新近的基因识别软件比先前的软件有着显著的提高 ,但是在外显子水平上的敏感性和特异性仍然不十分令人满意 .这是因为已有软件对于剪接位点 ,翻译起始等生物信号位点的识别还不够有效 .如果能够分别提高这些生物信号位点的识别效果 ,就能够提高整体的基因识别效率 .隐半马氏模型能够很好地刻画 3′剪接位点 (acceptor)的结构 .据此开发的一套对acceptor进行识别的算法在Burset/Guigo的数据集上经过检验 ,获得了比已有算法更好的识别率 .该模型的成功还使得我们对剪接点上游的分支位点和嘧啶富含区的概貌有了一定的认识。

The neutron-induced total cross sections of natural lead have been measured in a wide energy range(0.3 eV-20 MeV)on the back-streaming white neutron beamline(Back-n)at the china Spallation Neutron *** energy was determined by the neutron total cross-section spectrometer using the time-of-flight technique.A fast multi-cell fission chamber was used as the neutron detector,and a 10-mm-thick high-purity natural lead sample was employed for the neutron transmission *** on-beam background was determined using Co,In,Ag,and Cd *** excitation function of ^(nat)Pb(n,tot)reaction below 20 MeV was calculated using the TALYS-1.96 nuclear-reaction modeling *** present results were compared with previous results,the evaluated data available in the five major evaluated nuclear data libraries(i.e.,ENDF/B-VIII.0,JEFF-3.3,JENDL-5,CENDL-3.2,and BROND-3.1),and the theoretical calculation *** agreement was found between the new results and those of previous experiments and with the theoretical curves in the corresponding *** measurement obtained the neutron total cross section of natural lead with good accuracy over a wide energy range and added experimental data in the resonance energy *** provides more reliable experimental data for nuclear engineering design and nuclear data evaluation of lead.

BACKGROUND Acute-on-chronic liver disease(AoCLD)accounts for the majority of patients hospitalized in the Department of Hepatology or Infectious *** To explore the characterization of AoCLD to provide theoretical guidance for the accurate diagnosis and prognosis of *** Patients with AoCLD from the Chinese Acute-on-Chronic Liver Failure(ACLF)study cohort were included in this *** clinical characteristics and outcomes,and the 90-d survival rate associated with each clinical type of AoCLD were analyzed,using the Kaplan-Meier method and the log-rank *** A total of 3375 patients with AoCLD were enrolled,including 1679(49.7%)patients with liver cirrhosis acute decompensation(LC-AD),850(25.2%)patients with ACLF,577(17.1%)patients with chronic hepatitis acute exacer-bation(CHAE),and 269(8.0%)patients with liver cirrhosis active phase(LC-A).The most common cause of chronic liver disease(CLD)was HBV infection(71.4%).The most common precipitants of AoCLD was bacterial infection(22.8%).The 90-d mortality rates of each clinical subtype of AoCLD were 43.4%(232/535)for type-C ACLF,36.0%(36/100)for type-B ACLF,27.0%(58/215)for type-A ACLF,9.0%(151/1679)for LC-AD,3.0%(8/269)for LC-A,and 1.2%(7/577)for *** HBV infection is the main cause of CLD,and bacterial infection is the main precipitant of *** most common clinical type of AoCLD is *** diagnosis and timely intervention are needed to reduce the mortality of patients with LC-AD or ACLF.

本研究基于滇龙胆草转录组数据分析,使用RT-PCR技术克隆获得两条香叶醇10-羟化酶基因,命名为GrG10H1和GrG10H2。使用在线软件对其进行了生物信息学分析,两条基因全长分别为1548 bp、1482 bp。系统进化分析显示该基因与细胞色素P450家族中CYP76B亚家族亲缘关系最近。通过对该蛋白进行三维结构预测、同源模建和分子对接,分析了GrG10H蛋白活性口袋中可能参与催化功能发挥的关键氨基酸残基。荧光定量PCR表明GrG10H1基因在叶中表达水平最高,茎次之,根最低。而GrG10H2基因在根和叶中表达水平较高,在茎中几乎不表达。本研究还建立了能够生产龙胆苦苷的愈伤组织细胞培养体系,为进一步解析龙胆苦苷生物合成机制奠定了基础。

A strong animal survival instinct is to approach objects and situations that are of benefit and to avoid *** humans,a large proportion of mental disorders are accompanied by impairments in risk *** of the most important genes involved in mental disorders is disrupted-in-schizophrenia-1(DISC1),and animal models in which this gene has some level of dysfunction show emotion-related ***,it is not known whether DISC1 mouse models have an impairment in avoiding potential *** the present study,we used DISC1-N terminal truncation(DISC1-N^(TM))mice to investigate risk avoidance and found that these mice were impaired in risk avoidance on the elevated plus maze(EPM)and showed reduced social preference in a three-chamber social interaction *** EPM tests,c-Fos expression levels indicated that the nucleus accumbens(NAc)was associated with risk-avoidance behavior in DISC1-N^(TM)*** addition,in vivo electrophysiological recordings following tamoxifen administration showed that the firing rates of fast-spiking neurons(FS)in the NAc were significantly lower in DISC1-N^(TM)mice than in wild-type(WT)*** addition,in vitro patch clamp recording revealed that the frequency of action potentials stimulated by current injection was lower in parvalbumin(PV)neurons in the NAc of DISC1-N^(TM)mice than in WT *** impairment of risk avoidance in DISC1-N^(TM)mice was rescued using optogenetic tools that activated NAcPV ***,inhibition of the activity of NAcPV neurons in PV-Cre mice mimicked the risk-avoidance impairment found in DISC1-N^(TM)mice during tests on the elevated zero *** together,our findings confirm an impairment in risk avoidance in DISC1-N^(TM)mice and suggest that reduced excitability of NAc^(PV) neurons is responsible.
