Effects of long non-coding RNA Opa-interacting protein 5 antisense RNA 1 on colon cancer cell resistance to oxaliplatin and its regulation of micro RNA-137

BACKGROUND The incidence of colon cancer(CC)is currently high,and is mainly treated with chemotherapy.Oxaliplatin(L-OHP)is a commonly used drug in chemotherapy;however,long-term use can induce drug resistance and seriously affect the prognosis of patients.Therefore,this study investigated the mechanism of Opainteracting protein 5 antisense RNA 1(OIP5-AS1)on L-OHP resistance by determining the expression of OIP5-AS1 and micro RNA-137(miR-137)in CC cells and the effects on L-OHP resistance,with the goal of identifying new targets for the treatment of CC.AIM To study the effects of long non-coding RNA OIP5-AS1 on L-OHP resistance in CC cell lines and its regulation of miR-137.METHODS A total of 114 CC patients admitted to China-Japan Union Hospital of Jilin University were enrolled,and the expression of miR-137 and OIP5-AS1 in tumor tissues and corresponding normal tumor-adjacent tissues was determined.The influence of OIP5-AS1 and miR-137 on the biological behavior of CC cells was evaluated.Resistance to L-OHP was induced in CC cells,and their activity was determined and evaluated using cell counting kit-8.Flow cytometry was used to analyze the apoptosis rate,Western blot to determine the levels of apoptosisrelated proteins,and dual luciferase reporter assay combined with RNA-binding protein immunoprecipitation to analyze the relationship between OIP5-AS1 and miR-137.RESULTS OIP5-AS1 was up-regulated in CC tissues and cells,while miR-137 was downregulated in CC tissues and cells.OIP5-AS1 was inversely correlated with miR-137(P<0.001).Silencing OIP5-AS1 expression significantly hindered the proliferation,invasion and migration abilities of CC cells and markedly increased the apoptosis rate.Up-regulation of miR-137 expression also suppressed these abilities in CC cells and increased the apoptosis rate.Moreover,silencing OIP5-AS1 and up-regulating miR-137 expression significantly intensified growth inhibition of drug-resistant CC cells and improved the sensitivity of CC cells to LOHP.OIP5-AS1 targetedly inhibited miR-

.To the Editor:Lower respiratory tract infections(LRTIs)are among the deadliest diseases and remain one of the leading causes of death over the last 15 years,especially in low-income countries,and are of great concern to immunosuppressed individuals.Infections located in the peripheral lung field(PLF)are located within the outerthird elliptical regions around the hilum on computedtomography(CT)scans.[1]They are surrounded by normal lung parenchyma and are unlikely to be visualized and accurately diagnosed by bronchoscopy.

Co-Fe quantum dots coupled with ultrathin g-C_(3)N_(4) nanosheets as efficient and stable photo-Fenton catalysts

The Fenton reaction has been widely used in the environmental remediation.However,the sharp decline of photo-Fenton catalysts activity under neutral conditions is still an urgent problem to be solved.This study reports a Co/Fe bimetallic oxide quantum dots-coupled g-C_(3)N_(4)nanosheets(CoFeO QDs/g-C_(3)N_(4) NSs) composites with efficient degradation of organic pollutants under neutral conditions.Under the photo-Fenton condition,rhodamine B(RhB) degradation efficiency reached 98.32% within 90min for CoFeO QDs/g-C_(3)N_(4) NSs composites.The formed heterojunction between CoFeO QDs and g-C_(3)N_(4) NSs achieves enhanced charge transfer and efficient charge separation.Co-Fe bimetals make g-C_(3)N_(4) NSs easier to excite the production of·OH by H_(2)O_(2),achieving excellent degradation efficiency and cycle stability for organic pollutants in a wide pH range.Therefore,CoFeO QDs/g-C_(3)N_(4)NSs composites can be used as efficient and stable photoFenton catalysts to degrade organic contaminants in practical applications.

Effects of dissolved oxygen and nutrients from the Kuroshio on hypoxia off the Changjiang River estuary

