Social infrastructures such as dams are likely to be exposed to high risk of terrorist and military attacks,leading to increasing attentions on their vulnerability and catastrophic consequences under such events.This paper tries to develop advanced deep learning approaches for structural dynamic response prediction and dam health diagnosis.At first,the improved long short-term memory(LSTM)networks are proposed for data-driven structural dynamic response analysis with the data generated by a single degree of freedom(SDOF)and the finite numerical simulation,due to the unavailability of abundant practical structural response data of concrete gravity dam under blast events.Three kinds of LSTM-based models are discussed with the various cases of noise-contaminated signals,and the results prove that LSTM-based models have the potential for quick structural response estimation under blast loads.Furthermore,the damage indicators(i.e.,peak vibration velocity and domain frequency)are extracted from the predicted velocity histories,and their relationship with the dam damage status from the numerical simulation is established.This study provides a deep-learning based structural health monitoring(SHM)framework for quick assessment of dam experienced underwater explosions through blastinduced monitoring data.

Microglia serve as brain-resident myeloid cells that affect cerebral development, ischemia, neurodegeneration, and neuro-viral infection. MicroRNAs play a key role in central nervous system disease through post-transcriptional regulation. Indeed, evidence shows that microRNAs are one of the most important regulators mediating microglial activation, polarization, and autophagy, and subsequently affecting neuroinflammation and the outcome of central nervous system disease. In this review, we provide insight into the function of microRNAs, which may be an attractive strategy and influential treatment for microglia-related central nervous system dysfunction. Moreover, we comprehensively describe how microglia fight against central nervous system disease via multiple functional microRNAs.

Heterogenous nuclear ribonucleoprotein G is down-regulated in the spinal cord of the Tg(SOD1*G93A)1Gur(TG)amyotrophic lateral sclerosis mouse model.However,most studies have only examined heterogenous nuclear ribonucleoprotein G expression in the amyotrophic lateral sclerosis model and heterogenous nuclear ribonucleoprotein G effects in amyotrophic lateral sclerosis pathogenesis such as in apoptosis are unknown.In this study,we studied the potential mechanism of heterogenous nuclear ribonucleoprotein G in neuronal death in the spinal cord of TG and wild-type mice and examined the mechanism by which heterogenous nuclear ribonucleoprotein G induces apoptosis.Heterogenous nuclear ribonucleoprotein G in spinal cord was analyzed using immunohistochemistry and western blotting,and cell proliferation and proteins(TAR DNA binding protein 43,superoxide dismutase 1,and Bax)were detected by the Cell Counting Kit-8 and western blot analysis in heterogenous nuclear ribonucleoprotein G siRNA-transfected PC12 cells.We analyzed heterogenous nuclear ribonucleoprotein G distribution in spinal cord in the amyotrophic lateral sclerosis model at various time points and the expressions of apoptosis and proliferation-related proteins.Heterogenous nuclear ribonucleoprotein G was mainly localized in neurons.Amyotrophic lateral sclerosis mice were examined at three stages:preonset(60-70 days),onset(90-100 days)and progression(120-130 days).The number of heterogenous nuclear ribonucleoprotein G-positive cells was significantly higher in the anterior horn of the lumbar spinal cord segment of TG mice at the preonset stage than that of control group but lower than that of the control group at the onset stage.The number of heterogenous nuclear ribonucleoprotein G-positive cells in both central canal and surrounding gray matter of the whole spinal cord of TG mice at the onset stage was significantly lower than that in the control group,whereas that of the lumbar spinal cord segment of TG mice was significantly higher than that in the control gr

Obesity plays a crucial role in the development of non-alcoholic fatty liver disease (NAFLD). However, the underlying mechanism for the pathogenesis of obesity-associated NAFLD remains largely obscure. Although the “multiple hit” theory provides a more accurate explanation of NAFLD pathogenesis, it still cannot fully explain precisely how obesity causes NAFLD. The liver is the key integrator of the body's energy needs, receiving input from multiple metabolically active organs. Thus, recent studies have advocated the “multiple crosstalk” hypothesis, highlighting that obesity-related hepatic steatosis may be the result of dysregulated “crosstalk” among multiple extra-hepatic organs and the liver in obesity. A wide variety of circulating endocrine hormones work together to orchestrate this “crosstalk”. Of note, with deepening understanding of the endocrine system, the perception of hormones has gradually risen from the narrow sense (i.e. traditional hormones) to the broad sense of hormones as organokines and exosomes. In this review, we focus on the perspective of organic endocrine hormones (organokines) and molecular endocrine hormones (exosomes), summarizing systematically how the two types of new hormones mediate the dialogue between extra-hepatic organs and liver in the pathogenesis of obesity-related NAFLD.

Recent advances in CRISPR-based genome editing technology and its applications in cardiovascular research

The rapid development of genome editing technology has brought major breakthroughs in the fields of life science and medicine. In recent years, the clustered regularly interspaced short palindromic repeats(CRISPR)-based genome editing toolbox has been greatly expanded, not only with emerging CRISPR-associated protein(Cas) nucleases, but also novel applications through combination with diverse effectors. Recently, transposon-associated programmable RNA-guided genome editing systems have been uncovered, adding myriads of potential new tools to the genome editing toolbox. CRISPR-based genome editing technology has also revolutionized cardiovascular research. Here we first summarize the advances involving newly identified Cas orthologs, engineered variants and novel genome editing systems, and then discuss the applications of the CRISPR-Cas systems in precise genome editing, such as base editing and prime editing. We also highlight recent progress in cardiovascular research using CRISPR-based genome editing technologies, including the generation of genetically modified in vitro and animal models of cardiovascular diseases(CVD) as well as the applications in treating different types of CVD. Finally, the current limitations and future prospects of genome editing technologies are discussed.

