Profibrotic role of the SOX9-MMP10-ECM biosynthesis axis in the tracheal fibrosis after injury and repair

Fibroblast activation and extracellular matrix(ECM)deposition play an important role in the tracheal abnormal repair process and *** a transcription factor,SOX9 is involved in fibroblast activation and ECM ***,the mechanism of how SOX9 regulates fibrosis after tracheal injury remains *** investigated the role of SOX9 in TGF-b1-induced fibroblast activation and ECM deposition in rat tracheal fibroblast(RTF)***9 overexpression adenovirus(Ad-SOX9)and siRNA were transfected into RTF *** found that SOX9 expression was up-regulated in RTF cells treated with ***9 overexpression activated fibroblasts and promoted ECM *** SOX9 inhibited cell proliferation,migration,and ECM deposition,induced G2 arrest,and increased apoptosis in RTF ***-seq and chromatin immunoprecipitation sequencing(ChIP-seq)assays identified MMP10,a matrix metalloproteinase involved in ECM deposition,as a direct target of SOX9,which promotes ECM degradation by increasing MMP10 expression through the Wnt/b-catenin signaling ***,in vivo,SOX9 knockdown ameliorated granulation proliferation and tracheal fibrosis,as manifested by reduced tracheal *** conclusion,our findings indicate that SOX9 can drive fibroblast activation,cell proliferation,and apoptosis resistance in tracheal fibrosis via the Wnt/b-catenin signaling *** SOX9eMMP10 eECM biosynthesis axis plays an important role in tracheal injury and *** SOX9 and its downstream target MMP10 may represent a promising therapeutic approach for tracheal fibrosis.

ATP6AP2,a regulator of LRP6/β-catenin protein trafficking,promotes Wnt/β-catenin signaling and bone formation in a cell type dependent manner

Wnt/β-catenin signaling is critical for various cellular processes in multiple cell types,including osteoblast(OB)differentiation and *** how Wnt/β-catenin signaling is regulated in OBs remain ***6AP2,an accessory subunit of V-ATPase,plays important roles in multiple cell types/organs and multiple signaling ***,little is known whether and how ATP6AP2 in OBs regulates Wnt/β-catenin signaling and bone *** we provide evidence for ATP6AP2 in the OB-lineage cells to promote OB-mediated bone formation and bone homeostasis selectively in the trabecular bone *** knocking out(CKO)ATP6AP2 in the OB-lineage cells(Atp6ap2^(Ocn-Cre))reduced trabecular,but not cortical,bone formation and bone *** and cellular biochemical studies revealed that LRP6 and N-cadherin were reduced in ATP6AP2-KO BMSCs and OBs,but not *** in vitro and in vivo studies revealed impairedβ-catenin signaling in ATP6AP2-KO BMSCs and OBs,but not osteocytes,under both basal and Wnt stimulated conditions,although LRP5 was decreased in ATP6AP2-KO osteocytes,but not *** cell biological studies uncovered that osteoblastic ATP6AP2 is not required for Wnt3a suppression ofβ-catenin phosphorylation,but necessary for LRP6/β-catenin and N-cadherin/β-catenin protein complex distribution at the cell membrane,thus preventing their *** of activeβ-catenin diminished the OB differentiation deficit in ATP6AP2-KO *** together,these results support the view for ATP6AP2 as a critical regulator of both LRP6 and N-cadherin protein trafficking and stability,and thus regulatingβ-catenin levels,demonstrating an un-recognized function of osteoblastic ATP6AP2 in promoting Wnt/LRP6/β-catenin signaling and trabecular bone formation.

lignans are a powerful weapon for plants to resist stresses and have diverse bioactive functions to protect human *** the mechanisms of stereoselective biosynthesis and response to stresses of lignans is important for the guidance of plant ***,we identified the complete pathway to stereoselectively synthesize antiviral(-)-lariciresinol glucosides in Isatis indigotica roots,which consists of three-step sequential stereoselective enzymes DIR1/2,PLR,and ***1 was further identified as the key gene in respoJanuary 2024nse to stresses and was able to trigger stress defenses by mediating the elevation in lignan ***,the phytohormone-responsive ERF transcription factor LTF1 colocalized with DIR1 in the cell periphery of the vascular regions in mature roots and helped resist biotic and abiotic stresses by directly regulating the expression of *** systematic results suggest that DIR1 as the first common step of the lignan pathway cooperates with PLR and UGT71B2 to stereoselectively synthesize(-)-lariciresinol derived antiviral lignans in *** roots and is also a part of the LTF1-mediated regulatory network to resist *** conclusion,the LTF1-DIR1 module is an ideal engineering target to improve plant Defenses while increasing the content of valuable lignans in plants.

