Construction of a capsule associated protein 10 gene eukaryotic expression vector for RNA interference and confirmation of biologic relevance

Background The capsule associated protein 10 gene （caplO） is indispensible for the formation of the polysaccharide capsule, and is important in maintaining virulence of the Cryptococcus （C.） neoformans. In this study, we aimed to construct an short hairpin RNA （shRNA） expression vector targeting C. neoformans caplO gene expression and confirm its biologic relevance. Methods A pair of oligonucleotides targeting the cap10 cDNA sequence was designed and synthesized. It was cloned into the plasmid psilencer4.1-CMV neo to construct an eukaryotic shRNA expression vector. The vector was transfected into C. neoformans cells using the LiAc method. The expression of cap10 was assessed by real-time fluorescence quantitative PCR. Groups of C. neoformans cells were incubated with murine macrophage-like J774A.1 cells, and the phagocytic indexes and ratios were determined by the microscopic observation method. Results The expression of cap10 in C. neoformans cells transfected with ps4.1 nee-cap10 （（175 535.00±47 004.00） copies/μl） was lower than that of cells transfected with the empty vector （（512 698.89±32 318.02） copies/μl） and mock transfected cells （（562 931.66±65 928.41） copies/μl）. The average phagocytic ratio and phagocytic index of J774A.1 cells following incubation with C. neoformans were higher for cells transfected with ps4.1 neo-capl0 （0.21±0.02, （19.06±1.66）%） than for the control experimental group （0.08±0.02, （6.57±1.23）%） and the blank experimental group （（0.07±0.01）, （5.89±1.07）%）（P 〈0.05）. Conclusions The cap10 shRNA vector was successfully prepared and transfected into C. neoformans cells. The effect of RNA interference on the expression of the C. neoformans caplO gene is effective, and it can induce phagocytosis of C. neoformans.

Hybrid catalysts based on iron phthalocyanine(FePc)have raised much attention due to their promising applications in electrocatalytic oxygen reduction reaction(ORR).Various hybridization strategies have been developed for improving their activity and ***,the influence of different hybridization strategies on their catalytic performance remains *** this study,Fe Pc was effectively distributed on molybdenum disulfide(MoS_(2))forming Fe Pc-based hybrid catalysts,namely Fe Pc-MoS_(2),Fe Pc*-MoS_(2),and Fe Pc-Py-MoS_(2),respectively,to disclose the related *** direct hybridization,the stacked and highly dispersed Fe Pc on MoS_(2)resulted in Fe Pc-MoS_(2),and Fe Pc*-MoS_(2),respectively,in which the substrate and Fe Pc are mainly bound through van der Waals *** covalent hybridization strategy using pyridyl(Py)as a linker,Fe Pc-Py-MoS_(2)hybrid catalyst was *** and theoretical results disclosed that the linker hybridization of Fe Pc and MoS_(2)facilitated the exposure of Fe-N4 sites,maintained the intrinsic activity of Fe Pc by forming a more dispersed phase and increased the durability via Fe-N bonding,rendering the Fe Pc-Py-MoS_(2)an excellent ORR hybrid *** with van der Waals hybridized Fe Pc-MoS_(2)and Fe Pc*-MoS_(2)in alkaline media,the linker hybridized Fe Pc-Py-MoS_(2)showed an obviously enhanced ORR activity with a half-wave potential(E_(1/2))of 0.88 V vs RHE and an ultralow Tafel slope of 26 m V ***,the Fe Pc-Py-MoS_(2)exhibited a negligible decay of E_(1/2) after 50,000 CV cycles for ORR,showing its superior *** work gives us more insight into the influence of different hybrid strategies on Fe Pc catalysts and provides further guidance for the development of highly efficient and durable ORR catalysts.

