How incubation temperature affects hatchling performance in reptiles: an integrative insight based on plasticity in metabolic enzyme

Evaluating the effects of temperature variations on animals plays an important role in understanding the threat of climate *** effects of developmental temperature on offspring performance are critical in evaluating the effects of warming temperatures on the fitness of ovipa-rous species,but the physiological and biochemical basis of this developmental plasticity is largely *** this study,we incubated eggs of the turtle Pelodiscus sinensis at low(24℃),medium(28℃),and high(32℃)temperatures,and evaluated the effects of developmental temperature on offspring fitness,and metabolic enzymes in the neck and limb muscles of *** hatchlings from eggs incubated at the medium temperature showed better fitness-related performance(righting response and swimming capacity)and higher activities of metabolic enzymes(hexokinase,HK;lactate dehydrogenase,LDH)than hatchlings from the eggs incubated at high or low *** addition,the swimming speed and righting response were significantly correlated with the HK activities in limb(swimming speed)and neck(righting response)muscles,suggesting that the developmental plasticity of energy metabolic pathway might play a role in determining the way incubation temperature affects offspring *** the fitness-related performance and the activities of metabolic enzymes,we predict that the P sinensis from high latitude would not face the detrimental effects of climate warming until the average nest temperatures reach 32℃.

目的:探究布地奈德雾化吸入辅助阿奇霉素治疗对支原体肺炎(Mycoplasma pneumoniae pneumonia,MPP)患儿病情控制及C反应蛋白(C-reactive protein,CRP)、白细胞计数(White blood cells,WBC)的影响。方法:选取2023年9月~2024年1月本院接收的60例MPP患儿作为研究对象,以随机数字表法分为观察组与对照组,各30例。对照组口服阿奇霉素治疗,观察组在对照组基础上联合使用布地奈德雾化吸入;比较两组临床症状、病情控制情况及CRP、WBC水平,并统计不良反应发生率。结果:治疗后,观察组患儿咳嗽、发热消失时间与对照组患儿咳嗽、发热消失时间相比,观察组消失时间短(P0.05)。治疗后,两组CRP水平和WBC计数均明显下降,且观察组明显低于对照组(P0.05)。结论:布地奈德辅助阿奇霉素治疗MPP患儿,疗效应用显著,对于患儿临床症状具有较高疗效,无明显副作用,具有参考意义。

Pharmacologically inhibiting GluR2 internalization alleviates neuropathic pain

Neuropathic pain is of serious clinical concern and only about half of patients achieve partial relief with currently-available treatments,so it is critical to find new drugs for this ***,the cellsurface trafficking of pain-related receptors has been suggested as an important mechanism underlying persistent neuropathic ***,we used the short peptide GluA_（2-3y）,which specifically inhibits the GluA2-dependent endocytosis of a-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors,and tested its anti-nociceptive effect in the periaqueductal grey（PAG） of intact rats and rats with neuropathic ***-PAG injection of 0.15,1.5,7.5,and 15 pmol of GluA_（2-3y） induced dose-dependent increases in hindpaw withdrawal latencies to noxious thermal and mechanical stimuli in intact rats,suggesting that GluA2 cell-surface trafficking in the PAG is involved in pain ***,GluA_（2-3y） had much stronger anti-nociceptive effects in rats with neuropathic pain induced by sciatic nerve ***,the intra-PAG injection of 15 pmol GluA_（2-3y） had an analgesic effect similar to 10 ug（35nmol） morphine in rats with neuropathic *** together,our results suggested that GluA2 trafficking in the PAG plays a critical role in pain modulation,and inhibiting GluA2 endocytosis with GluA_（2-3y） has potent analgesic effects in rats with neuropathic *** findings strongly support the recent hypothesis that targeting receptor trafficking could be a new strategy for the treatment of neuropathic pain.

