Chimeric antigen receptor(CAR)-modified T-cell therapy has achieved remarkable success in the treatment of acute lymphoblastic leukemia(ALL).Measurable/minimal residual disease(MRD)monitoring plays a significant role in the prognostication and management of patients undergoing CAR-T-cell therapy.Common MRD detection methods include flow cytometry(FCM),polymerase chain reaction(PCR),and next-generation sequencing(NGS),and each method has advantages and limitations.It has been well documented that MRD positivity predicts a poor prognosis and even disease relapse.Thus,how to perform prognostic evaluations,stratify risk based on MRD status,and apply MRD monitoring to guide individual therapeutic decisions have important implications in clinical practice.This review assesses the common and novel MRD assessment methods.In addition,we emphasize the critical role of MRD as a prognostic biomarker and summarize the latest studies regarding MRD-directed combination therapy with CAR-T-cell therapy and allogeneic hematopoietic stem cell transplantation(allo-HSCT),as well as other therapeutic strategies to improve treatment effect.Furthermore,this review discusses current challenges and strategies for MRD detection in the setting of disease relapse after targeted therapy.

Learning-based approaches have made substantial progress in capturing spatially-varying bidirectional reflectance distribution functions(SVBRDFs)from a single image with unknown lighting and geometry.However,most existing networks only consider per-pixel losses which limit their capability to recover local features such as smooth glossy regions.A few generative adversarial networks use multiple discriminators for different parameter maps,increasing network complexity.We present a novel end-to-end generative adversarial network(GAN)to recover appearance from a single picture of a nearly-flat surface lit by flash.We use a single unified adversarial framework for each parameter map.An attention module guides the network to focus on details of the maps.Furthermore,the SVBRDF map loss is combined to prevent paying excess attention to specular highlights.We demonstrate and evaluate our method on both public datasets and real data.Quantitative analysis and visual comparisons indicate that our method achieves better results than the state-of-the-art in most cases.

Nd-Fe-B permanent magnets play a crucial role in energy conversion and electronic devices.The essential magnetic properties of Nd-Fe-B magnets,particularly coercivity and remanent magnetization,are significantly infuenced by the phase characteristics and microstructure.In this work,Nd-Fe-B magnets were manufactured using vacuum induction melting(VIM),laser directed energy deposition(LDED)and laser powder bed fusion(LPBF)technologies.Themicrostructure evolution and phase selection of Nd-Fe-B magnets were then clarified in detail.The results indicated that the solidification velocity(V)and cooling rate(R)are key factors in the phase selection.In terms of the VIM-casting Nd-Fe-B magnet,a large volume fraction of theα-Fe soft magnetic phase(39.7 vol.%)and Nd2Fe17Bxmetastable phase(34.7 vol.%)areformed due to the low R(2.3×10-1?C s-1),whereas only a minor fraction of the Nd2Fe14B hard magnetic phase(5.15 vol.%)is presented.For the LDED-processed Nd-Fe-B deposit,although the Nd2Fe14B hard magnetic phase also had a low value(3.4 vol.%)as the values of V(10-2m s-1)and R(1.45×106?C s-1)in the LPBF process resulted in the substantial formation of the Nd2Fe14B hard magnetic phase(55.8 vol.%)directly from the liquid,while theα-Fe soft magnetic phase and Nd2Fe17Bxmetastable phase precipitation are suppressed in the LPBF-processed Nd-Fe-B magnet.Additionally,crystallographic texture analysis reveals that the LPBF-processedNd-Fe-B magnets exhibit isotropic magnetic characteristics.Consequently,the LPBF-processed Nd-Fe-B deposit,exhibiting a coercivity of 656 k A m-1,remanence of 0.79 T and maximum energy product of 71.5 k J m-3,achieved an acceptable magnetic performance,comparable to other additive manufacturing

Mortality from cerebrovascular diseases in China:Exploration of recent and future trends

