Nowadays potential preclinical drugs for the treatment of nonalcoholic steatohepatitis(NASH)have failed to achieve expected therapeutic efficacy because the pathogenic mechanisms are underestimated.Inactive rhomboid protein 2(IRHOM2),a promising target for treatment of inflammation-related diseases,contributes to deregulated hepatocyte metabolism-associated nonalcoholic steatohepatitis(NASH)progression.However,the molecular mechanism underlying Irhom2 regulation is still not completely understood.In this work,we identify the ubiquitin-specific protease 13(USP13)as a critical and novel endogenous blocker of IRHOM2,and we also indicate that USP13 is an IRHOM2-interacting protein that catalyzes deubiquitination of Irhom2 in hepatocytes.Hepatocyte-specific loss of the Usp13 disrupts liver metabolic homeostasis,followed by glycometabolic disorder,lipid deposition,increased inflammation,and markedly promotes NASH development.Conversely,transgenic mice with Usp13 overexpression,lentivirus(LV)-or adeno-associated virus(AAV)-driven Usp13 gene therapeutics mitigates NASH in 3 models of rodent.Mechanistically,in response to metabolic stresses,USP13 directly interacts with IRHOM2 and removes its K63-linked ubiquitination induced by ubiquitin-conjugating enzyme E2N(UBC13),a ubiquitin E2 conjugating enzyme,and thus prevents its activation of downstream cascade pathway.USP13 is a potential treatment target for NASH therapy by targeting the Irhom2 signaling pathway.

中国古诗词语言凝练,富有音乐美,蕴含着丰富的内涵和意蕴,在翻译过程中因语言的差异往往很难再现原诗的精妙之处。以《清明》为例,原诗语言精练、优美,文化内涵丰富,景物与情感和谐统一,近代以来先后有多位翻译学家发表了其英译本。基于许渊冲教授“三美”原则,探讨《清明》的四种英译本运用了不同的翻译策略在音美、意美、形美三个方面传递原诗文化内涵,以期再现原诗精妙之处,从而促进中国古诗词的英译与传播。

Background:Retinal diseases can lead to severe visual impairment and even blindness,but current treatments are limited.For precise targeted therapy,the pathophysiological mechanisms of the diseases still need to be further explored.Iron serves an essential role in many biological activities and helps maintain the function and morphology of the retina.The vision problems caused by retinal diseases are affecting more and more people,the study of iron metabolism in retinal diseases possesses great potential for clinical application.Main text:Iron maintains a dynamic balance in the retina but in excess is toxic to the retina.Iron overload can lead to various pathological changes in the retina through oxidative stress,inflammation,cell death,angiogenesis and other pathways.It is therefore involved in the progression of retinal diseases such as age-related macular degeneration,glaucoma,diabetic retinopathy,retinitis pigmentosa,and hereditary iron overload.In recent years,iron chelators have been shown to be effective in the treatment of retinal diseases,but the exact mechanism is not yet fully understood.This question prompted further investigation into the specific mechanisms by which iron metabolism is involved in retinal disease.Conclusions:This review summarizes iron metabolism processes in the retina and mechanistic studies of iron metabolism in the progression of retinal disease.It also highlights the therapeutic potential of iron chelators in retinal diseases.

A strong adhesive block polymer coating for antifouling of large molecular weight protein

Some proteins secreted by microorganisms have large molecular weights. We report here an approach to prepare coating by multilayer polymers for antifouling of proteins, especially the proteins with a large molecular weight.Stainless steel was used as the model substrate. The substrate was first coated with a hybrid polymer film, which was formed by simultaneous hydrolytic polycondensation of 3-aminopropyltriethoxysilane and polymerization of dopamine(HPAPD). After grafting the macroinitiator 2-bromoisobutyryl bromide, the block polymer brushes PMMA-b-PHEMA were grafted. Three proteins were used to test protein adsorption and antifouling behavior of the coating, including recombinant green fluorescent(54 k Da), recombinant R-transaminase(2 × 90 k Da), and recombinant catalase(4 × 98 k Da). It is demonstrated that the block polymer brushes not only can prevent the adsorption of small molecular weight proteins, but also can significantly reduce the adsorption of the large molecular weight proteins.