The intrusion of the Kuroshio into the East China Sea(ECS)aff ects the development of hypoxia off the Changjiang(Yangtze)River estuary;however,quantitative analysis of its impacts is lacking.In this study,the Regional Ocean Modeling Systems(ROMS)model coupled with the Carbon,Silicate and Nitrogen Ecosystem(CoSiNE)model was used to investigate the relative importance of dissolved oxygen(DO)and diff erent nutrients(silicate,nitrate,and phosphate)in the Kuroshio on hypoxia in the ECS.Results show that changes in DO concentrations in the Kuroshio modify the distribution and intensity of hypoxia through direct onshore transport by hydrodynamic processes.An increase in Kuroshio DO concentration by 25%or 50%would result in a decrease of the maximum hypoxia extent(MHE)in the ECS by 76%or 86%,respectively,while a 25%decrease in Kuroshio DO would increase the MHE by up to 219%.The contribution of DO in the Taiwan Strait is almost negligible.In contrast to Kuroshio DO,nutrients aff ect hypoxia in the ECS through onshore transport by hydrodynamic and biochemical processes.Changes in phosphate and nitrate concentrations by 25%in the Kuroshio would change the MHE by up to 30%and 18%,respectively,accompanied by apparent changes in surface chlorophyll-a concentrations.The eff ect of silicate on hypoxia is negligible because a 25%change in silicate concentrations in the Kuroshio would result in less than 1%change in the MHE.Our results reveal a hierarchical rank of importance for environmental variables in the Kuroshio(i.e.,DO>phosphate>nitrate>silicate)in modifying the development of hypoxia in the ECS.

Nickel-rich LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2)(NCM811)is regarded as the promising cathode for lithium-ion batteries(LIBs).However,the challenges such as safety issues and poor cycling performance have seriously hindered its commercial applications.In order to overcome these difficulties,there has been extensive research and development of electrolyte modifications for high-energy-density LIBs with Ni-rich cathodes.Herein,this review introduces the research progress based on solvent additives,salt type additives and other electrolytes for LIBs with NCM811cathode materials and discusses how they control the interface stability.In particular,some recommendations for further modification of enhancing electrolyte stability and improving NCM811 electrochemical properties are summarized and proposed,which put forward new design rules for the screening and customizing ideal electrolyte additives for high performance NCM811 cathode-based LIBs.

Microstructure and Mechanical Behavior of Mg-Based Bimetal Plates with High Formability Sleeve by Co-extrusion

In this study,microstructure and mechanical behavior of two types of Mg-based bimetal plates with a high formability sleeve were systematically studied,with a great emphasis on the effect of the interface characteristic and the sleeve fraction on the plasticity of composite plates.The rule of mixtures(ROM)for elongation was also addressed.The results show that when there is no or thin diffusion layer with thickness of about 3μm,Mg-based bimetal plates have a good plasticity with elongation of about 19-24%,and the ROM predicted elongations are very close to the experimental ones.In contrast,with a diffusion layer about 95-155μm thick,Mg-based bimetal plates exhibit a poor plasticity with elongation of about 11-17%,and the experimental elongations largely deviate from the ROM predictions.The plasticity of Mg-based bimetal plates increases with increasing sleeve fraction.This study provides new insights on the plastic deformation of Mg-based bimetal composites with a high formability sleeve.

Spatiotemporal dynamics of cell abundance,colony size and intracellular toxin concentrations of pelagic and benthic Microcystis in Lake Caohai,China

Lake Caohai has experienced extensive Microcystis blooms in recent years,and to improve its water quality,the local government carried out a series of water control measures.To better understand the dynamics of both pelagic and benthic Microcystis and their characteristics in Lake Caohai,we conducted a 1-year investigation from December 2015to December 2016 to gain a seasonal outlook on the distribution and dynamics of cell abundance,colony size and intracellular microcystins(MCs)of Microcystis.The results indicated that the Microcystis bloom occupied primarily the northeastern region and then moved gradually from lakeshore to lake center.The perennial southwesterly winds and the water inflow from northeast to southwest in Lake Caohai determined the spatiotemporal distribution of pelagic Microcystis.Benthic Microcystis was mainly distributed in the northeastern region in summer,occupied the lake center in autumn and then occupied the southeastern region in winter,determined by the sedimentation of pelagic Microcystis and the death of benthic Microcystis.Small colonies(20-60μm)overwintered more easily in both water column and sediment.The concentrations of intracellular toxin of benthic Microcystis were observed to be significantly higher than those of pelagic Microcystis.This might be because Microcystis synthesized large amount of MCs to acclimate to an unfavorable benthic environment.This knowledge on the dynamics of Microcystis expands our understanding of mechanisms underpinning the formation of Microcystis blooms.