Coronary artery spasm （CAS） is found not only in Asian but also in white patients. CAS can lead to angina pecto- ris, myocardial infarction, ventricular arrhythmias and sud- den cardiac death. But CAS diagnosis is not necessarily easy, and it also remains highly challenging to therapy and prevention. We present a spontaneous extensive-CAS case with typical clinical features of acute myocardial infarction, despite the regular vasodilator treatment.

Rare-earth hexaborides(REB_(6))are vital raw materials for cathode materials and high temperature structural ceramics that are widely applied as high-frequency electron tubes and ceramics adaptive for extreme environment,respectively.In this work,single phase substitutional solid solution REB_(6)(LaB_(6),La_(0.75)Ce_(0.25)B_(6).La_(0.5)Ce_(0.5)B_(6),La_(0.25)Ce_(0.75)B_(6) and CeB_(6))powders were prepared with the raw materials of La_(2)O_(3),CeO_(2),B_(4)C and Al powders,after calcining at 1773 K for 4 h and the following alkaline leaching.All substitutional solid solution products have homogeneous distributions of La and Ce in particles.Through microscopic morphology analysis,it is discovered that the formation of solid solution is beneficial for reducing the particle size of product,relative to LaB_(6) and CeB_(6).Moreover,Al flux plays an important role in decarbonizing reaction,and C contents of all products are below 0.4 wt%.

We investigate the optimization of linear impulse systems with the reinforcement learning based adaptive dynamic programming(ADP)method.For linear impulse systems,the optimal objective function is shown to be a quadric form of the pre-impulse states.The ADP method provides solutions that iteratively converge to the optimal objective function.If an initial guess of the pre-impulse objective function is selected as a quadratic form of the pre-impulse states,the objective function iteratively converges to the optimal one through ADP.Though direct use of the quadratic objective function of the states within the ADP method is theoretically possible,the numerical singularity problem may occur due to the matrix inversion therein when the system dimensionality increases.A neural network based ADP method can circumvent this problem.A neural network with polynomial activation functions is selected to approximate the pre-impulse objective function and trained iteratively using the ADP method to achieve optimal control.After a successful training,optimal impulse control can be derived.Simulations are presented for illustrative purposes.

Hypothermia is an important protective strategy against global cerebral ischemia following cardiac arrest.However,the mechanisms of hypothermia underlying the changes in different regions and connections of the brain have not been fully elucidated.This study aims to identify the metabolic nodes and connection integrity of specific brain regions in rats with global cerebral ischemia that are most affected by hypothermia treatment.18F-fluorodeoxyglucose positron emission tomography was used to quantitatively determine glucose metabolism in different brain regions in a rat model of global cerebral ischemia established at 31–33℃.Diffusion tensor imaging was also used to reconstruct and explore the brain connections involved.The results showed that,compared with the model rats established at 37–37.5℃,the rat models of global cerebral ischemia established at 31–33℃had smaller hypometabolic regions in the thalamus and primary sensory areas and sustained no obvious thalamic injury.Hypothermia selectively preserved the integrity of the anterior forebrain mesocircuit,exhibiting protective effects on the brain during the global cerebral ischemia.The study was approved by the Institutional Animal Care and Use Committee at Capital Medical University(approval No.XW-AD318-97-019)on December 15,2019.

An improved rheo-optic in situ synchronous measurement system was employed to investigate the gelation behaviour and mechanism of waxy crude oil emulsions. By combining transmitted natural light and reflected polarized light microscopy, a multiangle composite light source was built to achieve the simultaneous observation of wax crystals and emulsified water droplets, as well as their dynamic aggregation process. Main outcomes on the microscopic mechanism were obtained by developed microscopic image processing method. It was found that the microstructure of W/O waxy crude oil emulsion has the evolution of “individual structure--homogeneous aggregate structure--heterogeneous coaggregate structure--floc structure” during the static cooling, which results in the four stages during gelation process. Different from previous studies, the aggregation of emulsified water droplets was found to be more significant and contributes to the formation and development of the wax crystals-emulsified water droplets coaggregate, which plays a decisive role in the further evolution of the gelled microstructure. Time series microscopic images show the dynamic aggregation of emulsified water droplets and wax crystals. Two different aggregation behaviours between wax crystals and water droplets were observed. That wax crystals can not only embed in gaps between adjacent water droplets and enhance the structure, but also surround the outside of the water droplets and continue to grow resulting in the interconnection of different coaggregates to form a larger floc structure. In addition, correlation between viscoelasticity and microstructure evolution of waxy crude oil emulsions of different water contents was discussed. With increasing water contents, the microstructure is changed from wax crystal flocculation structure as the main skeleton and the emulsified water droplets embedded in it, into the aggregation of emulsified water droplets occupying the main position.