【目的】探讨滋金补水膏对慢性阻塞性肺疾病(COPD)模型大鼠的治疗作用及机制。【方法】将96只大鼠随机分为正常组,模型组,滋金补水膏低、中、高剂量组,盐酸氨溴索组、滋金补水膏高剂量+pcDNA组,滋金补水膏高剂量+pcDNA-人浆细胞瘤转化迁移基因1(PVT1)组,每组12只。除正常组外,其他各组大鼠采用气道滴注脂多糖(LPS)联合烟雾暴露法构建COPD模型。建模成功后,进行给药干预。给药结束后,采用酶联免疫吸附分析(ELISA)检测大鼠血清中白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)水平;流式细胞术检测大鼠外周血中辅助性T细胞17/调节性T细胞(Th17/Treg)水平,苏木素-伊红(HE)染色法观察肺组织病理变化,实时定量聚合酶链反应(qRT-PCR)法检测血清和肺组织中长链非编码RNA(lncRNA)PVT1、miR-30b-5p表达,Western Blot法检测肺组织中维甲酸相关孤核受体γt(RORγt)、叉头框蛋白P3(Foxp3)蛋白表达,双荧光素酶报告基因实验验证lncRNA PVT1与miR-30b-5p的关系。【结果】与正常组比较,模型组肺泡间隔变大,支气管周围有大量炎症细胞浸润,Treg细胞比例、miR-30b-5p表达水平、Foxp3蛋白表达水平降低,IL-6、TNF-α水平,Th17细胞比例、Th17/Treg比值,lncRNA PVT1表达水平,RORγt蛋白表达水平升高(均P<0.05);与模型组比较,滋金补水膏低、中、高剂量组及盐酸氨溴索组大鼠肺组织病理损伤减轻、Treg细胞比例、miR-30b-5p表达水平、Foxp3蛋白表达水平升高,IL-6、TNF-α水平,Th17细胞比例,Th17/Treg比值,lncRNA PVT1表达水平,RORγt蛋白表达水平降低(均P<0.05)。lncRNA PVT1靶向调控miR-30b-5p表达。【结论】滋金补水膏可能通过调节lncRNA PVT1/miR-30b-5p轴促进Th17/Treg细胞平衡来抑制COPD大鼠炎症反应。

Effects of Central Administration of Glutamine and Alanine on Feed Intake and Hypothalamic Expression of Orexigenic and Anorexigenic Neuropetides in Broiler Chicks

Different amino acids have been shown to affect feed intake when injected directly into the central nervous system of birds. In the present study, we investigated the effects of L-glutamine and L-alanine on feed intake and the mRNA expression levels of hypothalamic neuropeptides involved in feed intake regulation in broiler chicks. L-Glutamine or L- alanine was intra-cerebroventricularly （ICV） administered to 4-d-old broiler chicks and the feed intake were recorded at various time points. Quantitative PCR was performed to determine the hypothalamic mRNA expression levels of neuropeptide Y （NPY）, agouti related protein （AgRP）, pro-opiomelanocortin （POMC）, melanocortin receptor 4 （MC4R） and corticotropin releasing factor （CRF）. Our results showed that ICV administration of L-glutamine （0.55 or 5.5 pmol） significantly increased feed intake up to 2 h post-administration period and the hypothalamic NPY mRNA expression levels, while it markedly decreased hypothalamic POMC and CRF mRNA expression levels. In contrast, ICV administration of L-alanine （4 lamol） significantly decreased feed intake for the first 0.5 h post-administration period, and reduced the hypothalamic AgRP mRNA expression levels, while it remarkablely enhanced the mRNA expression levels of MC4R and CRF. These findings suggested that L-glutamine and L-alanine could act within the hypothalamus to influence feed intake in broiler chicks, and that both orexigenic and anorexigenic neuropeptide genes might contribute directly to these effects.

