白车轴草多酚提取工艺优化

采用响应面法研究超声辅助提取白车轴草多酚工艺。先以白车轴草的茎、叶和花为原料,采用超声辅助法分别提取上述部位总多酚,并进行含量检测。结果表明白车轴草的花中多酚含量最高。再通过单因素试验考察提取液乙醇体积分数、提取时间和料液比对超声辅助提取白车轴草花中多酚得率的影响。以此为基础,采用响应曲面法优化白车轴草花中多酚提取工艺。结果表明,白车轴草花中多酚提取的最佳工艺参数为料液比1∶68(g/m L),提取时间43 min,乙醇体积分数43%。在此条件下,多酚得率为2. 55%,与预测得率2. 51%相近,且相对标准偏差为1. 57%。

Hepatitis C virus(HCV) infection usually induces chronic hepatic inflammation,which favors the initiation and progression of hepatocellular ***,microRNA-155 (miR-155) plays an important role in regulating both inflammation and ***, little is known about whether and how miR-155 provides the link between inflammation and cancer. In this study,we found that miR-155 levels were markedly increased in patients infected with HCV. miR-155 transcription was regulated by NF-κB,and p300 increased NF-κB-dependent miR-155 *** overexpression of miR-155 significantly inhibited hepatocyte apoptosis and promoted cell proliferation,whereas miR-155 inhibition induced G0/G1 *** miR-155 resulted in nuclear accumulation ofβ-catenin and a concomitant increase in cyclin D1, c-myc and ***-of-function and loss-of-function studies demonstrated that miR-155 promoted hepatocyte proliferation and tumorigenesis by increasing Wnt signaling in vitro and in vivo, and DKK1(Wnt pathway inhibitor) overexpression inhibited the biological role of miR-155 in ***,adenomatous polyposis coli(APC),which negatively regulates Wnt signaling, was identified as the direct and functional target of ***:HCV-induced miR-155 expression promotes hepatocyte proliferation and tumorigenesis by activating Wnt *** present study provides a better understanding of the relationship between inflammation and tumorigenesis,and thus may be helpful in the development of effective diagnosis and treatment strategies against HCV-HCC.

For the optical system of the telescope, with the increase in telescope size, the manufacture of monolithic primary becomes increasingly difficult. Instead, the use of segmented mirrors, where many individual mirrors （the segments） work together to provide an image quality and an aperture equivalent to that of a large monolithic mirror, is considered a more appropriate strategy. But with the introduction of the large telescope mirror comprised of many individual segments, the problem of insuring a smooth continuous mirror surface （co-phased mirrors） becomes critical. One of the main problems is the measurement of the vertical displacement between the individual segments （piston error）, for such mirrors, the segment vertical misalignment （piston error） between the segments must be reduced to a small fraction of the wavelength （＜100nm） of the incoming light. The measurements become especially complicated when the piston error is in order of wavelength fractions. To meet the performance capabilities, a novel method for phasing the segmented mirrors optics system is described. The phasing method is based on a high-aperture Michelson interferometer. The use of an interferometric technique allows the measuring of segment misalignment during the daytime with high accuracy, which is a major design guideline. The innovation introduced in the optical design of the interferometer is the simultaneous use of monochromatic light and multiwavelength combination white-light source in a direct method for improving the central fringe identification in the white-light interferometric phasing system. With theoretic analysis, we find that this multiwavelength combination technique can greatly increase the visibility difference between the central fringe and its adjacent side fringes, and thus it offers an increased signal resolution. We present a simultaneous time- and spectrum-resolved multifocal multiphoton microscopy （MMM） system using a novel field of view （FOV） zoom scanning protocol. The system employs a microlens array for producing discrete excitation spot array, a dispersive prism and a high repetition rate streak camera for simultaneous temporal and spectral resolutions. By combining a pair of galvo mirrors and a sample stage for fine and coarse scanning respectively, the resolution and FOV of the system can be changed without changing any optical elements. The system can be operated not only in low resolution and large FOV, but also in high resolution and small FOV applications, without compromising the performances of the optical elements in the system. By implementing a special system control protocol and image reconstruction algorithm, fluorescence images in different FOVs and resolutions can be obtained. This FOV zoom scanning protocol is demonstrated with two-photon excitation fluorescence imaging of a fluorescence resolution test target.

以实心砌体房屋为研究对象,建立了四层砌体房屋仿真模型,应用有限元软件ANSYS/LS-DYNA通过模态分析获取其固有频率及振型,根据模态分析结果设计非一致爆破地震波激励加载模型,分析砌体房屋在爆破振动作用下动态响应和变形特点。计算结果表明:结构响应随地震波振速幅值增大而增大,随主频的增大而减小;爆破地震波作用下底层门窗角落易形成集中应力而产生水平裂缝,但砌体产生破坏时其最大层间位移均未达到砌体结构设计规范控制值和各类文献建议值,说明结构是由于受拉导致的脆性破坏。由此可见,采用延性、层间位移角等地震工程常用的结构损伤评估指标来评价爆破地震对建筑物的破坏是不准确的。
