Radioiodine therapy for castration-resistant prostate cancer following prostate-specific membrane antigen promoter-mediated transfer of the human sodium iodide symporter

Radioiodine therapy, the most effective form of systemic radiotherapy available, is currently useful only for thyroid cancer because of the thyroid-specific expression of the human sodium iodide symporter （hNIS）. Here, we explore the efficacy of a novel form of gene therapy using prostate-specific membrane antigen （PSMA） promoter-mediated hNIS gene transfer followed by radioiodine administration for the treatment of castration-resistant prostate cancer （CRPC）. The androgen-dependent C33 LNCaP cell line and the androgen-independent C81 LNCaP cell line were transfected by adenovirus. PSMA promoter-hNIS （***-hNIS） or ***-hNIS containing the cytomegalovirus promoter （***-hNIS） or a control virus. The iodide uptake was measured in vitro. The in vivo iodide uptake by C81 cell xenografts in nude mice injected with an adenovirus carrying the hNIS gene linked to PSMA and the corresponding tumor volume fluctuation were assessed. Iodide accumulation was shown in different LNCaP cell lines after ***-hNIS and ***-hNIS infection, but not in different LNCaP cell lines after *** （***） infection. At each time point, higher iodide uptake was shown in the C81 cells infected with ***-hNIS than in the C33 cells （P 〈 0.05）. An in vivo animal model showed a significant difference in 1311 radioiodine uptake in the tumors infected with ***-hNIS, ***-hNIS and control virus （P 〈 0.05） and a maximum reduction of tumor volume in mice infected with ***-hNIS. These results show prostate-specific expression of the hNIS gene delivered by the PSMA promoter and effective radioiodine therapy of CRPC by the PSMA promoter-driven hNIS transfection.

Percent free prostatic-specific antigen （%fPSA） has been introduced as a tool to avoid unnecessary biopsies in patients with a serum PSA level of 4.0-10.0 ng ml^-1, however, it remains controversial whether %fPSA is effective in PSA range of 10.1-20.0 ng ml^-1 in both Chinese and Western population. In this study, the diagnostic performance of %fPSA and serum PSA in predicting prostate cancer （PCa） and high-grade PCa （HGPCa） was analyzed in a multi-center biopsy cohort of 5915 consecutive Chinese patients who underwent prostate biopsy in 22 hospitals across China from January 1, 2010 to December 31, 2013. The indication for biopsy was PSA〉4.0 ng ml^-1 or/and suspicious digital rectal examination. Total and free serum PSA determinations were performed by three types of electrochemiluminescence immunoassays with recalibration to the World Health Organization standards. The diagnostics accuracy of PSA, %fPSA and %fPSA in combination with PSA （%fPSA ＋ PSA） was determined by the area under the receivers operating characteristic curve （AUC）. %fPSA was more effective than PSA in men aged ≥60 years old. The AUC was 0.584 and 0.635 in men aged ≥60 years old with a PSA of 4.0-10.0 ng ml^-1 and 10.1-20.0 ng ml^-1, respectively. The AUC of %fPSA was superior to that of PSA in predicting HGPCa in patients ≥60 years old in these two PSA range. Our results indicated that %fPSA is both statistically effective and clinical applicable to predict prostate biopsy outcome in Chinese patients aged ≥60 years old with a PSA of 4.0-10.0 ng ml^-1 and 10.1-20.0 ng ml^-1.

Neutron-sensitive microchannel plates(nMCPs)have applications in neutron detection,including energy spectrum measurements,neutron-induced cross sections,and neutron imaging.10B-doped MCPs(B-MCPs)have attracted significant attention owing to their potential for exhibiting a high neutron detection efficiency over a large neutron energy *** spatial and temporal resolutions are useful for neutron energy-resolved ***,their practical applications still face many technical *** this study,a B-MCP with 10 mol%10B was tested for its response to wide-energy neutrons from eV to MeV at the Back-n white neutron source at the China Spallation Neutron *** neutron detection efficiency was calibrated at 1 eV,which is approximately 300 times that of an ordinary MCP and indicates the success of 10 B *** factors that caused the reduction in the detection efficiency were simulated and *** neutron energy spectrum obtained using B-MCP was compared with that obtained by other measurement methods,and showed very good consistency for neutron energies below tens of *** response is more complicated at higher neutron energy,at which point the elastic and nonelastic reactions of all nuclides of B-MCP gradually become *** is beneficial for the detection of neutrons,as it compensates for the detection efficiency of B-MCP for high-energy neutrons.