Non-Orthogonal Transmission for User and Control Plane Split Architecture in 5G Systems and Beyond: Performance Analysis and Design Insights

In next generation networks,mobility management will be a critical issue due to dense base station(BS)deployment,for which user and control plane split architecture provides a promising solution.Jointly designing such architecture with nonorthogonal transmission brings in more flexibility to further improve system efficiency.This paper proposes a non-orthogonal transmission design for user and control plane split architecture.In this design,user equipments(UEs)will select the BS providing the strongest received signal to associate its data channel,but constantly connect its control channel to the nearest macro-cell BS(MBS).Upon non-orthogonal transmission,an MBS can multiplex data traffics and control signals on the same resource.Stochastic geometry based analysis is carried out to investigate outage probability,which extends its regular definition by jointly considering data and control channels,and then mobility-aware outage rate.Numerical results show that:1)The proposed split architecture alleviates the increase in handover rate for ultra dense networking,compared with conventional architecture.2)Non-orthogonal transmission outperforms traditional orthogonal transmission in the split architecture,because it is capable of accommodating more control channels.3)By carefully adjusting power levels,minimum outage probabilities can be reached for macrocell UEs in the proposed design.

目的:探讨PDCA循环管理在医院消毒供应中心中的应用效果。方法:选取2019年10月—2020年4月我院消毒供应中心的复用医疗器械1800件作为研究对象,将2019年10月—2019年12月的复用医疗器械900件作为对照组,给予常规质量控制;将2020年1月—2020年4月的复用医疗器械900件作为观察组,实施PDCA循环管理。比较两组回收不及时率、清洗不合格率、包装不合格率、灭菌湿包率及ATP检测合格率。结果:观察组医疗器械的回收不及时率、清洗不合格率、包装不合格率、灭菌湿包率明显低于对照组。观察组100例抽样器械的ATP检测合格率明显高于对照组(P<0.05)。结论:医院消毒供应中心实施PDCA循环管理能提高消毒供应中心消毒灭菌质量,有效降低消毒供应中心器械的清洗及灭菌不合格率。

为了避免自动化生产线上机器人因多品种、多尺寸圆盘类工件夹持时的频繁换夹,提高机器人夹持稳定性和安全性,设计了一种自锁式欠驱动夹持器并进行了结构优化。基于欠驱动原理完成夹持器的结构设计,并通过夹持构型与夹持过程分析,确保了结构自锁和包络夹持的可行性;根据虚功原理及连杆机构的矢量封闭方程,构建了夹持器自适应抓取状态下的静力学建模;根据夹持器各指节夹持力平衡,建立了结构参数优化模型,并引入遗传算法实现了参数优化。数值测试结果表明,相比经验法和Fmincon算法,遗传算法优化的目标值误差分别减少70%、62.5%,且夹持器的夹持力最大偏差分别减少78.3%,61.4%,验证了文中欠驱动夹持器遗传优化设计的优越性,满足了夹持器通过各指节力平衡实现稳定夹持的要求。

A novel unsupervised method for new word extraction

New words could benefit many NLP tasks such as sentence chunking and sentiment analysis. However, automatic new word extraction is a challenging task because new words usually have no fixed language pattern, and even appear with the new meanings of existing words. To tackle these problems, this paper proposes a novel method to extract new words. It not only considers domain specificity, but also combines with multiple statistical language knowledge. First, we perform a filtering algorithm to obtain a candidate list of new words. Then, we employ the statistical language knowledge to extract the top ranked new words. Experimental results show that our proposed method is able to extract a large number of new words both in Chinese and English corpus, and notably outperforms the state-of-the-art methods. Moreover, we also demonstrate our method increases the accuracy of Chinese word segmentation by 10% on corpus containing new words.

Scattering coefficients of human skin in vivo with and without vitiligo were measured with optical coherence tomography(OCT).The experimental results show that there exist significant difference between the scattering coefficient of the epidermis of in vivo human skin with and without vitiligo disease.The results may be helpful for quantitatively diagnosing or evaluating the treatment of the disease.

以微晶纤维素为原料,高碘酸钠作为氧化剂,制备二醛基纤维素(DAC),考察各影响因素对二醛基纤维素制备的影响,采用FTIR、元素分析、XRD以及化学官能团测定等手段对二醛基纤维素的结构进行表征;探讨二醛基纤维素的醛基含量与尿素吸附容量之间的关系,并研究二醛基纤维素对尿素的吸附平衡和吸附动力学。结果表明:高碘酸钠与微晶纤维素的质量比为2.4∶1、反应温度为35℃、反应时间为3.5 h、反应介质的pH为2时,制得二醛基纤维素的醛基含量最大为97.74%。二醛基纤维素对尿素的吸附容量随醛基含量的增加先增大后减小,最大吸附容量为59.23 mg.g-1,是包醛氧淀粉尿素吸附容量的10倍(6 mg.g-1),是包醛酶淀粉吸附容量的4倍(15 mg.g-1),吸附平衡时间为4 h。吸附等温线符合Freundich方程。
