该研究通过探讨坛紫菜多糖浸泡的南美白对虾仁在微冻过程中肌原纤维蛋白氧化特性和结构特性的变化规律,从蛋白质角度揭示坛紫菜多糖抗冻保鲜的潜在机制。将虾仁分别用5 mg/mL坛紫菜多糖,蒸馏水(空白对照)和5 mg/mL焦磷酸钠(阳性对照)浸泡处理后于-3℃微冻贮藏,每5 d测定指标。结果表明,与对照组相比,坛紫菜多糖浸泡虾仁可显著延缓总巯基含量下降,防止羰基含量和表面疏水性上升,阻碍平均粒径增加和Zeta电位绝对值的下降(P<0.05),抑制肌原纤维蛋白的氧化和聚集。圆二色光谱分析表明,坛紫菜多糖显著抑制肌原纤维蛋白α-螺旋相对含量的降低(P<0.05)。内源荧光光谱分析表明,坛紫菜多糖能缓解肌原纤维蛋白中发色基团暴露,维持蛋白空间结构稳定性。扫描电镜结果显示,坛紫菜多糖能保持虾肌肉组织微观结构的完整性。该研究为坛紫菜多糖作为新型绿色无磷抗冻保鲜剂添加到水产品冷冻过程中保护蛋白质特性提供理论依据。

以琼胶和海藻酸钠为成膜基质,通过共混和层层自组装两种方法制备了琼胶/海藻酸钠共混膜和双层膜,并引入交联剂柠檬酸和阿魏酸,通过测定复合膜的机械强度、耐水性、热稳定性、傅里叶变换红外光谱(Fourier transform infrared spectrometer,FT-IR)和微观结构等指标,探究了不同制膜方式结合交联剂对复合膜性能的影响。结果表明,与共混膜相比,双层膜表现出更优异的机械性能和耐水性,添加交联剂后,膜的水蒸气透过率(P<0.05)显著降低,拉伸强度、热稳定性和不透明度提高。其中柠檬酸交联的双层膜综合性能最佳,即拉伸强度51.57 MPa,断裂伸长率26.02%,水溶性24.42%,溶胀率101.67%。FT-IR分析表明柠檬酸通过与琼胶和海藻酸钠发生酯化反应形成CO进行共价交联,而阿魏酸通过与成膜基质形成氢键进行非共价交联。扫描电镜分析表明琼胶和海藻酸钠的相容性良好,各膜表面均平整、光滑,而添加交联剂后,膜截面褶皱减少,膜结构更加紧密。由此表明,层层自组装技术和交联剂的结合使用可以制备出综合性能更好的复合膜。

Fermented foods are a potential source to produce novel dipeptidyl peptidase-IV inhibitory peptides(D4IPs).In this study,the fermented mandarin fish(Chouguiyu)was used to screen D4IPs and their formation mechanism was studied by metagenomics and peptidomics.A total of 400 D4IPs with DPP-IV inhibition structure and high hydrophobicity were *** correlation network map showed that Lactococcus,Bacillus,Lysobacter,Pelagivirga,Kocuria,Escherichia,Streptococcus,and Peptostreptococcus were significantly correlated with the most *** stable D4IPs,including KAGARALTDAETAT,GEKVDFDDIQK,VVDADEMYLKGK,and GQKDSYVGDEAQ were respectively from the precursor proteins parvalbumin,troponin,myosin,and actin,and were mainly formed by the hydrolysis of subtilisin(EC 3.***.21.62),aspartic proteinase(EC 3.***.23.1),thermolysin(EC 3.***.24.27),oligopeptidase B(EC 3.***.21.83),and proteinase P1(EC 3.***.21.96)from Bacillus,Kocuria,Lysobacter,Lactococcus,and *** inhibition mainly resulted from the hydrogen bond and salt bridge between D4IPs and DPP-IV *** study provides important information on the proteases and related microbial strains to directionally prepare D4IPs in Chouguiyu.