Background:Cerebrovascular disease(CVD)ranks among the foremost factors responsible for mortality on a global scale.The mortality patterns of CVDs and temporal trends in China need to be well-illustrated and updated.Methods:We collected mortality data on patients with CVD from Chinese Center for Disease Control and Prevention’s Disease Surveillance Points(CDC-DSP)system.The mortality of CVD in 2020 was described by age,sex,residence,and region.The temporal trend from 2013 to 2019 was evaluated using joinpoint regression,and estimated rates of decline were extrapolated until 2030 using time series models.Results:In 2019,the age-standardized mortality in China(ASMRC)per 100,000 individuals was 113.2.The ASMRC for males(137.7/10^(5))and rural areas(123.0/10^(5))were both higher when stratified by gender and urban/rural residence.The central region had the highest mortality(126.5/10^(5)),the western region had a slightly lower mortality(123.5/10^(5)),and the eastern region had the lowest mortality(97.3/10^(5)).The age-specific mortality showed an accelerated upward trend from aged 55-59 years,with maximum mortality observed in individuals over 85 years of age.The age-standardized mortality of CVD decreased by 2.43%(95%confidence interval,1.02-3.81%)annually from 2013 to 2019.Notably,the age-specific mortality of CVD increased from 2013 to 2019 for the age group of over 85 years.In 2020,both the absolute number of CVD cases and the crude mortality of CVD have increased compared to their values in 2019.The estimated total deaths due to CVD were estimated to reach 2.3 million in 2025 and 2.4 million in 2030.Conclusion:The heightened focus on the burden of CVD among males,rural areas,the central and western of China,and individuals aged 75 years and above has emerged as a pivotal determinant in further decreasing mortalities,consequently presenting novel challenges to strategies for disease prevention and control.

Chemotactic nanomotor for multimodal combined therapy of glioblastoma

Glioblastoma(GBM) is the most aggressive malignant brain tumor. Due to the infiltration and heterogeneity of GBM, the obstruction of the blood-brain barrier(BBB) and the unique immunosuppressive mechanism, it is hard to achieve significant effects of GBM treatment. Here, a kind of chemotactic nanomotor that loaded with glucose oxidase(GOx) and carboxylated cisplatin(Pt(IV)) prodrug on the L-arginine-derived polymer is proposed. The nanomotors are driven by catalysis of glucose decomposition and the positive chemotaxis towards the GBM microenvironment where inducible nitric oxide synthase and reactive oxygen species are highly expressed. This facilitates the BBB crossing and GBM targeting of the nanomotors. In addition, the released nitric oxide(NO) during propulsion as well as the loaded GOx and Pt(IV) can exert combined NO/starvation/chemotherapy. Meanwhile, it is able to induce and enhance the immune response through multiple pathways, thus better coping with the complexities of GBM treatment.

Synthesis and Properties of Rosin-Based Composite Acrylamide Hydrogels

Hydrogels have been widely applied in agricultural drought-resistance,pollution regulation,drug delivery and so on.Acrylamide(AM)is usually used as raw material to synthesize acrylamide hydrogels.However,inherently low mechanical strength greatly limits their applications in some special areas.Therefore,it is necessary to choose suitable functional monomers to optimize acrylamide hydrogels and improve their mechanical performances.In this paper,a novel acrylamide monomer modified by rosin was synthesized,and then polyacrylamide/rosinbased acrylamide(RAM)composite hydrogels were prepared via free radical polymerization using potassium persulfate as initiator,N,N′-methylene-bisacrylamide(MBA)as a crosslinker.The influence of RAM monomer was investigated in detail.The chemical structure,pore structure,swelling properties,thermal performances and mechanical properties of composite hydrogels were characterized by Fourier Transform Infrared spectrometer(FT-IR),thermogravimetric analysis(TG),scanning electron microscope(SEM),and universal testing,respectively.The results showed that the thermal stability and mechanical property of RAM hydrogels were improved significantly.The compressive strength of RAM hydrogels was increased to 3.5 times than that of AM hydrogels,and the tensile strength was 5.1 times compared with AM hydrogels as well.Moreover,RAM hydrogels exhibited a faster initial swelling rate due to the new pore structure formed after introducing the RAM monomer.

