On the basis of the uniform design method, six kinds of martensitic hot work die steels were designed. The phase transformation temperatures including Ac1 , Ac3 , and Ms were measured by DIL805A quenching dilatometer. The influences of the main elements on phase transformation temperatures were analyzed by quadratic stepwise regression analysis, and three corresponding equations were obtained. These equations, in which the interactions of the elements were considered, showed more effectiveness than the traditional ones. In addition, the thermal expansion coefficients of these steels in annealed state and quenched state were also obtained during the tests. The influences of chemical composition and temperature on the thermal expansion coefficient were analyzed; the equations obtained were verified by using several kinds of steels. The predicted values were in accordance with the results of the experiments.

In order to meet the demand of prehardened steel for large section plastic mould and save energy, a nonquenched prehardened （NQP） steel is developed. The temperature field of a large block is researched by finite element method simulation and 9 test steels are designed in the laboratory. Their microstructures and hardness are investigated when they are air cooled and control cooled at cooling rate similar to the simulation. The result shows that the hardness uniformity through section is closely correlated to bainitic hardenability for the NQP steel, and the hardness of one test steel （0.27C-1.95Mn-1.04Cr-0.45Mo-0.1V） fluctuates between HRC 40 and 43 under both cooling conditions. The test steel has better machinability compared with C45 steel, and the NQP steel is produced successfully in the factory based on the laboratory results. Its microstructure is bainite, and it is distributed uniformly through the size of 460 mm×800 mm×3 200 mm.

The selective hyperpolarization-activated cyclic nucleotide-gated（HCN） channel blocker 4-（N-ethyl-N-phenylamino）-1,2-dimethyl-6-（methylamino） pyrimidinium chloride（ZD7288） blocks the induction of long-term potentiation in the perforant path-CA3 region in rat hippocampus in vivo. To explore the mechanisms underlying the action of ZD7288, we recorded excitatory postsynaptic potentials in perforant path-CA3 synapses in male Sprague-Dawley rats. We measured glutamate content in the hippocampus and in cultured hippocampal neurons using high performance liquid chromatography, and determined intracellular Ca2＋ concentration（[Ca2＋]i） using Fura-2. ZD7288 inhibited the induction and maintenance of long-term potentiation, and these effects were mirrored by the nonspecific HCN channel blocker cesium. ZD7288 also decreased glutamate release in hippocampal tissue and in cultured hippocampal neurons. Furthermore, ZD7288 attenuated glutamate-induced rises in [Ca2＋]i in a concentration-dependent manner and reversed 8-Br-c AMP-mediated facilitation of these glutamate-induced [Ca2＋]i rises. Our results suggest that ZD7288 inhibits hippocampal synaptic plasticity both glutamate release and resultant [Ca2＋]i increases in rat hippocampal neurons.

AIM: To explore the possible mechanism of β-amyloid (Aβ)-induced apoptosis in rat cortical neurons and the protective effect of ginsenoside Rg1. METHODS: AO-EB staining was used to quantify the apoptotic cells. DNA fragmentation was observed by gel electrophoresis. The levels of cyclin-dependent kinases-4 (CDK4) and phosphorylated pRB were detected by Western blot. RT-PCR was used to examine the expression of E2FI mRNA. RESULTS: Treatment with Aβ1-40at the concentration of 20, 40, 80 mg/L for 48 h induced rat cortical neuron poptosis from 12.5％±1.5％(control) to 22.3％±1.4％, 38.8％±1.3％, 36.7％±1.4％, respectively. Pretreatment with Rg1at the dose of 0.5, 1, 2, 4, 8, 16 μmol/L for 24 h, then treatment with Aβ1-4040 mg/L for 24 h, the percentage of apoptotic neurons decreased from 38.8％±1.3％ to 14.5％±1.3％, 13.3％±1.0％, 11.6％±0.29％, 11.8％±1.0％, 6.2％±0.8％, 5.8％±0.8％, respectively. The paper presents experimental investigation results of crack pattern change in cement pastes caused by external sulfate attack(ESA).To visualize the formation and development of cracks in cement pastes under ESA,an X-ray computed tomography(X-ray CT)was used,i e,the tomography system of Zeiss Xradia 510 *** results indicate that X-CT can monitor the development process and distribution characteristics of the internal cracks of cement pastes under ESA with attack *** addition,the C3A content in the cement significantly affects the damage mode of cement paste specimens during sulfate *** damage of ordinary Portland cement(OPC)pastes subjected to sulfate attack with high C3A content are severe,while the damage of sulfate resistant Portland cement(SRPC)pastes is much smaller than that of OPC ***,a quadratic function describes the correlation between the crack volume fraction and development depth for two cement pastes immermed in sulfate solution.

＜正＞Purpose The present study was undertaken to examine the amplification and expression status of Cksl in breast cancer and its *** The amplification and expression status of Cksl gene was examined by FISH,Real-time PCR and Immunohistochemistry,*** interference was used to detect the effects of Cksl on migration,invasion, cell cycle progress and apoptosis of breast cancer cells. Results Cksl gene amplification was highly correlated with protein *** of Cksl was strongly associated with lymph node metastasis and poor prognosisr. Knockdown of Cksl expression by RNA interference inhibited the cell cycle progress,migration and invasion ability of breast cancer ***,overexpression of Cksl inhibited apoptosis of breast cancer cells through MEK-Erk pathway. Conclusion These data suggest that Cksl is an oncogene in the 1q21 amplicon and may play an important role for breast cancer development.

Dear Editor, Phenotypic drug discovery, which observes compound effects on physiology,has given way to targeting specific molecules, but has revived by using modern methods to find drugs based on therapeutic outcomes in disease models(Vincent et al., 2022). Osteoarthritis(OA) is a prevalent chronic joint disorder, primarily affecting knee joints, with osteoclast activation due to low-grade inflammation resulting in increased bone resorption and structural changes in the joint(Katz et al.,2021).