There is considerable potential for integrating transarterial chemoembolization(TACE),programmed death-(ligand)1(PD-[L]1)inhibitors,and molecular targeted treatments(MTT)in hepatocellular carcinoma(HCC).It is necessary to investigate the therapeutic efficacy and safety of TACE combined with PD-(L)1 inhibitors and MTT in real-world *** this nationwide,retrospective,cohort study,826 HCC patients receiving either TACE plus PD-(L)1 blockades and MTT(combination group,n=376)or TACE monotherapy(monotherapy group,n=450)were included from January 2018 to May *** primary endpoint was progression-free survival(PFS)according to modified *** secondary outcomes included overall survival(OS),objective response rate(ORR),and *** performed propensity score matching approaches to reduce bias between two *** matching,228 pairs were included with a predominantly advanced disease *** PFS in combination group was 9.5 months(95%confidence interval[CI],8.4-11.0)versus 8.0 months(95%CI,6.6-9.5)(adjusted hazard ratio[HR],0.70,P=0.002).OS and ORR were also significantly higher in combination group(median OS,19.2[16.1-27.3]vs.15.7 months[13.0-20.2];adjusted HR,0.63,P=0.001;ORR,60.1%vs.32.0%;Ptients with predominantly advanced HCC in real-world practice,with an acceptable safety profile.

The Large sky Area Multi-Object Fiber Spectroscopic Telescope（LAMOST） general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio（SNR） ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters： late A,FGK-type stars with high quality spectra（1 061 918 entries）, A-type stars（100 073 entries）, and M-type stars（121 522 entries）. This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. A description of the FITS structure of spectral files and parameter catalogs is also provided.

We present the observational results from a detailed timing analysis of the black hole candidate EXO 1846-031 during its outburst in 2019 with the observations of Insight-HXMT,NICER and *** outburst can be classified roughly into four different ***-C quasi-periodic oscillations(QPOs)observed by NICER(about 0.1-6 Hz)and Insight-HXMT(about 0.7-8 Hz)are also reported in this ***,we study various physical quantities related to QPO *** QPO rms-frequency relationship in the energy band 1-10 keV indicates that there is a turning pointing in frequency around2 Hz,which is similar to that of GRS 1915+105.A possible hypothesis for the relationship above may be related to the inclination of the source,which may require a high inclination to explain *** relationships between QPO frequency and QPO rms,hardness,total fractional rms and count rate have also been found in other transient sources,which can indicate that the origin of type-C QPOs is non-thermal.

In the archive of the Ground Wide Angle Camera(GWAC),we found 43 white light flares from 43 stars,among which,three are sympathetic or homologous flares,and one of them also has a quasi-periodic pulsation with a period of 13.0±1.5 *** these 43 flare stars,there are 19 new active stars and 41 stars that have available TESS and/or K2 light curves,from which we found 931 stellar *** also obtained rotational or orbital periods of 34 GWAC flare stars,of which 33 are less than 5.4 days,and ephemerides of three eclipsing binaries from these light *** with low resolution spectra from LAMOST and the Xinglong 2.16 m telescope,we found that L_(Hα)/L_(bol) are in the saturation region in the rotation-activity *** the LAMOST medium-resolution spectrum,we found that Star#3(HAT 178-02667)has double Hαemissions which imply it is a binary,and two components are both active *** stars have flare frequency distributions(FFDs)from TESS and/or K2 light *** FFDs show that the flares detected by GWAC can occur at a frequency of 0.5to 9.5 yr^(-1).The impact of flares on habitable planets was also studied based on these FFDs,and flares from some GWAC flare stars may produce enough energetic flares to destroy ozone layers,but none can trigger prebiotic chemistry on their habitable planets.

Study on the effect of moxibustion at “Shénquè”(神阙 CV 8) on the immune system of rats taking long-term exhaustive exercise

Objective To explore the effect of moxibustion at ＂Shénquè＂（神阙 CV 8） on the immune system of rats taking long-term exhaustive *** Thirty male SD rats were randomly divided into blank group,model group and moxibustion group with 10 rats in each *** the rats in blank group,all the rats were established into long-term exhaustive models through swimming again and *** moxibustion at CV 8 for 15 min was carried out in the rats in moxibustion group after *** treatment was conducted once every other day,and treatment for 10 times was *** exhaustive duration for each time was recorded,the levels of serum Ig A,Ig G,Ig M and ACP were detected 24 h after the 10 th exhaustive exercise,and the histomorphological changes of spleen were *** was adopted for detecting the Ig A level,and ELISA method was used for detecting the levels of Ig G,Ig M and *** The common exhaustive duration in moxibustion group was longer than that in model group,however,the time difference was statistically significant after the 5th exhaustive exercise（P0.01）.The levels of serum IgG and IgM in model group were lower than that in blank group（all P0.01）,and the serum ACP level in model group was higher than that in blank group（P0.01）,indicating that the splenic histomorphology was *** levels of serum Ig G and Ig M in moxibustion group were higher than that in model group（all P0.01）,and the serum ACP level in moxibustion group was lower than that in model group,showing that the splenic histomorphology was *** Moxibustion at CV 8 can improve the levels of immunoglobulin（Ig G,Ig A,Ig M） of tired body for a long time,decrease the ACP level,and repair the damaged spleen tissues.