Two putative fatty acid synthetic genes of BgFas3 and BgElo1 are responsible for respiratory waterproofing in Blattella germanica

Water retention is critical for physiological homeostasis and survival in terrestrial insects. While deposition of hydrocarbons on insect cuticles as a key measure for water conservation has been extensively investigated, we know little about other mechanisms for preventing water loss in insects. Here, we report two fatty acid synthetic genes that are independent of hydrocarbon production but crucial for water retention in the German cockroach Blattella germanica (L.). First, an integument enriched fatty acid elongase gene (BgElo1) was identified as a critical gene for desiccation resistance in B. germanica;however, knockdown of BgElo1 surprisingly failed to cause a decline in cuticular lipids. In addition, RNA interference (RNAi)-knockdown of an upstream fatty acid synthase gene (BgFas3) showed a similar phenotype, and transmission electron microscopy analysis revealed that BgFas3- or BgElo1-RNAi did not affect cuticle architecture. Bodyweight loss test showed that repression of BgFas3 and BgElo1 significantly increased the weight loss rate, but the difference disappeared when the respiration was closed by freeze killing the cockroaches. A water immersion test was performed, and we found that BgFas3- and BgElo1-RNAi made it difficult for cockroaches to recover from drowning, which was supported by the upregulation of hypoxia-related genes after a 10-h recovery from drowning. Moreover, a dyeing assay with water-soluble Eosin Y showed that this was caused by the entry of water into the respiratory system. Our research suggests that BgFas3 and BgElo1 are required for both inward and outward waterproofing of the respiratory system. This study benefits the understanding of water retention mechanisms in insects.

Development and validation of a predictive model for patients with post-extubation dysphagia

BACKGROUND:Swallowing disorder is a common clinical symptom that can lead to a series of complications,including aspiration,aspiration pneumonia,and malnutrition.This study aimed to investigate risk factors of post-extubation dysphagia(PED)in intensive care unit(ICU)patients with endotracheal intubation,and to develop a risk-predictive model for PED,which could serve as an assessment tool for the prevention and control of PED.METHODS:Patients retrospectively selected from June to December 2021 in a tertiary hospital served as the derivation cohort.Patients recruited from the same hospital from March to June 2022served as the external validation cohort for the predictive model.We used a combination of variable screening and least absolute shrinkage and selection operator(LASSO)regression to select the most useful candidate predictors and checked the multicollinearity of independent variables using the variance inflation factor method.Multivariate logistic regression analysis was performed to calculate the odds ratio(OR;95%confidence interval[95%CI])and P-value for each variable to predict diagnosis.The screened risk factors were introduced into R software to build a nomogram model.The performance of the model,including discrimination ability,calibration,and clinical benefit,was evaluated by plotting the receiver operating characteristic(ROC),calibration,and decision curves.RESULTS:A total of 305 patients were included in this study.Among them,235 patients(53PED vs.182 non-PED)were enrolled in the derivation cohort,while 70 patients(17 PED vs.53 nonPED)were enrolled in the validation cohort.The independent predictors included age,pause of sedatives,level of consciousness,activities of daily living(ADL)score,nasogastric tube,sore throat,and voice disorder.These predictors were used to establish the predictive nomogram model.The model demonstrated good discriminative ability,and the area under the ROC curve(AUC)was 0.945(95%CI 0.904-0.970).Applying the predictive model to the validation cohort demonstrated good discr

Fish oil alleviates liver injury induced by intestinal ischemia/reperfusion via AMPK/SIRT-1/autophagy pathway

AIM To evaluate whether fish oil(FO) can protect liver injury induced by intestinal ischemia/reperfusion(I/R) via the AMPK/SIRT-1/autophagy pathway.METHODS Ischemia in wistar rats was induced by superior mesenteric artery occlusion for 60 min and reperfusion for 240 min. One milliliter per day of FO emulsion or normal saline was administered by intraperitoneal injection for 5 consecutive days to each animal. Animals were sacrificed at the end of reperfusion. Blood andtissue samples were collected for analyses. AMPK, SIRT-1, and Beclin-1 expression was determined in lipopolysaccharide(LPS)-stimulated HepG2 cells with or without FO emulsion treatment.RESULTS Intestinal I/R induced significant liver morphological changes and increased serum alanine aminotransferase and aspartate aminotransferase levels. Expression of p-AMPK/AMPK, SIRT-1, and autophagy markers was decreased whereas tumor necrosis factor-α(TNF-α) and malonaldehyde(MDA) were increased. FO emulsion blocked the changes of the above indicators effectively. Besides, in LPS-stimulated HepG2 cells, small interfering RNA(siRNA) targeting AMPK impaired the FO induced increase of p-AMPK, SIRT-1, and Beclin-1 and decrease of TNF-α and MDA. SIRT-1 siRNA impaired the increase of SIRT-1 and Beclin-1 and the decrease of TNF-α and MDA.CONCLUSION Our study indicates that FO may protect the liver against intestinal I/R induced injury through the AMPK/SIRT-1/autophagy pathway.