Immunosuppressive microenvironments present critical problems in clinical chemotherapy.To regulate the tumor immune microenvironment for enhancing antitumor effect,a combination of immune checkpoint inhibitors(ICIs)with chemotherapeutics has been applied clinically.In this study,miriplatin(MiPt),the lipidic derivative of 5-fluorouracil(Fu-OA),as well as the programmed death ligand 1(PD-L1)target si RNA(siPD-L1)were integrated into Lip-Pt/Fu@siPD-L1 nanoparticles(NPs)for chemo-immunotherapy.In vitro results showed that Lip-Pt/Fu@siPD-L1 NPs could exhibit effective siRNA gene silencing and promote the phagocytosis of tumor cells by macrophages.Furthermore,in vivo results revealed that LipPt/Fu@siPD-L1 NPs showed significantly higher anti-tumor efficiency than that of the physical mixing of Mi Pt,5-fluorouracil,and Lip@siPD-L1 NPs(delivery of siPD-L1 by liposomes).The best anti-tumor efficiency of Lip-Pt/Fu@siPD-L1 NPs resulted from the synergistic immunotherapeutic effects of Mi Pt and siPD-L1 based on the inhibition of CD47 expression and the downregulation of PD-L1 in tumor cells,which elicited a robust anti-tumor immune response through the activation of macrophage phagocytosis and immune checkpoint inhibition.The Lip-Pt/Fu@siPD-L1 NPs provide a potential strategy for tumor chemo-immunotherapy.

An Optimal Bit-Rate Allocation Algorithm to Improve Transmission Efficiency of Images in Deep Space Exploration

As fixed compression ratio is used in traditional deep space exploration image transmission application,the same compression code rate is allocated to each image.However,since the information of each image in a space observation mission is nonuniform,the image with more information will inevitably lead to more compression distortion than the image with less information.Obviously,it’s not an efficient way to transmit information in terms of data importance or overall distortion.Therefore,we proposed a combinatorial optimal bit rate allocation algorithm to improve the efficiency of image transmission in space application.Different from traditional method,the rate-distortion model of wavelet coefficients for each image in a transmission task was built,and under the overall maximum transmission rate constraint,an bit-rate optimal allocation was applied for each image to minimize the overall distortion of a batch of images.The proposed algorithm can be widely used in image compression algorithm with embedded code stream characteristics,such as JPEG2000 and SPIHT.Experimental results shows that in the range of 2.6 to 10 compression ratio,the algorithm can reduce image distortion MSB by 40%~64%at the same overall transmission code rate,which equivalent to improvement of PSNR 3 dB to 5.4 dB.

目的探讨2例Gitelman综合征(GS)患者的遗传学病因。方法选取分别于2022年1月和6月就诊于临沂市人民医院的2例GS患者作为研究对象进行回顾性研究。收集患者的血清检测血清电解质水平及电解质排泄量资料。采集患者外周血样提取基因组DNA进行全外显子组测序,并进行Sanger测序验证。结果患者1为27岁女性,血清钠、钾、氯及镁水平以及24 h尿氯、钙水平均低于参考值。基因检测发现其携带SLC12A3:c.1456G>A(p.D486N)和SLC12A3:c.179C>T(p.T60M)复合杂合变异,前者遗传自其母亲,为已报道致病性变异。患者2为4岁男性,血钠、氯和镁水平低于参考值,血钾出现危急值。SLC12A3基因检出母源致病性c.602-16G>A和父源c.805_806insTTGGCGTGGTCTCGGTCA(p.V268_T 269insIGVVSV)复合杂合变异。根据美国医学遗传学与基因组学学会指南,患者2携带的变异均被判定为致病性变异(PVS1+PM2_Supporting+PP3;PVS1+PM2_Supporting+PM4)。结论上述SLC12A3变异考虑为2例GS患者的遗传学病因。
