A novel non-invasive identification of genome editing mutants from insect exuviae

With the wide application of genome editing in insects, a simple and efficient identification method is urgently needed to meet the increasing demand for mutation detection. Here, taking migratory locusts as a model system, we developed a non-invasive method to accurately identify genome-edited mutants by using DNA from insect exuviae. We compared the quantity and quality of genomic DNA from exuviae in five instar hoppers and found that the 1st instar exuviae had the highest DNA yield and content, while the 3rd instar exuviae had the best quality. Consensus genotypes were identified from genomic DNA of hoppers at different developmental stages in the same individuals. Moreover, we demonstrated that the amplification products from DNA extracted from locust exuviae are the consensus sequences with those from the hemolymph and foreleg pre-tarsus. Therefore, non-invasive samples provide the same genotyping results as minimally invasive and invasive samples of the same individuals. Furthermore, this identification method that uses genomic DNA from exuviae can be used for early screening of positive genome-edited individuals in each generation for adult crossing. In our study, the non-invasive identification method was not only simpler and provided results earlier than existing methods, but also had a better reproducibility and accuracy. This non-invasive identification approach using genomic DNA from exuviae can be adapted to meet the growing demand for genetic analysis and will find wide application in insect genome editing research.

Impact of enhanced recovery after surgery programs on pancreatic surgery:A meta-analysis

AIM To evaluate the impact of enhanced recovery after surgery(ERAS) programs on postoperative complications of pancreatic surgery. METHODS Computer searches were performed in databases(including PubM ed, Cochrane Library and Embase) for randomized controlled trials or case-control studies describing ERAS programs in patients undergoing pancreatic surgery published between January 1995 and August 2017. Two researchers independently evaluated the quality of the studies' extracted data that met the inclusion criteria and performed a metaanalysis using Rev Man5.3.5 software. Forest plots, demonstrating the outcomes of the ERAS group vs the control group after pancreatic surgery, and funnel plots were used to evaluate potential publication bias.RESULTS Twenty case-control studies including 3694 patients, published between January 1995 and August 2017, were selected for the meta-analysis. This study included the ERAS group(n = 1886) and the control group(n = 1808), which adopted the traditional perioperative management. Compared to the control group, the ERAS group had lower delayed gastric emptying rates [odds ratio(OR) = 0.58, 95% confidence interval (CI): 0.48-0.72, P < 0.00001], lower postoperative complication rates(OR = 0.57, 95%CI: 0.45-0.72, P < 0.00001), particularly for the mild postoperative complications(Clavien-Dindo Ⅰ-Ⅱ)(OR = 0.71, 95%CI: 0.58-0.88, P = 0.002), lower abdominal infection rates(OR = 0.70, 95%CI: 0.54-0.90, P = 0.006), and shorter postoperative length of hospital stay(PLOS)(WMD =-4.45, 95%CI:-5.99 to-2.91, P < 0.00001). However, there were no significant differences in complications, such as, postoperative pancreatic fistulas, moderate to severe complications(Clavien-Dindo Ⅲ-Ⅴ), mortality, readmission and unintended reoperation, in both groups.CONCLUSION The perioperative implementation of ERAS programs in pancreatic surgery is safe and effective, can decrease postoperative complication rates, and can promote recovery for patients.

Conductive Film with Flexible and Stretchable Capability for Sensor Application and Stealth Information Transmission

Flexible and wearable strain sensors for human-computer interaction,health monitoring,and soft robotics have drawn widespread attention to promising applications in the next generation of artificial intelligence devices.However,conventional semiconductor sensors are difficult to meet the requirements of flexibility and stretchability.Here,we reported a kind of novel and simple sensor based on layer-by-layer(LBL)method.Carbon nanotubes(CNTs)layer provides high ductility and stability in the process of tension sensing,while silver layer provides low initial resistance and fast reflecting in the process of tension sensing.LBL method ensures the uniformity of the conductive layer.The sensor has superior sheet resistance of 9.44Ω/sq.,high elongation at break of 104%.For sensing capability,the sensor has wide reflecting range of 60%,high gauge factor(GF)of 1000 up to 60%strain,fast reflecting time of 165 ms.Excellent reliability and stability have also been verified.It is also worth mentioning that the entire process does not require any expensive equipments,complicated processes or harsh experimental conditions.The above features provide an idea for large-scale application of flexible stretchable sensors.