Crashworthiness Design and Multi-Objective Optimization of Bionic Thin-Walled Hybrid Tube Structures

Thin-walled structures are widely used in cars due to their lightweight construction and energy-absorbing ***,issues such as high initial stress and lowenergy-absorbing efficiency *** study proposes a novel energy-absorbing structure inwhich a straight tube is combinedwith a conical tube and a bamboo-inspired bulkhead structure is *** configuration allows the conical tube to flip outward first and then fold together with the straight *** deformation mode absorbs more energy and less peak force than the conical tube sinking and flipping *** finite element numerical simulation,the specific energy absorption capacity of the structure is increased by 26%compared to that of a regular circular cross-section ***,the impact resistance of the bionic straight tapered tube structure is further improved through multi-objective optimization,promoting the engineering application and lightweight design of hybrid cross-section tubes.

Effects of Number of Bleed Holes on Shock-Wave/Boundary-Layer Interactions in a Transonic Compressor Stator

An extensive numerical investigation is conducted to characterize the flow separation control in a transonic compressor cascade with a porous *** bleed holes are arranged on the suction surface in a single row,two staggered rows and three staggered *** each bleed scheme,five bleed pressure ratios are examined at an inlet Mach number of *** results indicate that the aerodynamic performance of the cascade is significantly improved by the porous *** the single-row scheme,the maximum reduction in total pressure losses is 57%.For the two-staggered-row and three-staggered-row schemes,there is an optimal bleed pressure ratio of 1.0,and the maximum reductions in total pressure loss are 68% and 75%,*** low loss in the cascade is due to the well-controlled boundary *** new local supersonic region created by the bleed hole is the key reason for the improved boundary *** vortex induced by side bleeding provides another mechanism for delaying flow *** the bleed holes could create multiple local supersonic regions,which reduce the range of the adverse pressure gradient that the boundary layer needs to *** is the reason why cascades with more bleed holes perform better.

Modulation mechanism of electron energy dissipation on superlubricity based on fluorinated 2D ZIFs

Electron energy dissipation is an important energy dissipation pathway that cannot be ignored in friction ***-dimensional zeolite imidazole frameworks(2D ZIFs)and fluorine doping strategies give 2D Zn-ZIF and 2D Co-ZIF unique electrical properties,making them ideal materials for studying electron energy dissipation *** this paper,based on the superlubricity modulation of 2D fluoridated ZIFs,the optimal tribological properties are obtained on the 2D F-Co-ZIF surface,with the friction coefficient as low as *** experiments,density functional theory(DFT)simulation,and fluorescence detection are used to explain the mechanism of fluorine doping regulation of tribological properties from the two stages,namely energy transfer and energy ***,the energy will transfer into the friction system through the generation of electron–hole pairs under an external excitation,and release by radiation and non-radiation energy dissipation *** reduces energy transfer by altering the electronic properties and band structures of ZIFs,and slows down the charge transfer by enhancing the shielding efficiency,thus slowing the non-radiative energy dissipation rate during the energy release *** insights not only help us better understand the role of fluorine doping in improving tribological properties,but also provide a new way to further explore the electron energy dissipation pathway during friction.

Neutrophil-to-lymphocyte ratio associated with renal function in type 2 diabetic patients

BACKGROUND Type 2 diabetes mellitus(T2DM)is a leading risk factor for the development and progression of chronic kidney disease(CKD).However,an accurate and con-venient marker for early detection and appropriate management of CKD in in-dividuals with T2DM is *** studies have demonstrated a strong correlation between the neutrophil-to-lymphocyte ratio(NLR)and ***-theless,the predictive value of NLR for renal damage in type 2 diabetic patients remains *** study included 1040 adults aged 65 or older with T2DM from Shanghai's Community Health Service *** total number of neutrophils and lym-phocytes was detected,and NLR levels were *** was defined as an estimated glomerular filtration rate≤60 mL/min/1.73 m².Participants were di-vided into four groups based on NLR *** clinical data and biochemical characteristics were compared among groups.A multivariate logistic regression model was used to analyze the association between NLR levels and *** Significant differences were found in terms of sex,serum creatinine,blood urea nitrogen,total cholesterol,and low-density lipoprotein cholesterol among patients with T2DM in different NLR groups(Plinical and pathological parameters(P=0.0001,odds ratio=1.41,95%confidence intervals:1.18-1.68).CONCLUSION An elevated NLR in patients with T2DM is associated with higher prevalence of CKD,suggesting that it could be a marker for the detection and evaluation of diabetic kidney disease.