Curcumin protects against staurosporine toxicity in rat neurons

Objective Curcumin is extracted from the turmeric plant （Curcuma longa Linn.） and is widely used as a food additive and traditional medicine. The present study investigated the activity of curcumin against staurosporine （STS） toxicity in cell culture. Methods Rat hippocampal neurons in primary culture were exposed to STS （20 μmol/L） and treated with curcumin （20 μmol/L）. Cell viability was tested by MTT assay and reactive oxygen species （ROS） were measured using the MitoSOX red mitochondrial superoxide indicator. Western blot was used to assess changes in the levels of caspase-3 （Csp3）, heat shock protein 70 （Hsp70） and Akt. Results The results showed that curcumin protects against STS-induced cytotoxicity in rat hippocampal neurons. Csp3, Hsp70, Akt and ROS activation may be involved in this protection. Conclusion Curcumin could be a potential drug for combination with STS in cancer treatment to reduce the unwanted cytotoxicity of STS.

Relationship among eye condition sensitivities, photosensitivity and epileptic syndromes

Background Electroencephalogram （EEG） activity in normal subjects and epileptic patients is often closely related to the eye＇s status such as eye opened （EO）, eye closure （ECL） and eyes closed （EC）. ECL is the period immediately after closing of the eyes and only lasts for less than 3 seconds if the eyes remain closed. EC is the period as long as the eyes are closed. Epileptiform changes on EEG induced by ECL or EC are called the changes of ECL sensitivity （ECLS） or EC sensitivity （ECS）. ECLS occurs mainly but not exclusively in photosensitive patients and ECS has been seen rarely in photosensitive patients. This study aimed to investigate the relationships among ECLS, ECS, photosensitivity and epilepsy syndromes in children. Methods EEG records from child patients in the EEG Department of Peking University First Hospital during the period of May 2005 to May 2007 were examined for the presence of ECLS or ECS. Open-close eye tests and intermittent photic stimulations were carried out during video-EEG monitoring for examining ECLS, ECS and photosensitivity. Results Based on ECLS and ECS on their EEGs, 30 patients were divided into ECLS group （16 cases） and ECS group （14 cases）. There were more boys than girls in the two groups. The mean age of initial detection of ECLS and ECS was 10 years, and the average onset age of seizures was 9 years. The epilepsy syndromes in the ECLS group included idiopathic photosensitive occipital lobe epilepsy, Panayiotopoulos syndrome, symptomatic occipital lobe epilepsy, juvenile myoclonic epilepsy, juvenile absence epilepsy, eyelid myoclonia with absences, epilepsy with grand mal on awakening and pure photosensitive epilepsy with mainly generalized tonic clonic seizures. Those in the ECS group were juvenile myoclonic epilepsy, idiopathic photosensitive occipital lobe epilepsy, Panayiotopoulos syndrome and Gastaut type-idiopathic children occipital epilepsy. Post-miosis changes in the anterior chamber structures in primary and lens-induced secondary chronic angle-closure glaucoma

To evaluate post-miosis changes in the anterior chamber structures in various angle-closure glaucomas(ACG). Totally 14 eyes of primary chronic angle-closure glaucoma(PCACG), 12 eyes of lens-induced secondary chronic angleclosure glaucoma(LSACG) and 14 healthy eyes were recruited. After miosis, for PCACG group, intraocular pressure(IOP) and anterior chamber depth(ACD) changed not significantly, while anterior chamber angle widened significantly. LSACG group showed a significant increase in IOP, decrease in ACD, and narrowing in anterior chamber angle. Healthy eyes showed significant decreases in IOP and anterior chamber parameters. Thus, miosis could widen the anterior chamber angle of patients with PCACG, while increase the narrowing of anterior chamber angle and IOP of patients with LSACG. We should pay attention to the distinction between PCACG and LSACG patients and the proper administration of pilocarpine in the treatment of patients with chronic ACG.

