,and 95.21%,respectively,effectively segmenting the pig regions in the *** image features,namely the back area,body length,body width,average depth,and eccentricity,were *** pig depth images were used to build five regression algo-rithms(ordinary least squares(OLS),AdaBoost,CatBoost,XGBoost,and random forest(RF))for weight value *** achieved the best prediction result with a coefficient of determination(R^(2))of 0.987,a mean absolute error(MAe)of 2.96 kg,a mean square error(MSe)of 12.87 kg^(2),and a mean absolute percentage error(MAPe)of 8.45%.The results demonstrated that the machine learning models effectively predicted the weight values of the pigs,providing technical support for intelligent pig farm management.

Recent years have witnessed numerous technical breakthroughs in connected and autonomous vehicles (CAVs). On the one hand, these breakthroughs have significantly advanced the development of intelligent transportation systems (ITSs);on the other hand, these new traffic participants introduce more complex and uncertain elements to ITSs from the social space. Digital twins (DTs) provide real-time, data-driven, precise modeling for constructing the digital mapping of physical-world ITSs. Meanwhile, the metaverse integrates emerging technologies such as virtual reality/mixed reality, artificial intelligence, and DTs to model and explore how to realize improved sustainability, increased efficiency, and enhanced safety. More recently, as a leading effort toward general artificial intelligence, the concept of foundation model was proposed and has achieved significant success, showing great potential to lay the cornerstone for diverse artificial intelligence applications across different domains. In this article, we explore the big models embodied foundation intelligence for parallel driving in cyber-physical-social spaces, which integrate metaverse and DTs to construct a parallel training space for CAVs, and present a comprehensive elucidation of the crucial characteristics and operational mechanisms. Beyond providing the infrastructure and foundation intelligence of big models for parallel driving, this article also discusses future trends and potential research directions, and the ?S?goals of parallel driving.

AIM:To investigate the short-term efficacy and safety of inebilizumab for neuromyelitis optica spectrum disorders(NMOSD).MeTHODS:A total of 33 patients with NMOSD treated with inebilizumab(Group INB,n=15)or rituximab(Group RTX,n=18)in addition to high-dose glucocorticoids were *** groups underwent hormone shock therapy during the acute ***,Group INB received inebilizumab injections during the remission phase,while Group RTX received rituximab injections.A comparison of aquaporins 4(AQP4)titer values,peripheral blood B lymphocyte counts,and visual function recovery was conducted before and 8wk after ***,adverse reactions and patient tolerability were analyzed after using inebilizumab treatment ***:Following inebilizumab treatment,there was a significantly improvement in the visual acuity of NMOSD patients(Ped by a notable decrease in AQP4 titer values and B lymphocyte ratio(Peover,inebilizumab treatment showed a partial effect in preventing optic nerve atrophy(Pever,there were no significant differences in other therapeutic effects compared to rituximab,which has previously demonstrated substantial therapeutic efficacy(P>0.05).Furthermore,inebilizumab exhibited higher safety levels than that of rituximab ***:The combination of inebilizumab and high-dose glucocorticoids proves to be *** comparison to rituximab injections,inebilizumab displays better tolerance and ***,it demonstrates a partial effect in preventing optic nerve ***,it stands as an effective method to reduce the disability rates and improve the daily living ability of patients with NMOSD.

Testosterone is closely associated with lipid metabolism and known to affect body fat composition and muscle mass in ***,the mechanisms by which testosterone acts on lipid metabolism are not yet fully understood,especially in *** this study,cyp17a1-/-zebrafish(Danio rerio)exhibited excessive visceral adipose tissue(VAT),lipid content,and up-regulated expression and activity of hepatic de novo lipogenesis(DNL)*** assay for transposase accessible chromatinwithsequencing(ATAC-seq)results demonstrated that chromatin accessibility of DNL genes was increased in cyp17a1-/-fish compared to cyp17a1+/+male fish,including stearoyl-CoA desaturase(scd)and fatty acid synthase(fasn).Androgen response element(ARe)motifs in the androgen signaling pathway were significantly enriched in cyp17a1+/+male fish but not in cyp17a1-/-*** androgen receptor(ar)-/-and wildtype(WT)zebrafish administered with Ar antagonist flutamide displayed excessive visceral adipose tissue,lipid content,and up-regulated expression and activity of hepatic de novo lipogenesis *** Ar agonist BMS-564929 reduced the content of VAT and lipid content,and down-regulated acetyl-CoA carboxylase a(acaca),fasn,and scd ***,the rescue effect of testosterone on cyp17a1-/-fish in terms of phenotypes was abolished when ar was additionally ***,these findings reveal that testosterone inhibits lipid deposition by down-regulating DNL genes via Ar in zebrafish,thus expanding our understanding of the relationship between testosterone and lipid metabolism in teleosts.