Risk factors for avian influenza virus in backyard poultry flocks and environments in Zhejiang Province,China:a crosssectional study

Background:Human infection of avian influenza virus(AIV)remains a great concern.Although live poultry markets are believed to be associated with human infections,ever more infections have been reported in rural areas with backyard poultry,especially in the fifth epidemic of H7N9.However,limited information is available on backyard poultry infection and surrounding environmental contamination.Methods:Two surveillance systems and a field survey were used to collect data and samples in Zhejiang Province.In total,4538 samples were collected by surveillance systems and 3171 from the field survey between May 2015 and May 2017,while 352 backyard poultry owners were interviewed in May 2017 by questionnaire to investigate factors influencing the prevalence of avian influenza A virus and other AIV subtypes.RT-PCR was used to test the nucleic acids of viruses.ArcGIS 10.1 software was used to generate maps.Univariate and logistic regression analyses were conducted to identify risk factors for AIV infection.Results:Of the 428 poultry premises observed by the surveillance system,53(12.38%)were positive for influenza A virus.Of the 352 samples from poultry premises observed by field survey,13(3.39%)were positive for influenza A virus.The prevalence of AIV was unevenly distributed and the dominant subtype differed among cities.Eastern(Shaoxing and Ningbo)and southern(Wenzhou)cities exhibited a higher prevalence of AIV(16.33,8.94,and 7.30%respectively).Contamination of AIV subtypes was most severe in January,especially in 2016(23.26%,70/301).The positive rate of subtype H5/H7/H9 was 2.53%(115/4538).Subtype H5 was the least prevalent,while subtypes H7 and H9 had similar positivity rates(1.50 and 1.32%respectively).Poultry flocks and environmental samples had a similar prevalence of AIV(4.46%vs 5.06%).The type of live birds was a risk factor and the sanitary condition of the setting was a protective factor against influenza A contamination.Conclusions:AIV subtypes were prevalent in backyard poultry flocks and surrounding environmen

Joint Characteristics and Corrosion Properties of Bypass-Current Double-Sided Arc-Welded Aluminum 6061 Alloy with Al–Si Filler Metal

Sheets of aluminum 6061 alloy were welded using bypass-current double-sided arc welding with Al-Si filler wire to investigate the effect of Al-Si intermetallic compounds on the microstructure, microhardness and corrosion behavior of weld joint. Experimental results indicated that the Al4.5FeSi phase in the topside of the weld joint was finer than that in the backside and newly formed phase of Al0.5Fe3Si0.5 was observed in the backside. The formation of reinforcing phases of Al-Fe-Si in the weld improved the microhardness of the weld by about 18%. The corrosion resistance of the weld zone was greater than that of the base metal, while the corrosion current displayed opposite, and the corrosion resistance of the weld region was better than that of the base metal.

Recovery of natural killer cells is mainly in post-treatment period in chronic hepatitis C patients treated with sofosbuvir plus ledipasvir

