Plasma Homocysteine and Gene Polymorphisms Associated with the Risk of Hyperlipidemia in Northern Chinese Subjects

Objective To examine the relationship between occurrence of hyperlipidemia, plasma homocysteine and polymorphisms of methylenetetra hydrofolate reductase （MTHFR） gene and methionine synthase （MS） gene. Methods A total of 192 hyperlipidemia patients were selected and divided into hypercholesterolemia group, hypertriglyceridemia group, and combined hyperlipidemia group. Another 208 normal individuals were selected as control. Total plasma homocysteine （tHcy） concentration was measured by high-performance liquid chromatography （HPLC）. Lipid profiles were measured for all subjects The polymorphisms of MTHFR gene C677T and MS gene A2756G were analyzed by PCR-RFLP. Results The tHcy concentration in the combined hyperlipidemia patients was significantly higher than that in the control （15.95μmol/L vs 13.43 μmol/L, P〈0.05）. The prevalence of hyperhomocysteinemia （HHcy） in the combined hyperlipidemia group was significantly higher than that in the control （42.2% vs 23.0%, P=0.015）, with the odds ratio （OR） of 3.339 （95%CI： 1.260-8.849）. The hyperlipidemia patients with HHcy had a higher concentration of total cholesterol （TC） than that in the normal tHcy patients （5.67±0.95 mmol/L vs 5.47±0.92 retool/L, P=0.034）. There was no significant difference in genotype or allele frequencies of MTHFR C677T between the hyperlipidemic and control groups. The hyperlipidemia patients with MTHFR CT/TT genotype had a higher concentration of triglyceride （TG） than those with CC genotype （2.24±1.75 mmol/L vs 1.87±0.95 mmol/L, P〈0.05）. Individuals with CT/TT genotype had a higher concentration of tHcy than those with 677CC genotype both in the hyperlipidemia group （12.61±1.24μmol/L vs 11.20±1.37 μmol/L, P〈0.05） and in the control group （14.04±1.48 μmol/L vs 12.61±1.24 μmol/L, P〈0.05）. The percentage of MS 2756 GG/AG genotype in the combined hyperlipidemia group was significantly higher than that in the control （26.7% vs 13.0%, P=0.012）, with the OR of 3.121 （95%C1： 1.288-7.65/）. Alpha-decay properties of nuclei around neutron magic numbers

By combining experimental α-decay energies and half-lives, the α-particle preformation factor for nuclei around neutron magic numbers N of 126, 152, and 162 were extracted using the two-potential approach. The nuclei around the shell closure were more tightly bound than adjacent nuclei. Additionally, based on the WS4 mass model (Wang et al., Phys. Lett.B 734, 215 (2014)), we extended the two-potential approach to predict the α-decay half-lives of nuclei around N values of178 and 184 with Z of 119 and 120. We believe that our findings will serve as guidelines for future experimental studies.

SCFP, a novel fiber-specific promoter in cotton

To investigate the expression pattern of GhSCFP which was isolated from cotton fiber cDNA library, a 1006 bp upstream fragment of the gene was cloned by chromosome walking and fused to GUS and GFP respectively. Histochemical GUS and GFP fluorescence analysis revealed that the expression of the report genes driven by the promoter sequence was detectable only in outer layer cells during the seed development in the transgentic tobaccos. In transgenic cotton, strong GUS activity was observed in spherical protrusions on 0 dpa (days post anthesis) ovule surface, and in the 2―36 dpa fiber cells, while no GUS signals were detected in the root, leaves, stem, corolla, anther and stigma. Our data demonstrated that GhSCFP upstream sequence is a cotton fiber-specific promoter and this promoter will be useful in the molecular research on fiber cell development and in cotton fiber improvements by genetic modification.

Protective effects of cariporide on endothelial dysfunction induced by high glucose

Aim:To explore the effects of cariporide,a selective sodium-hydrogen antiporterinhibitor,on endothelial dysfunction induced by high ***:Acetyl-choline (ACh)-induced endothelium-dependent relaxation (EDR),sodium nitro-prusside (SNP)-induced endothelium-independent relaxation and biochemical pa-rameters including malondialdehyde (MDA),superoxide dismutase (SOD),andnitric oxide (NO) were measured in rat isolated ***:A 6-h incubationof aortic rings with high glucose (44 mmol/L) resulted in a significant inhibitionof EDR,but had no effects on endothelium-independent *** the 6-hincubation of aortic rings in the co-presence of cariporide (0.01,0.1,and 1 μmol/L)with high glucose,cariporide prevented the inhibition of EDR caused by highglucose in concentration-dependent ***,high glucose decreasedSOD activity and contents of NO,and increased MDA concentration in *** (1 μmol/L) significantly resisted the decrease of NO contentand SOD activity,and elevation of MDA concentration caused by high glucose inaortic *** (44 mmol/L) or cariporide (1 μmol/L) alone had no ef-fect on EDR,endothelium-independent relaxation and biochemical ***:Cariporide significantly prevented endothelial dysfunction inducedby high *** mechanisms of endothelial dysfunction induced by highglucose may involve the activation of sodium-hydrogen antiporter and the gen-eration of oxygen-free radicals,but it is not related to the change of osmolarity.

