Tumor microenvironment(TME)is the environment around the tumor cells,consisting of extracellular matrix,nonmalignant stromal cells,and various types of immune cells(like macrophages,B and T lymphocytes).1 TME has been recurrently reported to play important roles in the progression of cancers.1 The mechanism study of TME may offer opportunities for targeted/immune ***,how TME components impact the prognosis of cancer patients remains to be determined.

Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is the pathogen of the ongoing coronavirus disease 2019(COVID-19)global ***,by centralizing published cell-based experiments,clinical trials,and virtual drug screening data from the NCBI PubMed database,we developed a database of SARS-CoV-2 inhibitors for COVID-19,dbSCI,which includes 234 SARS-CoV-2 inhibitors collected from publications based on cell-based experiments,81 drugs of COVID-19 in clinical trials and 1305 potential SARS-CoV-2 inhibitors from bioinformatics *** provides four major functions:(1)search the drug target or its inhibitor for SARS-CoV-2,(2)browse target/inhibitor information collected from cell experiments,clinical trials,and virtual drug screenings,(3)download,and(4)submit *** entry in dbSCI contains 18 types of information,including inhibitor/drug name,targeting protein,mechanism of inhibition,experimental technique,experimental sample type,and reference *** summary,dbSCI provides a relatively comprehensive,credible repository for inhibitors/drugs against SARS-CoV-2 and their potential targeting mechanisms and it will be valuable for further studies to control COVID-19.

为了解决抗生素污染日益严峻和二氧化碳过量排放的问题,本研究以蛋白核小球藻为原料,海藻酸钠为固定剂,氯化钙为交联剂,制备蛋白核小球藻藻珠并将其用于抗生素去除与二氧化碳固定耦合。研究结果表明:当海藻酸钠为2%(质量分数,下同)和氯化钙为3%时,藻珠成球速度、机械强度和抗崩解性最佳。5%(体积分数,下同)和10%的二氧化碳显著促进了蛋白核小球藻的生长并增强了其对头孢拉定的处理能力,反应24 h后头孢拉定去除率分别为84.89%和84.31%,二氧化碳的吸收率分别为72.25%和71.71%。相比于二氧化碳5%和10%条件,二氧化碳20%条件下蛋白核小球藻对抗生素的处理和二氧化碳的吸收率均显著降低。二氧化碳5%和10%条件下的碳代谢相关酶1,5-二磷酸核酮糖羧化酶活性显著升高,能够使微藻细胞固定更多的二氧化碳。二氧化碳10%条件下的碳酸酐酶活性显著升高,能够催化更多的碳酸氢钠转变为二氧化碳,从而被微藻细胞吸收利用。

[目的]探讨脂肪间充质干细胞(ADMSC)外泌体(Exo)对心肌梗死(MI)后不良心室重塑的抑制作用和机制。[方法]观察心脏成纤维细胞经过氧化氢(H_(2)O_(2))处理后自噬和炎症表型的改变。MI大鼠经尾静脉注射等体积的生理盐水、ADMSC外泌体(MSC-Exo)、成纤维细胞外泌体(MEF-Exo),观察心脏成纤维细胞自噬相关16样蛋白1(ATG16L1)、自噬相关蛋白7(ATG7)和NOD样受体蛋白3(NLRP3)炎症小体的表达,炎症反应,心肌纤维化程度以及心功能。[结果]心脏成纤维细胞经H_(2)O_(2)处理后,自噬相关蛋白ATG16L1和ATG7表达显著降低(P<0.001),NLRP3表达显著升高(P<0.001),促炎细胞因子白细胞介素1β(IL-1β)和IL-18水平显著增加(P<0.001)。MI大鼠经MSC-Exo干预后,自噬相关蛋白ATG16L1和ATG7表达显著上调(P<0.001),NLRP3表达显著下调(P<0.001),血清IL-1β和IL-18水平显著降低(P<0.001),纤维化相关蛋白胶原蛋白Ⅰ和Ⅲ显著减少(P<0.001),心肌纤维化程度显著减轻(P<0.001),心功能明显改善(P<0.001)。[结论]脂肪MSC-Exo通过调控心脏成纤维细胞自噬和NLRP3炎症小体的平衡,发挥抑制MI后不良心室重塑的作用。
