Identification and functional analysis of CG3526 in spermatogenesis of Drosophila melanogaster

Spermatogenesis is a critical part of reproduction in insects;however,its molecular mechanism is still largely *** this study,we identified a testis-specific gene CG3526 in Drosophila *** analysis showed that CG3526 contains a zinc binding domain and 2 C2H2 type zinc fingers,and it is clustered to the vertebrate really interesting new gene(RING)family E3 ubiquitin-protein *** CG3526 was knocked down by RNA interference(RNAi),the testis became much smaller in size,and the apical tip exhibited a sharp and thin end instead of the blunt and round shape in the control *** importantly,compared to the control flies,only a few mature sperm were present in the seminal vesicle of C587-Gal4>CG3526 RNAi *** staining of the testis from CG3526 RNAi flies showed that the homeostasis of testis stem cell niche was disrupted,cell distribution in the apical tip was scattered,and the process of spermatogenesis was not ***,we found that the phenotype of CG3526 RNAi flies’testis was similar to that of testis of Stat92E RNAi flies,the expression level of CG3526 was significantly downregulated in the Stat92EF06346 mutant flies,and the promoter activity of CG3526 was upregulated by *** together,our results indicated that CG3526 is a downstream effector gene in the JAK-STAT signaling pathway that plays a key role in the spermatogenesis of Drosophila.

Oomycete, particularly Phytophthora species, causes the most devastating crop diseases, such as potato late blight,and threatens the sustainable crop production worldwide. Our previous studies identified Resistance to Phytophthora parasitica 1(RTP1) as a negative regulator of Arabidopsis resistance to multiple biotrophic pathogens and RTP1 lossof-function plants displayed rapid cell death and reactive oxygen species(ROS) production during early colonization of P. parasitica. In this study, we aim to decipher the mechanism of RTP1-mediated cell death, and identify a member of vaculoar processing enzymes(VPEs), γVPE, playing a role in rtp1-mediated resistance to P. parasitica and cell death occurrence. Our results showed up-regulation of the expression of γVPE as well as increased VPE/caspase 1-like protease activity in P. parasitica-infected rtp1 mutant plants. Besides, we found that the VPE/caspase 1-like protease activity was required for the cell death occurrence in Arabidopsis plants during the infection of P. parasitica as well as rtp1-mediated resistance to P. parasitica. Further pathogenicity assays on either Arabidopsis γvpe mutant plants or leaves of Nicotiana benthamiana with transient overexpression of γVPE demonstrated γVPE could positively affect plant resistance to P. parasitica. Together, our studies suggest that γVPE might function as an important regulator of plant defense and cell death occurrence in response to P. parasitica infection, and VPE/caspase 1-like protease activity is required for rtp1-mediated resistance to P. parasitica.

Destruxins, cyclohexadepsipeptidic mycotoxins isolated from the ento mopathogenic fungus Metarhizium anisopliae, inhibit innate insect immunity. However, their mechanism of action remains unclear. In this study, the effects ofdestruxins on changes in free calcium and hydrogen ions in the hemocytes ofExolontha serrulata, Bombyx mori and the Spodoptera litura SL1 cell line were detected using laser scanning confocal mi croscopy （LSCM）. An instant Ca2＋ influx of hemocytes induced by destruxins A and B （DA and DB） was recorded. The DA/DBdependent Ca2＋ influx was not influenced by the Ca2＋ channel inhibitors 2aminoethoxydiphenyl borane （2APB） and U73122. It also had an apparently different LSCM profile from that of the ionomycindependent Ca2＋ influx. However, the instant Ca2＋ influx was not seen in the SL1 cells; on the contrary, a slow, moderate enhancement of intracellular Ca2＋ was observed. Meanwhile, an instant intracellular free H＋ decrease aroused by DA and DB was found. DB at 20/zmol/L and DA at 690/zmol/L significantly reduced intracellular free H＋ levels. Furthermore, the vacuolar H＋ATPase （VATPase） inhibitor bafilomycin A1 had obvious effects on the decreases ofintracellular free H＋ in hemocytes. These results suggest that the mechanism of DA/DBdependent Ca2＋ influx is perhaps not related to Ca2＋ channels and ionophores; rather, the intracellular free H＋ decrease might be due to VATPase inhibition.

N^(6)-methyladenosine(m^(6)A)RNA is the most abundant modification of mRNA,and has been demonstrated in regulating various post-transcriptional *** studies have shown that m^(6)A methylation plays key roles in sex determination,neuronal functions,and embryonic development in Drosophila and ***,we analyzed transcriptome-wide profile of m^(6)A modification in the embryonic development of the destructive agricultural pest Spodoptera *** found that the 2 key mRNA m^(6)A methyltransferases SfrMETTL3 and SfrMETTL14 have high homologies with other insects and mammals,suggesting that SfrMETTL3 and SfrMETTL14 may have conserved function among different *** methylated RNA immunoprecipitation sequencing analysis,we obtained 46869 m^(6)A peaks representing 8587 transcripts in the 2-h embryos after oviposition,and 41389 m^(6)A peaks representing 9230 transcripts in the 24-h *** addition,5995 m^(6)A peaks were differentially expressed including 3752 upregulated and 2243 downregulated *** analysis with Gene Ontology and Kyoto Encyclopedia of Genes and Genomes suggested that differentially expressed m^(6)A peak-modified genes were enriched in cell and organ development between the 2-and 24-h *** conjoint analysis of methylated RNA immunoprecipitation-seq and RNA-seq data,we found that RNA m^(6)A methylation may regulate the transcriptional levels of genes related to tissue and organ development from 2-to 24-h *** study reveals the role of RNA m^(6)A epigenetic regulation in the embryonic development of ***,and provides new insights for the embryonic development of insects.

Trans-4- (N-2-hydroxyethyl-N-ethyl amino)-4ˊ-(diethyl amino)stilbene (DEAHAS) has been synthesized and characterized by 1H NMR, IR and elemental analysis. Linear absorption, single-photon induced fluorescence and two-photon induced fluorescence are experimentally studied. DEAHAS has an effective two-photon absorption cross-sections ofσ2 = 1.19×10-46 cm4·s/photon at 532 nm by using an open aperture Z-scan technique and exhibits a strong two-photon induced blue fluorescence of 430 nm when pumped with 800 nm laser irradiation.
