Proteomics to display tissue repair opposing injury response to LPS-induced liver injury

AIM: To examine the protein expression alterations in liver injury/repair network regulation as a response to gut-derived lipopolysaccharide (LPS) treatment, in order to anticipate the possible signal molecules or biomarkers in signaling LPS-related liver ***: Male BALB/c mice were treated with intraperitoneal (i.p.) LPS (4 mg/kg) and sacrificed at 0, 6, 24 and 30 h to obtain livers. The livers were stained with hematoxylin and eosin for histopathologic analyses. Total liver protein was separated by two-dimensional gel electrophoresis (2-DE). The peptide mass of liver injury or repair related proteins were drawn up and the protein database was searched to identify the ***: Observations were as follows: (1) TRAIL-R2 was down regulated in livers of LPS-treated mice. TNFAIP1 was significantly up regulated at 6 h, then down- regulated at 24, 30 h with silent expression during senescent stage.(2) The amount of metaxin 2 and mitochondria import inner membrane translocase subunit TIM8a (TIMM8A) was increased upon treatment with LPS, (3) P34 cdc2 kinase was significantly up-regulated 30 h after LPS administration with silent expression during senescent, 6, 24 h treated stage. (4) The amount of proteasome activator 28 alpha subunit (PA28), magnesium dependent protein phosphatase(MDPP) and lysophospholipase 2 was decreased 6 h after LPS treatment but recovered or up-regulated 24 and 30 h after LPS ***: LPS-treated mouse liver displaying a timedependent liver injury can result in expression change of some liver injury or repair related proteins.

Study on relationship between expression level and molecular conformations of gene drugs targeting to hepatoma cells in vitro

AIM: To increase exogenous gene expression level bymodulating molecular conformations of targeting gene ***: The full length cDNAs of both P40 and P35subunits of human interleukin 12 were amplified throughpolymerase chain reaction (PCR) and cloned into eukaryoticexpressing vectors pcDNA3.1(±) to construct plasmids of P(+)/IL-12, P(+)/P40 and P(-)/P35. These plasmids werecombined with ASOR-PLL to form two targeting gene drugs[ASOR-PLL-P(+)/IL-12 and ASOR-PLL-P(+)/P40 + ASOR-PLL-P(-)/P35] in optimal ratios. The conformations of these twodrugs at various concentrations adjuvant were examined underelectron microscope (EM) and the drugs were transfected intoHepG2 (ASGr+) cells. Semi-quantitative reverse transcriptionpolymerase chain reaction (RT-PCR) was performed withtotal RNA extracted from the transfected cells to determinethe hiL12 mRNA transcript level. The hiL12 protein in thecultured supernatant was measured with enzyme-linkedimmunosorbent assay (ELISA) 48 hours after ***: Targeting gene drugs, whose structures weregranular and circle-like and diameters ranged from 25 nmto 150 nm, had the highest hIL-12 expression level. ThehIL-12 expression level in the group co-transfected withASOR-PLL-P(+)/P4o and ASOR-PLL-P(-)/P35 was higher thanthat of ASOR-PLL-P(+)/IL-12 transfected ***: The molecular conformations of targetinggene drugs play an important role in exogenous geneexpression level, the best structures are granular and circle-like and their diameters range from 25 nm to 150 nm. Thesizes and linking styles of exogenous genes also have someeffects on their expression level.

COMBINED SURGICAL THERAPY FOR HEPATOLITHIASIS

Objective To evaluate the combined surgical therapy for calculi of intrahepatic duct. Methods One hundred and eight cases of hepatolithiasis treated in our hospital from January 1986 to September 2003 were summarized and analyzed retrospectively. The primary surgery included 57 cases of biliary tract exploration and cholangiolithotomy, 26 cases of cholangiojejunostomy, and 25 cases of partial hepatectomy. Of these cases, 156 operations were performed totally. There were 36 patients undergoing two or more operations for cholangiolithiasis problem. Fiberoptic choledochoscopy was employed to examine and remove the residual stones during and after operation in all cases. Results The residual stone rates were 57.89% (33/57), 26.92% (7/26), and 24.0% (6/25) for biliary tract exploration and cholangiolithotomy, cholangiojejunostomy, and partial hepatectomy, respectively. Fiberoptic choledochoscopy was utilized pre- and post-operatively for each patient, which was performed 2 to 9 times on each case. Following the combined treatment, the rate of residual stone after operation decreased to 12.96% (14/108). Conclusions The combined surgical therapy is valuable for resolving the problem of hepatolithiasis in most of the patients. Regarding the clinical outcomes of different surgical procedures, partial hepatectomy is superior to cholangiojejuno- stomy or biliary tract exploration and cholangiolithotomy. Fiberoptic choledochoscopy is also important to reduce the occurrence of residual stones and the rate of reoperation.

The Use of the Ratio between the Veno-arterial Carbon Dioxide Difference and the Arterial-venous Oxygen Difference to Guide Resuscitation in Cardiac Surgery Patients with Hyperlactatemia and Normal Central Venous Oxygen Saturation

Background：After cardiac surgery,central venous oxygen saturation （ScvO2） and serum lactate concentration are often used to guide resuscitation;however,neither are completely reliable indicators of global tissue *** observational study aimed to establish whether the ratio between the veno-arterial carbon dioxide and the arterial-venous oxygen differences （P（v-a）CO2/C（a-v）O2） could predict whether patients would respond to resuscitation by increasing oxygen delivery （DO2）.Methods：We selected 72 patients from a cohort of 290 who had undergone cardiac surgery in our institution between January 2012 and August *** selected patients were managed postoperatively on the Intensive Care Unit,had a normal ScvO2,elevated serum lactate concentration,and responded to resuscitation by increasing DO2 by 〉10%.As a consequence,48 patients responded with an increase in oxygen consumption （VO2） while VO2 was static or fell in ***：At baseline and before resuscitative intervention in postoperative cardiac surgery patients,a P（v-a）CO2/C（a-v）O2 ratio ≥1.6 mmHg/ml predicted a positive VO2 response to an increase in DO2 of〉1 0% with a sensitivity of 68.8% and a specificity of 87.5%.Conclusions：P（v-a）CO2/C（a-v）O2 ratio appears to be a reliable marker of global anaerobic metabolism and predicts response to DO2 ***,patients likely to benefit from resuscitation can be identified promptly,the P（v-a）CO2/C（a-v）O2 ratio may,therefore,be a useful resuscitation target.

随着信息时代的迅速发展,云计算数据访问安全已经成为了用户最关心的问题。身份认证技术是确保参与者在开放的网络环境中实现安全通信的一种重要手段,如何利用身份认证技术为云环境安全保驾护航,成为学者研究的热点。文中通过公钥基础设施(Public Key Infrastructure,PKI)颁发CA证书以在不同云服务间建立信任,将多个采用身份密码体制(Identity-Based Encryption,IBE)的云联合起来;采用分层身份加密体系,引入共享密钥技术,通过选取成环结构,提出一种PKI-IBE混合认证模型方案,并对方案的安全性进行分析,从理论上证明了云环境下PKIIBE(Public Key Infrastructure-Identity-Based Encryption)同层成环模型提供服务的可行性。同时文中设计了一种基于该模型的签密技术,通过公私密钥对实现云内认证以及跨云认证。安全性理论证明与性能分析表明,该方案在计算量稍增加的前提下,保证了足够的安全性,更加满足云环境下的用户分属不同云域的认证以及用户安全访问的需求,有效解决了云环境中数据访问的安全问题。