Conformation Disturbance of CopC by α-Naphthylamine

It is known that urea and guanidine hydrochloride（GuHC1） induce conformational change of proteins in a certain range of molar ratios. In our research, a-naphthylamine（NA） above 10^-4 mol/L at pH 7.0 was discovered to perturb the conformation of CopC, a copper resistant protein with a Greek fl-barrel motif; this was reflected by the greater fluorescence quenching and red-shifted emission peak of CopC. The conformation change of CopC was also verified in acrylamide collision experiment by comparing quenching levels of CopC in the presence or the abscence of NA. Comparison of emission change of CopC-Cu^2＋ with that of apo-CopC showed a consistent result with their denaturation in GuHCl. And decreasing fluorescence polarization of CopC with increasing NA concentration is consistent with a more extend conformation of CopC. Interaction mechanism was also explored.

Upregulated functional expression of Toll like receptor 4 in mesenchymal stem cells induced by lipopolysaccharide

Background The coordinated change of haematopoietic supporting microenvironment in bone marrow （BM） is crucial for innate immunity and inflammation. As the precursors of marrow stroma, BM derived mesenchymal stem cells （MSCs） promote haematopoietic function, but their roles in innate immunity or inflammation have not been investigated. Here we investigated the expression of Toll like receptor 4 （TLR-4） and the effect of lipopolysaccharide （LPS） on its expression in BM MSCs in vitro. Methods MSCs were harvested from adult rat＇s BM cells by density gradient centrifugation and adhesive culture. The purity of MSCs were identified with the cell morphological feature and osteogenic capacity, the phenotypes were tested by flow cytometry. Cultured MSCs were treated by LPS （1 μg/ml, 10μg/ml or 100μg/ml） for 24 hours. The relative expression levels of TLR-4 mRNA were detected by semiquantitative reverse transcription polymerase chain reaction and costimulatory molecules （CD80, CD86 and MHC-Ⅱ） expressed on MSCs were analyzed by flow cytometry. The levels of tumor necrosis factor-α （TNF-α） in supernatants were determined by enzyme linked immunosorbent assay. Results After incubation with LPS, MSCs expressed the higher levels of TLR-4 mRNA, costimulatory molecules and TNF-α than the untreated group： LPS 10 μg/ml was the most effective （P〈0.01）; the levels of TLR-4 mRNA, costimulatory molecules and TNF-α decreased when MSCs were exposed to 100 μg/ml LPS. Except for MHC-Ⅱ and TNF-α （P〉0.05）, the levels of CD80, CD86 and TLR-4 mRNA were significantly lower than that in the treated group of 10 μg/ml （P〈0.01）. Conclusion MSCs expressed TLR-4 mRNA. LPS activated the functional expression levels of TLR-4 in MSCs although the activity may depend on the concentration of LPS.

In vitro tissue engineering of lamellar cornea using human amniotic epithelial cells and rabbit cornea stroma

AIM:To reconstruct the lamellar cornea using human amniotic epithelial(HAE) cells and rabbit cornea stroma in vitro using tissue engineering technology.·METHODS:Human amnia taken from uncomplicated caesarean sections were digested by collagenase to obtain HAE cells,and the cells were cultured to *** corneal epithelial cells were removed by n-heptanol to make lamellar matrix *** second passage of HAE cells were cultured on the corneal stroma sheets for 1 or 2 days,then transferred to an air-liquid interface environment to culture for *** engineered lamellar cornea(TELC)morphology was observed by Hematoxylin-eosin(HE)staining;its ultrastructure was observed by transmission electron microscopy(TEM) and scanning electron microscopy(SEM);corneal epithelial cell-specific keratin3 and keratin 12 were detected with immunofluorescence microscopy.·RESULTS:HAE cells grew on the rabbit corneal stroma,forming a monolayer after 1-2 *** 4-5 layers of epithelial cells developed after 2 weeks of air-liquid interface cultivation,a result similar to normal corneal *** corneal stromal cells were significantly reduced after one week,then almost completely disappeared after 2 *** showed desmosomes between the epithelial cells;hemidesmosomes formed between the epithelial cells and the basement *** revealed that the HAE cells which grew on the lamellar cornea had abundant *** tissue-engineered cornea expressed keratin 3 and keratin 12,as detected by immunofluorescence assay.·CONCLUSION:Functional tissue-engineered lamellar corneal grafts can be constructed in vitro using HAE cells and rabbit corneal stroma.
