Identification and functional analysis of CG3526 in spermatogenesis of Drosophila melanogaster

Spermatogenesis is a critical part of reproduction in insects;however,its molecular mechanism is still largely *** this study,we identified a testis-specific gene CG3526 in Drosophila *** analysis showed that CG3526 contains a zinc binding domain and 2 C2H2 type zinc fingers,and it is clustered to the vertebrate really interesting new gene(RING)family E3 ubiquitin-protein *** CG3526 was knocked down by RNA interference(RNAi),the testis became much smaller in size,and the apical tip exhibited a sharp and thin end instead of the blunt and round shape in the control *** importantly,compared to the control flies,only a few mature sperm were present in the seminal vesicle of C587-Gal4>CG3526 RNAi *** staining of the testis from CG3526 RNAi flies showed that the homeostasis of testis stem cell niche was disrupted,cell distribution in the apical tip was scattered,and the process of spermatogenesis was not ***,we found that the phenotype of CG3526 RNAi flies’testis was similar to that of testis of Stat92E RNAi flies,the expression level of CG3526 was significantly downregulated in the Stat92EF06346 mutant flies,and the promoter activity of CG3526 was upregulated by *** together,our results indicated that CG3526 is a downstream effector gene in the JAK-STAT signaling pathway that plays a key role in the spermatogenesis of Drosophila.

We revisit the γ-ray emission above 300 Me V towards the massive star-forming region of Orion B by adopting14 yr observations with the Fermi Large Area Telescope and utilizing the updated software *** extended γ-ray emission region around Orion B is resolved into two components(region Ⅰ and region Ⅱ).The γ-ray spectrum of region I agrees with the predicted γ-ray spectrum assuming the cosmic ray(CR)density is the same as that of Alpha Magnetic Spectrometer(AMS-02)measured ***γ-ray emissivity of region II appears to be deficit at low energy band(E<3 GeV).Through modeling we find that CR densities exhibit a significant deficit below 20 Ge V,which may be caused by a slow diffusion inside the dense *** is probably caused by an increased magnetic field whose strength increases with the gas density.

AIM： To investigate the expression of Neurensin-2 （NRSN2） in hepatocellular carcinoma （HCC） and its prognostic values in predicting survival. METHODS： The expression and prognostic significance of NRSN2 in HCC was examined by performing immunohistochemical analysis using a total of 110 HCC clinical tissue samples, and Western blotting analysis to further confirm the result. RESULTS： Decreased NRSN2 expression was shown in 70.9% cases. Loss of NRSN2 expression in HCC was significantly related to tumor size （P = 0.006）. Larger tumor size was related to negative expression of NRSN2. Patients showing negative NRSN2 expression had a significantly shorter overall survival than those with positive expression （P = 0.008）. Multivariate Cox regression analysis indicated that NRSN2 expression level was an independent factor of survival （P = 0.013）. Western blotting analysis further confirmed decreased expression of NRSN2 in tumor tissues compared with non-tumorous tissues. CONCLUSION： Our study indicated that NRSIV2 could be a tumor suppressor gene for HCC and a candidate biomarker for long-term survival in HCC.

Serum levels of soluble MHC class I-related chain A (sMICA) are related with the prognosis of various types of cancer; however, few studies on the prognostic value of sMICA in hepatocellular carcinoma (HCC) have been reported. In this study, we retrospectively investigated the relationship between sMICA levels and clinical features of advanced HCC, and we assessed the prognostic value of sMICA in advanced HCC. Furthermore, the relationship of serum sMICA levels and natural killer group 2, member D (NKG2D) expression on natural killer (NK) cells was also evaluated. We detected sMICA levels in the serum of 60 advanced HCC patients using enzyme-linked immunosorbent assay (ELISA) and measured expression levels of NKG2D on NK cells using flow cytometry. We found that serum sMICA levels in HCC patients were in the range of 0.10 -6.21 ng/mL. Chi-square analyses showed that sMICA level was significantly related with only tumor size. Survival analysis showed that a high sMICA level was significantly related with poor prognosis among HCC patients. Multivariate analyses indicated that sMICA was an independent prognostic factor. In addition, the levels of CD56+NKG2D+NK cells were within the range of 11.2%-55.4%, and correlation analyses indicated that sMICA level was negatively correlated with the level of NKG2D+NK cells. Our results suggest that serum sMICA levels may be an independent prognostic factor for advanced HCC.

