Two-way Doppler measurement is a typical Earth-based radiometric technique for interplanetary spacecraft navigation and gravity science *** most widely used model for the computation of two-way Doppler observables is Moyer’s differenced-range Doppler(DRD) formula,which is based on a Schwarzschild approximation of the Solar-System ***,the computation of range difference in DRD formula is sensitive to round-off errors due to approximate numbers defined by the norm IEEE754 in all *** paper presented two updated models and their corresponding detailed instructions for the computation of the two-way Doppler observables so as to impair the effects of this type of numerical *** two models were validated by two case studies related to the Rosetta mission—asteroid Lutetia flyby and comet 67 P/Churyumov-Gerasimenko orbiting *** these two cases,the numerical noise from the updated models can be reduced by two orders-of-magnitude in the computed two-way Doppler *** results showed an accuracy from better than 6 × 10^(-3) mm s^(-1) at 1 s counting time interval to better than 3 × 10^(-5) mm s^(-1) at 60 s counting time interval.

Objective To investigate cognitive dysfunction in patients with carotid artery stenosis(CAS) and potential risk factors related to cognitive-especially memory-dysfunction. Methods Forty-seven patients with carotid artery stenosis were recruited into our study cohort. The Mini-Mental State Examination(MMSE) and the Montreal Cognitive Assessment(MoCA) were adopted to assess cognitive function, the Wechsler Memory Scale(WMS) to assess memory function, high-resolution MRI and enhanced ultrasound to evaluate carotid plaques, and computed tomography perfusion(CTP) imaging to evaluate intracranial blood perfusion. Single-factor analysis and multiple-factor regression analysis were used to analyze potential risk factors of cognitive impairment. Results Mini-Mental State Examination test results showed that 22 patients had cognitive impairment, and MoCA test results showed that 10 patients had cognitive impairment. Analysis of various risk factors indicated that the average memory quotient of female patients was higher than that of males(P = 0.024). The cognitive and memory performance of those with an educational background above high school were significantly better than those of participants with high school or lower(P = 0.045). Patients with abnormal intracranial perfusion performed worse on the MMSE test(P = 0.024), and their WMS scores were significantly lower(P = 0.007). The MMSE scores and the memory quotients were significantly lower in patients with a history of cerebral infarction(MMSE, P = 0.047, memory quotient score, P = 0.018). Conclusion A history of cerebral infarction and abnormal cerebral perfusion are associated with decline in overall cognitive function and memory in patients with carotid stenosis. Being female and having an educational background above high school may be protective factors in the development of cognitive dysfunction.

Objective To investigate the relationship between geranylgeranyl pyrophosphate synthase （GGPPS） gene polymorphisms and bone response to alendronate in Chinese osteoporotic women. Methods A total of 639 postmenopausal women with osteoporosis or osteopenia were included and randomly received treatment of low dose （70 mg per two weeks） or standard dose （70 mg weekly） of alendronate for one year. The six tag single nucleotide polymorphisms of GGPPS gene were identified. Bone mineral density （BMD）, serum cross-linked C-telopeptide of type I collagen （β-CTX）, and total alkaline phosphatase （ALP） were measured before and after treatment. GGPPS gene polymorphisms and the changes of BMD and bone turnover markers after treatment were analyzed. Results rs10925503 polymorphism of GGPPS gene was correlated to serumβ-CTX levels at baseline, and patients with TT genotype had significantly higher serum β-CTX level than those with TC or CC genotype （all P〈0.05）. No correlation was found between polymorphisms of GGPPS gene and serum total ALP levels, as well as BMD at baseline. After 12 months of treatment, lumbar spine and hip BMD increased and serum bone turnover markers decreased significantly （P〈0.01）, and without obvious differences between the low dose and standard dose groups （all P〉0.05）. However, GGPPS gene polymorphisms were uncorrelated to percentage changes of BMD, serum total ALP, and β-CTX levels （all P〉0.05）. Conclusion GGPPS gene polymorphisms are correlated to osteoclasts activity, but all tag single nucleotide polymorphisms of GGPPS gene have no influence on the skeletal response to alendronate treatment.

Articulated natural assemblages contain direct evidence of the element numbers,morphologies,positions and structures for reconstructing the feeding apparatuses of conodont animals,but these kind of materials are very rare in fossil *** we report ten new conodont natural assemblages from Member II of the Guanling Formation in Luoping County,eastern Yunnan Province,southwestern *** assemblages were obtained from the fossil-bearing layers of the Luoping Biota,dated to the early Middle Triassic Anisian stage(Pelsonian substage).These fossiliferous laminated limestones mainly consist of calcites and dolomites,with platy clay minerals and pyrites as subordinate components,indicating that the conodont assemblages were preserved in a lower energy and anoxic sedimentary *** new natural assemblages preserve the primary collapse orientations of the Nicoraella feeding apparatus from the Luoping Biota,showing the relative original positions of the S,M and P elements,that could be further used to refine the architecture of this apparatus in space variable M elements and the position unclear P1 *** previously reported three-dimensional and bilaterally symmetrical fused clusters,we statistically analyze the size of the positional homogenous elements within different sized materials,in particular S4,S3,or S2 elements,indicates that elements ontogenic changes do not affect the apparatus *** of Nicoraella apparatus remains stable in the process of ontogeny,so our reconstructed model is *** is the first time that ample types of materials have been used to restore a conodont apparatus within one genus,namely natural assemblages(articulated and disarticulated),fused clusters(articulated,disarticulated,compressed,and relatively three-dimensional),and abundant discrete elements,*** materials together record the most complete information on architecture in the gondolelloid apparatuses,thus enabling us