This study aimed to isolate and identify the novel potential Xanthine oxidase(XOD)inhibitory peptides derived from Trachinotus ovatus hydrolysate(TOH)(hydrolysis by neutral protease),and investigate their structure-function ***,TOH was separated to four fractions(F1–F4)by gel filtration chromatography and F4 was chosen with higher XOD inhibitory rate(85.16%±0.566,25 mg/mL)and lower molecular weight(95.73%,<1000 Da).F4 was further characterized by reversed-phase high performance liquid chromatography(RP-HPLC)coupled with Nano-HPLC-MS/MS and four peptides(FPAW,LLPW,WLLP and FHLP)were identified with IC50 value of 3.81±0.18 mM,4.17±0.12 mM,43.06±0.73 mM,and over 50.00 mM *** can be concluded that Trp-containing peptides especially located in C-terminal could inhibit XOD ***,the XOD-inhibiting actions of these peptides involvedπ-πstacking(Phe914 and Phe1009 in XOD),hydrogen bonding via molecular ***-Burk equation showed the inhibition type of FPAW was a mixed *** dichroism(CD)spectra indicated FPAW could combine and change the secondary structure of ***,two peptides of FPAW and LLPW might have potential to be used to alleviate hyperuricemia in the future.

Provenance of Conglomerate and Sandstone from Early Permian Shoushangou Formation in Xi Ujimqin,Inner Mongolia:Implications for Understanding Paleo-Asian Ocean Subduction

Quality deterioration induced by microbial community in Litopenaeus vannamei during superchilling storage were demonstrated in this study. In this study， 6 microorganisms were identified as the biomarkers by 16S rDNA sequencing and 71 differential metabolites were identified by untargeted metabolomics. The correlation analysis between microorganisms and differential metabolites revealed that Oceanicaulis， Aliihoeflea， Prauserella， Chelativorans and Pseudoalteromonas had the potential to be the important microorganisms of deterioration of superchilled shrimp. Moreover， the spearman correlation analysis revealed that L* value had highly significant correlation with Shewanella (P < 0.01)， and TBA value， TVC value total sulfhydryl content， TCA-soluble peptide and Ca2+-ATPase activity showed significantly correlation with Oceanicaulis， Prauserella， Chelativorans， Aliihoeflea and Pseudoalteromonas (P < 0.05). This study offers a better understanding for mechanisms behind quality degradation induced by microorganisms， which may help the quality maintaining of Pacific white shrimp during superchilling storage.

It has been revealed experimentally that astrocytes can participate in synaptic transmission by modulating and responding to the release of neurotransmitters with calcium elevations. Researches suggest that seizure-like discharges(SDs) or seizure-like firings(SFs) in neurons, characterizing neurological disorder, may arise locally in restricted areas(focal area) and then propagate throughout the brain. But the underlying mechanism remains unclear. To study the possible role astrocytes playing in the SDs propagation, we construct a minimal neuron-astrocyte network model by connecting a neurons chain and an astrocytes *** focal area is modelled by an IP3 reservoir which provides persistent IP3 out-flux. The study suggests that calcium wave propagation in astrocytes determines the propagation of SDs in the connected neurons. On the other hand, SDs in neurons allows the calcium wave propagates longer distance in the astrocytes, which suggests the mutually cooperating of astrocytes and neurons in accomplishing SD propagation. Furthermore, once SDs propagate and occupy the neuron network, it could not be terminated by recovery of the focal area. The results may imply that treatment of brain disorders should not only focus on local area but the whole neuron network.

Comparative Study on Antineoplastic Activity of Polysaccharide from Four Kinds of Sea Cucumber

The antineoplastic activity of polysaccharide was investigated in Stichopus chlorontus, Isostichopus badionotus, Stichopus horrens and Holothuria lessoni massin. Crude polysaccharide was prepared with enzyme hydrolyzation method and purified by anion exchange chromatography using DEAE-sepharose fast flow column. The effect of polysaccharide on cells apoptosis of SiHa and U87 was examined with cell counting kit-8 colorimetry method. Western blotting was used to analyze related proteins of cellular apoptosis including p53 and Bcl-2. Results showed that there were two main components in each sea cucumber polysaccharide, which could be eluted down by 1.0 mol/LNaCl solution. The four types of polysaccharide in the second component were named as SC-2, IB-2, HLM-2 and SH-2, respectively. They were used for comparing the antineoplastic activity. Results showed that SC-2, IB-2, HLM-2 and SH-2 could promote apoptosis of U87 and SiHa cells. SH-2 and HLM-2 were selected for the subsequent experiment to explore the additional effect of U87 and SiHa cells, The protein expressions of Bcl-2 and p53 decreased considerably with the increase of polysaccharide concentration in U87 cells. In SiHa cells, protein expressions of Bcl-2 and high dosage group of p53 decreased significantly, whereas no obvious decrease was observed in other groups. The polysaccharides are more effective in promoting apoptosis of U87 and SiHa cells from S. horrens and H. lessoni massin than from the rest species.