The paper addresses the decentralized optimal control and stabilization problems for interconnected systems subject to asymmetric *** with previous work,a closed-loop optimal solution to the control problem and sufficient and necessary conditions for the stabilization problem of the interconnected systems are given for the first *** main challenge lies in three aspects:Firstly,the asymmetric information results in coupling between control and estimation and failure of the separation ***,two extra unknown variables are generated by asymmetric information(different information filtration)when solving forward-backward stochastic difference ***,the existence of additive noise makes the study of mean-square boundedness an *** adopted technique is proving and assuming the linear form of controllers and establishing the equivalence between the two systems with and without additive noise.A dual-motor parallel drive system is presented to demonstrate the validity of the proposed algorithm.

The previous study conducted by Li(2022)demonstrates that the Goda graph can be adequately represented by a solitary-wave-like form across the entire range of relative water depth,with the exception of the extremely shallow ***,it remains uncertain whether this form is equally effective when applied to test data generated by irregular waves,as the Goda graph was initially developed based on overtopping data from regular wave ***,it is unclear whether this form is suitable for formulating overtopping discharge at composite vertical *** order to address these questions,a 2D overtopping experiment was conducted,incorporating both simple and composite types of vertical walls,with various relative water depths across the entire range,excluding the extremely shallow zone.A novel analysis procedure was developed,which proved to be highly productive and can be considered a general method for data ***,the study yielded two conclusions:(1)the solitary-wave-like form is remarkably effective in formulating overtopping test data generated by irregular waves,regardless of whether the vertical wall is simple or composite,and(2)the resulting formulas exhibit definitely better performance compared with existing formulas.

Background Clinical combination of some traditional Chinese medical herbs, including berberine, with irradiation is demonstrated to improve efficacy of tumor radiotherapy, yet the mechanisms for such effect remain largely unknown. The present study investigated the effect of berberine on apoptosis induced by X-rays irradiation and the relation between this effect and gap junction intercellular communication （GJIC）.Methods The role of gap junctions in the modulation of X-rays irradiation-induced apoptosis was explored by manipulation of connexin （Cx） expression, and gap junction function, using oleamide, a GJIC inhibitor, and *** In transfected HeLa cells, Cx32 expression increased apoptosis induced by X-rays irradiation, while inhibition of gap junction by oleamide reduced the irradiation responses, indicating the dependence of X-rays irradiation-induced apoptosis on GJIC. Berberine, at the concentrations without cytotoxicity, enhanced apoptosis induced by irradiation only in the presence of functional gap *** These results suggest that berberine potentizes cell apoptosis induced by X-rays irradiation, probably through enhancement of gap junction activity.

We investigate the uniqueness problems of meromorphic functions and their difference operators by using a new *** is proved that if a non-constant meromorphic function f shares a non-zero constant and ∞ counting multiplicities with its difference operators Δcf(z) and Δ_(c)^(2)f(z), thenΔcf(z)≡Δ_(c)^(2)f(z).In particular,we give a difference analogue of a result of *** method has two distinct features:(ⅰ) It converts the relations between functions into the corresponding *** makes it possible to deal with the uniqueness problem by linear algebra and combinatorics.(ⅱ) It circumvents the obstacle of the difference logarithmic derivative lemma for meromorphic functions of infinite order,since this method does not depend on the growth of the ***,the idea in this paper can also be applied to the case for several variables.