Relationship Between Polymorphism of Cystathionine beta Synthase Gene and Congenital Heart Disease in Chinese Nuclear Families

Objective To study the relationship between polymorphism of cystathionine beta synthase （CBS） gene and development of congenital heart disease （CHD）. Methods One hundred and twenty-seven CHD case-parent triads were recruited from Liaoning Province as patient group, and 129 healthy subjects without family history of birth defect were simultaneously recruited as control group together with their biological parents. For all subjects the polymorphism of CBS gene G919A locus was examined by PCR-ARMS method, Results The frequencies of three genotypes （w/w, w/m, and m/m） in control group were 27.2%, 58,4%, and 14.4%, respectively, with no significant difference in gender. A significant difference in the allele frequency was found between CHD patients and controls, the wild allele frequency was 67,9% in patients and 55.7% in controls CHD parents＇ genotype distribution was significantly different from that in controls. Further comparison of each type of CHD showed that genotype frequencies in several CHD subtypes were significantly different from those in their corresponding controls. The results of TDT analysis showed that no allele transmission disequilibrium existed in CHD nuclear families. Conclusions CBS gene G919A mutation is associated with the development of CHD, and the mutated allele may decrease the risk of CHD.

Analysis of causes of pain after kyphoplasty for osteoporotic vertebral compression fractures

For the treatment of osteoporotic vertebral compression fractures （OVCFs）, percutaneous kyphoplasty （PKP）enables rapid pain relief and spinal stabilization, as well as correction of kyphosis. Although serious complications such as pulmonary embolism and spinal cord and nerve damage may occur,1 recurrence of pain after PKP is unlikely to require medical attention. The present study analyzed the causes of post-operative residual pain by reviewing the clinical data of 16 from 121 patients with OVCFs who underwent PKP from August 2005 to April 2009. METHODS

Genome-wide and Interaction Linkage Scan for Nonsyndromic Cleft Lip with or without Cleft Palate in Two Multiplex Families in Shenyang,China

Objectives To identify the loci involved in nonsyndromic cleft lip with or without cleft palate （NSCL/P） in Northern Chinese people in Shenyang by using genomewide and interaction linkage *** Two multiplex families in Shenyang from North China were ascertained through probands with NSCL/*** of every member was drawn for DNA extraction and *** were available for 382 autosomal short tandem repeat （STR） markers from the ABI Prism Linkage Mapping Set version *** between markers and NSCL/P was assessed by 2-point parametric LOD scores,multipoint heterogeneity parametric LOD scores （HLODs）,and multipoint nonparametric linkage score （NPL）.Results The initial scan suggested linkage on Chromosomes 1,2,and *** subsequent fine mapping,1q32-q42 showed a maximum multipoint LOD score of 1.9（empirical P=0.013） and an NPL score of 2.35 （empirical P=0.053）.For 2p24-p25,the multipoint NPL increased to 2.94 （empirical P=0.007）.2-locus interaction analysis obtained a maximum NPL score of 3.73 （P=0.00078） and a maximum LOD score of 3 for Chromosome 1 （at 221 cM） and Chromosome 2 （at 29 cM）.Conclusion Both parametric and nonparametric linkage scores greatly increased over the initial linkage scores on 1q32-q42,suggesting a susceptibility locus in this *** linkage gave a strong evidence for a candidate region on chromosome *** superiority of 2-locus linkage scores compared to single-locus scores gave additional evidence for linkage on 1q32-q42 and 2p24-p25,and suggested that certain genes in the two regions may contribute to NCSL/P risks with interaction.

Palaeowind reconstruction from Lower Cretaceous wavedominated depositional records of the Tanan Depression,Northeast Asia

Palaeowind regime is an important component for reconstructing palaeoclimate and understanding ancient global *** applied the beach-ridge thickness technique(BTT)and the longshore-bar thickness technique(LTT)to reconstruct the Early Cretaceous palaeowind velocities based on depositional records of Tanan Depression in Northeast *** thickness of beach ridges and nearshore bars is the most important parameter for applying the BTT and the LTT *** identified 22 beach ridges and 23 nearshore bars based on drilling cores and wireline-log data in the Tanan *** velocities calculated by using the LTT method ranged from 10.84 to 13.66 m/s,showing a narrower range than those calculated by using the BTT method(7.58–15.93 m/s).We further analyzed the underlying factors causing the difference between the results derived from these two methods based on the sedimentary process of the beach ridge and the nearshore *** reconstructed wind regime was assumed to be the summer monsoon originating from the Mongol-Okhotsk Ocean before it had been completely enclosed,which provides valuable insight into the understanding of the atmospheric circulation in Northeast Asia during the Early Cretaceous.

Cold stress profoundly affects plant growth and development and is a key factor affecting the geographic distribution and evolution of *** have evolved adaptive mechanisms to cope with cold ***,through the genomic analysis of Arabidopsis,three Brassica species and 17 other representative species,we found that both cold-related genes(CRGs)and their collinearity were preferentially retained after polyploidization followed by genome instability,while genome-wide gene sets exhibited a variety of other expansion *** coldrelated regulatory network was increased in Brassicaceae genomes,which were recursively affected by *** combining our findings regarding the selective retention of CRGs from this ecological genomics study with the available knowledge of cold-induced chromosome doubling,we hypothesize that cold stress may have contributed to the success of polyploid plants through both increasing polyploidization and selectively maintaining CRGs during *** hypothesis requires further biological and ecological exploration to obtain solid supporting evidence,which will potentially contribute to understanding the generation of polyploids and to the field of ecological genomics.