AIM To investigate how natural killer(NK) cells are affected in the elimination of hepatitis C virus(HCV) by sofosbuvir/ledipasvir, two highly effective direct-acting antivirals(DAAs). METHODS Thirteen treatment-na?ve and treatment-experienced chronic hepatitis C(CHC) patients were treated with sofosbuvir/ledipasvir, and NK cells were detected at baseline, weeks 2, 4, 8 and 12 during therapy, and week post of treatment(Pt)-12 and 24 after the end of therapy by multicolor flow cytometry and compared with those from 13 healthy controls. RESULTS All patients achieved sustained virological response. There was a significant decline in CD56^(bright) NK cell frequencies at week 8(P = 0.002) and week 12(P = 0.003), which were altered to the level comparable to healthy controls at week Pt-12, but no difference was observed in the frequency of CD56^(dim) NK cells. Compared with healthy controls, the expression levels of NKG2A, NKp30 and CD94 on NK cells from CHC patients at baseline were higher. NKG2A, NKp30 and CD94 started to recover at week 12 and reached the levels similar to those of healthy controls at week Pt-12 or Pt-24. Before treatment, patients have higher interferon(IFN)-γ and perforin levels than healthy controls, and IFN-γ started to recover at week 8 and reached the normalized level at week Pt-12. CONCLUSION NK cells of CHC patients can be affected by DAAs, and phenotypes and function of NK cells recover not at early stage but mainly after the end of sofosbuvir/ledipasvir treatment.

Synthesis, chemical nuclease activity, and in vitro cytotoxicity of benzimidazole-based Cu(Ⅱ)/Co(Ⅱ) complexes

In this study, novel mono/di-nuclear Cu（p-2-bmb）（OH）（Cl O4）（1） and Co2（p-2-bmb）2Cl4（2）（p-2-bmb = 1-（（2-（pyridin-2-yl）-benzoimidazol-1-yl）methyl）-1H-benzotriazole） complexes with the nitrogen heterocyclic benzimidazole-based ligand were synthesized and characterized. The two complexes showed antiproliferative effects in various carcinoma cell lines, especially complex 1 in the SMMC7721 tumor cell line. Complex 1 was also able to pass through the cell membrane and enter the nucleus and mitochondrion. An analysis of in vitro chemical nuclease activity revealed that complex 1 partially intercalated to calf thymus DNA and exhibited strong unwinding activity against p BR322 superhelical plasmid DNA. The comet assay and flow cytometry analysis confirmed that 1 caused extensive DNA damage and arrested SMMC7721 tumor cells at G2/M phase of the cell cycle, leading to loss of mitochondrial membrane potential and apoptosis. These results suggest that these benzimidazole-based metal complexes could be potential anti-cancer agents.

Objective:To perform a retrospective cross-sectional study of the prevalence of apical periodontitis(AP) in general and root canal-filled teeth,as well as to assess some associated factors in Chinese adult subpopulations using cone beam computed tomography(CBCT).Materials and methods:CBCT images of 14756 teeth from 551 patients in the Department of Stomatology at Peking University Third Hospital were evaluated.AP was assessed and classified using the CBCT periapical index(CBCTPAI).The overall prevalence of AP and the prevalence of AP in root canal-filled teeth were reported.Logistic regression was used to determine the relationship between age,gender,tooth position,root canal fillings(RCF),coronal restorations and the prevalence of AP in overall teeth and root canal-filled teeth.Results:The overall prevalence of AP at the patient level was 41.6% and at the tooth level was3.1%.The prevalence of AP in root canal-filled teeth was as high as 57.3% among 372 teeth.Univariate and multiple logistic regression analyses indicated that in overall teeth,patients aged70 years or above(OR = 2.69,P = 0.002),maxillary teeth(OR = 1.77,P<0.001),molars(OR =1.87,P<0.001),teeth with RCF(OR = 23.99,P<0.001) and teeth with crown/post-core crown restorations(OR = 4.42,P<0.001) were more likely to have AP.In root canal-filled teeth,teeth with incomplete RCF(OR = 3.06,P<0.001) and teeth with crown/post-core crown restorations(OR = 2.20,P = 0.001) had a higher risk of persistent AP.However,there was no significant correlation between the quality of coronal restorations and the prevalence of AP in root canal-filled teeth.Conclusion:The prevalence of AP in Chinese subpopulations was comparable to that reported in other geographic locations.In overall teeth,the presence of AP was significantly associated with age,tooth position,tooth type,RCF,and type of coronal restorations.In root canal-filled teeth,the quality of the RCF and the type of coronal restorations were significant predictors of AP.CBCT databases are useful for cross-sectional st