Prokaryotic Expression of Gly-Gln Dipeptide and Its Bioactive Analysis: A Novel Method for Short Peptide Production

Small bioactive peptides, with diverse biological functions, have received increasing attention as physiologically beneficial substances in animal production. The main obstacle to wide application of small bioactive peptides is a lack of costeffective methods for mass production. In this study, we mass-production method for small bioactive peptides. used glycyl-glutamine （Gly-Gln） as a test case to develop a novel The oligonucleotide encoding Gly-Gln pro-peptide （Gpp） was designed and synthesized. Gpp includes 3 Gly-Gln dipeptides and 2 enzymatic sites for pepsin and trypase, allowing direct digestion and absorption of Gly-Gln in the gastrointestinal tract. The Gpp oligonucleotides were linked to generate an oligomeric oligonucleotide segment containing 12 tandem copies of Gpp. This 12Gpp segment was cloned and expressed in Escherichia coli vector pET32a. By optimizing culture conditions [0.1 mmol L^-1 isopropyl-β-D- thiogalactopyranoside （IPTG）, 50 μg mL^-1 ampicillin （Amp）, 30℃ for 12 h], the thioredoxin fusion peptides reached 40% of total bacterial protein. After purification, the fusion protein was fed to Kunming mice to determine its effect on mouse immune function. The results showed that similar to Gly-Gln dipeptide, Gpp polymer protein could significantly suppress the proliferation of T and B lymphocytes in blood and spleen, and additionally could significantly improve interleukin-2 （IL-2） and interleukin-6 （IL-6） secretion of blood and spleen lymphocytes. These effects were not observed in mice fed a 2 amino acids mix （glycine and glutamine）. These evidences indicated that an efficient digestion of Gpp polymer protein could be achieved when ingested into the animal gut. The expression system in this study provides a potential production method for not only Gly-Gln dipeptide but also other short bioactive peptides.

Isolation and Molecular Characterization of Toxoplasma gondii from Chickens in China

One strain of Toxoplasma gondii was successfully isolated from chickens in China by bioassay in mice. Antibodies and circulating antigens of T. gondii were assayed by the ELISA kits in 100 free range chickens from a rural area surrounding Funing, China. Fifty-three chickens were antibody-positive and 21 chickens were antigen positive. Hearts, brains, spleens, lungs, livers, and kidneys of 21 antibody or antigen-positive chickens were bioassayed in mice. One strain of T. gondii was isolated from 1 of 21 （4.76%） chickens. The isolated T. gondii killed all of the inoculated mice. Genotyping of this isolate using polymorphisms at the loci 5′-SAG2, 3′-SAG2, SAG3, cB21-4, L358, BTUB, and GRA6 revealed that it was Type I. These indicated that it was virulent for mice. This is the first report of isolation of T. gondii from chickens in China.

Despite the diverse roles of tripartite motif(Trim)-containing proteins in the regulation of autophagy,the innate immune response,and cell differentiation,their roles in skeletal diseases are largely *** recently demonstrated that Trim21 plays a crucial role in regulating osteoblast(OB)differentiation in ***,how Trim21 contributes to skeletal degenerative disorders,including osteoporosis,remains ***,human and mouse bone specimens were evaluated,and the results showed that Trim21 expression was significantly elevated in bone tissues obtained from osteoporosis ***,we found that global knockout of the Trim21 gene(KO,Trim2^(1-/-))resulted in higher bone mass compared to that of the control *** further demonstrated that loss of Trim21 promoted bone formation by enhancing the osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs)and elevating the activity of OBs;moreover,Trim21 depletion suppressed osteoclast(OC)formation of RAW264.7 *** addition,the differentiation of OCs from bone marrow-derived macrophages(BMMs)isolated from Trim21^(-/-)and Ctsk-cre;Trim21^(f/f)mice was largely compromised compared to that of the littermate control ***,YAP1/β-catenin signaling was identified and demonstrated to be required for the Trim21-mediated osteogenic differentiation of *** importantly,the loss of Trim21 prevented ovariectomy(OVX)-and lipopolysaccharide(LPS)-induced bone loss in vivo by orchestrating the coupling of OBs and OCs through YAP1 *** current study demonstrated that Trim21 is crucial for regulating OB-mediated bone formation and OC-mediated bone resorption,thereby providing a basis for exploring Trim21 as a novel dual-targeting approach for treating osteoporosis and pathological bone loss.