Assessment of intraocular pressure measured by Reichert Ocular Response Analyzer,Goldmann Applanation Tonometry,and Dynamic Contour Tonometry in healthy individuals

AIM: To investigate the accuracy of intraocular pressure (IOP) as measured by a Reichert Ocular Response Analyzer (ORA), as well as the relationship between central corneal thickness (CCT) and IOP as measured by ORA, Goldmann applanation tonometry (GAD, and dynamic contour tonometry (DCT). METHODS: A total of 158 healthy individuals (296 eyes) were chosen randomly for measurement of IOP. After CCT was measured using A-ultrasound (A-US), IOP was measured by ORA, GAT, and DCT devices in a randomized order. The IOP values acquired using each of the three tonometries were compared, and the relationship between CCT and IOP values were analyzed separately. Two IOP values, Goldmann-correlated IOP value (IOPg) and corneal-compensated intraocular pressure (IOPcc), were got using ORA. Three groups were defined according to CCT: 1) thin cornea (CCT580 mu m) groups. RESULTS: In normal subjects, IOP measurements were 14.95 2.99mmHg with ORA (IOPg), 15.21+/- 2.77mmHg with ORA(IOPcc),1522+/- 2.77mmHg with GAT,and 15.49+/- 2.56mmHg with DCT. Mean differences were 0.01+/- 2.29mmHg between IOPcc and GAT (P >0.05) and 0.28+/- 2.20mmHg between IOPcc and DC (P >0.05). There was a greater correlation between IOPcc and DCT (r =0.946, P =0.000) than that between IOPcc and GAT (r=0.845, P=0.000). DCT had a significant correlation with GAT (r=0.854, P=0.000). GAT was moderately correlated with CCT (r=0.296, P580 mu m) showed the least significant correlation between IOPcc and GAT (r=0.859, 0.000); while the thin cornea group (CCT< 520 mu m) had the most significant correlation between IOP

冷循环液温度对微波消融离体猪肝消融灶形态的影响

目的观察冷循环液温度对微波消融(MWA)离体猪肝消融灶形态的影响。方法将20块新鲜离体猪肝组织随机分为冰水冷循环组(A组)及常温冷循环组(B组),分别以不同消融功率(50、60、70 W)及消融时间(1、5 min)于各亚组,即A1与B1(50 W,1 min)、A2与B2(50 W,5 min)、A3与B3(60 W,1 min)、A4与B4(60 W,5 min)、A5与B5(70 W,1 min)及A6与B6(70 W,5 min)亚组内各消融10个靶区。对比A、B组相同条件亚组间及组内消融灶形态,包括消融灶纵向直径(LD)、横向直径(TD)、球形指数(RI)及体积(V)。结果相同消融功率及时间下,A组各亚组消融灶LD均小于B组(P均0.05)。冷循环液温度对2组消融灶TD(F=1.125)和V(F=3.332)的主效应均不显著(P均≥0.05)。相同冷循环液温度条件下,A1与A2、A3与A4、A5与A6各亚组间消融灶形态各参数差异均有统计学意义(P均<0.05),B组各亚组间亦然。结论以恒定消融功率及时间行MWA消融离体猪肝时,采用冰水冷循环可使消融灶形态更接近于球形;且随消融时间延长,消融灶范围越大,则RI越高。

The Main Biological Hazards in Animal Biosafety Level 2 Facilities and Strategies for Control

Concern about the biological hazards involved in microbiological research, especially research involving laboratory animals, has increased in recent years. Working in an animal biosafety level 2 facility （ABSL-2）, commonly used for research on infectious diseases, poses various biological hazards. Here, the regulations and standards related to laboratory biosafety in China are introduced, the potential biological hazards present in ABSL-2 facilities are analyzed, and a series of strategies to control the hazards are presented.

Global stability of an epidemic model for HIV-TB co-infection with infection-age

A nonlinear mathematical HIV TB model with infection-age is proposed in this paper. The basic reproduction numbers according to HIV and TB are respectively determined whether one of the diseases dies out or persists. The local and global stability of the disease-free and dominated equilibria are discussed by employing integral semigroup theory and Lyapunov functionals. The persistence of the system is also obtained by the persistence theories of the systems. The simulation illustrates the theoretical results.
